Discovery of an elliptical jellyfish galaxy with MUSE

  • We will present a discovery of an elliptical jellyfish galaxy in Abell 2670 (Sheen et al. 2017, ApJL, 840, L7). Our MUSE IFU spectra revealed a rotating gas disk in the center of the galaxy and long ionised gas tails emanating from the disk. Its one-sided tails and a tadpole-like morphology of star-forming blobs around the galaxy suggested that the galaxy is experiencing strong ram-pressure stripping in the cluster environment. Stellar kinematics with stellar absorption lines in the MUSE spectra demonstrated that the galaxy is an elliptical galaxy without any hint of a stellar disk. Then, the primary question would be the origin of the rich gas component in the elliptical galaxy. A plausible scenario is a wet merger with a gas-rich companion. In order to investigate star formation history of the system (the galaxy and star-forming blobs), we derived star-formation rate and metallicity from the MUSE spectra. Photometric UV-Optica-IR SED fitting was also performed using GALEX, SDSS, 2MASS and WISE data, to estimate dust and gas masses in the system. For a better understanding of star formation history and environmental effect of this galaxy, FIR/sub-mm follow-up observations are proposed.

Forskolin-Induced Stimulation of RGS2 mRNA in C6 Astrocytoma Cells

  • RGS is a negative regulator of G-protein signaling and can be identified by the presence of a conserved $120{sim}125$ amino acid motif, which is referred to as the RGS box. A number of RGSs are induced in response to a wide variety of stimuli. Increased levels of RGSs lead to significant decreases in GPCR responsiveness. To obtain further evidence of a role of RGS proteins in rat C6 astrocytoma cells, we first determined the expression profile of RGS-specific mRNA in C6 cells using reverse transcription-polymerase chain reaction (RT-PCR) with a poly dT18 primer and transcript-specific primers. We found that RGS2, RGS3, RGS6, RGS9, RGS10, RGS12, and RGS16 were differentially expressed in C6 astrocytoma cells. The highest expression rate was found for RGS3, followed by RGS16, RGS10 and RGS9, whereas the expression level for RGS2 was barely detectable. We next assessed whether forskolin regulated the expression of RGSs expressed in C6 astrocytoma cells. The present study found that forskolin dose-dependently stimulated the expression of RGS2 transcripts. This up-regulation of RGS2 gene was abrogated by H-89, potent and broad-spectrum protein kinase A (PKA) inhibitors. Actinomycin D completely inhibited the up-regulation of RGS2 gene induced by forskolin $(10{\mu}M)$, indicating that the regulation of RGS2 gene is controlled at the transcriptional level. In addition, forskolin did significantly activate transcriptional cAMP response element (CRE) in either HEK 293 cells or C6 cells and did not modulate the $NF-{\kappa}B$ and AP-l activity as measured by luciferase reporter gene assay. Finally, forskolin induced the expression of RGS2 mRNA in C6 astrocytoma cells, which depend on the PKA pathway and CRE transcriptional pathways.

Suppressive effects on the expression of cyclooxygenase-2 and inducible nitric oxide synthase by a natural sesquiterpenoid in lipopolysaccharide-stimulated mouse macrophage cells

