Activation of Antioxidant-Response Element (ARE), Mitogen- Activated Protein Kinases (MAPKs) and Caspases by Major Green Tea Polyphenol Components during Cell Survival and Death

  • Green tea polyphenols (GTP) have been demonstrated to suppress tumorigenesis in several chemical-induced animal carcinogenesis models, and predicted as promising chemopreventive agents in human. Recent studies of GTP extracts showed the involvement of mitogen-activated protein kinases (MAPKs) in the regulation of Phase II enzymes gene expression and induction of apoptosis. In the current work we compared the biological actions of five green tea catechins: (1) induction of ARE reporter gene, (2) activation of MAP kinases, (3) cytotoxicity in human hepatoma HepG2-C8 cells, and (4) caspase activation in human cervical squamous carcinoma HeLa cells. For the induction of phase IIgene assay, (-)-epigallocatechin-3-gallate (EGCG) and (-)-epicatechin-3-gallate (ECG) potently induced antioxidant response element (ARE)-mediated luciferase activity, with induction observed at 25 $\mu\textrm{m}$with EGCG. The induction of ARE reporter gene appears to be structurally related to the 3-gallate group. Comparing the activation of MAPK by the five polyphenols, only EGCG showed potent activation of all three MAPKs (ERK, JNK and p38) in a dose- and time-dependent manner, whereas EGC activated ERK and p38. In the concentration range of 25 $\mu\textrm{m}$ to 1 mM, EGCG and ECG strongly suppressed HepG2-ARE-C8 cell-growth. To elucidate the mechanisms of green tea polyphenol-induced apoptosis, we measured the activation of an important cell death protein, caspase-3 induced by EGCG, and found that caspase-3 was activated in a dose- and time-dependent manner. Interestingly, the activation of caspase-3 was a relatively late event (peaked at 16 h), whereas activation of MAPKs was much earlier (peaked at 2 h). It is possible, that at low concentrations of EGCG, activation of MAPK leads to ARE-mediated gene expression including phase II detoxifying enzymes. Whereas at higher concentrations of EGCG, sustained activation of MAPKs such as JNK leads to apoptosis. These mechanisms are currently under investigation in our laboratory. As the most abundant catechin in GTP extract, we found that EGCG potently induced ARE-mediated gene expression, activated MAP kinase pathway, stimulated caspase-3 activity, and induced apoptosis. These mechanisms together with others, may contribute to the overall chemopreventive function of EGCG itself as well as the GTP.

Role of Poly (ADP-ribose) Polymerase Activation in Chemical Hypoxia-Induced Cell Injury in Renal Epithelial Cells

  • The molecular mechanism of ischemia/reperfusion injury remains unclear. Reactive oxygen species (ROS) are implicated in cell death caused by ischemia/reperfusion in vivo or hypoxia in vitro. Poly (ADP-ribose) polymerase (PARP) activation has been reported to be involved in hydrogen peroxide-induced cell death in renal epithelial cells. This study was therefore undertaken to evaluate the role of P ARP activation in chemical hypoxia in opossum kidney (OK) cells. Chemical hypoxia was induced by incubating cells with antimycin A, an inhibitor of mitochondrial electron transport. Exposure of OK cells to chemical hypoxia resulted in a time-dependent cell death. In OK cells subjected to chemical hypoxia, the generation of ROS was increased, and this increase was prevented by the $H_2O_2$ scavenger catalase. Chemical hypoxia increased P ARP activity and chemical hypoxia-induced cell death was prevented by the inhibitor of PARP activation 3-aminobenzamide. Catalase prevented OK cell death induced by chemical hypoxia. $H_2O_2$ caused PARP activation and $H_2O_2-induced$ cell death was prevented by 3-aminobenzamide. Taken together, these results indicate that chemical hypoxia-induced cell injury is mediated by PARP activation through H202 generation in renal epithelial cells.

