도 건전엽과 도열병에 있어서의 Free Amino Acid 함량 비교연구 (Comparative studies on free amino acid content between healthy leaves and blasted leaves of rice plants.)

  • This experiment was undertaken the purpose of investigating the difference of the content of free amino acids in the diseased and healthy leaves of the resistance and susceptible rice varieties, to blast by means of paper chromatography. The results obtained are summarized as follows. (1) In the healthy and diseased leaves, the former was founded nine to tell sorts of free amino acids and later was observed eleven. In the both of leaves, alanine and valine were contained in larger amount, and aspartic acid, glycine and arginine were found relatively less in general. (2) In the diseased leaves, the amino acids were increased in quantity, especially glutamine and aspartic acid were remarkable, and cystine were found only in the diseased ones. (3) It seems to that the health leaves in all varieties were observed the amino acid contents, in notably increase and decrease, but in the diseased ones were not changed in amount. Tatacho, susceptible variety was more contained the amount of glutamine and aspartic acid than Zenith, resistance one.

용해조건 및 합금원소 첨가에 따른 고규소 내산주철의 기계적 성질 및 부식속도의 변화 (Variations of Mechanical Properties and Corrosion Rate with Melting Conditions and Alloying Elements in High Silicon Cast Irons)

  • High silicon cast irons(HSCI) with the high acid resistance have been used for the prevention of acid corrosion occurring in various structures under acid conditions. However, the HSCI is only known as one of materials which have high acid resistance, but few work has dealt with this material in domestic. Therefore, in this study, the acid resistance of various cast irons with alloying elements and melting conditions have been examined, and studied the influences of the matrix structures, mechanical properties and morphologies of graphite. The results obtained in this study are as follows : In case of melting temperature, the mechanical properties of specimen manufactured with high temperature of $1650^{\circ}C$ showed higher value because the inclusion and impurity were removed. In case of pouring temperature, the mechanical properties of specimen fabricated below $1350^{\circ}C$ of pouring temperature showed higher value because the amount of gas absorption from atmosphere decreased during the solidification time. The corrosion rate decreased with increase in Si content. On the other hand, Mn addition appeared an opposite trend with Si.

임상검체와 가축으로부터 분리된 대장균을 대상으로 Quinolone계 항균제 내성인자 분석 (Analysis of Quinolone Resistance Determinants in Escherichia coli Isolated from Clinical Specimens and Livestock Feces)

  • 퀴놀론 항균제가 사람과 동물에게 부적절하고 광범위하게 사용될 경우 항균제내성인자의 출현 및 확산이 가속화 될 수 있다. 본 연구에서는 돼지의 직장면봉 검체(N=40) 및 임상 검체로(N=25)부터 분리된 총 65균주의 nalidixic acid 내성 대장균을 대상으로 quinolone 내성 기전을 조사하였다. 항균제 감수성은 디스크 확산법에 의해 결정되었다. Quinolone 내성과 관련된 유전자와 돌연변이를 조사하기 위해 PCR 및 DNA sequencing이 수행되었다. 총 65균주의 nalidixic acid 내성 대장균 중 62균주가 gyrA, parC, parE 유전자에 돌연변이를 포함하고 있었는데, gyrA 유전자에 돌연변이를 포함하고 있는 균주는 62균주(95.4%)였고, 35균주(53.8%)가 parC 유전자에 돌연변이를 갖고 있었으며, 7균주(10.8%)가 parE 유전자에 돌연변이를 포함하고 있었다. 35균주는 gyrA 와 parC 유전자에 모두 돌연변이를 가지고 있는 것으로 나타났다. 총 65균주의 대장균을 대상으로 plasmid-mediated quinolone resistance (PMQR) determinants를 조사하였다. 65균주의 nalidixic acid 내성 대장균 중 13균주에서 qnrS 유전자가 검출되었으나 이 중 10균주는 gyrA, parC, parE 유전자에 돌연변이를 포함하고 있는 것을 나타났다. 본 연구에서는 사람과 돼지로부터 분리된 대장균이 quinolone 계열 항균제에 내성을 나타내는데 중요한 역할을 하는 기전이 gyrA, parC, parE 유전자에 염색체 돌연변이가 발생하는 경우임을 확인하였는데 이 돌연변이들은 치료목적 또는 동물의 성장촉진을 위한 항균제의 과다사용으로 유발될 수 있다.

