  • The gas-phase formation of polychlorinated naphthalenes (PCNs) and dibenzofurans (PCDFs) was experimentally investigated by slow combustion of the three chlorophenols (CPs): 2-chlorophenol (2-CP), 3-chlorophenol (3-CP) and 4-chlorophenol (4-CP), in a laminar flow reactor over the range of 550 to $750^{\circ}C$ under oxidative condition. Contrary to the a priori hypothesis, different distributions of PCN isomers were produced from each CP. To explain the distributions of polychlorinated dibenzofuran (PCDF) and PCN congeners, a pathway is proposed that builds on published mechanisms of PCDF formation from chlorinated phenols and naphthalene formation from dihydrofulvalene. This pathway involves phenoxy radical coupling at unsubstituted ortho-carbon sites followed by CO elimination to produce dichloro-9, 10-dihydrofulvalene intermediates. Naphthalene products are formed by loss of H and/or Cl atoms and rearrangement. The degree of chlorination of naphthalene and dibenzofuran products decreased as temperature increased, and, on average, the naphthalene congeners were less chlorinated than the dibenzofuran congeners. PCDF isomers were found to be weakly dependent to temperature, suggesting that phenoxy radical coupling is a low activation energy process. Different PCN isomers, on the other hand, are formed by alternative fusion routes from the same phenoxy radical coupling intermediate. PCN isomer distributions were found to be more temperature sensitive, with selectivity to particular isomers decreasing with increasing temperature.

  • Internet purchase is increasing according to the activation of Electronic Commerce. Currently, the hardware is main purchase items in the Electronic commerce. The on-line purchase of digital product, that is electronic file type of product, has many merits such as, does not cost in logistics, is ease of searching the product, and is possible of 'Try and Buy' system. But, actually digital products have not been major proudct because of tits piracy problem. The piracy problem of digital products comes from the lack of safety in DP license control technology. Current EC has no method to identify the object that has the right ot use the digital product. The protecting of object change is impossible and the protection of hacking is not enough and EC has any problems in efficient license control because of the incompletion of EC structure. In this paper, e propose the design and implementation of on-line DP circulation system, which can activate on-line circulation of DP in EC and protect its piracy, by using the new license control technology, that is Dynamic License control technology.

Comparison of Virulence Factors of Enterococci from Intestinal Drugs, Infant Feces and Clinical Isolates (정장제, 신생아 분변 및 병원에서 분리한 장구균의 병독성인자 비교)

  • Three isolates, E. faecium P1, P2 and P3, from intestinal drugs of three phamaceutical companies, four clinical vancomycin resistant isolates, E. faecium V1, V2, V3 and E. faecalis V4, and three isolates, E. faecalis DW01, DW07 and DW14, from infant feces were tested for the presence of virulence genes, ace, agg, esp, efaA, gelE, sprE, vanA and vanB as well as fsrABC, regulatory genes of gelE and sprE, cylMBA, cytolysin activation genes and cpd, cob and ccf, pheromone genes by PCR and for their phenotype activities such as protease, biofilm formation, cell clumping and hemolysis. The genes encoding cell surface adherence proteins, ace, agg, esp and efaA, were predominantly amplified from the vancomycin resistant strain V4 and the fecal isolates DW01, DW07 and DW14. Both protease and biofilm formation activity were detected only from E. faecalis V4 from which the PCR products of gelE and spreE as well as fsrABC were amplified. The pheromone genes were amplified from the V4, DW01, DW07 and DW14 strains and these strains showed clumping activity. Biofilm formation was observed from the strains DW01, DW07 and DW14, all of which produced PCR products of pheromone, and V4 as well. Whole cytolysin regulator genes were amplified from DW01, DW07 and DW14 and ${\beta}$-hemolysis activity was detected from these strains. Any virulence genes or activities except the pheomone gene ccf were not detected from the pharmaceutical isolates, E. faecium P1, P2 and P3.

