Panax ginseng C.A. Meyer(Araliaceae) has been traditionally used as an expensive and precious medicine in oriental countries for more than 5, 000 years. Ginseng saponin isolated from the root of Panax ginseng have been regarded as the main effective components responsible for the pharmacological and biological activities. Such as antiaging effects. antidiabetic effects anticancer effects. Protection against physical and chemical stress. Analgesic and antipyretic effects. Effects on the central nervous system, tranquilizing action and others. Thirty kinds of ginsenosides have been so far isolated from ginseng saponin and their chemical structures have been elucidated since 1960's. Among which protopanaxadiol type is 19 kinds. protopanaxatriol type. 10 kinds and oleanane type, one. Since ginsenosides are generally labile under acidic conditions ordinary acid hydrolysis is always accompanied by many side reactions, such as epimerization. hydroxylation and cyclization of side chain of the sapogenins Especially. it is well known that C-20 glycosyl linkage of ginsenoside was hydrolysed on heating with acetic acid to give an equilibrated mixture of 20(S) and 20(R) epimers. And also, the chemical transformations of the secondary metabolites have appeared during the steaming process to prepare red ginseng. Indicating demalonylation of malonyl ginsenosides, elimination of glycosyl residue at C-20 and isomerization of hydroxyl configuration at C-20. But these studies have not provided a comprehensive picture in explaning how these ginsenosides showed val'iotas pharmacological activities of ginseng. Though some of them have been involved in the mechanism of pharmacological actions. Recently, non-saponin components have received a great deal of attention for their antioxidant, anticancer antidiabetic, immunomodulating. anticomplementary activities and so on. To meet the demand for such wide applications, studies on the non-saponin components play an important role in providing a good evidence of pharmacological and biol ogical activities. Among the non-saponin constituents of Korean ginseng, polyacetylenes, phenols. Sesquiterpenes, alkaloids. polysaccharides oligosaccharides, oligopeptides and aminoglycosides together with ginsenosides of terrestrial part are mainly described.
We have isolated a water-extracted novel regulator for blood coagulation from an earthworm, Lumbricus rubellus. As a folk remedy, the earthworm has been known to facilitate blood circulation. After complete heat inactivation of endogenous proteases in the earthworm, an anticoagulant(s) was purified through ammonium sulfate fractionation and three consecutive gel permeation chromatography of Sephacryl S-300, Sephadex G-75, and G-150 by measuring activated partial thromboplastin time (APTT) The anticoagulant was further purified to 2,800 fold with a C4 reversed-phase HPLC This activity was stable under heat ($100^{\circ}C$ for 30 min) and acidic conditions (0.4 N HCl). The effects of this partially purified anticoagulant on thrombin were observed with various substrates such as N${\alpha}$-benzoyl-DL-arginine-p-nitroanilide (BApNA), H-D-phenylalanyl-L-pipecoyl-L-arginine-p-nitroanilide (S-2238), N${\alpha}$-p-tosyl-L-arginine methyl ester (TAME), and fibrinogen as a natural substrate. Only TAME hydrolysis, due to an esterase activity of the enzyme, was inhibited among the chromogenic substrates. In addition, the anticoagulant not only inhibited the conversion of fibrinogen to fibrin but also prolonged the fibrin clot formation monitored with the in vitro coagulation test. Based on these observations, we suggest the significance of measuring the ability of antithrombotic drugs to inhibit the esterase activity of thrombin. In this report, it was also shown that the earthworm indeed contained a water-extractable, heat- and acid-stable anticoagulant which could be used as a novel antithrombotic agent.
가정에서 제조된 청국장으로부터 lactose를 glucose와 galactose로 가수분해하는 ${\beta}$-galactosidase의 생산균을 분리하였다. 분리균 YB-1105는 형태적 특성, 생화학적 성질 및 16S rRNA 유전자 염기서열에 근거하여 Bacillus licheniformis로 확인되었다. B. licheniformis YB-1105의 배양상등액과 균체파쇄액에서 모두 ${\beta}$-galactosidase 활성이 관찰되었으며 이들은 모두 pH 6.5와 $50^{\circ}C$의 반응조건에서 paranitrophenyl-${\beta}$-D-galactopyranoside의 가수분해 활성이 최대로 나타났다. 그러나 균체파쇄상등액에 비해 배양상등액의 ${\beta}$-galactosidase 활성은 산성 pH와 고온에서 크게 영향을 받았다. 한편 두 분획의 가수분해 활성은 낮은 농도의 galactose에 의해도 급격하게 저해되었으나, glucose와 mannose는 고농도에 의해서는 약하게 저해를 받는 것으로 확인되었다.