Characteristics of the Catalysts Using Activated Carbon Nanofibers with KOH as the Support of Anode Catalyst for Direct Methanol Fuel Cell

  • Carbon nanofiber (CNF) grown catalytically was chemically activated with KOH to attain structural change of CNF. The structural changes of CNF through KOH activation were investigated by using BET and SEM. From the results of BET, it was found that KOH activation was effective to develop particular sizes of pores on the CNF surface, increasing the surface area of CNF. Activated CNF was applied as an anode catalyst support of fuel cell. The effects of different activation conditions including the activation temperature and the activation time on the specific surface area of the CNF activated with KOH were investigated to obtain appropriate structure as a catalyst support. The 60 wt% Pt-Ru catalyst prepared was observed by using TEM and XRD.

Comparison of the Changes in the Activation of the Quadriceps Muscle based on the Plantar Flexion Degree of the Ankle Joint in Healthy Young Females during the Stand-to-Sit movement

  • Purpose: The purpose of this study was to compare the changes in the muscle activation of the quadriceps muscle (rectus femoris, vastus lateralis, vastus medialis) during the stand-to-sit (StandTS) movement according to the plantar flexion angle of the ankle joint. Methods: A total of 22 healthy young females participated in this study. During the StandTS under the three conditions (plantarflexion angle 0°, 20°, and 45° of the ankle), electromyography (EMG) data (% maximum voluntary iso¬metric contraction) of the rectus femoris, vastus lateralis, and vastus medialis were recorded using a wireless surface EMG system. Results: There was a significant difference in the muscle activation of rectus femoris, vastus lateralis, and vastus medialis according to the plantar flexion angle (0°, 20°, and 45°) of the ankle. The muscle activation of the quadriceps was the highest at a 45° angle of plantarflexion and the lowest at 0°. One-way repeated ANOVA was used to analyze the muscle activation data of the lower extremity muscles according to the angle of the ankle joint. Conclusion: Based on the results of our study, it was confirmed that the muscle activity of the quadriceps can be increased even in the StandTS movement, which involves the eccentric contraction of the quadriceps muscle. This suggests that maintaining a plantar flexion posture for a long time, say by wearing high-heeled shoes, can quickly cause muscle fatigue in the lower-limb muscles, which can cause a decrease in balance ability leading to falls.

The Effects of Kinesio Taping on the Intestinal Activation (키네시오 테이핑이 장 활성도에 미치는 영향)

  • Background : The purpose of this study is to find out the impact of kinesio-taping method for female university students, who are vulnerable to constipation, on the activation of intestine. Methods : This study has been conducted for a total of three weeks including the pre-inspection, and it explained the purpose of the study upon selecting the adequate subjects who met the criteria for constipation among the female students. And It chose the nine students, who agreed to the experiment, as the subjects. It verified the frequency and time of excretion before experiment and the constipation ejaculation measure, and compared pre-arbitration with post-arbitration as for the time and frequency of excrement, the point of constipation ejaculation measure, etc. after two weeks since the application of tape. Result : The frequency of excrement after arbitration increased compared with pre-arbitration as result of the application of kinesio-taping method, and the time, which was taken for excrement, decreased. It was also shown that the point decreased in the point comparison for constipation ejaculation measure; thus, it obtained the result that showed the improvement of the level of constipation and all were statistically significant(p<.05). Conclusion : It is believed that kinesio-taping method has a positive impact on the activation of intestine of female university students.

Prediction of Creep Deformation and Short Time Rupture Life of AZ31 Magnesium Alloy below 0.5Tm (0.5Tm 이하에서의 AZ31 마그네슘합금의 크리이프 변형과 단시간 파단수명예측)

  • The initial strain, the applied stress exponent, the activation energy, and rupture time in AZ31 magnesium alloy have been measured in order to predict the deformation mechanism and rupture life of creep over the temperature range of 423-443K. Creep tests were carried out under constant applied stress and temperature, and the lever type tester and automatic temperature controller was used for it, respectively. The experimental results showed that the applied stress exponent was about 9.74, and the activation energy for creep, 113.6KJ/mol was less than that of the self diffusion of Mg alloy including aluminum. From the results, the mechanism for creep deformation seems to be controlled by cross slip at the temperature range of 423-443K. Also the higher the applied stress and temperature, the higher the initial strain. And the rupture time for creep decreased as quadratic function with increasing the initial strain in double logarithmic axis.

