Homoeopathically prepared Condurango 30C is traditionally used in amelioration of certain types of cancer by homeopathic practitioners. In this study, ability of Condurango 30C in amelioration of the conventional benzo[a]pyrene (BaP)-induced lung cancer in rat has been tested. After one month of scheduled oral feeding of BaP, lung cancer is routinely developed after four months in rats. Tumorbearing rats were then treated with Condurango 30C for the next one ($5^{th}$), two ($6^{th}$) and three ($7^{th}$) months, respectively, and sacrificed. Efficacy of post-cancer treatment by Condurango 30C was evaluated against controls (placebo) by different study parameters like: body and lung weights, number and diameter of lung tumour nodules, lung architecture, DNA damage, anti-oxidant activity and reactive oxygen species (ROS) accumulation. Administration of this homeopathic remedy caused increase of body weight and decrease of lung weight, decrease in number and diameter of lung tumour nodules, particularly after one and two months of drug treatment. BaP intoxication significantly increased lipid peroxidase (LPO) with concomitant decrease in activities of different antioxidants, while Condurango 30C administration certainly reduce their levels than normal and cancerous groups, notably after one and two months' of drug treatment. Condurango 30C showed capability to induce ROS-mediated cell death evidenced from the study of ROS activities at different time-points. Further, the remedy possibly achieved its anticancer goal through mediation of DNA-nicks that possibly led cancer cells to the apoptotic pathway. Thus, Condurango 30C has anticancer potential in BaP-induced lung cancer of rats via tissue damage recovery and ROS-mediated programmed cell death.
This paper describes the effects of sewage amount, temperature, and years in operation on the infiltration rate of a sewage disposal system. The self-purifying sewage disposal system, which is typically used in rural areas, consists of reeds and fine gravel. Water plants are planted on the gravel bed which provides the habitat for microbes. The basic process is that the gravel bed filters incoming sewage. Thus this system requires the smooth flow of sewage through the gravel. However, the efficiency of the disposal system will be lowered if the gravel bed is clogged with sewage sludge. A three year study shows that infiltration rate slows down significantly until the 7th day, depending on the sewage amount and the temperature. After the 7th day, the infiltration rate remains almost constant. In addition, the infiltration rate decreases as the temperature falls. It also decreases as the number of years in operation increase. But there is no significant change in the infiltration rate after the 7th day, independent of the temperature, the sewage amount, and years in operation. In order to take advantage of high infiltration rate, which improves the efficiency of the disposal system in its early stages, having two gravel beds and using them alternatively will be efficient. This operation method is called intermittent load and makes the disposal system last longer. The water plant roots above the gravel bed make the effective filtration possible because they delay accumulation of the sewage sludge and stabilize the filtration ability.

Cleaning up the landfill soil by phytoremediation in association with biomass production and utilization of biosolid as a soil amendment will be an attractive green technology. In order to examine this integrated green technology, in the current study of pot trial, heavy metal removal rate and biomass production were determined following cultivation of three different plant species in the landfill soil incorporated with biosolid at two different levels (25 ton $ha^{-1}$ and 50 ton $ha^{-1}$). Among the three plant species including Indian mustard (Brassica juncea), giant sunflower (Helianthus giganteus. L), and giant cane (Arundo donax. L), sunflower appeared to produce the largest biomass yield (19.2 ton $ha^{-1}$) and the produced amounts were magnificently increased with biosolid treatment compared to the control (no biosoild treatment). The increased production associated with biosolid treatment was common for other plant species and this was attributed to the biosolid originated nutrients as well as the improved soil physical properties due to the organic matter from biosolid. The elevated heavy metals in soil which was originated from the incorporated biosolid were Cu and Zn. Based on the phytoavailable amount of heavy metals from biosolid, the removed amount by plant shoots were 95% and 165% for Cu and Zn, respectively, when sunflower was grown. This indicated that mitigation of heavy metal accumulation in soils achieved by the removal of metal through sunflower cultivation enables the successive treatment of biosolid to soils. Moreover, sunflower showed heavy metal stabilization ability in the rhizosphere resulting in alleviation of metal release to ground water.
