Rice is widely cultivated in various regions throughout Asia. Over a five-year period, we investigated the effects of supplemental UVB radiation on the growth and yield of Japanese rice cultivars in the field. The findings of that study indicated that supplemental UVB radiation has inhibitory effects on the growth and grain development. Furthermore, we investigated the sensitivity to UVB radiation of rice cultivars of 5 Asian rice ecotypes, and found that rice cultivars vary widely in UVB sensitivity. The aim of our study is improving UVB resistance in plants by bioengineering or breeding programs. In order to make it, there is need to find the molecular origin of the sensitivity to UVB. Cyclobutane pyrimidine dimer (CPD) is major UV-induced DNA lesions. Plants possess two mechanisms to cope with such DNA damage. The first is the accumulation of UV-absorbing compounds. Our previous data showed that the steady-state CPD levels in leaves of rice grown under chronic radiation in any culture were not so greatly influenced by the increased UV-absorbing compounds content, although there was a significant positive correlation between the CPD levels induced by challenge UVB exposure and the UV-absorbing compounds content. The other is the repair of DNA damage. Photorepair is the major pathway in plants for repairing CPD. We found that the sensitivity to UVB could seriously correlate with the low ability in CPD photorepair in rice plants. These results suggest that photo lyase might be an excellent candidate for restoration by way of selective breeding or engineering in rice.
Poly(lactide)s(PLA) is an attractive material to solve the problem of waste plastic accumulation in nature because of its biodegradability. The lactide exists in three stereoisomeric configurations: L-lactide, D-lactide, and meso-lactide. PLA stereocomplexes, formed by the mixing of two enantiomers, poly(L-lactide)(PLLA) and poly(D-lactide)(PDLA), have many favorable characteristics because the stereocomplex showed $50^{\circ}C$ higher melting point than each enantiomeric polymer and the resistance toward degradation increased. In this study, we investigated the influence of the composition and the optical purity of each component on the formation of stereocomplexes. Also, the nanofibers of stereochemical PLA and their blends were prepared by electrospinning method. The properties of the obtained fibers were analyzed by differential scanning calorimetry and scanning electron microscopy. The results showed that a degree of stereocomplex was controlled by change of optical purity of each component. The enzymatic degradation of the fibers were strongly dependent on the stereocomplex.
The global regulator, GacS (global antibiotic and cyanide sensor kinase), was required for the increased resistance to hydrogen peroxide occurring as cultures of the rhizobacterium, P. chlororaphis O6, matured. Specific stationary-phase peroxidase and catalase isozymes were absent in the GacS mutant, whereas a manganese-superoxide dismutase isozyme was expressed earlier and to a great extent than wild type. In the wild type cell, transcript accumulation of rpoS was higher in late logarithmic-phase cells than cells from mid logarithmic- or stationary-phase. Transcripts from rpoS in the GacS mutant were reduced in each of these growth phases compared to the wild type expression. The down stream sequence from rpoS lacked sequences encoding a small RNA, rsmZ, found in other pseudomonads and implicated in control of genes activated by the GacS system. These findings suggest that GacS-mediated regulation of RpoS plays role in control of oxidative stress in P. chlororaphis O6 by as yet an unknown mechanism.
When plants are infected by plant pathogens, typical disease symptom termed lesion, appears in compatible interaction. Whereas, in incompatible interactions, only small speck of lesions are visible on the leaf surfaces. Hypersensitive response (HR) of plant which is the result of infection by incompatible pathogens, is a well known defense response inducing rapid cell death resulting in complete resistance. However, some rice mutants show spontaneous disease symptoms during the growth stages without interaction with pathogens. We investigated the spontaneous cell death mutant called Blast Lesion Mimic(BLM) generated by EMS mutation, on the relationship with the hypersensitive response as well as resistant characteristics. Accumulation of phenolic compounds were detected around the lesions as lesions develop on leaf surface. Activation of PR gene was detected before the lesion appeared, and that result indicates the defense-related response are started earlier than lesion formation. The BLM mutant showed resistant response to inoculation of Magnaporthe grisea KJ201 with which the wild type Hwacheong is totally susceptible. Informations on the formation of spontaneous lesions and detail analysis of lesion mimic mutants and related genes are very limited to date. It is really important to understand the phenomenon of the defense-related lesion formation for developing resistant cultivar for rice blast pathogens
Mechanical transmission of barley seedlings with barley yellow mosaic virus (BaYMV) is generally inefficient and is the major constraint for testing cultivar resistance to the virus. To explore mechanical transmission, BaYMV-infected barley plants were grown at different conditions and used as inoculum sources to seedlings of susceptible barley cultivar Baegdong. Extracts prepared from BaYMV-infected Baegdong plants at 47, 53, 74, and 90 days after symptom appearance (DASA) and grown at 10 and $12^{\circ}C$ gave 10, 30, 68 and 76% infection, respectively on inoculated susceptible barley cv. Baegdong seedlings. While Jinyangbori, another susceptible cultivar obtained 95% infection rate inoculated with extracts from 90 DASA disease source and grown at $10/12^{\circ}C$. However, low infection rates were obtained when the virus sources were grown in a greenhouse at $15-18^{\circ}C$. Our results indicate that longer incubation period and lower temperature are required for virus accumulation and stability.
