Callus Induction and Plant Regeneration from Mature Embryos in Oat

  • Mature embryos of five oat genotypes were cultured to develop an efficient method of callus induction and plant regeneration. Murashige and Skoog(MS) and N6 media supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D) and kinetin were used for callus induction. Percentage of callus induction showed significant among the combinations of plant growth regulators. Callus induction showed high efficiency in medium containing 3 mg/$\ell$ of 2,4-D. The high frequency of callus induction was obtained in Gwiri37. For plant regeneration, calli induced from mature embryos were transferred onto MS and N6 media supplemented with combinations of 6-benzyladenine (BA) and naphthaleneacetic acid (NAA) for 5 weeks. Percentage of plant regeneration showed high in MS medium containing 0.2 mg/$\ell$ of NAA and 1 mg/$\ell$ of BA. The callus initiation medium affected the subsequent plant regeneration. Treatment with 3 mg/$\ell$ of 2,4-D, and 3 mg/$\ell$ of 2,4-D and 3 mg/$\ell$ of kinetin in callus induction media showed high frequency for plant regeneration. Plant regeneration frequency among the genotypes showed significant. Especially, Gwiri37 showed high regeneration frequency. Regenerated shoots were treated with 200, 350 and 500 mg/$\ell$ of indole-3-butyric acid (IBA) transferred onto half-strength MS medium without plant growth regulators. Treatment of shoots with IBA induced root formation rapidly.

Endogenous retinoic acid mediates the early events in salamander limb regeneration

  • Urodeles including newt and salamander have remarkable regenerative capacity during the postembryonic life span. Some of the unique features are the formation of the well-developed wound epidermis and the active dedifferentiation process in the early phase of regeneration. These are regarded as key events for the successful regeneration since no further regenerative activity is possible without them. In this study, we investigated the role of retinoic acid (RA) in salamander limb regeneration by blocking RA synthesis using disulfiram, an inhibitor of aldehyde dehydrogenase that oxidizes retinal to RA. Disulfiram treatment resulted in delaying the limb regeneration processes via inhibition of wound epidermis formation and dedifferentiation process. When RA was administered after disulfiram treatment, the inhibitory effect of disulfiram was rescued. In addition, disulfiram treatment after the dedifferentiation stage resulted in the mild retardation of limb regeneration, suggesting that RA might also be involved in the blastema outgrowth. Furthermore, salamander MMP-9 gene expression was also inhibited by disulfiram treatment. Collectively, our findings indicate that endogenous RA may play an important role(s) in the early phase of limb regeneration by regulating the expression of molecules responsible for the modification of intracellular and extracellular environment during salamander limb regeneration.

Study of continuous regeneration of carbon dioxide sorbent using amino acid salt (아미노산염을 이용한 이산화탄소 흡수제 연속 재생에 관한 연구)

  • In this study, the regeneration efficiency of the amino acid salt absorbent which can be applied to carbon dioxide absorption / regeneration process was confirmed. The regeneration efficiency has a great influence on the economical judgment of the process. so, continuous regeneration experiment was conducted to establish economical process. The amino acid salts used in the experiments are Potassium L-lysinate and Potassium L-alaninate. Each amino acid and potassium hydroxide(KOH) were mixed at a 1: 2 molar ratio. In order to confirm the regeneration efficiency of the absorbent, carbon dioxide was absorbed in the two materials, and the carbon dioxide desorption experiment was carried out by heating. The initial reaction rate was L-alanine was faster. Over time, L-lysine, desorption higher concentrations of carbon dioxide. L-lysine showed higher regeneration efficiency than L-alanine, (L-alanine 47.26% and L-lysine 62.11%). As a result of the continuous regeneration experiment using the L-lysine having good absorption and regeneration efficiency, it was confirmed that the regeneration efficiency decreases as the number of regeneration increases.