Mycolic acid-containing gram-positive bacteria, so called nocardioform actinomycetes, have become a great interest to environmental microbiologists due to their metabolic versatility, multidegradative capacity and potential for bioremediation of priority pollutants. For example, Rhodococcus rhodochrous N75 was able to metabolize 4-methy1catechol via a modified $\beta$-ketoadipate pathway whereby 4-methylmuconolactone methyl isomerase catalyzes the conversion of 4-methylmuconolactone to 3-methylmuconolactone in order to circumvent the accumulation of the 'dead-end' metabolite, 4-methylmuconolactone. R. rhodochrous N75 has also shown the ability to transform a range of alkyl-substituted catechols to the corresponding muconolactones. A novel 3-methylmuconolactone-CoAsynthetase was found to be involved in the degradation of 3-methylmuconolactone, which is not mediated in a manner analogous to the classical $\beta$-ketoadipate pathway but activated by the addition of CoA prior to hydrolysis of lactone ring, suggesting that the degradative pathway for methylaromatic compounds by gram-positive bacteria diverges from that of proteobacteria. Mycobacterium sp. Strain PYR-l isolated from oil-contaminated soil was capable of mineralizing various polyaromatic hydrocarbons (PAHs), such as naphthalene, phenanthrene, pyrene, fluoranthrene, 1-nitropyrene, and 6-nitrochrysene. The pathways for degradation of PAHs by this organism have been elucidated through the isolation and characterization of chemical intermediates. 2-D gel electrophoresis of PAH-induced proteins enabled the cloning of the dioxygenase system containing a dehydrogenase, the dioxygenase small ($\beta$)-subunit, and the dioxygenase large ($\alpha$)-subunit. Phylogenetic analysis showed that the large a subunit did not cluster with most of the known sequences except for three newly described a subunits of dioxygenases from Rhodococcus spp. and Nocardioides spp. 2-D gel analysis also showed that catalase-peroxidase, which was induced with pyrene, plays a role in the PAH metabolism. The survival and performance of these bacteria raised the possibility that they can be excellent candidates for bioremediation purposes.
The potential ability of Button mushroom compost (BMC) to solubilize heavy metals was estimated with metal contaminated soils collected from abandoned mines of Boryeong area in South Korea. The bacterial strains in BMC were isolated for investigating the mobilization of metals in soil or plant by the strains and identified according to 16S rRNA gene sequence analysis. When metal solubilization potential of BMC was assessed in a batch experiment, the BMC was found to be capable of solubilizing metals in the presence of metals (Co, Pb and Zn) and the results showed that inoculation of BMC could increase the concentrations of water soluble Co, Pb and Cd by 35, 25 and 45% respectively, than those of non-inoculated soils. BMC-assisted growth promotion and metal uptake in sunflower (Helianthus annuus) was also evaluated in a pot experiment. In comparison with non-inoculated seedlings, the inoculation led to increase the growth of H. annuus by 27, 25 and 28% respectively in Co, Pb and Zn contaminated soils. Moreover, enhanced accumulation of Co, Pb and Zn in the shoot and root systems was observed in inoculated plants, where metal translocation from root to the above-ground tissues was also found to be enhanced by the BMC. The apparent results suggested that the BMC could effectively be employed in enhancing phytoextraction of Co, Pb and Zn from contaminated soils.
The ability of the total extract from Physalis angulata; three fractions after partitioning with n-hexane, ethyl acetate (TBE), and water; and four withanolides (compounds 1 - 4) to phosphorylate 5'-adenosine monophosphate-activated protein kinase (AMPK) and acetyl-CoA carboxylase (ACC) in HepG2 cells was evaluated. The TBE fraction (50 ㎍/mL) activated p-ACC and p-AMPK expression most strongly. Compounds 1 - 4 (10 μM) upregulated p-ACC expression at different levels. Compound 4 induced the most significant changes in p-AMPK expression, followed by 1 and 2. Sterol regulatory element-binding proteins (SREBPs) play a functional role in the transcriptional regulation of the lipogenic pathway, including fatty acid synthase (FAS) and ACC. The effects of compounds 2 and 4 (10 μM) on FAS and SREBP-1c expression under high glucose conditions (30 mM) in HepG2 cells were evaluated further. Both dose-dependently inhibited FAS and SREBP-1c expression as well as lipid accumulation (1 - 10 μM) were compared to high-concentration glucose control, which upregulated FAS and SREBP-1c. These results suggest that compounds 2 and 4 upregulate AMPK, suppress FAS and SREBP-1c, and have potential effects on glucose and lipid metabolism.
