New Antioxidant Sources; Tinged Autumnal Leaves of Maple and Cherry Trees

  • Living systems constantly suffer from atmospheric reactive oxygen species and also produce inevitably them by the course of metabolism. Therefore, antioxidants play important roles in protecting the systems from various diseased caused by them. In this study, we investigated various tinged autumnal leaves as antioxidant sources. Among the, the red leaf extracts of Acer Palmatum THUNBERG.(Aceraceae: maple tree) and Prunus Donarioum Sieb. Var. spontanea Makino(Rosaceae : cherry trees) showed the highest anti-oxidativities. The major antioxidants in their red leaves were isolated and identified as vitexin from maple leaves and isoscutellarein-4'-O-$\beta$-glucopyranoside from cherry leaves. Finally, we evaluated the efficacy of skin care products containing the extracts by human use study. As a result, the tinged leaves of maple and cherry trees were evaluated as good antioxidant sources on the bases of antioxidativities, cytotoxicities, cell proliferation effects and human use study.

신나무 유래 Acertannin의 인체 모유두 세포 Apoptosis 조절 효능 (Modulative Effect of Human Hair Dermal Papilla Cell Apoptosis by Acertannin from the Barks and Xylems of Acer ginnala Maxim)

  • We isolated gallotannin, 2,6-digalloyl-1,5-anhydroglucitol, known as acertannin (1), from the barks and xylems of Acer ginnala Maxim. It is a genus of Acer species of shrubs in the family Aceraceae. A. ginnala grows in Korea, Japan and Mongolia. We accomplished the structure elucidation by confirming that the result of $^1H$,$^{13}C-NMR$,MS spectrum data was similar to previous references. We measured DPPH and ABTS radical scavenging activity in vitro to evaluate anti-oxidative activities on acertannin isolated from A. ginnala. Acertannin from A. ginnala exhibited potent DPPH and ABTS radical scavenging activities. We examined the antioxidant and apoptosis modulative effects. This examination shows that A. ginnala has not only 1,1-diphenyl-2-picryhydrazyl(DPPH) radical scavenging activity and ABTS radical scavenging activity, but also human hair dermal papilla cell protection effects. These results indicate that the barks and xylems of A. ginnala might be developed as a potent anti-oxidant, hair growth agent, and ingredient for related new functional cosmetic materials.

산겨릅나무 줄기에서 페놀성 글루코사이드의 분리 (Isolation of Phenolic Glucosides from the Stems of Acer tegmentosum Max)

  • 산겨릅나무(A. tegmentosum)의 성분 연구를 위하여 줄기를 MeOH로 추출하여 $CH_2Cl_2$, n-BuOH 및 $H_2O$ 순으로 분획하였다. n-BuOH fraction에 대하여 silica gel과 RP-18 column chromatography를 행하여 2종의 화합물을 분리하였다. 이들 화합물의 구조는 spectral data를 문헌치와 비교하여 methyl gallate $4-O-{\beta}-D-glucoside(1)$와 salidroside(2)로 동정하였다. 이들 화합물은 산겨릅나무에서 처음으로 분리되었다.

  • 예로구터 그 수액이 건강보조식품으로 사용되어온 고로쇠나무의 줄기를 대상으로 항산화 활성을 가지는 성분을 찾기위하여 실험에 착수하였으며, 그 MeOH추출물의 ETOAc분획을 대상으로 Column chromatography를 실시하여 scopoletin, isoscopoletin, (-) -epicatechin등 3종의 화합물을 분리하고 항산화 활성을 측정하였다. 이중 scopoletin과 isoscopoletin은 BHA나 ${\alpha}-tocopherol$보다 활성이 약하였으나 $(RC_{50}\;:\;120,\;110\;{mu}g)$, (-)-epicatechin의 항산화활성 $(RC_{50}\;:\;7.5\;{mu}g)$은 BHA나 ${\alpha}-tocopherol$보다 뚜렷하게 높았다.

Chemical Constituents from Acer mandshuricum and Their Effects on the Function of Osteoblastic MC3T3-E1 Cells

  • A new compound, 4-methoxyl 5-hydroxymethyl benzoic 3-O-$\beta$-D-glucopyranoside (1), has been isolated from the leaves and stems of Acer mandshuricum, along with nine known compounds (2-10). Their structures were determined by a variety of spectroscopic analyses. The effect of compounds 1-10 on the function of osteoblastic MC3T3-E1 cells was examined by determining alkaline phosphatase (ALP) activity, collagen synthesis, and mineralization. Compound 1 significantly increased the function of osteoblastic MC3T3-E1 cells; $5.0\;{\mu}M$ of 1 increased ALP activity, collagen synthesis, and mineralization of MC3T3-E1 cells to 114.7, 119.5, and 108.2% (P < 0.05) of the basal value, respectively. In addition, compounds 2-10 also potently increased the function of osteoblastic MC3T3-E1 cells.

