In September 2017, the rhizospheric soil of Tetragonia tetragonoides (Pall.) Kuntze was further sampled. One hundred and thirty eight species of microorganisms were isolated from the soil. Indole-3-acetic acid (IAA) production, siderophore production, and phosphate degradation were examined in order to confirm bacterial growth from isolated microorganisms. As a result, most strains were able to produce auxins or siderophores and to solubilize phosphate. In addition, 138 isolated strains were treated with tobacco extract and conferred pathogen resistance to host plants upon treatment. As a result, 35 strains that were able to reduce pathophysiology by more the 60% were selected. Among them, 6 strains with high induced systemic resistance (ISR) activity were found. All of these strains belong to the genus Bacillus according to the 16S rDNA sequence analysis. Bacillus aryabhattai KUDC6619 showed outstanding effects with reduced infection in tobacco and pepper plants. Probably, these Bacillus species play a beneficial role by association with T. tetragonoides for its survival in the harsh conditions found on the island of Dokdo.
Analytical methods for the determination of the derivatives of the herbicide (${\pm}$)-2-(4-chloro-2-methylphenoxy)propionic acid (MCPP) by capillary column gas chromatography with mass spectrometer (GC-MS) and electron-capture detection (GC-ECD) were studied. A successful procedure was introduced for the ester preparation using $H_2SO_4$, as the catalyst and the alcohol 2,2,2-trichloroethanol (TCE) or 2,2,2-trifluoroethanol (TFE). The identificaiton and elucidation of MCPP by GC-MS spectrometry following the esterification with diazomethane, $BF_3$/methanol, $H_2SO_4$/methanol, TCE, TFE, or pentafluorobenzyl bromide (PFB) were carried out. A comparison of the response-sensitivities among those MCPP esters was made with GC-ECD. Although the methylation product of MCPP was confirmed by GC-MS, its low sensitivity to the ECD limited the detection of MCPP. TCE, TFE, and PFB derivatization methods resulted in a high rate of MCPP esterifications and very sensitive ECD molecular responses. Based on efficiency, convenience, worker safety, and least sample contamination, TFE esterificaiton was considered as the superior method for MCPP analysis to the other methods of derivatization. An accurate method is described for quantifying MCPP in soil leachates by GC-ECD at very low concentrations without the requirement of a complicated clean-up process. As a result, MCPP residues at concentrations of less than $0.1{\mu}g$ in 100ml soil leachate were detected.

Due to rapid consumption of hydrogen peroxide, large amount of hydrogen peroxide is required when Fenton reaction is applied to the contaminated soil. In this study, acetate was employed as a ligand of $Fe^{2+}$ to enhance the efficiency of removal of phenanthrene by securing the stability of hydrogen peroxide. 0.5 ${\sim}$ 3 times of acetate (2${\sim}$12mM) was added to compare with molar concentration of $Fe^{2+}$. Low initial concentration of hydrogen peroxide was 0.7% to eliminate side effect of removal efficiency. The results showed that hydrogen peroxide lifetime was lasted up to 72 hours, or more than 50 times of normal lifetime. Phenanthrene removal efficiency was improved up to 70% due to stabilized hydrogen peroxide. Ferrous ion was oxidized to ferric ion and oxidation-reduction was repeated during the reaction. Finally ferric ion was reduced to ferrous by $HO_2$. It was confirmed that, due to the influence of hydrogen peroxide, pH was acid region and it remained at the range of 4 ${\sim}$ 5 when 8 mM or more of acetate was added. Acetate which was used as the ligand of Fe was also decomposed by Fenton reaction. The decomposition time of acetate was slower than phenanthrene. Therefore, it was able to come to the conclusion that phenanthrene was superior to acetate at the competition for decomposition. Through the results of this study, it was able to identify the possibilities to improve the efficiency of Fenton reaction in the contaminated soil and its economic feasibility, and to move to more realistic technique through research expanded to neutral pH region.
The purposes of this study were to investigate vegetation structure and to present management plan of mountain type green space in Kangwhado, Inchon. The actual vegetation in survey sites(11,331ha) was divided into 19 community types. It was consisted of secondary forest(92.32%) which was Quercus acutissima, Pinus densiflora-Q. acutissima, and Q. mongolica community so on. Artificial planting forest area, such as Robinia pseudoacacia and Pinus rigida forest and others, was 5.40%(612ha) and it was less than cases in other cities in the Metropolitan area. According to the classification by TWINSPAN, 57 survey plots were divided into seven community types; P rigida(community A), Q. acutissima(community B) P. densiflora-Q. acutissima(community C), Q. acutissima-P. densiflora (community D), P. densiflora-Carpinus laxiflora-Q. serrata-Q. acutissima(community E), Q. serrata-Q. mongolica(community F), and Zelkova serrata-Acer mono(community G). From this result, ecological succession trend of vegetation in this area seems to be change from P. densiflora forest through Q. acutissima forest to Q. mongolica, Q. serrata, and C. laxiflora forest. It was similar to the ordinary successional trend of temperate deciduous forest in middle area, Korea. In addition, this study area was showed acid soil(pH 4.17). Therefore, there is a need for managing the soil environment for effective vegetation management.
