Effect of Recombinant Human Epidermal Growth Factor(DWP 401) on Gastric Secretion and Ulcers in Rats (재조합 인간 상피세포 성장인자(DWP 401)의 흰쥐 위액분비 및 궤양에의 작용)

  • The effects of human epidermal growth factor(EGF) which was produced by recombinant DNA technique was investigated on gastric secretion, gastric lesion and ulcer models in rats. The EGF showed significant inhibition of secretion of gastric juice and total acid output, at 0.4mg/kg, id and also inhibited Shay ulceration at 0.4mg/kg, id in rats. The lesion induced by absolute ethanol was significantly reduced by oral administration of EGF at 0.4mg/kg. Likewise, EGF caused significant inhibition of indomethacin induced gastric ulcer at oral doses of 0.2 and 0.4mg/kg. The EGF produced dose-dependent inhibition of gastric ulcer induced by acidified aspirin, but showed no significant inhibition at oral doses of 0.1, 0.2 and 0.4mg/kg. The chronic gastric ulcer induced by injection of 20% acetic acid solution was significantly reduced by oral doses of 0.1 and 0.4mg/kg of EGF. Duodenal ulcer induced by mepirizole was dose-dependently inhibited by oral doses of 0.1, 0.2 and 0.4mg/kg of EGF. These data suggest that EGF possesses pronounced inhibitory action in gastric ulcer and duodenal ulcer of rats.

  • Antiulcer effects of Artemisia herba extract (DA-9601) were evaluated in various types of experimental gastric ulcer induced in rats. And the effects of DA-9601 on mucus, basal and stimulated gastric acid secretion were also investigated in rats. DA-9601 (12.5∼400 mg/kg, p.o.) prevented the formation of gastric ulcers induced by 60% EtOH in 150 mM HC1, restraint water immersion stress, platelet activating factor (PAF), aspirin in 150 mM HCI with Pylorus-ligation and indomethacin. DA-9601 (4∼400 mg/kg, p.o.) significantly accelerated the healing rate of acetic acid-induced gastric ulcer and significantly stimulated mucus secretion in a dose-dependent manner. DA-9601 (20∼200 mg/kg, i.d.), however, did not inhibit basal gastric acid secretion in pylorus ligated rats and DA-9601 (200 mg/kg, i.d.) failed to influence histamine-, pentagastrin- and carbachol- stimulated gastric acid secretion. These results suggest that DA-9601 has inhibitory action on gastric lesion and ulceration through increasing mucus secretion in the stomach of rats without influencing basal and stimulated gastric acid secretion.

Buccal Mucosal Ulcer Healing Effect of rhEGF/Eudispert hv Hydrogel

  • We have studied the effect of rhEGF on the buccal mucosal ulcer healing. rhEGF was rapidly degraded upon incubation with the hamster buccal mucosal homogenates; The degradation of rhEGF was significantly inhibited by sodium lauryl sulfate (SLS). Eudispert hv hydrogel and Polycarbophil 974P hydrogel were prepared for rhEGF delivery and their mucoadhesiveness was measured by the $Instron^R$ method. The mucoadhesive force of Eudispert hv was significantly greater than that of Polycarbophil 974P. Moreover, rhEGF in Eudispert hv hydrogel remained stable for about 2 months. To evaluate the ulcer healing effect of rhEGF, the buccal mucosal ulcer was induced in golden hamsters using acetic acid. At 24 h after administration of rhEGF/Eudispert hv hydrogel, the ulcerous area was decreased compared with rhEGF solution and, as a result, the curative ratio was $36.8\pm5.68$%. By the addition of SLS (0.5%) to Eudispert hv hydrogel, the curative ratio increased 1.5 times. The mechanism of the action was probably due to a combination of protection of the drug against proteases present in mucosa and prolongation of the release of rhEGF from the formulation at the site of action.