Clubroot disease is one of the most wide-spread and devastating diseases in the cultivation of Chinese cabbage. To develop a protein marker for resistance to clubroot disease in Chinese cabbage, a comparative proteome analysis was performed between a sensitive line, 94SK, and a resistant line, CR Shinki DH. Three proteins of two fold or higher accumulation that are specific to each line were found 3 days after innoculation of the Plasmodiphora brassicae. They are glutamine synthetase, malate dehydrogenase/oxidoreductase and fructose-bisphosphate aldolase in the 94SK and actin, phosphoglycerate kinase, and Cu/Zn superoxide dismutase in the CR Shinki line. From the comparison of the synthesized proteins in the 94SK and the CR Shinki, CR Shinki was found to produce more ATP-binding protein for the ABC transporter while 94SK showed a higher level of pathogenesis-related protein 1 production. All of these proteomic variations may lead to the development of molecular markers to accelerate the breeding process.

This study was performed to develop an LRFD (Load Resistance Factored Design) Code for Domestic Larch. To accomplish his, we evaluated bending, compression, tension and shear strength. The results of the strength evaluation were utilized to verify the distribution and code conversion. For bending, tension and compressive strength, the Weibull distribution was well-fitted, but for shear strength we observed a normal distribution. For evaluating the bending and compressive strength, a full-sized specimen was used. A small clear specimen was used to test tension and shear strength. Compressive strength in particular was found to be affected by tight knots, although there was little difference between grades. In the code conversion, the design value of the LRFD was larger than the existing allowable stress value in the Korean Building Code. However, the allowable stress in this study was about two times higher than the value listed in the Korean Building Code. This result induced the difference between the soft and hard conversions. For greater reliability, the accumulation of additional data is necessary and further studies should be performed
This study was conducted to investigate host and non-host disease resistances of kimchi cabbage plants to bacterial infection. Kimchi cabbage leaves responded differently to infections with a virulent strain of Xanthomonas campestris pv. campestris (Xcc) 8004 and two strains (85-10 and Bv5-4a.1) of non-host bacteria X. campestris pv. vesicatoria (Xcv). Non-host bacteria triggered a rapid tissue collapse of the leaves showing as brown coloration at the infected sites, highly increased ion leakage, lipid peroxidation and accumulation of UV-stimulated autofluorescence materials at the inoculated sites. During the observed interactions, bacterial proliferations within the leaf tissues were significantly different. Bacterial number of Xcc 8004 progressively increased within the inoculated leaf tissues over time, while growths of two non-host bacteria Xcv strains were distinctly limited. Expressions of pathogenesis-related genes, such as GST1, PR1, BGL2, VSP2, PR4 and LOX2, were differentially induced by host and non-host bacterial infections of X. campestris pathovars. These results indicated that rapid host cellular responses to the non-host bacterial infections may contribute to an array of defense reactions to the non-host bacterial invasion.
We displayed four types of Solanum nigrum metallothionein (SMT) for the first time on the surface of Saccharomyces cerevisiae using an α-agglutinin-based display system. The SMT genes were amplified by RT-PCR. The plasmid pYES2 was used to construct the expression vector. Transformed yeast strains were confirmed by PCR amplification and custom sequencing. Surface-expressed metallothioneins were indirectly indicated by the enhanced cadmium sorption capacity. Flame atomic absorption spectrophotometry was used to examine the concentration of Cd2+ in this study. The transformed yeast strains showed much higher resistance ability to Cd2+ compared with the control. Strikingly, their Cd2+ accumulation was almost twice as much as that of the wild-type yeast cells. Furthermore, surface-engineered yeast strains could effectively adsorb ultra-trace cadmium and accumulate Cd2+ under a wide range of pH levels, from 3 to 7, without disturbing the Cu2+ and Hg2+. Four types of surfaceengineered Saccharomyces cerevisiae strains were constructed and they could be used to purify Cd2+-contaminated water and adsorb ultra-trace cadmium effectively. The surface-engineered Saccharomyces cerevisiae strains would be useful tools for the bioremediation and biosorption of environmental cadmium contaminants.
A seismic damaged bridge may be hit again by a strong aftershock or another earthquake in a short interval before the repair work has been done. However, discussions about the impact of the unrepaired damages on the residual earthquake resistance of a steel bridge are very scarce at present. In this paper, nonlinear time-history analysis of a steel arch bridge was performed using multi-scale hybrid model. Two strong historical records of main shock-aftershock sequences were taken as the input ground motions during the dynamic analysis. The strain response, local deformation and the accumulation of plasticity of the bridge with and without unrepaired seismic damage were compared. Moreover, the effect of earthquake sequence on crack initiation caused by low-cycle fatigue of the steel bridge was investigated. The results show that seismic damage has little impact on the overall structural displacement response during the aftershock. The residual local deformation, strain response and the cumulative equivalent plastic strain are affected to some extent by the unrepaired damage. Low-cycle fatigue of the steel arch bridge is not induced by the earthquake sequences. Damage indexes of low-cycle fatigue predicted based on different theories are not exactly the same.
