Seasonal Variation of Phenolic Component Contents in the Stems of Korean Acanthopanax senticosus (계절 변화에 따른 한국산 가시오가피 줄기의 페놀성 성분 함량 분석)

  • Objectives: The aim of this study was to compare the total phenolic content of Korean Acanthopanax senticosus stems harvested in different seasons and provide basic data for developing functional reinforcement products based on the optimal harvesting time. Methods: Each sample harvested in different seasons was extracted and concentrated twice for 2 hours using 70% ethyl alcohol. Phenolic compounds were analyzed using high-performance liquid chromatography for simultaneous multi-component analysis of 14 compounds, including syringaresinol and so on. Results: The results showed that the stem of Korean Acanthopanax senticosus harvested in winter (November 29th) (EAS-5) had the highest phenolic content of 1.038%. The stem of Korean Acanthopanax senticosus harvested in autumn (October 1st) (EAS-4) showed the second-highest phenolic content of 0.764%, followed by the stem of Korean Acanthopanax senticosus harvested in spring (February 2nd) (EAS-1) with a content of 0.390%. On the other hand, the stem of Korean Acanthopanax senticosus harvested in the summer (June 2nd) (EAS-3) showed the lowest content at 0.342%. In conclusion, the stem of Korean Acanthopanax senticosus harvested in winter (EAS-5) showed the highest phenolic compound content. Conclusions: Considering the extraction yield and the total phenolic content, as well as the concentrations of key functional components such as eleutheroside B, chlorogenic acid, and syringaresinol in the 70% ethyl alcohol extract of Korean Acanthopanax senticosus, it is suggested that the stems of Korean Acanthopanax senticosus harvested during the winter season are suitable for the development of novel materials with enhanced anti-obesity functionality.

Studies for Component Analysis and Antioxidant Effect, Antimicrobial Activity in Acanthopanax senticosus HARMS (가시오갈피 (Acanthopanax senticosus) 열매의 성분 분석과 항산화 효과 및 항균활성)

  • In the present studγ, we investigated the component analγsis, measurements of antioxidant activities and anti-microbial activities from fruit of Acanthopanax senticosus HARMS in order to detect the biological activities and develop novel functional resources. Different solvents extraction ratios were different according to different solvent extraction of Acanthopanax senticosus HARMS. Composition analysis we highly composed as 72.33%. The minerals of different organs were highly composed of potasium as 5951.3 mg/100g. The monosaccharldes are composed of the arabinose, xylose, mannose, galactose, glucose. Antioxidamt activity was measured in water extracts: $57.3\;{mu}/ml$, ethanol extracts $630.1\;{\mu}g/ml$, methanol extracts: $248.5\;{\mu}g/ml$, 71% ethanol extracts: $198.97\;{\mu}g/ml$, 75% methanol exracts: $96.77\;{\mu}g/ml$, chloroform extracts. $1194.83\;{\mu}g/ml$ at $IC_{50}$ value. The antimicrobial activities were observed in extracts from Acanthopanax senticosus HARMS against am negative bacteria and gram positive bacteria.

Investigation of Sound Absorption Ability of Acanthopanax senticosus Wastes

  • This study aims to investigate the sound absorption ability of Acanthopanax senticosus wastes as an eco-friendly sound-absorbing material. The sound absorption coefficient was examined with different heights of A. senticosus wastes filling (40, 60, 80, and 100 mm) in impedance tubes. The sound absorption peaks shifted to a lower frequency as the height of A. senticosus wastes inside the tubes increased. The sound absorption ability at filling heights of 80 and 100 mm was obtained as 0.3M and 0.5M grades, respectively, based on KS F 3503. The results suggest that A. senticosus wastes exhibit good sound absorption ability and can therefore be used as an efficient, eco-friendly sound-absorbing material.

Discrimination and Genetic Diversity of Acanthopanax senticosus Using RAPD Markers (RAPD마커를 이용한 가시오갈피의 기원판별 및 유전적 다양성)

  • Acanthopanx senticosus (Araliaceae) is a long-lived woody species primarily distributed throughout East Asia. This species is regarded as medically and ecologically important woody plants in Korea. To identify the variation of the RAPD patterns between domestic and foreign A. senticosus species, 22 random primers were applied to Korean A. senticosus and A. senticosus for. inermis, Chinese and Russian A. senticosus. Six primers of them could be used to discriminate the origins and 58 polymorphisms among 92 scored DNA fragments. Six bands are specific for Korean A. senticosus and A. senticosus for. inermis. Especially, three primers, OPD04, OPD11 and OPE10, were useful to differentiate between domestic and foreign Acanthopanax species. RAPD analysis was a useful method to discriminate among A. senticosus populations or accessions and Korean accessions are distinct genetically.