가정에서 제조된 된장을 산성조건에서 계대 배양한 후 균체외 mannanase 를 생산하는 Bacillus sp. YB-1401를 분리하였다. 분리균의 당이용능을 비롯한 생화학적 특성은 Brevibacillus laterosporus와 유사도(61.1%)가 가장 높은 반면에 16S rRNA 유전자의 염기서열은 B. amyloliquefaciens와 유사도가 가장 높았다. 분리균 YB-1401의 mannanase 생산성은 mannans에 의해 급격하게 증가하였으며, 특히 곤약(4%)이 첨가된 배지에서 약 265 U/ml로 최대 생산성을 보였다. 분리균의 mannanase는 $55^{\circ}C$와 pH 5.5 반응조건에서 최대활성을 보였으며 pH 3.5-11.0의 범위에서 1시간 방치하였을 때 실활이 거의 일어나지 않았다. 또한 LBG와 guar gum 및 mannooligosaccharides를 mannanase로 분해하였을 때 주된 반응산물로 mannobiose와 mannotriose가 생성되었으며 mannose도 소량 생성되었다.
The covalent interactions of toluenediisocyanate (TDI) with macromolecules were investigated both in vitro and in vivo. In vitro incubations of 2,4- and 2,6-TDI with DNA or proteins resulted in dose-dependent formation of TDI-protein and TDI-DNA adducts. TDI-treated DNA was highly resistant to enzymatic digestion and thermal hydrolysis, but was readily hydrolyzed under acidic conditions by releasing its corresponding toluenediamine (TDA), suggesting that TDI caused the crosslinking of DNA. Reaction of TDI with albumin and globin resulted in the formation of several adducts, and some adducts were formed in blood of TDI-treated rats in a dose-dependent fashion. Administration of TDI to rats resulted also in a dose-dependent binding of TDI to hepatic tissue. Levels of TDI-albumin adducts were 10 times higher than those of TDI-globin adducts; the biological half lives of TDI-albumin and TDI-globin adducts were 1.2 and 12.5 days, respectively. Globin adducts were detected up to 28 days after the treatment. Hepatic TDI protein adducts were persistent for a substantial period whereas the levels of hepatic TDI-DNA adduct were decreased rapidly. These results indicate that the isocyanato group of TDI is not readily hydrolyzed under physiological conditions, is transported to other organs, and is bound to DNA and/or proteins without further metabolic activation. As the adducted products degrade in the body, TDA is released and introduced to the liver. TDA may additionally bind to hepatic tissue after metabolic activation. Thus, the toxic effect of TDI exposure is considered to persist during the lifetime of the adducted biological macromolecules.

CMCase produced by recombinant E. coli JM109 (pCEH#4) containing CMCase gene from Cellulomonas sp. YE-5 was purified to 24.3 fold and 2.6% yield by ammoniumsulfate precipitation, DEAE-cellulose column chromatography and gel filtration on Sephadex G-100. The optimum pH and temperature for CMCase activity were pH 7.0 and 5$0^{\circ}C$. The enzyme was stable between pH 5.0 and 10.0, and up to 6$0^{\circ}C$. The molecular weight of he enzyme was estimated to be approximately 40,000 daltons by SDS-PAGE. Analysis of the amino acid composition showed that the enzyme contained many glycines and acidic amino acids. The enzyme was an endo-type CMCase and the final enzyme reaction product from hydrolysis of Cm-cellulose by the enzyme was cellobiose. {TEX}$K_{M}${/TEX} value determined with CM-cellulose was 1.28mM.

A new ${\alpha}$-amylase-encoding gene (amySL3) of glycoside hydrolase (GH) family 13 was identified in soda lake isolate Alkalibacterium sp. SL3. The deduced AmySL3 shares high identities (82-98%) with putative ${\alpha}$-amylases from the genus Alkalibacterium, but has low identities (<53%) with functionally characterized counterparts. amySL3 was successfully expressed in Escherichia coli, and the recombinant enzyme (rAmySL3) was purified to electrophoretic homogeneity. The optimal temperature and pH of the activity of the purified rAmySL3 were determined to be $45^{\circ}C$ and pH 7.5, respectively. rAmySL3 was found to be extremely halophilic, showing maximal enzyme activity at a nearly saturated concentration of NaCl. Its thermostability was greatly enhanced in the presence of 4 M NaCl, and it was highly stable in 5 M NaCl. Moreover, the enzyme did not require calcium ions for activity, and was strongly resistant to a range of surfactants and hydrophobic organic solvents. The major hydrolysis products of rAmySL3 from soluble starch were maltobiose and maltotriose. The high ratio of acidic amino acids and highly negative electrostatic potential surface might account for the halophilic nature of AmySL3. The extremely halophilic, calcium-independent, and surfactant-resistant properties make AmySL3 a promising candidate enzyme for both basic research and industrial applications.