  • Cognitive model, that is cognitive architecture, is the model expressed with computer program to show the process how human solve a certain problem and it is continuously under investigation through various fields of study such as cognitive engineering, computer engineering, and cognitive psychology. In addition, the much extensive applicability of cognitive model usually helps it to be used for quantitative prediction of human Behavior or Natural programming of human performance in many HCI areas including User Interface Usability, artificial intelligence, natural programming language and also Robot engineering. Meanwhile, when a system designed, an usability test about conceptual design of interface is needed and in this case, analysis evaluation using cognitive model like GOMS or ACT-R is much more effective than empirical evaluation which naturally needs products and subjects. In particular, if we consider the recent trend of very short-end term between a previous technology development and the next new one, it would take time and much efforts to choose subjects and train them in order to conduct usability test which is repeatedly followed in the process of a system development and this finally would bring delays of development of a new system. In this study, we predicted quantitatively the human behavior processes which contains cognitive processes for menu selection in touch screen interface through ACT-R, one of the common method of usability test. Throughout the study, it was shown that the result using cognitive model was equal with the result using existing empirical evaluation. And it is expected that cognitive model has a possibility not only to be used as an effective methodology for evaluation of HCI products or system but also to contribute the activation of HCI cognitive modeling in Korea.

Anti-proliferative Effects by Aqueous Extract of Cordyceps Militaris in Human Leukemic U937 Cells (동충하초 추출물에 의한 U937 인체 백혈병 세포의 성장억제 효과)

  • Cordyceps militaris is a medicinal fungus, which has been used for patient suffering from cancer in Oriental medicine. It was reported previously that C. militaris extracts are capable of inhibiting tumor growth, however, the anti-poliferative effects of human cancer cells have not been poorly understood. In this study, to elucidate the growth inhibitory mechanisms of human cancer cells by treatment of aqueous extract of C. militaris (AECM) we investigated the anti-proliferative effects of AECM in human leukemia U937 cell line. AECM treatment inhibited the growth of U937 cells and induced the apoptotic cell death in a concentration-dependent manner, which was associated with morphological changes. We observed the up-regulation of cyclin-dependent kinase (Cdk) inhibitor p21(WAF1/CIP1) by p53-independent manner and activation of caspase-3 in AECM-treated U937 cells, however, the activity of caspase-9 was remained unchanged. Additionally, AECM treatment caused a dose-dependent inhibition of the expression of telomere regulatory gene products such as human telomere reverse transcriptase (hTERT) and telomerase-associated protein-1 (TEP-1). Taken together, these findings suggest that AECM-induced inhibition of human leukemic cell proliferation is associated with the induction of apoptotic cell death via modulation of several major growth regulatory gene products, and C. militaris may have therapeutic potential in human lung cancer.

미소조류의 응용연구(개관) (Applied Researches on Microalgae(Overview))

  • Trends in the applied microalgal researches were described as those in two separate periods. During the first period (earlie. than 1970) . most researches were oriented to using the whole microalgal cells as feed, food, fertilizer, and agent for the treatment of organic waste water. Since early 1970's (the second period) the number of researches on the production of specific cell metabolites has grown so rapidly. Many researchers endeavored after the very expensive 'natural products' form microalge such as vitamins, amino acids, $\beta-carotene$, phycofluor, pharmaceuticals, biologically active compounds, $H_2$ gas, and deutrated chemicals. On the other hand, the applied microalgal researches in Korea are still in the early stage of developments, and urgent activation of the related researchers is quite important to meet the future needs in the microalgal products. Systems for the management of the management of the microalgal clonal cultures from Korean waters should be established soon, which will support many microalgal researchers in Korea.

