저장 조건에 따른 녹차 추출물의 acetylcholinesterase(AChE) 저해 효과 (Acetylcholinesterase Inhibitory Effect of Green Tea Extracts According to Storage Condition)

  • 잎녹차 추출물 보다는 분말녹차 추출물의 경우가 그리고 저장된 sample보다는 fresh sample의 AChE 저해활성이 상대적으로 높았고 이는 생리활성 물질의 추출효율성과 생리활성 물질의 특성 변화 측면에서 기인되는 것으로 판단된다. 또한 본 연구팀에서 구성한 저장 조건에서, 4주간의 짧은 저장기간이 3개월 저장조건보다는 우수한 효소 저해활성도를 나타냈다. 수분활성도 측면에서는 수분활성도 0.81의 조건을 갖는 sample에서의 효과가 비교적 우수했고, 3개월 저장하는 동안의 저장온도 조건은 효소 저해 활성도에 큰 차이를 보이는 요소로 작용하지 못했다. 다만 상온이나 냉장조건 보다는 냉동상태의 저장조건이 다소 우수한 것으로 판단된다. 결국 생리활성물질을 보다 효율적으로 추출할 수 있는 녹차의 상태와 처리 또는 섭취 농도가 더욱 중요한 것으로 사료되고, 추출 효율을 증가시키기 위해서는 잎 보다는 분말의 상태가 더 우수한 것으로 판단된다. 따라서 녹차 추출물은 acetylcholine성 신경세포의 기능을 강화시켜줄 수 있는 AChE 저해제로서 활용가치가 높을 것이다.

Development of Selective Butyrylcholinesterase Inhibitors Using (R)-Lipoic Acid-Polyphenol Hybrid Molecules

  • A series of hybrid molecules between (R)-lipoic acid (ALA) and the acetylated or methylated polyphenol compounds were synthesized and their in vitro cholinesterase [acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE)] inhibition activities were checked. The $IC_{50}$ values of all hybrid molecules for a BuChE inhibition were lower than those of the single parent compounds. Specifically, ALA-acetyl protected caffeic acid (11, ALA-AcCA) was shown as an effective inhibitor of BuChE ($IC_{50}=0.5{\pm}0.2\;{\mu}M$) and also had a great selectivity for BuChE over AChE (more than 800 fold). Inhibition kinetic study indicated that 11 is a mixed inhibition type. Its binding affinity ($K_i$) value to BuChE is $1.52{\pm}0.18\;{\mu}M$.

  • A series of hybrid molecules between (${\alpha}$)-lipoic acid (ALA) and benzyl piperazines were synthesized and their in vitro cholinesterase [acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE)] inhibitory activities were evaluated. Even though the parent compounds did not show any inhibitory activity against cholinesterase (ChE), all hybrid molecules showed BuChE inhibitory activity. Some hybrid compounds also displayed AChE inhibitory activity. Specifically, ALA-1-(3-methylbenzyl)piperazine (15) was shown to be an effective inhibitor of both BuChE ($IC_{50}=2.3{\pm}0.7{\mu}M$) and AChE ($IC_{50}=30.31{\pm}0.64{\mu}M$). An inhibition kinetic study using compound 15 indicated a mixed inhibition type. Its binding affinity ($K_i$) value to BuChE is $2.91{\pm}0.15{\mu}M$.

  • Donepezil hydrochloride is a reversible acetylcholinesterase inhibitor that is used in the treatment of Alzheimer’s disease to improve the cognitive performance. It shows different crystalline forms including hydrates. Therefore, it is very important to confirm the polymorphic forms in the formulations of pharmaceutical materials because polymorphs of the same drug often exhibit significant differences in solubility, bioavailability, processability and physical/chemical stability. In this paper, four different forms of donepezil hydrochloride were prepared and characterized using X-ray powder diffraction, Fourier transform infrared, and solid-state nuclear magnetic resonance (NMR) spectroscopy. This study showed that solid-state NMR spectroscopy is a powerful technique for obtaining structural information and the polymorphology of pharmaceutical solids.

