Micropropagation of Juvenile and Mature Tree of Corylopsis coreana by Axillary Bud Culture (액아배양에 의한 유묘 및 성숙 히어리나무의 기내번식)

  • We have developed an in vitro micropropagation system via shoot formation from axillary buds using nodal segments of Corylopsis coreana. Explants from both juvenile tree (one-year-old greenhouse stock seedlings) and mature tree (ten-years-old tree in nursery) were compared with regard to propagation efficiency. Combined treatment of both BA and zeatin were effective on shoot proliferation since the best result was obtained on MS medium supplemented with 0.5∼3.0 mg/L zeatin and 0.2 mg/L BA. Generally, juvenile explants were better in both shoot proliferation and growth than mature explants. However, as the duration of in vitro culture was proceed to 6 months, explants from mature tree also produced three shoots per explant. Distinctive differences in rooting and adaptability to soil of shoots obtained from mother trees. Whereas shoots originated from juvenile explants rooted as high as 97%, those from adult explants showed 62% rooting. Similar result was also observed in soil acclimatization. The plantlets derived from juvenile plants survived 67%, while only 48% of those from adult trees survived. The results showed a possibility of the micropropagation of Corylopsis coreana through shoot formation from axillary buds. In addition, the advance of the research still remain to enhance the frequency of acclimatization of plantlets from mature trees for practical application.

Effects of Growth Regulators and Culture Environment on ex vitro Rooting and Acclimatization of Apple Rootstock in vitro Propagated (기내배양 사과 대목의 기외 삽목 시 발근과 순화에 미치는 배양조건 및 생장조절물질의 효과)

  • Growth of M.9 (Malus domestica Bark. cv) and M.26 (Malus domestica Bark. cv) of dwarf apple rootstock, cultured on MS agar medium in a vessel with ventilating stopper (VS) and then in vivo rooting and acclimatization under combined-treatment by some materials with IBA, were investigated. Concentration of $CO_2$ and ethylene in the vessel with VS was lower then in the vessel with non-VS. Change of temperature and humidity in the vessel with VS was repeated by light condition. Stomatal pares of tissue in the vessel with VS were immediately closed after plantlets were exposed to room humidity but those in the vessel with non-VS were opened after 20 minutes exposure to room humidity. Leaf area and chloroplast index of tissue in the vessel with VS was higher then in the vessel with non-VS. In vivo rooting ratio and acclimatization ratio of M.9 and M.26 was highest in 300mg/L IBA+3% sucrose dip-treatment among other combined- treatments.

The Effect of Plant Growth Regulators on Regeneration Rate during Tissue Culture of Hop (Humulus lupulus L.) (홉(Humulus lupulus L.) 조직배양 시 재분화율에 미치는 식물생장조절제의 영향)

  • Hops enhance beer flavor and bitterness, acting as a preservative. In recent years, the booming trend of craft beer has prompted the introduction of foreign hop varieties for cultivation and production in South Korea. This study focuses on developing efficient in vitro culture condition of the hop (Humulus lupulus L.) variety 'Cascade' for treatments of plant growth regulators, i.e. IAA and Cytokinin. Using Auxin IAA and Cytokinin 2iP, Zeatin, BAP, and TDZ on MS medium as plant growth regulators, the experiment involved removing three nodes from the shoot apex. In vitro hop culture showed the highest shoot proliferation rate when only IAA was added, with approximately 21% higher compared to the combination with Cytokinin. Notably, IAA 0.1 mg/L + BAP 1 mg/L resulted in a superior shoot proliferation rate of around 91%. IAA 0.1 mg/L + BAP 1 mg/L was advantageous for shoot elongation. Callus induction occurred with TDZ, while control or IAA-only conditions exhibited shoot and root growth. Cytokinin addition led to callus formation and increased weight. Assessing survival and soil adaptation during in vitro hop seedling acclimatization involved maintaining near 100% humidity initially, gradually reducing it over three weeks. When transferred outdoors, 9 out of 10 seedlings acclimated successfully, confirming a 90% acclimatization rate.

  • in order to develop a new technique for the removal of arsenic compoundsfrom acidified water, the removal of arsenic compounds by an acidophilic moss, Jungermannia vulcanicola Steph. was investigated in this study. The result of vial tests for arsenic removal is dependent on the biological activity of moss. The presence of phosphate inhibited the arsenic removal. And the acclimatization of moss by the media containing arsenic increased the its capability of arsenic remova.

Effect of Genotype and Explant on Somatic Embryogenesis and Acclimatization of Acanthopanax senticosus (가시오갈피의 수집종과 배양조직에 따른 체세포배발생 및 재분화 식물체의 순화)

  • Callus induction and embryogenesis were studied in three different genotypes of Acanthopanax senticosus, to develop a protocol for somatic embryogenesis and acclimatization. Young leaf, stem, node, petiole, peduncle, flower and root explants were collected from 3-year old trees of A. senticosus accessions (Korea, Russia and Japan). Callus was obtained from all cultured explants but showed the higher rate of callus formation in flower cultured. For the three A. senticosus accessions, callus was well formd on MS media containing 2mg/ l of 2,4-D and 2mg/ l of TDZ, 4mg/ l of 2,4-D and 1mg/ l of TDZ than other treatments. For three A. senticosus accessions, when callus transferred to MS medium with 2,4-D, embryogenic cell formed. For A. senticosus accessions Korea, embryogenic cells were obtained on callus induced from petiole, stem, node and root explants, and induction rate was lower than 3%. 200mg of embryogenic callus was transferred to MS free liquid medium and somatic embryos of heart stage were obtained after 45days of culture. When somatic embryo of germination stage were transferred to solid medium, most of the embryos were regenerated into plantlets on 1/4 MS medium. Normal plants with both shoots and roots were transferred to greenhouse soil and were successfully acclimatized.
