The mosquito-larvicidal Cry4B protein from Bacillus thuringiensis subsp. israelensds was expressed in Escherichia coli. Upon activation by trypsin, the 130-kDa protoxin was processed into the 65-kDa active toxin containing two polypeptide fragments of ca. 47 and ca. 20 kDa. These two polypeptides are products of internal cleavages on the exposed loop connecting helices 5 and 6 in the seven-helical bundle domain. PCR-based mutagenesis was employed to introduce an additional cleavage site into the loop connecting helices 3 and 4. A series of amino acid changes were introduced into the targeted loop, resulting in seven mutant protoxins. Upon digestion with trypsin, a group of mutants with arginine introduced into the loop (EPRNQ, EPNRNQ, EPRNP, ESRNP and SSRNP) produced polypeptide products similar to those of the wild type (EPNNQ). When the loop, SSRNP, was expanded by an insertion of either asparagine (NSSRNP) or valine (VSSRNP), an additional cleavage was detected with proteolytic products of 47,12 and 6 kDa. This cleavage was confirmed to be at the introduced arginine residue by N-terminal sequencing. The mosquito larvicidal assay against Aedes aegypti demonstrated a relatively unchanged toxicity for the mutants without cleavage and reduced toxicity for those with an additional cleavage.

The microwave plasma and catalytic reaction have been employed to investigate the activation of methane to hydrogen and higher hydrocarbons at low gas temperature. The catalytic activity of Fe, Ni, Pt Pd metal catalysts were also studied in this reaction system. With increasing plasma power at a $CH_{4}$ flow rate of 20 ml/min, C2+ products increased from 29 to $42\%$, whereas hydrogen from 60 to $65\%$. When catalysts were loaded below the plasma region, the selectivitity of ethylene md acetylene increased but the yield of C2+ products remained constant. The usage of ECR electric fie3d and Pd-Ni bimetal catalyst produced a minimum C2+ yield of $64\%$.

Therefore, in this study, consumers of the craft cultural products awareness and marketability to domestic craft industry status and enhance the use of the show. The purpose of this study was to document a phenomenon of rural tourism in Korea by Investigating characteristics of 50 villages that had participated in a competition of 'stimulation of rural village' The results are as follows: 1) The degrees of activation of rural tourism varied according to marketing efforts made by individual provincial(local) government. The results of contest indicated that provinces such as Gangwon-do, Chungnam-do, and Gyunggi-do showed highest performance among other competing local governments; 2) The results revealed that the total profit generated from direct sale of farm products outnumbered profits gained from sales of lodging and foods. This indicated that there is a room for improvement with respect to boosting more sales on lodging and food; 3) The regression analysis indicated a significant relationship between profits earned from sales of experiential tourism product and those from direct sale of farm products(multi-correlation coefficient: 0.38); 4) It was identified that Gyunggi and Ganwon provinces were ranked first in sales of foods and those of lodging, respectively; 5) Finally, it was showed that among the 50 participating villages, only 16 ones hold festivals by means of attracting tourists from outside.

Objective : Accumulation of advanced glycation end-products (AGE) and mitochondrial glycation is importantly implicated in the pathological changes of the brain associated with diabetic complications, Alzheimer disease, and aging. The present study was undertaken to determine whether sildenafil, a type 5 phosphodiesterase type (PDE-5) inhibitor, has beneficial effect on neuronal cells challenged with AGE-induced oxidative stress to preserve their mitochondrial functional integrity. Methods : HT-22 hippocampal neuronal cells were exposed to AGE and changes in the mitochondrial functional parameters were determined. Pretreatment of cells with sildenafil effectively ameliorated these AGE-induced deterioration of mitochondrial functional integrity. Results : AGE-treated cells lost their mitochondrial functional integrity which was estimated by their MTT reduction ability and intracellular ATP concentration. These cells exhibited stimulated generation of reactive oxygen species (ROS), disruption of mitochondrial membrane potential, induction of mitochondrial permeability transition, and release of the cytochrome C, activation of the caspase-3 accompanied by apoptosis. Western blot analyses and qRT-PCR demonstrated that sildenafil increased the expression level of the heme oxygenase-1 (HO-1). CoPP and bilirubin, an inducer of HO-1 and a metabolic product of HO-1, respectively, provided a similar protective effects. On the contrary, the HO-1 inhibitor ZnPP IX blocked the effect of sildenafil. Transfection with HO-1 siRNA significantly reduced the protective effect of sildenafil on the loss of MTT reduction ability and MPT induction in AGE-treated cells. Conclusion : Taken together, our results suggested that sildenafil provides beneficial effect to protect the HT-22 hippocampal neuronal cells against AGE-induced deterioration of mitochondrial integrity, and upregulation of HO-1 is involved in the underlying mechanism.

