Protective effect of resveratrol on the oxidative stress-induced inhibition of gap junctional intercellular communication in HaCaT keratinocyte

  • The aim of this study was to investigate the effect of resveratrol on the oxidative stress-induced inhibition of gap junctional intercellular communication (GJIC) in HaCaT keratinocyte. Anti-oxidative activity of resveratrol was measured by a,a-diphenyl-b-picrylhydrazyl (DPPH) assay and dichlorodihydrofluorescein diacetate oxidation assay. GJIC of HaCaT keratinocyte was assessed using the scrape loading/dye transfer technique. Western blots and reverse transcription-polymerase chain reaction were also analyzed for Connexin 43 protein and mRNA expression, respectively. Resveratrol scavenged directly the stable DPPH radical over a concentration range of 4 mg/$\mell$ (78.2 $\pm$ 2.7% of control) to 500 mg/$\mell$ (29.9 $\pm$ 4.2% of control) and prevent to increase the intracellular fluorescence induced by oxidative stress significantly. Ultraviolet A irradiation (UVA) and 12-O-tetradecanoylphorbol-13-acetate markedly reduced GJIC, which was restored by resveratrol. There were no significant differences in the level of Connexin 43 protein and mRNA expression among any of the experimental groups. Our data suggests that resveratrol has the protective effect on the oxidative stress-induced inhibition of gap junctional intercellular communication in HaCaT keratinocyte and this protection is likely due to the scavenging of reactive oxygen species.

In vitro anti-oxidant, anti-wrinkle and whitening effect of solvent fraction from takli-san (탁리산 용매 분획물의 in vitro 항산화, 주름 억제 및 미백 효과)

  • In this study, the activity of Takli-san for anti-oxidation, anti-wrinkle and whitening effect was verified and its applicability as a cosmetic material was confirmed. 1-1-diphenyl-2-picryl-hydrazyl radical scavenge, 2,2'-azinobis(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenge, elastase and tyrosinase inhibitory activity were investigated by solvent fraction, and ethyl acetate fraction of water extract from Takli-san (TW-EA) showed the highest inhibitory activity. Western blot was performed to confirm anti-wrinkle and whitening effect in the cells, and it was investigeted that the TW-EA inhibition effect was superior to that of the positive contol used for the comparison test. Through the results of the experiments, the applicability as a cosmetic material of Takli-san was verified.

Tyrosinase Inhibitors Isolated from the Edible Brown Alga Ecklonia stolonifera

  • Extracts from seventeen seaweeds were determined for tyrosinase inhibitory activity using mushroom tyrosinase with L-tyrosine as a substrate. Only one of them, Ecklonia stolonifera OKAMURA (Laminariaceae) belonging to brown algae, showed high tyrosinase inhibitory activity. Bioassay-guided fractionation of the active ethyl acetate (EtOAc) soluble fraction from the methanolic extract of E. stolonifera, led us to the isolation of phloroglucinol derivatives [phloroglucinol (1), eckstolonol (2), eckol (3), phlorofucofuroeckol A (4), and dieckol (5)]. Compounds 1~5 were found to inhibit the oxidation of L-tyrosine catalyzed by mushroom tyrosinase with $IC_{50}$ values of 92.8, 126, 33.2, 177, and 2.16 ${\mu}g$ /mL, respectively. It was compared with those of kojic acid and arbutin, well-known tyrosinase inhibitors, with $IC_{50}$ values of 6.32 and 112 ${\mu}g$ / mL, respectively. The inhibitory kinetics analyzed from Lineweaver-Burk plots, showed compounds 1 and 2 to be competitive inhibitors with $K_i$ of $2.3{\times}10^{-4}\;and\;3.1{times}10^{-4}$ M, and compounds 3~5 to be noncompetitive inhibitors with $K_i$ of $1.9{\times}10^{-5},\;1.4{\times}10^{-3}\;and\;1.5{\times}10^{-5}$ M, respectively. This work showed that phloroglucinol derivatives, natural compounds found in brown algae, could be involved in the control of pigmentation in plants and other organisms through inhibition of tyrosinase activity using L-tyrosine as a substrate.