Biomass is originally photosynthesized from inorgainic compounds such as $CO_2$, minerals, water and solar energy. Recent studies have shown that anaerobic bacteria have the ability to convert recalcitrant biomass such as cellullosic or chitinoic materials to useful compounds. The biomass containing agricultural waste, unutilized wood and other garbage is expected to utilize as feed, food and fuel by microbial degradation and other metabolic functions. In this study we isolated several anaerobic, cellulolytic and chitinolytic bacteria from rumen fluid, compost and soil to study their related enzymes and genes. The anaerobic and cellulolytic bacteria, Clostridium thermocellum, Clostridium stercorarium, and Clostridium josui, were isolated from compost and the chitinolytic Clostridium paraputrificum from beach soil and Ruminococcus albus was isolated from cow rumen. After isolation, novel cellulase and xylanase genes from these anaerobes were cloned and expressed in Escherichia coli. The properties of the cloned enzymes showed that some of them were the components of the enzyme (cellulase) complex, i.e., cellulosome, which is known to form complexes by binding cohesin domains on the cellulase integrating protein (Cip: or core protein) and dockerin domains on the enzymes. Several dockerin and cohesin polypeptides were independently produced by E. coli and their binding properties were specified with BIAcore by measuring surface plasmon resonance. Three pairs of cohesin-dockerin with differing binding specificities were selected. Two of their genes encoding their respective cohesin polypeptides were combined to one gene and expressed in E. coli as a chimeric core protein, on which two dockerin-dehydrogenase chimeras, the dockerin-formaldehyde dehydrogenase and the dockerin-NADH dehydrogenase are planning to bind for catalyzing $CO_2$ reduction to formic acid by feeding NADH. This reaction may represent a novel strategy for the reduction of the green house gases. Enzymes from the anaerobes were also expressed in tobacco and rice plants. The activity of a xylanase from C. stercorarium was detected in leaves, stems, and rice grain under the control of CaMV35S promoter. The digestibility of transgenic rice leaves in goat rumen was slightly accelerated. C. paraputrificum was found to solubilize shrimp shells and chitin to generate hydrogen gas. Hydrogen productivity (1.7 mol $H_2/mol$ glucos) of the organism was improved up to 1.8 times by additional expression of the own hydrogenase gene in C. paraputrficum using a modified vector of Clostridiu, perfringens. The hydrygen producing microflora from soil, garbage and dried pelletted garbage, known as refuse derived fuel(RDF), were also found to be effective in converting biomass waste to hydrogen gas.

In Korea, over three hundreds of the coal mines were closed or abandoned due to the depression of the mining industry since the late 1980s. Many of them locate in the steep mountain valleys and the coal mine wastes had been disposed without a proper treatment From these mines, enormous amounts of coal mine overburdens have been abandoned in the slopes and the ample amounts of acid mine drainage (AMD) from either portal or overburdens have been discharging directly to the streams, causing the detrimental effects on soil and water qualities. Objectives of this research were to reclaim the coal mine overburdens using the lime waste cake from the soda ash production by stabilizing the overburden slopes, introducing the vegetation alleviate the environmental problems caused by the closed coal mines. The percentages of the grass distribution ratio (%) and the surface coverage ($\textrm{cm}^2$) in each treatment plot were determined during June to August after seed spraying grasses such as orchard grass (Dactylis glomerata L), Kentucky Bluegrass (Poa pratensis L.) and Eulalia (Miscanthus sinensis Anderss) at the end of May. The grasses covered only 15.5 % of the coal overburden plot at the early stage but the coverage was increased with time to 33% in August. Growth of such grasses was enhanced with the combined treatments of lime waste and topsoil resulting in the increased surface coverage by the grasses. The Increment of the surface coverage from June to August was higher with lime waste treatments. The distribution percentages and surface coverage were highest when the lime wastes were treated at 25 % of the lime requirement. This might be related with the high salt contents in the hire wastes. Results demonstrated that the amounts of lime wastes at 25% of the lime requirement were sufficient for neutralizing the acidic coal overburden and introducing the re-vegetation. Either layering between the coal waste and topsoil or mixing with coal overburdens could be adopted as the lime waste treatment method. The combined treatment of lime wastes and topsoil was recommended for re-vegetation in the coal overburden slopes. The lime wastes from the soda ash production might have a potential to be recycled for the reclamation of the abandoned coal mines to alleviate the environmental problems associated with coal mine waste.