Burkholderia anthina R-4183, Burkholderia diffusa R-15930 and Burkholderia stabilis LMG 14294 isolated from green house soils (Gongju-Gun area, South Korea) were characterized and their phosphate solubilizing ability was assessed. Under in vitro culture conditions, all three species were proved to be effective in solubilizing phosphates in varying degrees. Strain Burkholderia anthina exhibited the highest phosphate solubilization in NBRIP medium ($665{\mu}g\;ml^{-1}$) followed by Burkholderia diffusa ($630{\mu}g\;ml^{-1}$) and Burkholderia stabilis ($578{\mu}g\;ml^{-1}$). However, solubilization of $FePO_4$ and $AlPO_4$ was found to be poor in all the strains. Acidification by means of gluconic and oxalic acids accumulation in the culture medium could be the possible mechanism responsible for phosphate solubilization. Glucose at the rate of 3% was found be the best carbon source for Burkholderia anthina while other two Burkholderia species showed maximum phosphate solubilization at 2% of glucose. In the case of nitrogen sources, ammonium and nitrate were equally effective in solubilizing phosphates by Burkholderia species. Despite a slight decrease in phosphate solubilization observed at increasing temperature, all three Burkholderia species could withstand a temperature of $30-35^{\circ}C$, pH at the range of 7-9 and the presence of NaCl (up to 2.5%) without much compromising the phosphate solubilization. As shown with potted mung bean seedlings, all the three isolates could enhance soil fertility and plant growth indicating their great potential to be used as bio-inoculants.

On the foundation of a database of genome sequences and protein analyses, the ability to clone a gene based on a peptide analysis is becoming more feasible and effective for identifying a specific gene and its protein product of interest. As such, the current study conducted a protein analysis using 2-D PAGE followed by MALDI- TOF and ESI-MS to identify a highly expressed gene product of C. parasitica. A distinctive and highly expressed protein spot with a molecular size of 47.2 kDa was randomly selected and MALDI-TOF MS analysis was conducted. A homology search indicated that the protein appeared to be a fungal enolase (enol). Meanwhile, multiple alignments of fungal enolases revealed a conserved amino acid sequence, from which degenerated primers were designed. A screening of the genomic $\lambda$ library of C. parasitica, using the PCR amplicon as a probe, was conducted to obtain the full-length gene, while RT-PCR was performed for the cDNA. The E. coli-expressed eno 1 exhibited enolase enzymatic activity, indicating that the cloned gene encoded the C. parasitica enolase. Moreover, ESI-MS of two of the separated peptides resolved from the protein spot on 2-D PAGE revealed sequences identical to the deduced sequences, suggesting that the cloned gene indeed encoded the resolved protein spot. Northern blot analysis indicated a consistent accumulation of an eno1 transcript during the cultivation.
Melanin is a major factor that darkens skin color as one of the defense systems to prevent the harmful effects of UV light. However, darkened skin from the localized or systemic accumulation of melanin is viewed in many cultures as an esthetic problem. Consequentially, searching for anti-melanogenic agents from natural sources is very popular worldwide. Previous screening of fermented rice products, obtained from various rice cultivars fermented with different sources of loog-pang (Thai traditional fermentation starter), revealed that the highest ability to reduce the melanin content in B16F10 melanoma cells was from unpolished black rice fermented with a defined starter mixture of microbes isolated from loog-pang E11. The aim of this study was to investigate the mechanism of the fermented unpolished black rice (FUBR) on the inhibition of melanogenesis in B16F10 melanoma cells. The strongest reduction of cellular melanin content was found in the FUBR sap (FUBRS). The melanin reduction activity was consistent with the significant decrease in the intracellular tyrosinase activity. The FUBRS showed no cytotoxic effect to B16F10 melanoma or Hs68 human fibroblast cell lines. It also significantly reduced the transcript and protein expression levels of tyrosinase, tyrosinase-related protein 1 (TYRP-1), TYRP-2, and microphthalmia-associated transcription factor. Furthermore, it induced a significantly increased level of phosphorylated ERK, p38 and Akt signaling pathways, which likely contributed to the negative regulation of melanogenesis. From these results, a model for the mechanism of FUBRS on melanogenesis inhibition was proposed. Moreover, these results strongly suggested that FUBRS possesses anti-melanogenesis activity with high potential for cosmeceutical application as a skin depigmenting agent.