연료전지 스택 작동환경에서의 열노화 특성을 살펴보기 위하여 다양한 고무 compound의 내열 및 압축 영구 줄음률의 평가를 실시하고, 스택의 장시간 운전을 통해 접합할 수 있는 대상액인 $H_2SO_4$, $H_2O$, LLC (Ethylene glycol : $H_2O=50:50$)에 대하여 장시간 평가를 실시하였다. NBR과 EPDM은 시간이 경과할수록 침적액의 변색하는 것을 알 수 있었으며, VMQ는 $H_2SO_4$ 분위기에서 시간에 따라 고무가 노화되는 것을 TGA, SEM, EDS 분석을 통하여 확인하였다.

Sulfur is one of the inorganic elements used by plants to develop and produce phytoalexin to resist certain diseases. This study reported a method for preparing a material for plant disease resistance. Sulfur nanoparticles (SNPs) stabilized in the chitosan-Cu2+ (CS-Cu2+) complex were synthesized by hydrolysis of Na2S2O3 in an acidic medium. The obtained SNPs/CS-Cu2+ complex consisting of 0.32% S, 4% CS, and 0.7% Cu (w/v), contained SNPs with an average size of ~28 nm as measured by transmission electron microscopy images. The X-ray diffraction pattern of the SNPs/CS-Cu2+ complex showed that SNPs had orthorhombic crystal structures. Interaction between SNPs and the CS-Cu2+ complex was also investigated by ultraviolet-visible. Results in vitro nematicidal effect of materials against Meloidogyne incognita showed that SNPs/CS-Cu2+ complex was more effective in killing second-stage juveniles (J2) nematodes and inhibiting egg hatching than that of CS and CS-Cu2+ complex. The values of LC50 in killing J2 nematodes and EC50 in inhibiting egg hatching of SNPs/CS-Cu2+ complex were 75 and 51 mg/l, respectively. These values were lower than those of CS and the CS-Cu2+ complex. The test results on the nematicidal effect against M. incognita on coffee pots showed that the SNPs/CS-Cu2+ complex was 100% effective at a concentration of 150 mg/l. Therefore, the SNPs/CS-Cu2+ complex could be considered as a biochemical material with potential for agricultural applications to control root-knot nematodes.
도서관이나 문서보관소에서 책이나 서류를 보존하는데 있어 중요한 문제점 중의 하나는 종이의 열화이다. 그러므로 이러한 문제를 해결하기 위해서는 종이의 열화의 주요 원인을 규명할 필요가 있다. 몇몇의 학자들이 이에 대한 연구를 수행한 결과 종이 열화의 주요 원인으로서 종이 섬유내 셀룰로오스의 산 촉매 가수분해로 밝혀졌다. 일반적으로 종이제조시 첨가되는 첨가제로 인해 산성지의 노화율이 중성지의 노화율에 비해 더 높다. 따라서 종이내 존재하는 산을 제거해 줄 필요가 있다. 종이의 탈산화 처리는 열화속도를 감소시켜 산성지의 수명을 3, 4배정도 늘릴 수 있다고 한다. 최근에는 기존의 탈산화 처리방법과는달리 대량의 책과 서류의 탈산화를 위한 효과적인 탈산화 방법의 필요성이 인식되어져 왔다. 따라서 본 논문에서는 종이 제조시 사용되는 첨가제가 노화에 미치는 영향을 알아보고,효과적인 종이의 노화 방지를 위해 산성지에 가스상 에탄올아민류(모노에탄올아민, 디에탄올아민, 트리에탄올아민)를 사용하여 탈산처리를 시도하여 보았다. 첨가제가 종이의 노화에미치는 영향을 실험한 결과, 종이의 노화율이 알럼+로진>알럼>AKD> 무처리 순으로 나타났다. 또한 가스상 에탄올아민류 탈산처리 실험결과, 탈산처리율이 모노에탄올아민>디에탄올아민>트리에탄올아민 순으로 나타났다. 그러나 처리 후 종이에 약간의 백색도와 내절도의 감소가 나타났다. 이러한 문제를 해결하기 위해 가스상 에탄올아민류들을 조합 처리하여 탈산처리를 시도해 본 결과, 백색도와 내절도의 감소없이 효과적인 탈산처리 효과를 얻을 수 있었다.ird limit" is characterized by the production of reduction of $H_2O$$_2$, which is reduced by wall effect.
