연료전지 전극 반응을 위한 카본 담지 표면 합금의 전기촉매 활성 (Electrocatalytic activity of carbon-supported near-surface alloys (NSAs) for electrode reaction of fuel cell)

  • There is a worldwide interest in the development and commercialization of polymer electrolyte membrane fuel cells [PEMFCs] for vehicular and stationary applications. One of the major objectives is the reduction of loaded electrode materials, which is comprise of the Pt-based noble metals. In this paper, a novel chemical strategy is described for the preparation and characterization of carbon-supported and surface-alloys, which were prepared by using a successive reduction process. After preparing Au colloid nanoparticles, the supporting of Au colloid nanoparticles occurred spontaneously in the carbon black-dispersed aqueous solution. Then nano-scaled active materials were formed on the surface of carbon-supported Au nanoparticles. The structural and electrochemical analyses indicate that the active materials were deposited on the surface of Au nanoparticles selectively and that an alloying process occurred during the successive reducing process. The carbon-supported & surface-alloys showed the higher electrocatalytic activity than those of the particle-alloys and commercial one [Johnson-Matthey] for the reaction of methanol and formic acid oxidation. The increased electrocatalytic activity might be attributed to the effective surface structure of surface-alloys, which have a high utilization of active materials for the surface reaction of electrode.

Isolation and Characterization of Intestinal Immune System Modulating and Anticancer Active Fractions from the Herbal Prescriptions

  • The prescriptions (DB-1-DB-5) were prepared with the active herbal medicines, Acanthopanax senticosus, Glycyrrhiza uralensis, Polygonatum odoratum, and Cichorium intybus. The most active crude polysaccharide fraction (DB-2-3), which was isolated through the fractionation of hot-water extract from DB-2, was significantly reduced by periodate oxidation (52.7 and 63.7%) on intestinal immune system modulating and anticancer activity. When DB-2-3 was further fractionated by column chromatographies, DB-2-3IIc-2 showed the most potent activities. In addition, DB-2-3IIc stimulated the proliferation of bone marrow cells via Peyer's patch in dose-dependent pattern by oral administration. The metastasis of colon 26-M3.1 lung carcinoma had significantly inhibited in mice fed DB-2-3IIc at 1 mg/mouse (43.8%). DB-2-3IIc-2 mainly contained uronic acid (46.1%) and 42.5% of neutral sugar with a small amount of protein (7.6%), and component sugar analysis also showed that DB-2-3IIc-2 was composed Ara, Gal, and GalA (molar ratio; 0.50:0.63:1.00). It may be suggested that activities of DB-2-3IIc-2 are resulted from pectic polysaccharides containing a polygalacturonan moiety with side chain of neutral sugars, such as Ara and Gal.

Antioxidant and Bioactive Films to Enhance Food Quality and Phytochemical Production during Ripening

  • Antioxidant films are one active packaging technology that can extend food shelf-life through preventing lipid oxidation, stabilizing color, maintaining sensory properties and delaying microbial growth in foods. Because raw, fresh and minimal processed foods are more perishable during storage or under display conditions than further processed foods, they rapidly lose their original quality. Foods are susceptible to physical, chemical, and biochemical hazards to which packaging films can be effective barriers. Although films incorporated natural (tocopherols, flavonoids and phenolic acids) or synthetic antioxidants (BHT, BHA, TBHQ, propyl gallate) have been extensively tested to improve quality and safety of various foods, food applications require addressing issues such as physical properties, chemical action, cost, and legal approval. Increased interest in natural antioxidants as substitutes for synthetic antioxidants has triggered research on use of the new natural antioxidants in films and coatings. Use of new components (phytochemicals) as film additives can improve food quality and human health. The biosynthesis of plant phenolics can potentially be optimized by active coatings on harvested fruits and vegetables. These coatings can trigger the plants natural proline-linked pentose phosphate pathway to increase the phenolic contents and maintain overall plant tissue quality. This alternate metabolic pathway has been proposed by Dr. K. Shetty and is supported by numerous studies. A new generation of active food films will not only preserve the food, but increase food's nutritional quality by optimizing raw food biochemical production of phytochemicals.

  • 산화-환원 활성 단백질중에 하나인 설피레독신과의 결합 단백질을 효모 Two-hybrid 기법을 이용하여 탐색한 결과, 알파-만노시다제가 설피레독신과 특이적으로 결합함을 밝혔다. 알파-만노시다제는 D-만노스 당을 비환원성 말단으로부터 유리시키는 가수분해 효소로서, 세포 원형질에 다량체 형태로 존재한다. 본 연구에서는 설피레독신과 알파-만노시다제간의 단백질결합을 설피레독신의 새로운 생리기능 관점에서 토의했다.