  • Prostaglandins (PGs) and nitric oxide (NO) produced by inducible cyclooygenase (COX-2) and nitric oxide synthase (iNOS), respectively, have been implicated as important mediators in the process of inflammation and carcinogenesis. On this line, the potential COX-2 or iNOS inhibitors have been considered as anti-inflammatory and cancer chemopreventive agents. In our continuing efforts of searching for novel cancer chemopreventive agents from natural products, we isolated natural sesquiterpenoids as potential COX-2 and iNOS inhibitors in cultured lipopolysaccharide (LPS)-activated mouse macrophage RAW 264.7 cells. Alantolactone, a natural eudesmane-type sesquiterpenoid, exhibited a potent inhibition of COX-2 (IC50 = 0.4 $\mu\textrm{g}$/$m\ell$) and iNOS activity (IC50 = 0.08 $\mu\textrm{g}$/$m\ell$) in the assay system determined by PGE2 and NO accumulation, respectively. The inhibitory potential of alantolactone on the PGE2 and NO production was well coincided with the suppression of COX-2 and iNOS protein and mRNA expression in LPS-induced macrophages. Furthermore, alantolactone inhibited NF-kB but not AP-l binding activity on nuclear extracts evoked by LPS-stimulated macrophage cells, suggesting the possible involvement of NF-kB in the regulation of COX-2 and iNOS expression. In further study with COX-2-expressing human colon HT-29 cells, alantolactone inhibited the cell proliferation, down-regulated COX-2, and inhibited the ERK phosphorylation in the early time. These results suggest that a natural sesquiterpenoid alantolactone might be a potential lead candidate for further developing COX-2 or iNOS inhibitor possessing cancer chemopreventive or anti-inflammatory activity

Inulin stimulates NO synthesis via activation of PKC-$\alpha$ and protein tyrosine kinase, resulting in the activation of NF-$textsc{k}$B by IFN-ν-primed RAW 264.7 cells

  • Inulin, an active component of Chicorium intybus root, has been shown to stimulate the growth of bifidobacteria, and inhibit colon carcinogenesis. NO mediates a number of the host-defense functions of activated macrophages, including antimicrobial and tumoricidal activity. We examined the effect of inulin on the synthesis of NO in RAW 264.7 cells. Inulin alone had no effect, whereas inulin with IFN-ν synergistically increased the NO production and inducible NO synthase (iNOS) expression in RAW 264.7 cells. Synergy between IFN-ν and inulin was mainly dependent on inulin-induced TNF-${\alpha}$ secretion. Also, protein kinase C (PKC)-${\alpha}$ was involved in the inulin-induced NO production. Inulin-mediated NO production was inhibited by the protein tyrosine kinase (PTK) inhibitor, tyrphostin AG126. Since iNOS gene transcriptions have been shown to be under the control of the NF -$\kappa$B/Rel family of transcription factors, we assessed the effect of inulin on NF -$\kappa$B/Rel using an EMSA. Inulin produced strong induction of NF-$\kappa$B/Rel binding, whereas AP-l binding was slightly induced in RAW 264.7 cells. Inulin stimulated phosphorylation and degradation of I$\kappa$B-${\alpha}$. These results suggest that in IFN-ν-primed RAW 264.7 cells inulin might stimulate NO synthesis via activation of PKC-${\alpha}$ and PTK, resulting in the activation of NF-$\kappa$B.

Effect of the Hesperetin and Naringenin on $pp60^{v-src}$-induced $NF-{\kappa}B$ Activation ($pp60^{v-src}$에 의한 $NF-{\kappa}B$ 활성화에 대한 헤스페레틴과 나린제닌의 저해 효과)

  • The effects of hesperetin and naringenin on $NF-{\kappa}B$ activation were investigated in normal rat kidney cells transformed by temperature sensitive Rous Sarcoma Virus (tsNRK). The flavonoids, naringenin and hesperetin, significantly reduced v-Src-induced $NF-{\kappa}B$ activation as well as phosphorylation of Akt and GSK-3 in tsNRK cells, whereas these compounds did not effect on platelet-derived growth factor (PDGF)-induced $NF-{\kappa}B$ activation in $NIH3T3{\gamma}l$ cells. In addition, the DNA binding activity of SP-I was also reduced but that of AP-1 was not affected by the compounds. Our study suggests that Src-induced $NF-{\kappa}B$ activation could occur via Akt-GSK-3 pathway without $IkB{\alpha}$ degradation and that naringenin and hesperetin could be used in the treatment of cancer through the inhibition of $NF-{\kappa}B$ activation.

