Effects of Sipjeondaebotang on the white rat Hypothyroidism Induced by PTU(6-n-propyl-2-thiouracil) (PTU로 유발된 흰쥐의 갑상선 기능저하증에 미치는 십전대보탕(十全大補湯)의 효과)

  • Objectives : This study was to evaluate Sibjeondaebo-tang’s(Shiquan dabutang) effect to rat hypothyroidism which induced by 6-n-propyl-2-thiouracil(PTU)Methods : Aqueous extracts of Sibjeondaebo-tang(Shiquan dabutang)(21.29%) were administered by oral dose of 300 and 150 mg/kg(rat’s body weight) for 42 days from 2 weeks before start of PTU treatment. During this moment the body weight, thyroid gland weights, liver weight, serum thyroid hormone, lipid content and liver antioxidant defense system were measured with histopathological changes of thyroid glands, Liver and these result were compared with LevoT4 0.5mg/kg treated ratsResults : As a result of PTU treatment, marked decreases of body weight, liver weight, serum thyroid hormone levels of T3, T4, triglyceride content and liver CAT activation were observed with marked increases of thyroid gland weight, thyroid gland hormone TSH, activate liver H2O2 and SOD, serum Total cholesterol, HDL and AST amount were admitted. In addition, marked hyperplasia of follicular cells with reduce of follicular colloid contents and sizes were showed at histopathological inspections. These PTU-induces histopathological changes related to hypothyroidism were significantly reduced by treatment of both different dosages of Sibjeondaebo-tang(Shiquan dabutang).Conclusions: This study suggest that Sibjeondaebo-tang(Shiquan dabutang) extracts have favorable effect on the thyroid hormone productions with beneficial effect on hypothyroidism medicated by modulatory effects on antioxidant defense system.

  • In Korea, a huge amount of radioactive concrete waste will be generated through decommissioning of nuclear facilities in the near future; therefore, optimum technology for the treatment of concrete waste should be reviewed thoroughly and the future direction of technology development should be discussed. In this paper, many domestic and foreign examples of generation of radioactive concrete waste were pieced together and the characteristics of radioactive concrete waste were examined. Moreover, we reviewed trends in technology development by analyzing the examples of various studies and practical applications of treatment technologies, such as mechanical decontamination, chemical decontamination, volume reduction, recycling and solidification, and also tried to understand the limitations of existing technologies and determine a direction for technical improvement.

  • Lysine is known as a limiting amino acid in rice. In addition, it is considered to be important in that it is easily non-activated by the browning reaction during processing or storage. The present study was designed to utilize a kinetic approach to analyse the effect of temperature and water activity on available lysine loss in rice. Simplified kinetic models were used to obtain the various kinetic parameters for available lysine loss in rice subjected to accelerated shelf-life tests (ASLT). These kinetic parameters were then used to predict protein quality loss under the non-steady state storage. The predicted losses were compared to the actual losses. As expected, available lysine loss was increased with increased temperature and water activity. The activation energies and $Q_{10}$ values for available lysine loss ranged from 4.03 to 5.10 Kcal/ mole and 1.22 to 1.27, respectively, The shelf-lives at $25^{\circ}C$, the time to reach 25% loss of the available lysine, which was derived from the accelerated shelf-life tests showed 67 to 107 days according to $a_w$'s. The amount of loss for the fluctuating condition was greater than that occurring at the mean temperature of $45^{\circ}C$. Actually, the differences in effective temperature for the fluctuating storage were between about 4 and $6^{\circ}C$. In predicting the extent of loss using constant state data, the predicted shelf-lives were 2 to 7 days shorter than the actual storage values.

Effect of Lipofectin on Antigen-presenting Function and Anti-tumor Activity of Dendritic Cells (수지상세포의 항원제시 능력 및 항암활성에 미치는 Lipofectin의 영향)

