Isolation of the Inositol Phosphoceramide Synthase Gene (AUR1) from Stress-Tolerant Yeast Pichia kudriavzevii

  • This study is the first report of the entire nucleotide sequence of an inositol phosphoceramide synthase gene from the stress-tolerant yeast Pichia kudriavzevii (PkAUR1). Sequence analysis revealed an open reading frame that spans 1,443 bp and encodes a 480-amino-acid-residue protein with the highest sequence similarity (41.7%) to Aur1 from Spathaspora passalidarum. A phenotypic assay with transformed S. cerevisiae and P. kudriavzevii indicated that two amino acid residues, Phe166 and Gly249, play crucial roles in the resistance to aureobasidin A, which is consistent with previous reports for other fungal Aur1s.   • We present [$Fe\;{\small{II}}$] ${\lambda}1.257{\mu}m$ spectra toward the interacting binary UY Aur with 0".14 angular resolution, obtained with the Near infrared Integral Field Spectrograph (NIFS) combined with the adaptive optics system Altair of the GEMINI observatory. In the [$Fe\;{\small{II}}$] emission, UY Aur A (primary) is brighter than UY Aur B (secondary). The blueshifted and redshifted emission between the primary and secondary show a complicated structure. The radial velocities of the [$Fe\;{\small{II}}$] emission features are similar for UY Aur A and B: ${\sim}-100km\;s^{-1}$ and ${\sim}+130km\;s^{-1}$ for the blueshifted and redshifted components, respectively. Considering the morphologies of the [$Fe\;{\small{II}}$] emissions and bipolar outflow context, we concluded that UY Aur A drives fast and widely opening outflows with an opening angle of ${\sim}90^{\circ}$ while UY Aur B has micro collimated jets.

  • The BVR CCD full light curves of ZZ Aur were obtained as one of the near-contact binary observing programs at the Sobaek Mt. Observatory during Feb. 2000 - Feb. 2001. From the present observations, three primary and seven secondary times of minimum light were obtained. The photometric solutions have been found using the Wilson and Divenny model.

  • 공생별 AG Dra와 UV Aur에 대해 지난 10여년간 보현산천문대 1.8m망원경과 고분산 에셀 분광기 BOES(BOao Echelle Spectrograph)로 분광관측을 수행해 왔다. 최근 2017년 11월 - 2018년 6월 관측에서, AG Dra의 Fe II 방출선과 UV Aur $H{\alpha}$ 방출선이 예년과 다른 변화 모습을 보이고 있음을 찾아내었다. 이에 대한 변화 원인을 공전주기, 밝기 변화 또는 다른 이유와 연관지어 설명하고자 한다.

  • We carried out near-IR photometric and optical spectroscopic observations of V582 Aur, which is a FU Orionis type object, to investigate any periodic and/or aperiodic variations. We obtained light curves on the scale of a night and a year, in J, H and Ks bands with KASINICS (KASI Near Infrared Camera System) attached to the BOAO (Bohyun-san Optical Astronomy Observatory) 1.8-m reflector in Youngcheon, South Korea and examined photometric variations on the two time scales. So far we have not found any periodic brightness variations on the scale of a night. On the other hand, we have found that there seems to be a periodic brightness variation with a period of approximately 45 days. In addition, high-resolution optical spectroscopic observations of V582 Aur were performed from February 2013 to May 2014 with the high-resolution echelle spectrograph BOES attached to the BOAO 1.8-m reflector. We analyzed several spectral lines to understand the physical state of V582 Aur. The P Cyg profiles are clearly shown in the $H{\alpha}$ line and Na I D line.

Characterization of Xanthomonas axonopodis pv. glycines plasmids

  • To characterize plasmids in Xanthomonu axonopodis pv. glycines, we isolated plasmids pAG1 from the strain AG1 and pXAG81 and PXAG82 from the strain Bra, respectively, and sequenced three plasmids. The size of plasmids, pAG1, pXAG81, and pXAG82 was 15,149-base pairs (bp), 26,727-bp, and 1,496-bp, respectively Fifteen and twenty six possible open reading frames (ORFs) were present in pAG1 and pXAG81, respectively. Only one ORF homologous to a rep gene of Xylella fastidiosa was present in pXAG82. pAG1 contained genes homologous to avrBs3, tnpA, tnpR, repA, htrA, three parA genes, M.XmaI, R.XmaI, and six hypothetical proteins. pXAG81 contained genes homologous to avrBs3, tnpA, tnpR, repA, htrA, two parA genes, pemI, pemK, mobA, mobB, mobC, mobD, mobE, trwB, traF, traH, ISxac2, and eleven hypothetical proteins. Based on DNA sequence analysis, we presume that pXAG81 is a conjugal plasmid. Interestingly, we found 0.5-kb truncated avirulence gene similar to aurXacE3 on the right border of avrBs3 homolgs of pAG1 and pXAG81. Two hundred twenty five isolates were analyzed to find aurBS3 or tra gene homologs by Southern hybridization. The numbers of avrBs3 homolog varied from 3 in AG1 to 8 in AG166. Two hundred seventeen isolates appeared to can conjugative plasmids (pXAG81 type), and thirty eight isolates appeared to carry non-conjugative plamids (pAGl type). This indicated that aurBs3 gene homologs might be spread by conjugation in X. axonopodis pv. glycines.