Removal of Contaminants Deposited on Surfaces of Matrices by Using Low-Temperature Argon Plasma Treatment (저온 아르곤 플라즈마처리를 이용한 모재 표면의 오염물 제거)

  • The possibility of a low-temperature argon plasma treatment as a mean of restoration technology for contaminated invaluable archive materials and artefacts, and evidencing documents was investigated along with an oxygen plasma treatment for comparison. For this purpose, the degree of color changes, ${\Delta}E^*ab$, and surface morphological changes due to plasma treatments as an evaluation of removal performance of artificial contaminants such as brilliant green dye and carbon deposit on cellulose acetate and plain paper as matrices, respectively, were measured and analyzed using a spectrophotometer and a field emission scanning electron microscope. Compared to the argon plasma treatment with sputtering characteristic, that of the oxygen plasma with characteristic of an oxidation reaction has shown superior results in removing the contaminants; the oxygen plasma has proven to damage the matrices significantly due to its oxidative characteristic, and post-plasma reactions has anticipated to be also detrimental to the surfaces, whereas, the problems caused by the counterpart has resulted in being negligible and rather has thought to be an appropriate mean for delicate restoration and/or removal operations of contaminants.

Effects of $LiMn_{2}O_{4}$ Addition on $CO_{2}$ Decomposition Using Spinel Phase $Fe_{3}O_{4}$ (스피넬상 $Fe_{3}O_{4}$를 이용한 $CO_{2}$ 분해에서 $LiMn_{2}O_{4}$ 첨가효과)

  • The spinel $Fe_{3}O_{4}$ powders were synthesized using 0.2 $M-FeSO_4{\cdot}7H_{2}O$ and 0.5 M-NaOH by oxidation in air and the spinel $LiMn_{2}O_{4}$ powders were synthesized at 480 $^{\circ}C$ for 12 h in air by a sol-gel method using manganese acetate and lithium hydroxide as starting materials. The synthesized $LiMn_{2}O_{4}$ powders were mixed at portion of 5, 10, 15 and 20 wt% of $Fe_{3}O_{4}$ powders using a ball-mill. The mixed catalysts were dried at room temperature for 24 hrs. The mixed catalysts were reduced by hydrogen gas at 350 $^{\circ}C$ for 2 h. The carbon dioxide decomposition rates of the mixed catalysts were 90% in all the mixed catalysts but the decomposition rate of carbon dioxide was increased with adding $LiMn_{2}O_{4}$ powders to $Fe_{3}O_{4}$ powders.

The Isolation and Antioxidative Effects of Vitexin from Acer palmatum

  • Free radicals and reactive oxygen species (ROS) caused by UV exposure or other environmental factors are critical players in cellular damage and aging. In order to develop a new antiphotoaging agent, this work focused on the antioxidant effects of the extract of tinged autumnal leaves of Acer palmatum. One compound was isolated from an ethyl acetate soluble fraction of the A. palmatum extract using silica gel column chromatography. The chemical structure was identified as apigenin-8-C-beta-D-glucopyranoside, more commonly known as vitexin, by spectral analysis including LC-MS, FT-IR, UV, $^{1}H-$, and $^{13}C-NMR$. The biological activities of vitexin were investigated for the potential application of its anti-aging effects in the cosmetic field. Vitexin inhibited superoxide radicals by about $70\%$ at a concentration of $100\;{\mu}g/mL$ and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals by about $60\%$ at a concentration of $100\;{\mu}g/mL$. Intracellular ROS scavenging activity was indicated by increases in dichlorofluorescein (DCF) fluorescence upon exposure to UVB $20\;mJ/cm^2$ in cultured human dermal fibroblasts (HDFs) after the treatment of vitexin. The results show that oxidation of 5-(6-)chloromethyl-2',7'-dichlo-rodihydrofluorescein diacetate ($CM-H_{2}DCFDA$) is inhibited by vitexin effectively and that vitexin has a potent free radical scavenging activity in UVB-irradiated HDFs. In ROS imaging using a confocal microscope we visualized DCF fluorescence in HDFs directly. In conclusion, our findings suggest that vitexin can be effectively used for the prevention of UV-induced adverse skin reactions such as free radical production and skin cell damage.

  • The separation of sulfone components using light cycle oil(LCO) after oxidation was carried out by solvent extraction method using various polar solvents such as water, n-methyl-2-pyrrolidone(NMP), dimethyl sulfoxide, ethyl acetate, acetonitrile, dimethyl formamide, and methyl alcohol. It was found that phase separation between LCO layer and solvent occurred under mixed solvent adding a proper amount of water. The mixture solvent of NMP and water was a promising extraction solvent due to the selective removal and high distribution coefficient of sulfone component in LCO. 99.5% over of sulfur contents in LCO can be removed by 4 stages equilibrium extraction.