  • Background: Dendritic cells (DC) are professional antigen-presenting cells in the immune system and can induce T cell response against virus infections, microbial pathogens, and tumors. Therefore, immunization using DC loaded with tumor-associated antigens (TAAs) is a powerful method of inducing anti-tumor immunity. For induction of effective anti-tumor immunity, antigens should be efficiently introduced into DC and presented on MHC class I molecules at high levels to activate antigen-specific $CD8^+$ T cells. We have been exploring methods for loading exogenous antigens into APC with high efficiency of Ag presentation. In this study, we tested the effect of the cationic liposome (Lipofectin) for transferring and loading exogenous model antigen (OVA protein) into BM-DC. Methods: Bone marrow-derived DC (EM-DC) were incubated with OVA-Lipofectin complexes and then co-cultured with B3Z cells. B3Z activation, which is expressed as the amount of ${\beta}$-galactosidase induced by TCR stimulation, was determined by an enzymatic assay using ${\beta}$-gal assay system. C57BL/6 mice were immunized with OVA-pulsed DC to monitor the in vivo vaccination effect. After vaccination, mice were inoculated with EG7-OVA tumor cells. Results: BM-DC pulsed with OVA-Lipofectin complexes showed more efficient presentation of OVA-peptide on MHC class I molecules than soluble OVA-pulsed DC. OVA-Lipofectin complexes-pulsed DC pretreated with an inhibitor of MHC class I-mediated antigen presentation, brefeldin A, showed reduced ability in presenting OVA peptide on their surface MHC class I molecules. Finally, immunization of OVA-Lipofectin complexes-pulsed DC protected mice against subsequent tumor challenge. Conclusion: Our data provide evidence that antigen-loading into DC using Lipofectin can promote MHC class I- restricted antigen presentation. Therefore, antigen-loading into DC using Lipofectin can be one of several useful tools for achieving efficient induction of antigen-specific immunity in DC-based immunotherapy.

  • Despite the economic downturn, the amount of clothing being discarded is increasing due to the fast fashion. The purpose of this study is to propose a fashion sharing service development to settle the rational consumption culture along with the abandoned clothes problem. Shared fashion rental service has attracted as a solution to environmental problems not only when clothes are incinerated but also in manufacturing process, but there is a lack of precedent research. Therefore, this study is meaningful in that it has developed two development plans based on the results of analysis of service case based on three factors by Rachel Botsman. First, active community development. Second, it is the creation of goods through activation of the shared economy. In this study, there are limitations that can't be proved through actual application, so more research is needed through empirical studies on actual users in the future.

Inhibition of cell growth and induction of apoptosis by bilobalide in FaDu human pharyngeal squamous cell carcinoma

  • Bilobalide isolated from the leaves of Ginkgo biloba has several pharmacological activities such as neuroprotective, anti-inflammatory, and anticonvulsant. However, the effect of bilobalide on cancer has not been clearly established. The main purpose of this study was to investigate the effect of bilobalide on cell growth and apoptosis induction in FaDu human pharyngeal squamous cell carcinoma. This was examined by 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay, nuclear 4′,6-diamidino-2-phenylindole dihydrochloride staining, DNA fragmentation analysis, and immunoblotting. Bilobalide inhibited the growth of FaDu cells in dose- and time-dependent manners. Treatment with bilobalide resulted in nuclear condensation and DNA fragmentation in FaDu cells. Furthermore, it promoted the proteolytic cleavage of procaspase-3/-7/-8/-9 with increase in the amount of cleaved caspase-3/-7/-8/-9. Bilobalide-induced apoptosis in FaDu cells was mediated by the expression of Fas and the activation of caspase-8, caspase-3, and poly (ADP-ribose) polymerase. Immunoblotting revealed that the antiapoptotic mitochondrial protein Bcl-2 was downregulated, but the proapoptotic protein Bax was upregulated by bilobalide in FaDu cells. Bilobalide significantly increased Bax/Bcl-2 ratio. These results suggest that bilobalide inhibits cell proliferation and induces apoptosis in FaDu human pharyngeal squamous cell carcinoma via both the death receptor-mediated extrinsic apoptotic pathway and the mitochondrial-mediated intrinsic apoptotic pathway.

A study on the Compliance and Educational Demand of Renal Transplantation Patient (신장이식 환자의 치료지시 이행정도와 교육 요구도에 관한 연구)

  • This study was started for the purpose of providing the basic data for continous managment of kidney transplantation patients after discharge. This study was conducted on 180 patients who received renal transplants at three hospital( B, M, P) pusan, korea. The data collection was done for june 1, to August 31, 2002. General characteristics, renal transplantaton characteristics, physical characteristics, the level of compliance and the degree of educational demand were done by the number and percentage, the mean, standard deviation. The level of compliance and educational demand followed by the characteristics of general and kidney transplantation were analyzed by t-test and ANOVA. The result were as fallows; 1. Man was higher than woman such as 60.0%, Mean age was 42.5 years old, Average total duration of after operation was 5.5 years. 2. Cases of systolic blood pressure over 140mmHg were 10.0%, cases of diastolic blood pressure over 90mmHg were 22.8% and obesity factor in BMI was 15.6%. The person who daily water intake amount is 5000cc was 0.6%, the case that daily urine output is below 1000cc was 8.9%, and the case that urine output is zero was 0.6%. 3. The mean score of compliance was 77.47 point, The score in medication part was highest such as 4.67 point, that in stress situation was lowest such as 3.50 point. 4. The average score of educational demand was 154.02 point, and physical state part was 4.36 points highest, activation part was 3.48 points lowest. As a role of nurse Confirmation of compliance is very important encourage to make good through regular hospital visitation, point out the noncompliance part and then increase compliance of renal transplantation patient As well there will be maintain the normal kidney function to satisfy educational demand through continous education.