Recently, the fresh logistics recognizes to be created the high value-added part for logistics companies. Because the fresh logistics, which provides the system of temperature control and sanitary management, is able to be created more better high value-added products than the existed logistics system. In this research, we suggest the fresh logistics cluster for the proposal of activation in the field of food industry, which apply good activity for fresh logistics, and show the present condition and matters domestic fresh logistics market. This study did analyze the status & volume of perishable cargo which come from the imported Chinese fresh products representatively, at Incheon port and assume to build the fresh logistics cluster complex at Songdo FEZ, which is available developing and located near by Incheon port. The actual intensive study should be progressed for Korean logistics companies in order to optimize to the suitable fresh logistics system, need to set up the policy and management of statistical data concerning fresh logistics system.

Fundamental problems of e-commerce are exaggerated advertising of products, lack of trust in products or suppliers, and false reviews. As a solution, I have merged the concept of trust service embedded in social network service(SNS) with commercial domain to develop a new type of service called "Reliable SNS Commerce Service". The contents developed in this paper are as follows: first, online community functions for users to provide services; second, commerce functions; and third, functions for linking SNS and commerce. Through the reliability information presented in this paper, the seller provides more reliable and objective purchase information to the buyer about the sales items, thereby contributing to the sales by increasing the probability of the actual purchase. The buyer can purchase the higher-quality products with confidence. The service providers can gain the reputation as a reliable site for purchasing members. In conclusion, this paper provides a positive effect to all the participants, which will contribute to the development of a new commerce market and activation of electronic commerce.

Because of the radical changes in the domestic and foreign economic circumstances Koreaa fisheries is confronted with difficulties. Also migration of labor to other industries and rise of wage level in Korean fisheries deteriorate managerial conditions. But in Korea which has little natural resourses fisheries is still more important. These roles of fisheries in national economy mentioned above are to be excuted forward. In spite of the radical change in the economic circumstances at the end of the UR fisheries if crucial in Korea as a industry. For this our fisheries is to be made to compete with foreign fisheries. Through the cheap price and high quality our fisheries must be came to compete with foreign fisheries and meet the people's needs for marine products. For this it is necessary to maintain high productivity and competitive power. Now with the exception of a portion of the deep - sea fishing, our fisheries is generally paltry, Especially inshore fishery which is the main stock in our fisheries is very paltry and so productivity and competetive power are very low. So to develop our fisheries which has s comparative disadvantage active polices that follows below are to be promoted on a large scale. \circled1 improvement of structure \circled2 augmentation of productivity in fishing ground by making fisheries resourses \circled3 enlargement of finantial and monetary assistance \circled4 effective administration of fisheries cooperative union \circled5 activation of R&D etc These polices which need to be scientific and comprehensive are very valuable. Especially without making fishieries resources we cannot expect economy of scale, promotion of productivity and development of fisheries. And we do also endeavour to gather the results of the study and investigation about fisheries domestic and foreign and do ceaselessly put these to practical use systimatically.

Vibrio harveyi, one of the major causal agent of vibriosis, affects a diverse range of marine vertebrates and invertebrates over a wide geographical area. The aims of this study were to investigate the characteristics of extracellular products (ECPs) from an isolate of the pathogenic non‐luminous V. harveyi and the luminous V. harveyi. And ECPs of V. harveyi were examined the pathogenicity to the black rockfish, Sebastes schlegeli, and histopathological traits of internal organs injected by ECPs. Four strains of V. harveyi cultures produced ECPs showing various enzymatic activites (caseinase, gelatinase, phospholipase, lipase, haemolysin). The ECPs showed strong cytotoxicity on macrophages of black rockfish, Sebastes schlegeli and olive flounder, Paralichthys olivaceus. Especially, the virulence of ECPs from the isolate of V. harveyi FR 2 was higher in the intraperitoneally injected black rockfish. Also, the ECPs of V. harveyi FR 2 caused the expansion of sinusoids in the liver, the activation of ellipsoid in the spleen and the sloughing of the epidermal cell in the intestine. It was suggested that the ECPs from V. harveyi play an important role in the pathogenicity process of the V. harveyi.

This study focuses on online communities and the ritual conversations of users when participating in sharing activities. The study aims to understand the social and psychological phenomena that occur between users within the context of Erving Goffman's impression management theory. Case studies and a content analysis were conducted by collecting posts and comments related to fashion products in the sharing activities category on Naver Cafe "Family Sale." On the one hand, the study identified various disposition motives among givers, including a desire for recognition, self-expression, activation of the community, emotional sympathy, goodwill, play, and simple disposition. On the other hand, receivers' purchase motives included the need for a product, reciprocation based on a sense of belonging, play, gift-giving, and simple response. Analyzing the posts of givers and the comments of receivers of fashion products using impression management strategies and dramaturgical analysis, the study interpreted users' impression management and revealed propensities in fashion consumption: fashionability, conspicuousness, value orientation, and economic feasibility. Through ritual conversations, users managed to attain emotional stability on an individual level, while they reinforced collective bonds on a social level. They fulfilled their roles with their own narratives to achieve personal and collective goals in a non-face-to-face situations and non-monetary transactions. This study is significant in that it examines normative communication in an online community and user relationships to understand a recent phenomenon in the fashion industry.