Micro-shear bond strength of resin-modified glass ionomer and resin-based adhesives to dentin (상아질 위치에 따른 접착성 수복재의 미세전단결합강도에 관한 연구)

  • 이 연구의 목적은 수종 상아질 접착 시스템과 복합 레진 및 resin-modified glass ionomer를 상아질 표면에 접착하여 재료 및 상아질내 위치에 따른 미세전단결합강도를 측정, 비교하는 것이다. 상아질 접착 시스템으로는, 3-step인 Scotchbond Multi-Purpose Plus, 2-step인 Single Bond와 자가 부식형 시스템인 Clearfil SE Bond를, 1-step인 Prompt L-Pop을 사용하였다. 이와 함께 hybrid type의 복합 레진인 Clearfil AP-X와 2250을 사용하였으며 resin-modified glass ionomer로는 Fuji II LC를 사용하였다. 상악 소구치를 치아의 근원심 중앙부를 절단하여 상아질면을 노출시켰다. 5개 실험군으로 분류하고 상아질면을 위치에 따라 치관부의 occlusal $\frac{1}{3}$, middle $\frac{1}{3}$, cervical $\frac{1}{3}$과 치근부로 구분지어 시편을 부착하였다. 미세전단결합강도측정는 Universal testing machine(EZ-test; Shimadzu, Japan)에서 측정하였다. Occlusal 1/3부위에서는 SE가 가장 높은 값을, SM과 SB간에는 유의차가 없었으며, PL, GI순으로, Middle 1/3부위에서는 $SM{\;}{\geq}{\;}SE{\;}{\geq}{\;}SB{\;}{\geq}{\;}PL{\;}{\geq}{\;}GI$순으로, cervical 1/3부위에서는 SM, SE, SB간에 유의차가 없었다. Root dentin에서는 SM이 가장 높은 값을 보였으며 SE, PL, GI간에 유의차가 없었다. SE만이 치관부 상아질에 비해 치근부에서 유의할만한 결합강도의 감소를 나타냈다(p<0.05). GI는 치관부 상아질에서는 다른 군에 비해 유의성 있게 낮은 결합강도를 보였으나 치근부에서는 SE, PL과 유의차가 없었다.

Bi-flavonoids are Superior to Mono-flavonoid in Inhibiting Amyloid-${\beta}$ Toxicity and Fibrillogenesis through Accumulating Nontoxic Oligomer-like Structures

  • Polymerization of monomeric amyloid-${\beta}$ peptides ($A{\beta}$) into soluble oligomers and insoluble fibrils is one of the major pathways triggering the pathogenesis of Alzheimer's disease (AD). Using small molecules to prevent the polymerization of $A{\beta}$ peptides can, therefore, be an effective therapeutic strategy for AD. In this study, we investigated the effects of mono- and bi-flavonoids on $A{\beta}42$ toxicity and fibrillogenesis and found that the bi-flavonoid, taiwaniaflavone (TF) effectively and specifically inhibits $A{\beta}$ toxicity and fibrillogenesis. Compared to TF, the mono-flavonoid apigenin (AP) is less effective and less specific. Our data showed that differential effects of the mono- and bi-flavonoids on $A{\beta}$ fibrillogenesis correlate with their varying cytoprotective efficacies. We also found that other bi-flavonoids, namely 2',8"-biapigenin, amentoflavone, and sumaflavone, can also effectively inhibit $A{\beta}$ toxicity and fibrillogenesis, implying that the participation of two mono-flavonoids in a single bi-flavonoid molecule enhanced their activity. Bi-flavonoids, while strongly inhibited $A{\beta}$ fibrillogenesis, accumulated nontoxic $A{\beta}$ oligomeric structures, suggesting that these are off-pathway-oligomers. Moreover, TF abrogated the toxicity of preformed $A{\beta}$ oligomers and fibrils, indicating that TF and other bi-flavonoids may also reduce the toxicity of toxic $A{\beta}$ species. Altogether, our data clearly show that bi-flavonoids, possibly due to the possession of two $A{\beta}$ binders separated by an appropriate size linker, are likely to be promising therapeutics to suppress $A{\beta}$ toxicity.