A Comparative Study on Domestic and International Evaluation Criteria of Commissioning for Sustainable Building Certification (국내외 녹색건축인증에서의 커미셔닝 평가기준에 대한 비교 연구)

  • Ministry of Land, Infrastructure and Transport established Heating and cooling energy conservation plan of apartment for 2017, reducing 90% compared to energy consumption of 1990. Also, at 2015 United Nations Climate Change Conference, the Ministry announced to reduce 37% of estimated $CO^2$ emission(850 million ton) at 2030. To satisfy this, it is urgent to reduce $CO^2$ emission due to the building, which takes majority amount of total $CO^2$ emission, and public interest for the commissioning of intensified building performance is uprising. However, the building commissioning at construction is not enacted, and not activated due to the lackness of promotion and will of general contractor. Furthermore, commissioning is not mandatory for obtaining G-SEED certification. Therefore, this paper describes effectiveness of commissioning with comparison of commissioning evaluation criteria of G-SEED with LEED and BREEAM, to propose making commissioning as mandatory process and drive changes in perception among stakeholders for activation of domestic building commissioning.

Optimization of 150kW Cogeneration Hybrid System (150kW급 열병합발전 하이브리드 시스템 최적화 연구)

  • The importance of the more efficient cogeneration system is emphasized. Also the more clean energy is needed at recent energy system. The cogeneration system using Lean burn engine is more preferred to the system using Rich burn engine because of the electrical efficiency. Although the cogeneration system using Lean burn engine is economically preferred, because of the NOx emission level, the system using Rich burn engine with 3-way catalyst can only be used in Korea. The NOx regulation level is 50ppm at oxygen level 13%. The cogeneration hybrid system using Lean burn engine is up to be optimized because of the large amount of the extra-fuel at the after-burner system. The after-burner system at different concept was applied. The reduction time for the activation temperature of the DeNOx catalyst was achieved by making a hole between the combustor and boiler. Because of the lowered fuel consumption, the lowered temperature level was optimized by blocking the hole of the boiler The optimized cogeneration hybrid system consumes $76Nm^3/h$ LNG to produce 150kW electricity compared to before optimization $103Nm^3/h$ LNG. The system was accurately evaluated and the result is following ; 90% total efficiency, below 10 ppm NOx, 50ppm CO, 25ppm HC. The cogeneration hybrid system can meet the current NOx level and exhaust gas regulation. It can achieve the clean combustion gas and efficient cogeneration system.

Effects of GP extract on oxidative stress and contact dermatitis in NC/Nga Mice induced by DNCB (가감평위산(加減平胃散)이 산화적 손상과 접촉성 피부염에서의 면역 조절작용에 미치는 영향)

  • To evaluate the effects of GP on contact dermatitis, we examined the composition of immune cells from drain lymph node in DNCB-induced contact dermatitis murine model NC/Nga mice. And the amount of pathologic cytokines of spleen and antioxidant activity were investigated. The results were summarized as followers; 1. GP did not show cytotoxic effect on mLFC in vitro. 2. GP did not have hepatotoxicity in vivo in the level of ALT, AST. 3. GP decreased the production of DPPH and in a dose-dependent. 4. GP significantly decreased total cell number of DLN in DNCB-induced NC/Nga mice compared to the untreated control group. 5. GP significantly decreased the number of CD3+, CD19+, CD4+, CD8+, CD3+/CD69+ and CD4+/CD45+ in DLN of DNCB-induced NC/Nga mice compared to the untreated control group. 6. GP significantly reduced the level of IL-4 and IFN-$\gamma$ in splenocytes of DNCB-induced NC/Nga mice compared to the untreated control group. Taken together above results, GP have therapeutic effects on contact dermatitis by regulating T cell activation. This study warranted further investigations of molecular mechanisms of GP on contact dermatitis.