Protoplast Formation and Regeneration from Mycelia of Phytophthora capsici (Phytophthora capsici의 균사체(菌絲體)로부터 원형질체(原形質體) 형성(形成)과 재생(再生))

  • ABSTRACT: Factors responsible for protoplast formation and regeneration of Phytophthora capsici were examined. Protoplasts were successfully liberated from the mycelial culture by digestion for 6-9 hrs with Novozym 234 in 0.35 M $CaCl_2$, (pH 5.7) as osmotic stabilizer. Young rapidly-growing mycelium (24 hrs old) showed highest protoplast yields. High concentrations of Novozym 234 were effective in releasing protoplasts from the mycelium. The combination of 0.4 M mannitol and 0.1 M $CaCl_2$ was optimal osmotic stabilizers for protoplast regeneration. The synthetic Henninger media containing all nutritional elements gave the best regeneration rate. The protoplast regeneration was greatly inhibited in the media which were not supplement with amino acids or ${\beta}-sitosterol$. Certain amino acids such as L-aspartic acid and L-glutamic acid remarkably enhanced protoplast regeneration. However, the addition of microelements did not affect protoplast regeneration.

A comparative clinical study on oxidized titanium implants and sandblasted large-grit acid etched implants in soft bone

  • Purpose: The aim of this retrospective study was to compare the survival rate of oxidized titanium implants and sandblasted large-grit acid etched implants in soft bone. Methods: 201 oxidized titanium implants were inserted in 84 patients between May 1999 and May 2004. 120 sandblasted large-grit acid etched implants were inserted in 74 patients between December 2000 and May 2004. The patients were followed-up 0${\sim}$5 years in ITI group or 0${\sim}$6 years in BRA group, respectively. The following information was collected from the patient records: age, gender, systemic disease, implant type, number, length and diameter of the implants, their location in the jaws, bone quantity, the number of failed implants, the causes of failure, and advanced surgery for bone augmentation. Results: In the oxidized titanium implants, 8 implants showed early failure, and 1 implant showed late failure, respectively. The cumulative survival rate was 95.48%. In the sandblasted large-grit acid etched implants, 1 implant showed late failure and cumulative survival rate was 99.10%. The cumulative survival rate and the survival rates in the case of the advanced procedure during the implant placement were not significantly different in both groups. Conclusions: Oxidized titanium implants and sandblasted large-grit acid etched implants can be used successfully in soft bone regardless of the surgical methods used during the implant placement. Electrospun poly(D,L-lactic acid)/gelatin membrane using green solvent for absorbable periodontal tissue regeneration

  • Electrospinning was performed using an eco-friendly solvent composed of acetic acid, ethyl acetate and distilled water to investigate the effect of gelatin concentration on mechanical properties and cytotoxicity of absorbable poly(D,L-lactic acid) (PDLLA)/gelatin blend membrane. The tensile stress, strain at break, and WUC of the PDLLA/gelatin (97/3) scaffold at 26 wt% concentration were determined to be 3.9 ± 0.7 MPa, 37 ± 1.3 %, and 273 ± 33 %, respectively. FT-IR results revealed that PDLLA and gelatin were bound only by van der Waals interactions. The cell viability of PDLLA/gelatin membranes containing 0 %, 1 %, 2 %, 3 %, and 4 % gelatin were more than 100 %, which makes all membranes highly suitable as a barrier membrane for absorbable periodontal tissue regeneration due to their marketed physical properties and biocompatibility.

Lysosomal acid phosphatase mediates dedifferentiation in the regenerating salamander limb

  • In this study, monoclonal antibodies against lysosomal acid phosphatase (LAP) of a salamander, Hynobius leechii, were used to determine the spatial and temporal expression of the LAP in the regenerating limbs. The Western blot and immunohistochemical analysis in the limb regeneration revealed that LAP was highly expressed at the dedifferentiation stage, especially in the wound epidermis and dedifferentiating limb tissues such as muscle and cartilage. With RA treatment, the LAP expression became upregulated in terms of both level and duration in the wound epidermis, blastemal cell and dedifferentiating limb tissues. In addition, in situ activity staining of LAP showed a similar result to that of immunohistochemistry. Thus, the activity profile of LAP activity coincides well with the expression profile of LAP during the dedifferentiation period. Furthermore, to examine the effects of lysosomal enzymes including LAP on salamander limb regeneration, lysosome extract was microinjected into limb regenerates. Interestingly, when the lysosome extract was microinjected into limb regenerates with a low dose of RA($50\;{\mu}g/g$ body wt.), skeletal pattern duplication occurred frequently in the proximodistal and transverse axes. Therefore, lysosomal enzymes might cause the regenerative environment and RA plays dual roles in the modification of positional value as well as evocation of extensive dedifferentiation for pattern duplication. In conclusion, these results support the hypothesis that dedifferentiation is a crucial event in the process of limb regeneration and RA-evoked pattern duplication, and lysosomal enzymes may play important role(s) in this process.