Antigastritic and Antiulcerative Effect of Pulmuone Healthy Aloe Gel (생약복합제 Pulmuone Healthy Aloe Gel의 위장질환 개선 효과)

  • Present study was performed for the development of a new supplementary product with gastroprotective effect. The preliminary screening were conducted for the effects of HCl-ethanol-induced gastric lesions in rats. Samples were aloe gel, active hexose correlated compound (AHCC) mentioned that have GI protective property and pulmuone healthy aloe gel (PHAG) that mixture of natural products from Pulmuone company. Aloe gel significantly inhibited HCl-ethanol-induced gastric lesions at the oral dose of 5 ml/kg. AHCC showed the strongest effectiveness at the oral dose of 1,200 mg/kg. PHAG also showed the significant effects at the oral dose of 10, 20 g/kg. In pylorus ligated rats, the treatments of aloe gel, AHCC and PHAG showed decrease in the volume of gastric secretion and acid output. And aloe gel, AHCC and PHAG significantly suppressed the aspirin-induced ulcer and chronic ulcer in pylorus ligated rats. The treatments of aloe gel and PHAG significantly reduced acetic acid-induced ulcer at the oral dose of 5 ml/kg and 10 g/kg for 12 days. In this study; we have found that PHAG had significant improvement in acute gastritis and ulcer at the dose of 20 g/kg and in chronic gastritis and ulcer at the dose of 10 g/kg. Also we evaluated the anti-bacterial activity against H. pylori treated with aloe gel, AHCC and PHAG. PHAG had a equivalent anti bacterial activity with ampicillin against H. pylori at the dose of 1 g/kg.

Stability and Effectiveness of Laser and Liquiband Treatments in the Rat Model of Oral Ulcer

  • Background and Objectives This study was undertaken to assess the stability and efficacy of laser therapy (808 nm), Liquiband (a commercial topical skin adhesive product), and a combination treatment, for application in oral ulcers. Materials and Methods The oral ulcer rat animal model was used to determine the efficacy of photobiomodulation, Liquiband, or combination therapy. Ulcers were induced by injecting 60% acetic acid in the oral mucosa. Three days after ulcer confirmation, the chemically induced ulcers were treated with either laser (808 nm), Liquiband, or a combination of both, every two days for 8 days (4 sessions). Combination therapy was performed by first treating with laser followed by application of the Liquiband. Reduction in ulcer area size was subsequently determined, and animals were sacrificed at 5 and 10 days after the last administration, for histological observation of the extracted oral ulcer tissue samples. Results In this study, treatment with either laser (808 nm) or commercial Liquiband product as well as the combined treatment significantly enhanced oral ulcer healing in the rat animal model. The effect of laser treatment is mainly attributed to collagen synthesis, whereas application of the Liquiband promotes vascularization and formation of granulation tissue. Our results indicate that further optimization of the combined therapy could synergistically and significantly enhance oral ulcer healing. Conclusion Laser or Liquiband treatment of oral ulcer in the rat animal model exert different outcomes, but both methods confirm significant effects relative to the untreated group. The combined treatment group with laser and Liquiband showed marked oral ulcer healing, but further optimization is required to get highly significant results and establish the potential synergistic effect of the combination therapy, as compared to the individual enhancing effects of healing oral ulcers.

Enhancing Effect of the Combined Preparation Containing Antacid and Aceglutamide Aluminium on Defensive Factors in Chronic Ulcer Model of the Rat (흰쥐의 만성궤양모델에서 Aceglutamide aluminium를 함유한 복합제산제의 점막방어인자 증강작용)

  • Antacid(AM, 600 mg/kg=aluminium hydroxide, magnesium hydroxide, and simethicone with a ratio of 1 : 1 : 0.1) and aceglutamide aluminium(AGA, 263 mg/kg)-Effect of the combined preparation containing on the gastric mucosal hexosamine, sialic acid, and aluminium contents adhered to the gastric wall of the rat was investigated. Severe ulcers were produced in rats by injecting of $30\;{\mu}l$ acetic acid(30%) into the subserosal layer of one position in the corpus. When given orally for 15 consecutive days, AM(1,200 mg/kg), AGA(525, 1,050 mg/kg), and the combined preparation significantly decreased the ulcer area. AGA(525, 1,050 mg/kg) and the combined preparation also increased the amount of hexosamine and sialic acid in the intact and ulcerated areas. On the other hand, the contents of hexosamine and sialic acid were not affected by AM (600, 1,200 mg/kg). The amount of aluminium adhered to the gastric wall of the rat was higher in the combined preparation when compared to the AM(600 mg/kg) and AGA(263 mg/kg). The aluminium contents adhered may play an important role protecting mucosa from aggresive action of gastric juice and potenting defensive factors through the increase of mucosa-forming components by AGA.

