L-trans-pyrrolidine-2,4-dicarboxylate (PDC) induces Excitotoxic and Oxidative Neuronal Death in Cultured Cortical Neurons

  • L-trans-pyrrolidine-2,4-dicarboxylate (PDC) is a potent inhibitor of glutamate transporters. In our current study, we investigated whether the neuronal death induced by PDC involves mechanisms other than excitotoxicity in mixed mouse cortical cultures. Cortical cultures at 13-14 days in vitro were used and cell death was assessed by measuring the lactate dehydrogenase efflux into bathing media. Glutamate and PDC both induced neuronal death in a concentration-dependent manner but the neurotoxic effects of glutamate were found to be more potent than those of PDC. Treatment with 10, 100 and 200 ${\mu}$M PDC equally potentiated 50 ${\mu}$M glutamate-induced neuronal death. The neuronal death induced by 75 ${\mu}$M glutamate was almost abolished by treatment with the NMDA antagonists, MK-801 and AP-5, but was unaffected by NBQX (an AMPA antagonist), trolox (antioxidant), BDNF or ZVAD-FMK (a pan-caspase inhibitor). However, the neuronal death induced by 200 ${\mu}$M PDC was partially but significantly attenuated by single treatments with MK-801, AP-5, trolox, BDNF or ZVAD-FMK but not NBQX. Combined treatments with MK-801 plus trolox, MK-801 plus ZVAD-FMK or MK-801 plus BDNF almost abolished neuronal death, whereas combined treatments with trolox plus ZVADFMK, trolox plus BDNF or ZVAD-FMK plus BDNF did not enhance the inhibitory action of any single treatment with these drugs. These results demonstrate that the neuronal death induced by PDC involves not only in the excitotoxicity induced by the accumulation of glutamate but also the oxidative stress induced by free radical generation. This suggests that apoptotic neuronal death plays a role in PDCinduced oxidative neuronal injury.

Search of Beacon in Low Power Wireless Interface (저전력 무선접속에서 비콘 탐색)

  • In IEEE 802.11b wireless network, stations synchronize themselves to the beacons periodically sent by the access point(AP) when they are running in low power mode. In case of missing beacon due to noise or traffic from neighboring wireless network stations must be awake until they get the next beacon, which causes energy consumption in stations. In this paper, we propose a scheme searching next beacon consuming little energy. The problems of missing beacon in low power mode of IEEE 802.11b wireless interface are described and a new method to reduce energy consumption is proposed. The proposed method is simulated with the network simulator, ns2, and compared with the low power mode of the IEEE 802.11b. The result measured in terms of station's wakeup time shows some enhancement in energy consumption when some errors occur in receiving frames.

ON LOCAL SPECTRAL PROPERTIES OF RIESZ OPERATORS

  • In this paper we show that if T ∈ L(X) and S ∈ L(X) is a Riesz operator commuting with T and XS(F) ∈ Lat(S), where F = {0} or F ⊆ ℂ ⧵ {0} is closed then T|XS(F) and T|XT(F) + S|XS(F) share the local spectral properties such as SVEP, Dunford's property (C), Bishop's property (𝛽), decomopsition property (𝛿) and decomposability. As a corollary, if T ∈ L(X) and Q ∈ L(X) is a quasinilpotent operator commuting with T then T is Riesz if and only if T + Q is Riesz. We also study some spectral properties of Riesz operators acting on Banach spaces. We show that if T, S ∈ L(X) such that TS = ST, and Y ∈ Lat(S) is a hyperinvarinat subspace of X for which 𝜎(S|Y ) = {0} then 𝜎*(T|Y + S|Y ) = 𝜎*(T|Y ) for 𝜎* ∈ {𝜎, 𝜎loc, 𝜎sur, 𝜎ap}. Finally, we show that if T ∈ L(X) and S ∈ L(Y ) on the Banach spaces X and Y and T is similar to S then T is Riesz if and only if S is Riesz.

Numerical Simulations of Dynamic Response of Cased Reactive System Subject to Bullet Impact (총탄 충격이 가해진 반응 시스템의 파괴 거동에 관한 수치적 연구)

  • Safety of reactive systems is one of the most important research areas in the field of weapon development. A NoGo response or at least a low-order explosion should be ensured to prevent unexpected accidents when the reactive system is impacted by high-velocity projectile. We investigated the shock-induced detonation of cased reactive systems subject to a normal projectile impact to the cylindrical surface based on two-dimensional hydrodynamic simulations using the I&G chemical rate law. Two types of energetic materials, namely LX-17 and AP-based solid propellant, were considered to compare the dynamic responses of the reactive system when subjected to the threshold impact velocity. It was found that shock-to-detonation transition phenomena occurred in the cased LX-17, whereas no full reaction occurred in the propellant.

Sensitivity Analysis of dVm/dtMax_repol to Ion Channel Conductance for Prediction of Torsades de Pointes Risk (다형 심실빈맥의 예측을 위한 dVm/dtMax_repol의 이온채널 전도도에 대한 민감도 분석)