In vitro regeneration from cotyledon explants in figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), a rootstock for Cucurbitaceae

  • An efficient plant regeneration system has been developed for figleaf gourd (Cucurbita ficifolia Bouch$\'{e}$), which is exclusively used as a rootstock for cucumber. The protocol is based on results obtained from a series of culture experiments involving different parts of the cotyledons and various media. The culture of cotyledon explants was critical for the enhancement of shoot regeneration frequency. The lower parts of the cotyledon excised at the plumule base were found to display a markedly enhanced production of adventitious shoots compared to other cotyledon regions. Culture in silver nitrate-supplemented Murashige and Skoog (MS) medium was not beneficial for shoot regeneration and suppressed root regeneration. Efficient shoot regeneration was obtained on MS medium containing 1.0 $mg\;l^{-1}$ zeatin and 0.1 $mg\;l^{-1}$ indole-3-acetic acid. Regenerated shoots successfully elongated and rooted in medium containing 0.1 $mg\;l^{-1}$ 1-naphthalene-acetic acid after 10-15 days of subculturing. The plantlets were satisfactorily acclimatized in a greenhouse and grew into normal plants without any morphological alterations.

Improvement of Regeneration Efficiency from Mature Embryo and Leaf Base Segment in Korean Oat Genotypes

  • Mature embryo and leaf base segment of Korean oat were used as materials in an experiment to check plant regeneration efficiency. MS media supplemented with 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin, and picloram were used for callus induction from mature embryos and leaf base segments. Three mg/l of 2,4­D and 3 mg/l of picloram in callus induction medium showed high frequency for plant regeneration from mature embryos. Leaf base segments were transferred to callus induction medium and incubated at $25^{\circ}C$ in 16/8 hr light/dark cycle for 3 weeks. Callus induction from leaf base segments of Malgwiri showed high efficiency in medium containing 3 mg/l of 2,4-D and 1 mg/l of kinetin $(91.8\%)$. In case of Samhangwiri, the combinations of phytohormones did not show significant difference. Regeneration from leaf base segments showed high frequency in shoot medium containing 1 mg/l of antiauxin, tri-iodobenzoic acid (TIBA) and 1 mg/l of 6-benzyladenine (BA). Calli induced from leaf base segments of Samhangwiri and Malgwiri in media containing 3 mg/l of 2,4-D and 3 mg/l of picloram showed high regeneration frequency. It appears that the callus initiation medium may be an important factor for subsequent plant regeneration.

Clinical study of guided bone regeneration of extracted socket with PLA/PGA membrane and silk fibroin membrane (PLA/PGA 차폐막과 실크 피브로인 차폐막을 이용한 발치와의 골유도재생술의 비교연구)

  • Purpose: This study was designed to compare the bond regeneratiom effects of treatment using silk fibroin membrane ( Nanogide-S$^{(R)}$ ) resorbable barrier with control group treated by polyactic acid / polylacticglycolic acid membrane(Biomesh$^{(R)}$ ) Methods: 44 severe bone loss on extraction socket from 44 patients were used in this study. In experimental group 22 sites of them were treated by silk fibrin membrane as and the other 22 sites were treated by polyactic acid/ polylacticglycolic acid membrane as a control group. Clinical parameters including recovered bone width, length and radiographic parameter of vertical length were evlauated at base line and 3 months after surgery. Results: 1) Severe bone width, length was significantlly decreased in two group. 2) Bone width, length was significantlly decreased in two group. 3) Decreased bone width, length and radiographic examination differences between group. Conclusions: On the basis of these results, silk fibrin resorbable membrane has similar bone regeneration ability to polyactic acid / polylacticglycolic acid membrane in guided bone regeneration for severe bone loss defect on extraction socket.
