  • IASL(iodo acetamide) and MSL(maleimide) disordered the orderly helix arrangement of myosin in the rest state of spin level. Especially the effect of IASL was great. Equatorial reflection(10,11) change inferred that myosin head was moved to the vicinity of actin filament by spin level. The intensity change of 143 $\AA$ and 72 $\AA$ could offer infor-mation of the mass projection of population of myosin heads along the filament axis. The slope of intensity profile of the mass projection of 143 $\AA$ and reflection of IASL is appeared and that of MSL is appeared sharply. The dec-rease of 215 $\AA$ reflection intensity the periodical characteristic of 143 $\AA$ reflection by spin label. The raise of MSL actin reflection at 51 $\AA$ and 59 $\AA$ in the actin reflection change refers that the shifted myosin head binds a certain actin or changes an actin structure by spin label effect. Because iodo acetamide has a tendency to decease the actin reflection, actin dose not bind myosin head. From this result, we could conclude that LASL and MSL are spin labeled on SH of myosin head and disordered the helix arrangement of actin.

  • The synaptic vesicle is a specialized structure in presynaptic terminals that stores various neurotransmitters. The actin filament has been proposed for playing an important role in mobilizing synaptic vesicles. To understand the role of actin filament on synaptic vesicle pooling, we characterized synaptic vesicles and actin filament after treatment of brain-derived neurotrophic factor (BDNF) or Latrunculin A on primary cultured neuron from rat embryo hippocampus. Western blots revealed that BDNF treatment increased the expression of synapsin I protein, but Latrunculin A treatment decreased the synapsin I protein expression. The increased expression of synapsin I after BDNF disappeared by the treatment of Latrunculin A. Three-dimensional (3D) tomography of synapse showed that more synaptic vesicles localized near the active zone and total number of synaptic vesicles increased after treatment of BDNF. But the number of synaptic vesicle was 2.5-fold reduced in presynaptic terminals and the loss of filamentous network was observed after Latrunculin A application. The treatment of Latruculin A after preincubation of BDNF group showed that synaptic vesicle number was similar to that of control group, but filamentous structures were not restored. These data suggest that the actin filament plays a significant role in synaptic vesicles pooling in presynaptic terminals.

  • Barley stripe mosaic virus (BSMV) induces massive actin filament thickening at the infection front of infected Nicotiana benthamiana leaves. To determine the mechanisms leading to actin remodeling, fluorescent protein fusions of the BSMV triple gene block (TGB) proteins were coexpressed in cells with the actin marker DsRed: Talin. TGB ectopic expression experiments revealed that TGB3 is a major elicitor of filament thickening, that TGB2 resulted in formation of intermediate DsRed:Talin filaments, and that TGB1 alone had no obvious effects on actin filament structure. Latrunculin B (LatB) treat-ments retarded BSMV cell-to-cell movement, disrupted actin filament organization, and dramatically decreased the proportion of paired TGB3 foci appearing at the cell wall (CW). BSMV infection of transgenic plants tagged with GFP-KDEL exhibited membrane proliferation and vesicle formation that were especially evident around the nucleus. Similar membrane proliferation occurred in plants expressing TGB2 and/or TGB3, and DsRed: Talin fluorescence in these plants colocalized with the ER vesicles. TGB3 also associated with the Golgi apparatus and overlapped with cortical vesicles appearing at the cell periphery. Brefeldin A treatments disrupted Golgi and also altered vesicles at the CW, but failed to interfere with TGB CW localization. Our results indicate that actin cytoskeleton interactions are important in BSMV cell-to-cell movement and for CW localization of TGB3.

  • The effect of temperature on the structure change of the SH of myosin head have been investigated with improved resolution by x-ray diffraction using synchrotron radiation. The movement of myosin head and conformational change of contractile molecules were occurred in the muscle contraction. IASL (iodo acetamide) and MSL (maleimide) disordered the orderly helix arrangement of myosin in the rest state of spin level. The temperature effect on the structure change was great at the UL in the equatorial reflection. But those of IASL and MSL were minor. Equatorial reflection (10, 11) change inferred that myosin head was moved to the vicinity of actin filament by temperature change (from $25^{\circ}C$ to $0^{\circ}C$) at UL, but spin level was not changed. The intensity change of 143 $\AA$ and 72 $\AA$ could offer information of the mass profection of population of myosin heads along the filament axis. The slope of intensity profile of the mass profection of 143$\AA$ and reflection of MSL is appeared sharply and those of UL and IASL were not changed. The decrease of MSL actin reflection at 51 $\AA$ and 59 $\AA$ in the actin reflection change refers that the shifted myosin head binds a certain actin or changes an actin structure. From these results, we could conclude that IASL and MSL were spin labeled on SH of myosin head and disordered the helix arrangement of actin.

  • Saccharomyces cerevisiae contains 10-nm tilament ring which lies just under the inner surface of the plasma membrane within the mother-bud neck. Although H)-nm filaments may he involved in cellular morphogenesis. their role and organization are not clear. Here we report the production of antihodies specific for the CDel2 protein hy use of gene fusion techniques. and studies on the organization and function of IO-nm filaments using these antibodies. The CDCl2 protein arc translated through the whtlle cell cycle and present in the cytosol. 'They are polymerized just before bud emergence and unpolymerized alier cytokinesis. and do not have organizational relationship with actin. Thc possible role of 10-nm filaments is the determination of bud emergence site and completion of cytokinesis.

  • Motivation: Polymerized actin-based cytoskeletal structures are crucial in shape, dynamics, and resilience of a cell. For example, dynamical actin-containing ruffles are located at leading edges of cells and have a significant impact on cell motility. Other filamentous actin (F-actin) bundles, called stress fibers, are essential in cell attachment and detachment. For this reason, their mechanistic understanding provides crucial information to solve practical problems related to cell interactions with materials in tissue engineering. Detecting and counting actin-based structures in a cellular ensemble is a fundamental first step. In this research, we suggest a new method to characterize F-actin wrapping fibers from confocal fluorescence image stacks. As fluorescently labeled F-actin often envelope the fibers, we first propose to segment these fibers by diminishing an energy based on maximum flow and minimum cut algorithm. The actual actin is detected through the use of bilateral filtering followed by a thresholding step. Later, concave actin bundles are detected through a graph-based procedure that actually determines if the considered actin filament is enclosing the fiber.

• Actin filament is a major cytoskeleton in synapses and highly enriched in the presynaptic and postsynaptic compartments. Their roles in synaptic vesicle recycling and synaptogenesis have been extensively studied but functional evidence whether actin filaments are involved in these processes is as yet lacking. Dysfunction in synaptic vesicle recycling causes various diseases such as Alzheimer's disease, Schizophrenia, Bipolar disease, depression etc.