  • A series of 1-aryl-lH-pyrazolo[4,3-c]quinolines and 2-aryl-2H-pyrazolo[4,3-c]quinolines are prepared by reacting 3-acyl-4-chloroquinolines in ethanol or 3-acyl-4(lH)-quinolone in acetic acid with appropriate hydrazines as possible anti-ulcer agents. A regiospecific synthesis of 1-aryl-lH-pyrazolo[4,3-c]quinolines is also achieved. The central pyridine ring could be easily reduced by catalytic hydrogenation.

Therapeutic Effects on Gastrointestinal Diseases of a New Combined Ranitidine Preparations, DWP302 (라니티딘을 함유한 새로운 의약조성물(DWP302)의 위장질환 치료효과)

  • The eight combined products composed of ranitidine.HCI, tripotassium dicitrato bismuthate and sucralfate were prepared with various ratios and studied in therapeutic effects of them on various gastrointestinal diseases. These were induced in rats with the porous ligation, ethanol-HCI, acetic acid and cysteamine method, etc. In all experimental setting, the effect of the combined treating was more pronounced than the effect of each drug alone. Specially, the combined treatment consisted of ranitidine : tripotassium dicitrato bismuthate sucralfate ratio of 1.5 : 2 : 6 showed the most powerful therapeutic effect on acute gastric ulcer model and revealed a significant acceleration of the healing on chronic gastroduodenal ulcer model. And that, therapeutic doses of ranitidine, tripotassium dicitrato bismuthate arid sucralfate given in combination had an additive or, in some case, synergistic effect. From the above results, this combined treatment may useful to heal the gastrointestinal diseases that aren't cured well by treatment of each them alone.

An experimental study on the effect of Samchulgunbitang affecting gastro-intestine and central nervous system (삼출건비탕(蔘朮健脾湯)이 위장관(胃腸管)에 미치는 영향(影響)에 관(關)한 실험적(實驗的) 연구(硏究))

  • An experimental study was done to investigate the spontaneous movements to the isolated ileum with liquid extracts of Samchulgunbitang. Then the action of gastric ulcer, gastric-juice secretion, the free acidity, total acidity, pepsin output, the transport ability in the intestine, analgesic effect and sleeping time were measured. The following results were obtained; 1. As to the spontaneous movements in the isolated ileum, the effect of contraction against suppression was recognized. 2. The effects of contraction against suppression induced by acetylcholine chloride and barium chloride were recognized on the gastric funds strip significantly. 3. The preventive effect of Samchulgunbitang on the pylorus-lightedulcer in rat was recognized significantly. 4. The anti-ulcer effect of Samchulgunbitang was not recognized on the gastric ulcer caused by indomethacin. 5. The effects of decreasing on the secretion gastric juice, the free acidity, total acidity and pepsin output of Samchulgunbitang were recognized significantly. 6. The transport rate in the small intestine of Samchulgunbitang was decreased. 7. The transport rate in the large intestine of Samchulgunbitang was increased. 8. The analgesic effect of Samchulgunbitang caused by acetic acid was recognized significantly. 9. The sleeping time caused by pentobarbital-Na of Samchulgunbitang was prolonged significantly. According to the results, it is considered that the Samchulgunbitang has effects of gastric ulcer, chronic gastritis, hyper-acidity, gastroptosis such as abdominal discomfort, gastric acid, indigestion and anorexia.

Studies on Protective Effect of Bupleurum falcatum Extract (SHI-1909) against Experimental Inflammtory Bowel Disease Model (시호추출물 (SHI-1909)의 염증성 대장염 실험동물 모델에 대한 치료효과)

  • The Efficacy of SHI-1909 was investigated in comparision with predinisolone in acetic acid and Picrylsulfonic acid solution (TNBS)-induced rat inflammatory bowel disease (IBD) for 5 days. 7% Acetic acid and 5% TNBS solution were administered with polyethylene (P.E) tube inserted to rats intracolon, which causing colitis to the rats. The acetic acid and TNBS control group (the saline treated colitic rat) exhibited ulceration and inflammation of the distal colon with formation of granuloma and pathologic connections. We checked the inflammatory parameters like rat's weight, food intake quantity change during administration. After 5 days, we sacrificed the rats and checked the colon's length, ulcer and pathologic condition. Oral treatment with SHI-1909 resulted in significant recovery of macroscopic parameters like weight and diet intake change. Especially, SHI-1909 had a more potent effect than prednisolone on macroscopic colonic damage score. We can suggest that SHI-1909 could be a promising drug in the treatment of IBD.