Effects of Water Extract of Smoke-dried Skipjack Tuna on Memory in a Scopolamine-induced Amnesia Animal Model

  • Natural products have been used to treat many neurological illnesses such as Alzheimer's disease. In the present study, the effects of the water extract of smoke-dried skipjack tuna (WSST), which is used as a traditional seasoning in Japan, as well as its fractions on acetylcholinesterase (AChE) inhibition in vitro and on memory in scopolamine-induced amnesia mice in vivo were evaluated. Bio-Rad P-2 gel permeation chromatography revealed the presence of 7 peaks and AChE significantly inhibited peak 3 and 5. When in vivo behavioral studies were conducted, a passive avoidance test revealed that treatment with 50 and 100 mg/kg WSST as well as with fraction 3 and 5 improved the loss in memory retention induced by scopolamine. These results suggest that skipjack tuna extract and its fractions improve memory deficits and that these substances are suitable for use in healthy foods designed to improve memory deficits induced by aging and Alzheimer's disease.

Anxiolytic effects of an acetylcholinesterase inhibitor, physostigmine, in the adult zebrafish

  • Anxiety in zebrafish can be determined by examining their bottom-dwelling and light-avoidance behavior. This study determines the effects of physostigmine and scopolamine on anxiety in zebrafish by measuring swimming frequency for three horizontal layers and three vertical columns of a water test tank illuminated by a light source located above the central surface of the tank. In the 1 h session, zebrafish in the control group preferred the bottom layer the most and the center column the least. Zebrafish treated with 2-20 ${\mu}M$ physostigmine were more likely to prefer the to layer than controls, and there were significant pairwise differences between physostigmine-treated zebrafish and controls, indicating the anxiolytic effect of physostigmine. Further, 10 and $20{\mu}M$ physostigmine-treated zebrafish no longer avoided the center column. Scopolamine had no anxiolytic effect on bottom-dwelling and light-avoidance behaviors but suppressed the anxiolytic effect of physostigmine. In terms of their preference for various zones formed by layers and columns, zebrafish in the control group preferred the bottom left and right zones the most. Physostigmine had a positive effect on the preference for the top center zone, which was suppressed by scopolamine pretreatment. The results suggest that the level of anxiety in zebrafish can be reduced by activating acetylcholinergic neurotransmitter systems, which is mediated in part by muscarinic receptors.

한약재 물 추출물의 생리활성 검색 및 MPTP-유도 신경독성에 대한 대황의 보호효과 (Screening of the Biologoical Activity from Water Extracts of the Medicinal Plants and the Protective Effect of R. palmatum on MTPT-induced Neurotoxicity)

  • This present study was designed to screen medicinal plants for the treatment of brain diseases such as Parkinson's disease or aging. We tested the effects of the water extracts from 38 species medicinal plants on antioxidant capacity, monoamine oxidase B (MAO-B) inhibitory activity, acetylcholinesterase (AChE) inhibition and antiperoxidation activity in vitro. The water extracts from 38 species were tested on their antioxidant activity using radical scavenging effects against ABTS+. The water extract of C. sappan was showed the highest antioxidant capacity, the antioxidant activity at 1 Jig of herbal extract being 0.38mM TE. Lipid peroxidation in brain homogenates induced by NADPH and ADP-Fe/sup 2+/ was strong inhibited by C. sappan and R. palmatum extracts. Among the 38 medicinal plants investigated, R. palmatum showed significant biological activity (antioxidant capacity, MAO-B inhibiory activity, and AChE inhibitory activity). The protective efficacy of R. palmatum water extract on 1-methyl-4­phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism and its possible mechanism were studied in C57BL/6 mice. Treatment of R. palmatum water extract protected biomacromolecules such as lipids from oxidative damage induced by MPTP. The content of MDA in brain tissue was decreased significantly by R. palmatum extract. These results suggest that R. palmatum water extract plays on effective role in attenuating MPTP-induced neurotoxicity in mice. This protective effect of R. palmatum might be estimated the result from the inhibitory activity on monoamine oxidase B and the enhancement of antioxidant activity.