WASTE CLASSIFICATION OF 17×17 KOFA SPENT FUEL ASSEMBLY HARDWARE

  • Metal waste generated from the pyroprocessing of 10 MtU of spent fuel was classified by comparing the specific activity of a relevant radionuclide with the limit value of the specific activity specified in the Korean acceptance criteria for a lowand intermediate-level waste repository. A Korean Optimized Fuel Assembly design with a 17${\times}$17 array, an initial enrichment of 4.5 weight-percent, discharge burn-up of 55 GWD/MtU, and a 10-year cooling time was considered. Initially, the mass and volume of each structural component of the assembly were calculated in detail, and a source term analysis was subsequently performed using ORIGEN-S for these components. An activation cross-section library generated by the KENO-VI/ORIGEN-S module was utilized for top-end and bottom-end pieces. As a result, an Inconel grid plate, a SUS plenum spring, a SUS guide tube subpart, SUS top-end and bottom-end pieces, and an Inconel top-end leaf spring were determined to be unacceptable for the Gyeongju low- and intermediate-level waste repository, as these waste products exceeded the acceptance criteria. In contrast, a Zircaloy grid plate and guide tube can be placed in the Gyeongju repository. Non-contaminated Zircaloy cladding occupying 76% of the metal waste was found to have a lower level of specific activity than the limit value. However, Zircaloy cladding contaminated by fission products and actinides during the decladding process of pyroprocessing was revealed to have 52 and 2 times higher specific activity levels than the limit values for alpha and $^{90}Sr$, respectively. Finally, it was found that 88.7% of the metal waste from the 17${\times}$17 Korean Optimized Fuel Assembly design should be disposed of in a deep geological repository. Therefore, it can be summarized that separation technology with a higher decontamination factor for transuranics and strontium should be developed for the efficient management of metal waste resulting from pyroprocessing.

Anti-Inflammatory Principles from the Fruits of Evodia rutaecarpa and Their Cellular Action Mechanisms

  • The fruits of Evodia rutaecarpa Benth (Rutaceae) has long been used for inflammatory disorders and some anti-inflammatory actions of its constituents such as dehydroevodiamine, evodiamine and rutaecarpine were previously reported. Since the pharmacological data is not sufficient to clearly establish the scientific rationale of anti-inflammatory medicinal use of this plant material and the search for its active principles is limited so far, three major constituents (evodiamine, rutaecarpine, goshuyuamide II) were evaluated for their anti-inflammatory cellular action mechanisms in the present study. From the results, evodiamine and rutaecarpine were found to strongly inhibit prostaglandin $E_2$ synthesis from lipopolysaccharide-treated RAW 264.7 cells at $1-10{\mu}M$. Evodiamine inhibited cyclooxygenase-2 induction and NF-kB activation, while rutaecarpine did not. On the other hand, goshuyuamide II inhibited 5-lipoxygenase from RBL-1 cells $(IC_{50}=6.6{\mu}M)$, resulting in the reduced synthesis of leukotrienes. However, these three compounds were not inhibitory against inducible nitric oxide synthase-mediated nitric oxide production from RAW cells up to $50{\mu}M$. These pharmacological properties may provide the additional scientific rationale for anti-inflammatory use of the fruits of E. rutaecarpa.

Differentiation and identification of ginsenoside structural isomers by two-dimensional mass spectrometry combined with statistical analysis

  • Background: In the current phytochemical research on ginseng, the differentiation and structural identification of ginsenosides isomers remain challenging. In this paper, a two-dimensional mass spectrometry (2D-MS) method was developed and combined with statistical analysis for the direct differentiation, identification, and relative quantification of protopanaxadiol (PPD)-type ginsenoside isomers. Methods: Collision-induced dissociation was performed at successive collision energy values to produce distinct profiles of the intensity fraction (IF) and ratio of intensity (RI) of the fragment ions. To amplify the differences in tandem mass spectra between isomers, IF and RI were plotted against collision energy. The resulting data distributions were then used to obtain the parameters of the fitted curves, which were used to evaluate the statistical significance of the differences between these distributions via the unpaired t test. Results: A triplet and two pairs of PPD-type ginsenoside isomers were differentiated and identified by their distinct IF and RI distributions. In addition, the fragmentation preference of PPD-type ginsenosides was determined on the basis of the activation energy. The developed 2D-MS method was also extended to quantitatively determine the molar composition of ginsenoside isomers in mixtures of biotransformation products. Conclusion: In comparison with conventional mass spectrometry methods, 2D-MS provides more direct insights into the subtle structural differences between isomers and can be used as an alternative approach for the differentiation of isomeric ginsenosides and natural products.