Ameliorating Effect of Gardenia jasminoides Extract on Amyloid Beta Peptide-induced Neuronal Cell Deficit

  • The brains of Alzheimer's disease (AD) patients are characterized by large deposits of amyloid beta peptide ($A{\beta}$). $A{\beta}$ is known to increase free radical production in nerve cells, leading to cell death that is characterized by lipid peroxidation, free radical formation, protein oxidation, and DNA/RNA oxidation. In this study, we selected an extract of Gardenia jasminoides by screening, and investigated its ameliorating effects on $A{\beta}$-induced oxidative stress using PC12 cells. The effects of the extract were evaluated using the 2',7'-dichlorofluorescein diacetate (DCF-DA) assay and the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) reduction assay. To find the active component, the ethanol extract was partitioned with hexane, chloroform, and ethyl acetate, respectively, and the active component was purified by silica-gel column chromatography and HPLC. The results suggested that Gardenia jasminoides extract can reduce the cytotoxicity of $A{\beta}$ in PC 12 cells, possibly by reducing oxidative stress.

사과주(酒) 발효과정중(醱酵過程中) Polyphenol성(性) 물질(物質) 및 Polyphenol Oxidase 활성(活性) Band의 변화(變化) (Changes of Polyphenols and Polyphenol Oxidase Active Bands during Apple Wine Fermentation)

  • 사과주(酒)의 효소(酵素)에 의(依)한 갈변(褐變)을 조사하기 위하여 갈변(褐變)과 관련된 polyphenol성(性) 물질 및 polyphenol oxidase 활성 band의 변화를 사과주(酒) 제조기간동안 조사한 결과는 다음과 같다. Total phenol 의 감소율은 $SO_2$ 처리에 의해서 현저히 둔화되었고, catechol의 산화(酸化)는 온도(溫度)와 pH가 높아짐에 따라 촉진되었으나, pH 5.0 이하에서는 $4^{\circ}C$에서 산화(酸化)가 일어나지 않았다. Polyacrylamide gel 전기영동으로 사과에는 Rm값이 각각 0.21, 0.30, 0.41, 0.51인 a, b, c, d 4 개의 polyphenol oxidase활성을 가진 band가 관찰되었다. 이들중 a, c 2 개의 band는 발효(醱酵) 5 일째까지 계속 관찰되었으며, 발효(醱酵) 6 일째부터는 c band만 관찰되었다. 이 band는 $60^{\circ}C$, 30분간(分間)의 살균(殺菌) 처리 후에도 계속 남아있었다.

RuTi 촉매의 소성온도가 NH3-SCO 반응활성에 미치는 영향 (The Effect of Calcination Temperature of RuTi Catalysts on the Reaction Activity of NH3-SCO)

  • 본 연구에서는, NH3-SCO (selective catalytic oxidation) 반응에서 RuTi 촉매 제조 시 소성온도에 따른 영향을 확인하였다. RuTi 촉매는 습윤 함침법을 이용하여 제조되었고, 공기 분위기에서 400~600 ℃로 4 h 동안 소성되었다. 촉매는 RuTi x00로 표기되었으며, x00는 소성온도를 의미한다. XRD, TEM, H2-TPR 분석에 따르면, RuTi x00 촉매는 소성온도가 증가할수록 활성금속의 분산도가 감소하는 것을 나타내었다. XPS, NH3-TPD 분석을 통하여, 낮은 분산도를 갖는 촉매는 표면 흡착 산소 종(Oβ) 및 NH3 흡착량이 감소하는 특성을 나타내었다. 따라서 RuTi 400 촉매는 TiO2 표면에 활성금속이 가장 잘 분산되었으며, NH3 제거 효율이 가장 우수하였다.

Effect of Medicinal Plant Extract Incorporated Carrageenan Based Films on Shelf-Life of Chicken Breast Meat