  • This study was designed to validate Spreading Activation Theory (SAT) for an evaluation methodology for menu structure through Eye-Tracking approach. When a visual search is on the way, more eye fixations and time are necessary to visually process complex and vague area. From the aspect of recognition, well-designed menu structures were hypothesized to have fewer numbers of fixations and shorter duration because well-designed menu structures reflecting the users' mental model would be well matched with the product's menu structure, resulting in reducing the number of fixations and duration time. The results show that the shorter reaction times for SAT had significantly fewer numbers of fixation and shorter duration time as the hypothesis for this study stated. In conclusion, SAT was proved to be an effective evaluation methodology for menu structure with the eye tracking equipment. In addition, using SAT instead of the real performance experiment would be useful for designing user-centered systems and convenient information structures because SAT was proven to be the theoretical background for design and evaluation of menu structures.

Effects of Morphology, Reproductive Cycle, Incubation Time and Activation of Oocytes on Developmental Rate of Embryos Fertilized in vitro (난자의 형태, 번식주기, 배양시간 및 활성화 처리가 개 난자의 체외수정후 발생에 미치는 영향에 관한 연구)

  • The study was carried out to investigate the effects of morphology, reproductive cycle, incubation time and activation of oocytes in vitro maturation of canine oocytes and development of canine IVM/IVF embryos. The results were summarized as follows: 1. The developmental rates to 16 cells of fresh, salts and 4$^{\circ}C$-stored oocytes with and without cumulus cells were 14.3%, 5.0% and 7.5%, 2.8% and 5.7%, 0.0%, respectively. The rate of oocytes with cumulus cells(5.7%~14.3%) was higher than that of denuded oocytes(0.0%~5.0%). 2. The developmental rate to If cells of in vitro cultured oocytes recovered from ovaries collected at different stages of the reproductive cycle were 0.0%, 10.7%, 1.5%, respectively. 3. The developmental rate to 16 cells of fresh oocytes with cumulus cell cultured for 24, 32 and 48 hrs in $CO_2$ incubator were 0.0%, 5.3%, 11.8%, respectively. The rate of oocytes cultured for 48 hrs was higher than that oocytes cultured for 24 and 32 hrs. 4. The development to If cells treated activation and non-activation oocytes were 15.0%, 6.7%, respectively. The rate of oocytes treated activation was higher than that oocyte treat non-activation.

Correlation between Faster Response Time and Functional Activities of Brain Regions during Cognitive Time Management (인지적 시간관리에 필요한 기능적 뇌 활성 영역과 반응시간의 상관관계)

  • Purpose: This study was designed to determine the correlation between faster response time and functional activities of brain regions during cognitive time management. Methods: Twelve healthy subjects participated in this experiment. Subjects performed the serial reaction time task (SRTT), which was designed by the Superlab program, during fMRI scanning. When the 'asterisk' appeared in the 4 partition spaces on the monitor, the subject had to press the correct response button as soon as possible. Results: fMRI results showed activation of the left primary sensorimotor cortex, both premotor areas, the supplementary motor area, posterior parietal cortex and cerebellum. There were significant correlations, from moderate to strong, between faster reaction time and BOLD signal intensity in activated areas. Conclusion: These results suggest that motor skill learning to be needed cognitive time management is associated with greater activation of large scale sensorimotor networks.

The Effect of Surface Treatment on Creep Behaviors of Mg Alloy (마그네슘 합금의 크리이프 거동에 표면처리가 미치는 영향)

  • The apparent activation energy, the applied stress exponent, and rupture life have been measured from creep experiments over the range of $200^{\circ}C$ to $220^{\circ}C$ and the applied stress range of 64MPa to 94MPa. The materials were used AZ31 magnesium alloys treated by plasma electrolytic oxidation of $20{\mu}m$ and $40{\mu}m$ at surface to investigate the its influence on creep behavior, and creep tests were carried out under constant applied stress and temperature. The experimental results showed that the dipper the thickness of surface treatment the higher the activation energy and stress exponent. And the higher temperature and applied stress, the lower stress exponent and activation energy, respectively. Also the dipper the thickness of surface treatment the longer creep rupture time.