The acid mine drainage (AMD) and landfill leachates released into the subsurface environment can result in serious environmental problems like soil and groundwater contamination. The AMD and the leachates of landfill were known to contain many heavy metals. In this study, the author assessed the reactivity and ability of the FeS coated-ALC for the removal of contaminants (As, Cd, Cu, Pb, Ni, Zn, Al) in AMD and leachates in landfill. The synthetic nano-FeS and Autoclaved Lightweight Concrete (ALC) were used as reactive materials in the permeable reactive barriers(PRBs). The result of batch test indicated that synthetic nano-FeS can remove 99% of heavy metals for the 1hr of reaction time except for As and Ni(about 90%). However, the 80% of As and Ni was removed in column 1(FeS coated-ALC). The column 2(Ore FeS) removed more than 99% of heavy metals. The pH of the column 1 was increased from 3.51 to 6.39~6.50, and the pH with column 2 was increased from 3.51 to 9.20. As the result of this study, the author can surmise that the synthetic nano-FeS coated ALC will use as a very good reactive material of the PRBs to treat the contaminated groundwater with AMD and leachate of landfill.
This study was carried out to investigate the differences in germination and growth of Quamoclit coccinea Moench under various temperatures, seeding depths, and levels of shading and soil moisture for effective weed management. Seed dormancy of Q. coccinea Moench was over 1 year; best results were obtained when seeds were soaked in sulfuric acid for 15 minutes in order to break the dormancy. Germination rates of Q. coccinea Moench ranged from 69 to 73% at $25-35^{\circ}C$ and 26% at $15^{\circ}C$. The germination rates ranged from 70-84% at 2, 3, 5, 7, and 8 cm of seeding depths. In addition, the germination rates were 7% and 13% at 12 cm and 15 cm of seeding depths, respectively, and showed normal growth at the both seeding depths. Q. coccinea Moench showed a high germination rate regardless of shading levels, but shoot fresh weight varied depending on the level of shading as follows: 20%>no shading=shading 35%>shading 50%>shading 75%=shading 90%. Q. coccinea Moench did not germinate when soil had a saturation rate of either 30% or 100%. However, 60-83% of seeds germinated with optimal growth when soil had saturation rates of 60% and 80%.

We examined the nutritive value of common grass species in the semi-arid rangelands of Borana in southern Ethiopia using local experience based herbage preference (LEBHP) perception and laboratory techniques. Local pastoralists in the study area were asked to identify common grass species and rank them according to the species' preferences and palatability to cattle. The pastoralists listed a total of 15 common grass species which were then sampled during the main rain and cold dry seasons and analyzed for crude protein (CP), Neutral Detergent Fiber (NDF), Acid Detergent Fiber (ADF) and ash content to verify pastoralists' claim regarding the quality of individual species. The relative feed value (RFV) and dry matter digestibility (DMD) were also calculated using NDF and ADF contents. Spearman's rank correlation was used to examine possible relationships between laboratory results and pastoralists' experience on grass quality. Cenchrus ciliaris, Chrysopogon aucheri, Digitaria milanjiana, Eragrostis papposa and Panicum maximum were the top five species based on LEBHP perception. There were indications of inconsistency in terms of LEBHP perception among the different pastoral communities. The chemical composition of all grass species showed significant (p<0.05) variation between sites, seasons and species. The results showed that the CP values for the Borana rangelands were in the range of 8.7% in the main rain season to 5.1% for the cold dry season. The fiber constituents were relatively low in the main rain season compared to the cold dry season. Overall, Digitaria milanjiana had the highest CP (16.5%) content, while the least was recorded with Heteropogon contortus (10.8) and Aristida adoensis (9.8%) during the main rain season. It seems that the spatial variability of landscapes within the wider geographical regions, soil properties and texture, and land-use patterns probably contributed to site differences in species quality. Generally, the RFV of individual grass species was significantly (p<0.05) varied between and within sites. The ranking of species by pastoralists according to their preferences by cattle was highly correlated with the chemical composition of laboratory results of individual grass species with 'r' values for CP (0.94), ash (0.95), NDF (-0.98), ADF (-0.93) and ADL (-0.93). We suggest the complimentary use of LEBHP and laboratory techniques in evaluating the nutritive quality of rangeland forage species for sustainable animal production.
Two hundred and sixty-eight bacterial strains were isolated from pine mushroom (Tricholoma matsutake) basal soil. In the course of screening for antifungal activity against seven plant pathogenic fungi (Alternaria panax, Botrytis cinerea, Colletotrichum gloeosprioides, Fusarium oxysporum, Phytopthora capsici, Pythium ultimum, Rizoctonia solani) of isolates, strain KM1-15 showed strong antibiotic activity against Alternaria panax and Colletotrichum gloeosprioides. In determining its relationship on the basis of 16S rDNA sequence, KM1-15 strain was most closely related to Bacillus $koguryoae^T$ (AY904033) (99.62%). When assayed with the API 50CHE Kit, unlike Bacillus koguryoae, it is positive for utilization of L-arabinose, cellobiose, inulin, and D-turanose. Results of cellular fatty acid analysis showed that major cellular fatty acids were 15:0 anteiso (35.78%) and 17:0 anteiso (17.97%). In particular, hydroxyl fatty acids such as 13:0 iso 3-OH, 14:0 iso 3-OH, 15:0 iso 3-OH, and 17:0 iso 3-OH were only restricted to strain KM1-15. DNA G+C content was 43.7 mol% and quinone system was MK-7 (100%) in strain KM1-15.