  • A practical method applicable to the synthesis of aromatic dihydrazines was proposed by reducing tetrazonium salt in strong mineral acid media. By diazotizing p-phenylenediamine with sodium nitrite in a medium of concentrated hydrochloric acid or 45 % perchloric acid at $-5 {\sim} -10{\circ}C$ and reducing the tetrazonium salt with stannous chloride, p-phenylenedihydrazine (PPDH) was separated in the form of hydrochloride as colorless fine needles. Since PPDH was subject to oxidation and unstable, the free base could not be isolated. PPDH${\cdot}$2HCl was decomposed at $^180{\circ}C$ without showing sharp melting point. It behaved largely as aromatic monohydrazines, and reacted immediately with aldehydes and ketones in acetate buffer, giving generally yellow to brownish condensation products, dihydrazones. This suggests that PPDH will react with dicarbonyl compounds producing high molecular polymers or cyclization products.

Characterization and Photocatalytic effect of ZnO nanoparticles synthesized by spray-pyrolysis method

  • ZnO shows a direct band gap of 3.37eV, large exciton binding energy (~60 meV), high oxidation ability, high sensitivity to many gases, and low cost, and it has been used in various applications such as transparent electrodes, light emitting diodes (LEDs), gas sensors and photocatalysts. Among these applications ZnO as photocatalyst has considerably attracted attention over the past few years because of its high activities in removing organic contaminants generated from industrial activities. In this research, ZnO nanoparticles were synthesized by spray-pyrolysis method using the zinc acetate dihydrate as starting material at synthesis temperature of $900^{\circ}C$ with concentration varied from 0.01 to 1.0M. The physical and chemical properties of the synthesized ZnO nanoparticles were examined by X-ray Diffraction (XRD), Scanning Electron Microscopy (SEM), Fourier Transformation Infrared (FT-IR), and UV-vis spectroscopy. The Miller indices of XRD patterns indicate that the synthesized ZnO nanoparticles showed a hexagonal wurtzite structure. With increased precursor concentration, a primary, secondary particle sizes of ZnO nanoparticles increased by 0.8 to $1.5{\mu}m$ and 15 to 35nm, and their crystallinity was improved. Methyleneblue (MB) solution ($1{\mu}M$) as a test comtaminant was prepared for evaluating the photocatalytic activities of ZnO nanoparticles synthesized in different precursor concentration. The results show that the photocatalytic efficiency of ZnO nanoparticles was gradually enhanced by increased precursor concentration.

Inhibitory Effect of Pinus rigida × Pinus taeda on Melanogenesis in B16 F10 Cells

  • The cone of Pinus rigida × Pinus taeda (PRT), a plant in the Pinaceae family, has long been used in traditional medicine to treat hemostasis, bruises, and burns. Previous research has shown that regulating oxidation-reduction reactions in reactive oxygen species can help inhibit melanogenesis, the process of melanin synthesis, which is a common target for addressing hyperpigmentation. Inhibiting tyrosinase is also known to be effective in this regard. Based on these findings, we conducted an investigation into the inhibitory effect of the ethyl acetate fraction of PRT (ERT) on melanogenesis in B16 F10 cells. We know that the expression levels of melanin biosynthesis-related proteins, including tyrosinase, TRP-1, and TRP-2, are regulated by MITF (microphthalmia-associated transcription factor) and cAMP, with cAMP affecting the activity of protein kinase A (PKA). PKA can reduce melanogenesis, and CREB reduces the phosphorylation of melanin-producing enzymes. In addition, the MAPK signaling pathway, composed of ERK, JNK, p38, and other factors, is also known to play a role in the inhibition of melanogenesis in melanocytes. Our immunoblotting results showed that ERT inhibited the expression of melanin production-related proteins (tyrosinase, TRP-1, TRP-2, and MITF) that were significantly increased by a-MSH treatment to promote melanin production. Furthermore, the phosphorylation levels of factors related to cAMP/PKA/CREB and MAPK signaling pathways were significantly reduced without affecting the total form. In conclusion, we believe that treatment with ERT can inhibit melanin synthesis by modulating the phosphorylation of cAMP/PKA/CREB and MAPK signaling pathways at the cellular level. These findings suggest the potential of ERT as a raw material for functional cosmetics and pharmaceuticals, thanks to its antioxidant activity and ability to inhibit melanogenesis. We thought that these findings of ERT as a natural plant resource will inspire further research and development in this area.