Different Mechanisms of Induced Systemic Resistance and Systemic Acquired Resistance Against Colletotrichum orbiculare on the Leaves of Cucumber Plants

  • Defense mechanisms against anthracnose disease caused by Colletotrichum orbiculare on the leaf surface of cucumber plants after pre-treatment with plant growth promoting rhizobacteria(PGPR), amino salicylic acid(ASA) or C. orbiculare were compared using a fluorescence microscope. Induced systemic resistance was mediated by the pre-inoculation in the root system with PGPR strain Bacillus amylolquefaciens EXTN-1 that showed direct antifungal activity to C. gloeosporioides and C. orbiculare. Also, systemic acquired resistance was triggered by the pre-treatments on the bottom leaves with amino salicylic acid or conidial suspension of C. orbiculare. The protection values on the leaves expressing SAR were higher compared to those expressing ISR. After pre-inoculation with PGPR strains no change of the plants was found in phenotype, while necrosis or hypersensitive reaction(HR) was observed on the leaves of plants pre-treated with ASA or the pathogen. After challenge inoculation, inhibition of fungal growth was observed on the leaves expressing both ISR and SAR. HR was frequently observed at the penetration sites of both resistance-expressing leaves. Appressorium formation was dramatically reduced on the leaves of plants pre-treated with ASA, whereas EXTN-1 did not suppress the appressorium formation. ASA also more strongly inhibited the conidial germination than EXTN-1. Conversely, EXTN-1 significantly increased the frequency of callose formation at the penetration sites, but ASA did not. The defense mechanisms induced by C. orbiculare were similar to those by ASA. Based on these results it is suggested that resistance mechanisms on the leaf surface was different between on the cucumber leaves expressing ISR and SAR, resulting in the different protection values.

알루미늄의 아노다이징과 나노 다이아몬드 분말 봉공처리에 의한 내식성과 내마모성 향상에 관한 연구 (Study on Improvement of Corrosion Resistance and Wear Resistance by Anodizing and Sealing Treatment with Nano-diamond Powder on aluminum)

  • In this study, in order to improve corrosion resistance and wear resistance of aluminum, surface treatment was made by anodizing with oxalic acid solution and sealing with nano-diamond powder. Average size of nano-diamond powder was 30nm. Anodizing with oxalic acid made many pores in the aluminum oxide layer. Pore size and oxide thickness were investigated by scanning electron microscope (SEM). Pore size increased as temperature increased and voltage increased. It was possible to make oxide layer with pore diameter more than 50 nm. Oxide thickness increased as temperature and voltage and treatment time increased. Oxide layer with above $10{\mu}m$ thickness was made. Aluminum oxide layer with many pores was sealed by water with nano-diamond powder. Surface morphology was investigated by SEM. After sealing treatment with nano-diamond powder, corrosion resistance, wear resistance and hardness increased.

Cronobacter sakazakii의 산 저항성 (Acid Resistance of Cronobacter sakazakii)