  • This study was performed to examine the possibility of water extracts for several medicinal plants, such as Amomum tsao-ko, Alpinia oxyphylla, and Citrus unshiu, as an active packaging ingredient for prevention of lipid oxidation. Chicken breast meats were packed with medicinal plant extracts incorporated carrageenan based films and their physico-chemical and microbial properties during storage at $5^{\circ}C$ were investigated. In chicken meat samples packed with A. tsao-ko (TF) or A. oxyphylla (OF) extract incorporated carrageenan based films, pH value, thiobarbituric acid reactive substances (TBARS), and the population of total microbes were significantly lower than those of the negative control (film of no extract was incorporated, CF) after 5 d of storage (p<0.05). Especially, TBARS value of TF ($0.12{\pm}0.01$ mg malonaldehyde/kg meat) was significantly lower than chicken meat samples packed with positive control (ascorbic acid incorporated film, AF, $0.16{\pm}0.01$ mg malonaldehyde/kg meat) at 3 d of storage, and it means TF has enough antioxidative activity to prevent the lipid oxidation of chicken meat. However, there was no consistent effect on VBN values of chicken meats packed with medicinal plant extracts incorporated films during storage. Based on the obtained results, it is considered that A. tsao-ko extract has potential for being used as a natural antioxidant ingredient in active packaging areas.

크롬 산화물 촉매를 이용한 Vinyl Chloride의 산화 분해반응 (Catalytic Oxidation of Vinyl Chloride on Chromium Oxide Catalysts)

  • 알루미나에 담지한 $CrO_x$ 촉매를 이용하여 $200{\sim}400^{\circ}C$의 온도 구간에서 공기 중 vinyl chloride의 산화반응을 조사하였다. 탄소를 포함하는 주생성물은 CO 및 $CO_2$로, 온도의 증가에 따라 $CO_2$의 선택도는 증가하고 CO는 감소하였다. 이로부터 $CrO_x$ 촉매상에서 vinyl chloride는 먼저 CO로 일차산화 후 다시 $CO_2$로 완전산화됨을 알 수 있었다. 반응물에 HCl을 첨가하였을 때 vinyl chloride의 전환율은 변화가 없었으나 $CO_2$의 선택도는 감소하였는데 이는 주반응생성물인 HCl이 vinyl chloride의 $CO_2$로 완전산화를 방해함을 의미한다. 반응물에 물이 없는 조건하에서 vinyl chloride를 산화시킬 때 상당량의 $Cl_2$가 생성되었으나 물을 첨가 시 $Cl_2$는 검출되지 않았다. Vinyl chloride의 촉매산화반응에 대한 $CrO_x$ 담지촉매와 몇가지 귀금속 및 다른 전이 금속 산화물의 활성을 비교하였는데 $CrO_x$ 담지촉매의 vinyl chloride 분해활성은 1% Pt 담지촉매를 제외한 다른 촉매에 비해 높았으며, $275^{\circ}C$에서 12% $CrO_x/Al_2O_3$의 반응속도를 기준으로 각 촉매의 활성을 비교시 1% $Pt/Al_2O_3$ 보다 1.2배정도 활성이 떨어지나 다른 촉매에 비하여 각각 3배에서 8배 가량 높은 활성을 나타내었다.

L-Ascorbic Acid의 산화특성에 따른 화장품 용기 개발 및 유효성 분석 (A Development and Validation of Cosmetic Container Based on L-Ascorbic Acid Oxidation Property)

  • 피부 미백, 콜라겐 합성 및 노화방지 등에 좋은 효능을 나타내는 대표적인 항산화제인 L-ascorbic acid를 사용하여 다양한 보관 조건(분말 상, 수용액 상, 온도 변화, 자외선 조사 및 외부공기 유입 등)에서 산화특성을 조사하였으며, 열에 의한 산화 및 분해특성을 조사하였다. 또한 이중 공간을 갖는 파우치를 사용하여 L-ascorbic acid를 분말 상태 및 에센스와 혼합한 상태에서 각각 보관 조건에 따른 산화특성을 분석하여 파우치의 유효성을 검증하였다. 열 특성조사를 위해서는 TGA, DSC 및 FT-IR을 사용하여 분석하였고, 산화특성 조사를 위해서는 UV-visible spectrophotometer와 산화환원 적정법을 병행 시행하였다. 실험 결과, L-ascorbic acid는 금속이온, 하이드록시이온 등이 많이 함유된 수용액 상에서 고온, 자외선 조사 및 외부공기가 유입되는 상태가 가장 빠른 산화조건인 것으로 나타났고, 순수한 분말 상으로 보관할 경우에는 가열, 자외선 조사 및 외부공기 유입 시에도 오랜 기간 동안 산화되지 않는 특성이 있었다. 이러한 결과를 바탕으로, L-ascorbic acid와 같은 산화 안정성이 떨어지는 활성 성분을 분리하여 보관할 수 있도록 설계 제작 중인, 진공포장이 가능한 이중 공간 구조의 파우치를 사용할 경우 화장품 보관기간을 획기적으로 증가시킬 수 있음을 확인하였다.