Convergence Behavior Analysis of The Maximally Polyphase Decomposed SAP Adaptive Filter (최대 다위상 분해 부밴드 인접투사 적응필터의 수렴거동 해석)

  • Applying the maximally polyphase decomposition and noble identity to the adaptive filter in subband structure, the conventional fullband affine projection algorithm is translated to the subband affine projection (SAP) algorithm. The Maximally polyphase decomposed SAP (MPDSAP) algorithm is a special version of the SAP algorithm, and its adaptive sub-filters have unity projection dimension. The weight updating formular of the MPDSAP is similar to that of the NLMS algorithm, so it may be more proper algorithm than other AP-type algorithms for many practical applications. This paper presents a new statistical analysis of the MPDSAP algorithm. The analytical model is derived for autoregressive (AR) inputs and the nonunity adaptive gain in the subband structure with the orthonormal analysis filters (OAF), The pre-whitening by the OAF allows the derivation of a simple-analytical model for the MPDSAP with the AR inputs and the nonunity adaptive gain.

Case study of application on pelvic manipulation which low back pain patient in unilateral weight bearing due to pelvic imbalance (골반 불균형에 의한 편측체중지지 요통환자의 골반도수교정 적용사례)

  • Purpose: to recognized that influence of decrease low back pain, change pelvic structure and balance control on unilateral weight bearing after application on pelvic manipulation which low back pain patient in unilateral weight bearing due to pelvic imbalance. Methods: The patient with low back pain in unilateral weight bearing due to pelvic imbalance was 39year female. one subject received intervention of pelvic manipulation on sidelying position and reaching exercise on sitting position which during 2 weak at the 3 time per a weak, each 30 minutes. outcomes measured were Facia l Action Coding System(FACS), Radiograph(Lumbar-Spine Anteroposterior AP.), Pressure Scan. Results: The results of this study were summarized below : 1. FACS score were Pre: min.4 - max.6 and Post: min.2 - max.4. 2. Radiograph measured Ilium width were Pre: Lt.14cm, Rt.12.7cm and Post: Lt.13.4cm, Rt.13cm which discrepancy of Ilium height were Pre: 1cm and Post: 0.2cm. 3. Pressure scan measured Pre: Lt. 36.8%, Rt.40.2% and Post: Lt.41.3%, Rt.36.2%. Conclusion: Pelvic manipulation applied a patient with low back pain in unilateral weight bearing due to pelvic imbalance suggest that can decrease low back pain, change pelvic structure and balance control on unilateral weight bearing.

Effect of Addition of Pentaerythritol Triacrylate and Silane Coupling Agents on the Properties of Waterborne Polyurethane (Pentaerythritol Triacrylate와 실란커플링제의 첨가가 수분산 폴리우레탄의 물성에 미치는 영향)

  • Acrylic terminated polyurethane prepolymers were synthesized by capping the NCO groups of polyurethane prepolymers, prepared from isophorone diisocyanate (IPDI), polycarbonate diol (PCD) and dimethylol propionic acid (DMPA), with pentaerythritol triacrylate (PETA). Subsequently, silylated acrylic terminated prepolymers were prepared by capping the NCO groups of acrylic terminated polyurethane prepolymers with different types of silane coupling agents, glycidoxypropyl trimethoxysilane (GPTMS) or aminopropyl triethoxysilane (APS). The average particle size of pure waterborne polyurethane solution, measured by the dynamic light scattering method, was increased from 14.3 nm to 208.6 nm by adding PETA and APS. Also, the coating film of silylated acrylic terminated waterborne polyurethane showed better abrasion resistance and pencil hardness than that of pure waterborne polyurethane.