  • 본 연구는 표준균주 C. sakazakii(CAFM2)와 4개의 야생균주(EB 1, EB 5, EB 7, EB 41)를 사용하여 pH를 조정한 산성 환경에서의 저항성을 살펴보았다. C. sakazakii는 pH 4.5 TSB에서 5시간, 10시간 동안 적응한 경우, pH 4.5 TSB에서 적응하지 않은 경우와 비교하여 EB 7을 제외한 모든 균주가 pH 2.5 TSB에서의 D값이 유의적으로 높게 나타났다(p<0.05). 따라서 약산성 환경에서 적응한 균주는 강산성 환경에서의 저항성이 증가하였다. C. sakazakii는 pH 4.5 TSB에서 배양한 경우, pH 7.2 TSB에서 배양한 경우와 비교하여 모든 균주가 pH 2.5 TSB에서의 D값이 유의적으로 높게 나타났다(p<0.05). 따라서 약산성 환경에서 배양한 균주는 강산성 환경에서의 저항성이 증가하였다. 본 연구를 통해 C. sakazakii는 약산성 환경에 적응한 경우 강산성 환경에서의 저항성이 증가하는 것으로 나타났다. 이러한 acid adaptation response는 C. sakazakii와 관련된 식품 가공 산업에서의 hurdle technology를 적용하는데 있어서 안전성 확보와 관련된 시사점을 제공할 수 있을 것으로 판단된다.

  • 본 연구에서는 산 및 황산염 저항성에 미치는 광물질 혼화재의 영향을 평가하기 위한 것이다. 이를 위하여 물-결합재비 32% 및 43%에 대해서 광물질 혼화재 종류에 따라 이들을 혼입한 콘크리트 공시체를 제작하였다. 재령 182일 동안의 침지실험 결과로부터, 콘크리트의 산 및 황산염에 대한 저항성은 광물질 혼화재를 혼입한 콘크리트가 플레인 콘크리트보다 훨씬 우수한 것으로 나타났다.

솔잎재 콘크리트의 응력-변형과 내산성에 관한 실험적 연구 (Experimental Study on the Stress-Strain and Acid-Resistance of Pine Needle Ash Concrete)

  • This study examines the stress-strain and acid-resistance of pine needle ash (PNA) concrete. Materials used for this experiment are PNA , normal portland cement, natural fine and coarse aggregate. Test results show that the highest static modulus of elasticity are achieved by 5 % PNA filled PNA concrete. Acid-resistance of PNA concrete is increased with increase of the content of PNA. It is 1.29 times of the normal cement concrete for 5 % PNA filled PNA concrete and 2.57 times fo r15% PNA filled PNA concrete, based on the elased days for 25% mass loss of original mass immersed in the 5% H2SO4 solution. Accordingly, PNA concrete will greatly improve the properties of concrete.

산성비 분위기에서 교량용 강재 SWS400의 용접부 부식에 관한 연구 (A study of corrosion of welded bridge steel SWS400 in the acid-rain environment)

  • Corrosion behavior of welded SWS400 steel used for bridges was studied in a range of the acid-rain environment using immersion, potentiodynamic polartization, polarization resistance, and galvanic corrosion tests. The SWS400 steel exhibited active corrosion behavior in the range of acid-rain environment, i.e. no passivation. As the results of immersion corrosion test, Tafel extrapolation method, and polarization resistance measurement, the average corrosion rats of the steels were 0.31-0.72 mm/year in the pH of 4-5, and 0.17 mm/yera in the pH 6, respectively. The steel showed a resistance to corrosion in the pH 6. The observed active behavior of SWS400 steel in chloride-containing environment indicated that the chloride ions exerts a detrimental influence on the formation of passive films. Galvanic corrosion was observed between the weld and the base metals because the weld is anodic to the base metal.

  • This study is performed to evaluate an elastic properties and acid-resistance of concrete using pine needle ash(PNA). Materials used for this experiment are PNA , normal portland cement, natural fine and coarse aggregate. Test results show that the highest ultrasonic pulse velocity , dynamic and static modulus of elasticity is achieved by 5% PNA filled PNA concrete, which has showed similar with those of thei normal cement concrete. Acid-resistance of PNA concrete is increased with increase of the contnet of PNA , it is 1.29 times of the normal cement concrete by 5% PNA fille PNA concrete an d2.57 times by 15% PNA filled PNA concrete . Accordingly , PNA concrete wil greatly improve the properties of concrete.