파밤나방(Spodoptera exigua (Hiibner)) 살충제 감수성 진단장치모형 (Diagnostic Device Model for Insecticide susceptibilities of Beet Armyworm, Spodoptera exigua(Hubner))

  • 파밤나방(Spodoptera exigua (Hiibner))의 살충제 감수성을 판별할 수 있는 간편한 진단 장치가 개발되어 야외 집단을 대상으로 판별능력을 시험했다. 이 진단장치의 기본설계원리는 에스테라제와 아세틸콜린에스테라제의 효소활력이 파밤나방의 살충제 감수성과 밀접한 관련성이 있음을 이용하였다. 네가지 형태의 진단장치(ED, EM, AD 및 AM)가 고안되었고 판별효소(E-에스테라제, A-아세틸콜린에스테라제)와 억제제(D-dichlorvos, M-monocrotophos)의 종류에 따라 분류되었다. 판별억제제의 농도는 각각 ED는 1mM, EM는 10mM, AD는 100mM 및 AM는 100mM이었다. 억제되지 않은 저항성 개체는 두 효소 모두에서 적색을 보이나 억제된 감수성 개체의 시료는 무색으로 나타난다. 한 개체의 파밤나방으로부터 채취된 혈액과 머리시료가 네가지 판별장치 검색장치에 한꺼번에 이용되었다. 검색장치의 실증검증을 위해 11개의 실내와 야외 집단들을 대상으로 살충제 감수성과 검색장치 판별력을 비교하였다. 이들 집단은 파밤나방 방제에 이용되고있는 3가지 살충제들(bifenthrin, methomyl, chlorpyrifos-methyl)에 대해 네 종류의 판별장치는 상이한 약제 감수성을 보였다. ED는 bifenthrin, AD는 methomyl 및 ED와 AM은 chlorpyrifos-methyl에 대하여 약제 감수변이를 판별하는 데는 각각 효과적이었다. 이상의 본 연구에서는 개발된 검색장치는 간편하고 신속하게 특정집단의 약제 저항성개체의 빈도수를 알려줌으로 효과적인 방제방법을 선정하는데 이용될 수 있다. 특히, 각 지역의 지도소에서 이 검색장치를 이용할 경우 객관적이고 합리적인 현장 농민에게 지도할 수 있겠다.

노화촉진마우스를 이용한 복분자의 기억력 감퇴 개선 효과 (Rubus coreanus Miquel Improves on Impairment of Memory in Senescence-Accelerated Mouse (SAM))

  • 본 연구에서는 복분자 섭취가 노화에 의한 기억력 감퇴에 개선 효과를 나타내는지 조사하기 위해 노화촉진마우스 (SAM)를 사용하여 평가하였다. 28주령 SAMP8 마우스에 복분자 시료를 100 mg/kg body weight/day로 8주간 투여한 후 수동회피시험과 수중미로시험을 실시하여 기억력 개선효과를 평가하였다. 수동회피시험에서 노화 촉진 동물(SAMP8)은 대조동물(SAMR1)에 비해 밝은 방에서 체류시간이 현저히 감소하였다. 복분자를 섭취한 노화 촉진 동물의 밝은 방에서의 체류 시간은 복분자를 섭취하지 않은 동물에 비해 현저히 증가하였다. 수중미로실험에서 대조 동물에 비해 노화 촉진 동물의 평균 탈출잠복기는 길었고, 복분자를 섭취한 경우 평균 탈출잠복기가 유의적으로 감소하였다. 뇌의 대뇌피질과 해마 조직에서 대조군에 비해 노화 촉진 동물의 acetylcholinesterase 활성이 증가하였다. 복분자를 섭취한 노화 촉진 동물의 해마 조직 내 acetylcholinesterase는 복분자를 섭취하지 않은 동물에 비해 유의적으로 감소하였다. 이 결과들은 복분자가 노화촉진마우스에서 노화에 의한 기억력 감퇴에 대한 개선 효과를 나타내며, 이는 콜린성 신경전달에 중요한 역할을 하는 acetylcholinesterase 활성 감소를 통해 이루어짐을 나타낸다. 향후 더 많은 연구가 수행 되어야 하나, 본 연구는 복분자가 노인의 기억력 개선제 개발에 좋은 기능성 소재로 활용할 수 있음을 제시한다.
