Inhibition Effect of Achyranthes japonica N. Root Extract on Cathepsin B (우슬뿌리 추출물의 Cathepsin B에 대한 저해효과)

  • This study was carried out to investigate the cathepsin B inhibition effect by Achyranthes japonica N. root extract in vitro. The methanol/$H_{2}O$(4:1, v/v) extract was fractionated by ethyl acetate(F1), chloroform(F2), chloroform/methanol(3:1, v/v)(F3) and methanol(F4). The yield of F4 in Achyranthes japonica N. root was $8.27\%$. As an index material of Achyranthes japonica N. root, 20-hydroxy ecdysone was detected by TLC, and HPLC and it's content was $0.33\%$. Three isolates(F1, F3, F4) showed the cathepsin B inhibition activity, and F4 showed the highest inhibition activity among them. In the inhibition activity on cathepsin B, leupeptin, 20-hydroxy ecdysone and F4(at the same concentration of 20-hydroxy ecdysone.) were 92, 88 and $97\%$ on BANA($N{\alpha}$-benzoyl-DL-arginine ${\beta}$-naphthylamide) substrate, and 62, 36 and $67\%$ on CLN($N{\alpha}$-CBZ(carbobenzlyoxy)-L-lysine p-nitrophenyl ester HCI) substrate, respectively.

Influence of Nitrogen Application Rate on Growth and Dry Matter Yield of Achyranthes japonica Nakai (질소시비량이 쇠무릎의 생육 및 건물수량에 미치는 영향)

  • To determine the optimum N rate m Jeju island for utricle and root production of Achyranthes japonica Nakai, a medicinal plant, the plants were grown at two plant densities $(50\;and\;100\;hills/m^2$, two plants per hill; mall plots) at six N application rates (0, 6, 12, 18, 24, and 30 kg/10a; split plots) in 2001. There was no significant interaction between plant density and N application rate for all measured agronomic characters. Main root length and roots per hill were 5 and 52% greater, respectively, but N content of stover was lower under lower plant density compared to higher plant density. The other characters were not affected by plant density. N application rate did not significantly affect mall stem diameter, spikes per hill, spike length, utricles per spike, mall root length and diameter, and utricle N content. As N rate increased from 0 to 30 kg/10a, SPAD values and stover N content increased linearly from 35.0 to 40.5 and 1.09 to 1.38%, respectively, and plant height, branches per hill, stover, utricle and root dry matter yields, roots per hill, and top N yield increased quadratically. Spikes per hill were increased in a cubic manner with increased N application rates. N application rate for the maximum dry matter yield of utricle and root in A. japonica was estimated to be 21 kg N/10a.

Effect of Organic Fertilizers on Growth and Yield of Achyranthes japonica N. (유기질(有機質) 비종(肥種)이 쇠무릎 생육(生育) 및 수량(收量)에 미치는 영향(影響))

  • This experiment was carried out to investigate the effect of several organic fertilizers on the growth and root yield of Achyranthes japonica N. from 1995 to 1996. Four organic fertilizers were applied: rice straw manure (RSM), fermented rice straw manure (FRSM), mixed oil cake manure (MOCM) and mighty soil manure (MSM). Organic matter, available phosphate, $K_2O$, CaO, and MgO of soil increased in all plots with the addition of organic fertilizers, specifically with MOCM application. The highest contents of total nitrogen were 6.16% for MOCM and the available phosphate contents were very high in all of organic fertilizers except MSM. RSM and MSM with C/N ratios of 22.5, 17.7, respectively, were easily decomposed but C/N ratios of FRSM and MOCM, which were considered as irresolvable organic fertilizers, were 40.9 and 8.4, respectively. FRSM and MOCM applications increased emergence rate and improved the growth characters of shoot and root parts of plants compared to those of N - P - K fertilization. The highest dry root yield resulted from FRSM 20% and MOCM 26% treatment. There were significantly positive correlations between the growth characters of shoot, root parts of plants and dry root yield in A. japonica grown under organic fertilizer applications.

Effect of Phosphorous Fertilizer Rate on Growth, Dry Matter Yield, and Phosphate Recovery in Achyranthes japonica (인산시비량이 쇠무릎의 생육, 건물수량 및 인산회수율에 미치는 영향)

  • To determine the optimum $\textrm{P}_2\textrm{O}_5$ rate for dry matter production of Achyranthes japonica Nakai a medicinal plant, in Jeju island, plants were grown in 2001 under ten fertilizer treatments comprising of two N (90, and 180kg/ha) and five $\textrm{P}_2\textrm{O}_5$ (0, 100, 200, 300, and 400 kg/ha) levels. Plant height leaf length and width, stem diameter, and main root length and diameter were not significantly affected by $\textrm{P}_2\textrm{O}_5$ rate while $\textrm{P}_2\textrm{O}_5$ fertilization of 100 kg/ha increased the number of branches, spikes, and roots per hill by 18,38 and 43%, respectively, compared with the unfertilized $\textrm{P}_2\textrm{O}_5$ treatment and decreased with further increases in $\textrm{P}_2\textrm{O}_5$ rate. The number of roots per hill was greatest with the application of 200kg $\textrm{P}_2\textrm{O}_5$. The highest utricle dry matter yield (2.92 t/ha) was obtained with the application of 100kg $\textrm{P}_2\textrm{O}_5$. However, as $\textrm{P}_2\textrm{O}_5$ rate increased from 0 to 100 and 200kg/ha, root dry matter yield increased from 2.36 to 3.55, and 3.80 t/ha, and then decreased to 3.14, and 2.86 t/ha at 300, and 400kg $\textrm{P}_2\textrm{O}_5$ rates, respectively. As $\textrm{P}_2\textrm{O}_5$ rate increased from 100 to 400 kg/ha, $\textrm{P}_2\textrm{O}_5$ recovery in aboveground parts decreased from 34.1 to 7.1%.

  • This experiment was conducted to investigate the effects of planting density on the growth characteristics and root yield of Achyranthes japonica N. from 1995 to 1996. Stem diameter, no. of branch and fresh weight of above-ground parts per plant were reduced by increasing the planting density, but stem length, length and diameter of main root increased at high density, $25{\times}5cm$. The heighest percent of large roots was 71 % at $25{\times}5cm$ planting density. The dry root yield per 10a at $25{\times}5cm$ planting density was 7% higher than 306kg of $20{\times}5cm$ planting density, but root yields were lower in other planting density compared to $25{\times}5cm$ planting density. The root dry weight showed negative correlation with stem diameter, no. of branch and fresh weight of above-ground parts per plant but showed positive correlation with stem length, length and diameter of main root.

  • This experiment was carried out to investigate the effects of black P.E. film mulching by different sowing dates on the growth and yield of Achyranthes japonia NAKAI. Emergence rate and the growth increment were increased in black polyethylene (P.E.) film mulching of sowing date at April 1. Dried root yield was highest by 3,190kg/ha in black P.E. film mulching and earlist sowing at April 1 with 20% increase compared to that of non mulching at April 1 planting.