Suppression of Fat Accumulation and Improvement of Glucose Tolerance in High-fat Diet-induced Obese Mice Treated with an Acanthopanax senticosus Extract (고지방식이 유도 비만 마우스에서 가시오가피(Acanthopanax senticosus)의 지방축적 억제 및 인슐린 저항성 개선 작용)

  • Although it has recently been reported that Acanthopanax senticosus has a protective effect against the development of metabolic disorders such as diabetes and hyperlipidemia, its underlying molecular mechanisms have remained to be elucidated. In this study, we observed that treatment of an extract of A. senticosus (ASE) reduced body weight and adiposity and improved glucose tolerance in high-fat diet (HFD)-induced obese mice. HFD-induced weight gain in white adipose tissues and liver was also significantly reduced in ASE-treated HFD-fed mice, which was found to be contributed by suppression of mRNA expression of genes involved in fatty-acid uptake and fat synthesis in white adipose tissue. ASE treatment also attenuated M1 macrophage activation, increased regulatory T ($T_{reg}$) cell population, and modulated leptin/adiponectin profile that might alleviate chronic inflammation to protect against HFD-induced insulin resistance. Our findings suggest a therapeutic potential of ASE for prevention and treatment of type 2 diabetes without adverse effects such as weight gain and dyslipidemia.

Effects of Egg Yolk from Hens Fed with Acanthopanax senticosus Extract on Physical Endurance and Reproductive Parameters in Rats

  • Acanthopanax senticosus, a member of the Araliaceae used as an invigorant in traditional Korean medicine, is known to relieve physical fatigue. The principal objective of this study was to evaluate the effects of eggs laid by hens fed on Acanthopanax senticosus extracts on physical endurance and reproductive parameters. Male Sprague-Dawley rats at 4 wk of age were divided into 3 groups of 15 rats each: group A (14% egg yolk powder from hens fed with Acanthopanax senticosus extracts+86% AIN-93G), group B (14% plain egg yolk powder + 86% AIN-93G) and group C (7% fat source + 93% AIN-93G), and studied for 5 wk. The rats' physical endurance was measured via forced swimming tests. According to the results, 53.3% rats of group A swam for longer than 20 min, in group B and C, only 48.9% and 46.7% of the rats achieved this. In terms of reproductive parameters, sperm motility was significantly higher in group A than in groups B and C (p<0.05). However, no differences in sperm count were detected among the groups. Additionally, the serum testosterone levels of groups A and B were higher than that of group C. Our findings suggest that rats fed with egg yolk powder from hens fed on Acanthopanax senticosus extracts may improve athletic endurance and reproductive parameters in rats.

Pharmacognostical Studies on "Ga Si O Gal Pi" (가시오갈피의 생약학적 연구)

  • "Ga Si O Gal Pi" is one of the Korean crude drugs used mainly to cure arthritis, acute gastritis, and beriberi. With regard to the botanical origin of "Ga Si O Gal Pi", it has been considered to be Acanthopanax species of Araliaceae, but there was no pharmacognostical confirmation on it. To clarify the botanical origin of "Ga Si O Gal Pi", the anatomical characteristics of the branch of Acanthopanax species growing wild in Korea and Japan, A. chiisanensis, A. divaricatus, A. koreanum, A. rufinerve, A. senticosus, A. seoulense, A. sessiliflorus, A. sieboldiaum were studied. As a result, it was clarified that "Ga Si O Gal Pi" was the branch and rarely the bark of branch of Acanthopanax senticosus.

Enhancing Effect of Acanthopanax senticosus Extracts on Mouse Spleen and Macrophage Cells Activation (가시오가피 물 추출물에 의한 마우스 비장세포 및 대식세포 활성의 항진효과)

  • Acanthopanax senticosus is an herb that has been used as a traditional remedy and medicine source. Its anti-inflammatory and, anti-oxidative effects have been reported in previous studies. This study aimed to investigate the effect of Acanthopanax senticosus water extracts on mouse macrophage cell in vitro. Mouse splenocyte proliferation increased after application of Acanthopanax senticosus water extract supplement of 5, 10, 50, 100, 250, 500, $1,000{\mu}g/mL$ after 48 h pre-treatment with a mitogen (ConA or LPS). The production of cytokines secreted by LPS and non LPS stimulated macrophages was detected by ELISA assay using a cytokine kit. Cytokine production (IL-2, IFN-${\gamma}$, and TNF-${\alpha}$) increased after water extract supplementation. The result of this in vitro study, showed that splenocyte proliferation and cytokine production by activated peritoneal macrophages were increased after Acanthopanax senticosus water extract in the range of $500{\sim}1,000{\mu}L/mL$. Thus, it is suggested that supplementation with Acanthopanax senticosus water extracts may enhance immune function by regulating splenocyte proliferation and enhancing cytokine production by activated macrophage.

The Comparison of Phytochemical Components from the Berry of Acanthopanax Species (오가피속 식물 열매의 식물화학 성분 비교)

  • This study was carried out to obtain the basic information that can be used to index Acanthopanax in eleven species of China and Korea. The phytochemical components from the berry of Acanthopanax species, were measured by the HPLC analysis. Protocatechuic acid, eleutheroside B, eleutheroside E, scopolin, rutin, hyperoside, chiisanoside, oleanolic acid were found in ethanol extracts from the berry of Acanthopanax species. Total phenolic compound of Acanthopanax sessiliflorum f. chungbuensis berry (0.682%) was about seven times higher than those of Acanthopanax divaricatus f. distigmatis berry (0.091%). As a result, the order of the eleutheroside E content was 1) Acanthopanax sessiliflorum f. chungbuensis (0.554%), 2) A. divaricatus var. albeofructus (0.501%), 3) A. divaricatus f. flavi-flos (0.452%). And also, the order of the chiisanoside content was 1) Acanthopanax senticosus var. subinermis (8.434%), 2) A. seoulense (0.94%), 3) A. divaricatus f. flavi-flos (0.798%).