Weakly Complementary Cycles in 3-Connected Multipartite Tournaments

  • The vertex set of a digraph D is denoted by V (D). A c-partite tournament is an orientation of a complete c-partite graph. A digraph D is called cycle complementary if there exist two vertex disjoint cycles $C_1$ and $C_2$ such that V(D) = $V(C_1)\;{\cup}\;V(C_2)$, and a multipartite tournament D is called weakly cycle complementary if there exist two vertex disjoint cycles $C_1$ and $C_2$ such that $V(C_1)\;{\cup}\;V(C_2)$ contains vertices of all partite sets of D. The problem of complementary cycles in 2-connected tournaments was completely solved by Reid [4] in 1985 and Z. Song [5] in 1993. They proved that every 2-connected tournament T on at least 8 vertices has complementary cycles of length t and ${\mid}V(T)\mid$ - t for all $3\;{\leq}\;t\;{\leq}\;{\mid}V(T)\mid/2$. Recently, Volkmann [8] proved that each regular multipartite tournament D of order ${\mid}V(D)\mid\;\geq\;8$ is cycle complementary. In this article, we analyze multipartite tournaments that are weakly cycle complementary. Especially, we will characterize all 3-connected c-partite tournaments with $c\;\geq\;3$ that are weakly cycle complementary.

Determination of Longitude and Latitude of Kongju National University Observatory (공주대학교 천문대의 경 ${\cdot}$ 위도 결정)

  • The longitude and latitude of the Kongju National University Observatory was determined by using TM-1A theodolite and GPS(model: 4000SSI, GPS 45). In the observation using theodolite TM-1A observed the meridian transit time(KST) and meridian altitude of the 2 stars, ${\alpha}$ Aur and ${\alpha}$ Boo. In the observation using GPS measured the longitude and latitude by receiving data of 6 GPS satellites. The longitude and latitude of the Kongju National University Observatory was determined to 127$^{\circ}$8'33'.16 and 36$^{\circ}$28'14'.20, respectively.

  • A long-term project(ten-year ; 1982-92) for the photoeletric observation in the UBV passbands of selected eclipsing binaries with P$\geq$10 days has initiated at Yonsei University Observatory using 40-cm and 61-cm reflectors. the instrumentatio used and the observation techniques and the reduction procedures applied to this investiagation are described. Out of 39 program stars, successful results have been obtained during our first two years for the 11 stars listed below, with the orbital period in days in the parenthesis, and the incomplete light curves of these stars are presented. SX Cas (36.57) AQ Cas (11.72) UU Cnc (96.71) RY Gem (9.30) V373 Cas (13.42) NY Cep (15.28) RX Cas (32.32) V396 Cas (15.28) ZZ Cnc (25.60) Zet Aur (982.2) Eps Aur (9885.) For the rest of the stars, the observations made in the first two years are not sufficient to attempt any meaning light curve construction ; some of the data are too fragmented and others show large scatter.

  • To construct an amylolytic industrial strain of Saccharomyces cerevisiae, the glucoamylase cDNA gene (GAl) from Aspergillus awamori was expressed under the control of the alcohol dehydrogenase gene promoter (ADC1p) and integrated into the chromosomes of industrial S. cerevisiae. An integrative cassette lacking bacterial ampicillin resistance gene but containing the GA1 gene, $\delta$ sequences of Ty1 retrotransposon as target sites for homologous recombination and S. cerevisiae aureobasidin A resistance gene (AUR1-C) as the selection marker was constructed to obtain a strain eligible for commercial use. Industrial S. cerevisiae transformed with this 15-integrative cassette efficiently secreted glucoamylase into the medium and grew on starch as the sole carbon source. The transformants were mitotically stable for 100 generations in nonselective medium.