속리산의 식물상 및 식물지리학적 해석 (Flora of Songni Mountain and Its Phytogeographical Interpretation)

  • 속리산 지역내에 생육하고 있는 목본식물을 대상으로 현지조사를 하였으며, 총 44과 89속 140종 10변종 및 2품종이 조사되었다. 한편 국내의 몇 지역과 비교하여 속리산 식물상의 식물지리학적인 분포특성을 파악하고자, 백두산, 설악산, 지리산 및 속리산 등 내륙지역 4개지역, 어유정도, 덕적군도, 고군산군도 및 제주도 등 도서지역 4개 지역등 총 8개 지역의 식물상과 상호 비교하였다. 속리산의 식물상은 설악산 및 지리산의 식물상과 가장 유사하였으며, 단풍나무과, 두릅나무과, 참나무과, 피나무과 및 느릅나무과와 같은 북방계 식물 및 노박덩굴과, 메꽃과, 대극과, 갈매나무과, 가지과, 돌나물과, 쐐기풀과 등과 같은 남방계 식물이 유입되어 있는 것으로 해석되었다.

산겨릅나무 간 보호 화합물 salidroside의 추출조건 최적화 (Optimization of Extraction Conditions of Salidroside, Hepatoprotective Compound from Acer Tegmentosum)

  • Salidroside is a major phenolic glycoside of Acer tegmentosum (Aceraceae) and known to be a hepatoprotective compound. Extraction conditions are one of the most important factors to save time and cost from an economic point of view. For this reason, this study was conducted to optimize the extraction condition for maximum yield of salidroside. For optimization, three extraction factors such as ethanol concentration (%), extraction temperature (℃), and solvent to material ratio (mg/mL) were tested and optimized for maximum yield of salidroside using response surface methodology (RSM). The optimal condition was obtained as an ethanol concentration of 53.4%, an extraction temperature at 67.11℃ and a solvent to material ratio (mg/mL), 195.55 mg/ml. The salidroside yield under optimal conditions was found to be 1.59 mg/g dried samples, which were well-matched with the predicted value of 1.56 mg/g dried samples. These results will provide useful information about optimized extraction conditions for the development of salidroside as hepatoprotective therapeutics.

산청목 메탄올 추출물의 진통 효과 (Anti-nociceptive Activity of Acer tegmentosum MeOH Extract)

  • Acer tegmentosum which is belongs to Aceraceae has been widely used as a traditional medicine for the treatment of lots of diseases including pain management. In this study, we evaluated the anti-nocicepitve effects of methanolic extract of A. tegmentosum (MAT) in mice using various pain models. MAT presented strong and dose-dependent anti-nociceptive activities on thermal nociception models such as tail-immersion test and hot plate test. Moreover, acetic acid-induced chemical nociception was signigicantly reduced by MAT treatment. We could confirm MAT's central and peripheral analgesic properties by formalin test. We also found that the pre-treatment of opioid receptor antagonist did not alter the MAT's anti-nociception, suggesting opioid receptor is not involved in analgesic activity of MAT. Based on our results, we could conclude that MAT may be possibly used as an anti-nociceptive agent for the treatment of various nociceptive pains.

Chemical Constituents from the Leaf and Twig of Acer okamotoanum Nakai and their Cytotoxicity

  • As a result of cytotoxic compounds against cancer cell lines from natural sources, senven compounds were isolated from the leaf and twig of Acer okamotoanum Nakai. The compounds (1-7) were identified as ethyl gallate (1), methyl gallate (2), gallic acid (3), trans $resveratrol-3-O-{\beta}-D-glucopyranoside$ (4), acertannin (5), nikoenoside (6), and fraxin (7) by physicochemical and spectroscopic data (including mp, UV, IR, MS, $^1H-NMR,\;^{13}C-NMR$, DEPT, and HMBC) in comparison with those of published papers. All the compounds were tested for their cytotoxic activity against L1210, HL-60, K562, and B16F10 cancer cell lines in vitro by MTT assay method. Compounds 1-3 and 5 showed cytotoxic activity against all tested cell lines with $IC_{50}$ values ranged from 12.5 to $72.2\;{\mu}M$. Of the compounds, methyl gallate (2) exhibited the most potent cytotoxic activity against L1210, HL-60, K562, and B16F10 tumor cells with $IC_{50}$ values of 12.5, 48.3, 22.8, and $22.8\;{\mu}M$, respectively. Other compounds did not show any cytotoxic activity against four cancer cell lines.

고로쇠나무 종자 수확시기에 따른 발아특성 및 초기생육 (Germination Characteristics and Early Growth of Acer pictum subsp. mono (Maxim.) H. Ohashi by Harvesting Dates)

  • Background : Acer pictum is a deciduous tree belonging to the family Aceraceae. This study was conducted to collect basic data on mass propagation techniques by surveying the germination and growth characteristics of A. pictum seeds. Methods and Results : A. pictum seeds were harvested in 2014 on August 19, September 5, September 29 and October 31. The seeds were then sown on the same dates they were harvested. A portion of seeds harvested in August 2014 were stored at low temperatures and subsequently sown on March 11, 2015. The germination rate of A. pictum seeds was highest in seeds that were stored harvested on October 31. Mean germination time (MGT) was shortest and germination velocity (Rs) was fastest in seeds harvested on October 31. Root collar diameter, total root length, and dry weight were also highest from seeds collected October 31. Conclusions : A. pictum seeds harvested on October 31 had highest germination and growth rates compared with seeds harvested on earlier dates. It is expected that these results demonstrating optimum harvesting and sowing dates will be applicable to future seedling production for this tree species.