  • Early afterdepolarization (EAD), a significant cause of fatal ventricular arrhythmias including Torsade de Pointes (TdP) in long QT syndromes, is a depolarizing afterpotential at the plateau or repolarization phase in action potential (AP) profile early before completing one pace. AP duration prolongation is related to EAD but is not necessarily accounted for EAD. Several computational studies suggested EAD can form from an abnormality in the late plateau and/or repolarization phase of AP shape. In this sense, we hypothesized the slope during repolarization has the characteristics to predict TdP risk, mainly focusing on the maximum slope during repolarization (dVm/dtmax_repol). This study aimed to predict the sensitivity of dVm/dtmax_repol to ion channel conductances as a TdP risk metric through a population simulation considering multiple effects of simultaneous reduction in six ion channel conductances of gNaL, gKr, gKs, gto, gK1, and gCaL. Additionally, we verified the availability of dVm/dtmax_repol for TdP risk prediction through the correlation analysis with qNet, the representative TdP metric. We performed the population simulations based on the methodology of Gemmel et al. using the human ventricular myocyte model of Dutta et al. Among the sixion channel conductances, dVm/dtmax_repol and qNet responded most sensitively to the change in gKr, followed by gNaL. Furthermore, dVm/dtmax_repol showed a statistically significant high negative correlation with qNet. The dVm/dtmax_repol values were significantly different according to three TdP risk levels of high, intermediate, and low by qNet (p<0.001). In conclusion, we suggested dVm/dtmax_repol as a new biomarker metric for TdP risk assessment.

Effect of $K^+-channel$ Blockers on the $A_1-adenosine$ Receptor-Coupled Regulation of Electrically-Evoked Norepinephrine Release in the Rat Hippocampus (흰쥐 해마에서 Norepinephrine 유리를 조절하는 $A_1-adenosine$ 수용체의 역할에 미치는 $K^+$ 통로 차단제의 영향)

  • Since it has been reported that the depolarization-induced NE release is inhibited by activation of presynaptic $A_1-adenosine$ heteroreceptor in hippocampus, a large body of experimental data on the post-receptor mechanism of this process has been accumulated. But, the post-receptor mechanism of presynaptic $A_1-adenosine$ receptor on the NE release has not been clearly elucidated yet. Therefore, it was attempted to clarify the participation of $K^+-channel$ in the post-receptor mechanisms of the $A_1-adenosine$ receptor-mediated control of NE release in this study. Slices from rat hippocampus were equilibrated with $^3H-norepinephrine$ and the release of the labelled products was evoked by electrical stimulation (3 Hz, 5 $VCm^{-1}$, 2 ms, rectangular pulses), and the influence of various agents on the evoked tritium-outflow was investigated. Adenosine, in concentrations ranging from $1{\sim}30\;{\mu}M$, decreased the NE release in a dose-dependent manner, without affecting the basal rate of release. 4AP $(1{\sim}30{\mu}M)$, a specific A-type $K^+-channel$ blocker, increased the evoked NE release in a dose-related fashion, and the basal rate of release is increased by 10 and $30{\mu}M$. TEA $(1{\sim}10{\mu}\;M)$, a nonspecific $K^+-channel$ blocker, increased the evoked NE release in a dose-dependent manner without affecting basal release. The adenosine effects were significantly inhibites by 3 ${\mu}M$ 4AP and 10 mM TEA treatment. 4AP $(30{\mu}M)-$ and TEA (10 mM)-induced increments of evoked NE release were completely abolished in $Ca^{++} free, but these were recoverd in low $Ca^{++} medium. And the effects of $K^+-channel$ blockers in low $Ca^{++} medium were inhibites and abolishes by $Mg^{++} (4 mM) adding and TTX $(0.3{\mu}M)$ adding medium, respectively. These results suggest that the decrement of the evoked NE-release by $A_1-adenosine$ receptor is mediated by 4AP and TEA sensitive $K^+-channel$.

The Quality and Antioxidant Properties of Cookies containing Aronia Powder (아로니아 분말을 첨가한 쿠키의 품질특성 및 항산화 활성)

  • This study investigated the quality characteristics of cookies containing Aronia powder, which is well known for its various functions and biological activity. In this study, we assessed the quality characteristics and antioxidant activity of cookies containing various concentrations (0, 2, 4, 6 and 8%) of Aronia powder(AP). To analyze quality characteristics, pH of dough, moisture content, color(L, a, b), hardness, and sensory properties were measured. Qualities, such as moisture content, redness and yellowness and hardness increased significantly (p<0.001). whereas pH and lightness of the cookies significantly decreased (p<0.001). The antioxidant activity measured by DPPH and ABTS radical scavenging activities were significantly higher than the control, and it proportionally increased as the amount of AP increased (p<0.001). The results of sensory properties showed that cookies made with 2% and 4% AP did not differ significantly from the control in color, taste, and overall quality. Texture scores for the 2% and 4% AP groups ranked significantly higher than those of the other groups. Taken together, the results of this study suggest that AP is a beneficial ingredient for increasing the consumer acceptability and functionality of cookies. The quality characteristics of the 4% added samples exhibited significantly similar or higher values as compared to those of the controls, raising the possibility of developments in health-functional cookies. The results of the sensory evaluation produced very significant values for color, appearance, texture, and overall quality of instrumental analysis.

Cloning And Characterization of Pathogen-Inducible EREBP-Like Transcription Factor(CaNR19) From Hot Pepper (Capsicum annuum L.)

  • An EREBP/AP2-type transcription factor (CaPFl) was isolated by DDRT-PCR following inoculation of soybean pustule pathogen Xanthomonas axonopodis pv. glycines Bra which induces HR on pepper leaves. Genomic Southern blot analysis revealed that the CaPFl gene is present as a single copy within the hot pepper genome. The deduced amino acid sequence of CaPFl has two potential nuclear localization signals, a possible acidic activation domain, and an EREBP/AP2 motif that could bind to a conserved cis- element present in promoter region of many stress-induced genes. The mRNA level of CaPFl was induced by both biotic and abiotic stresses. We observed higher-level transcripts in resistance-induced pepper tissues than diseased tissues. Expression of CaPFl is also induced upon various abiotic stresses including ethephon, MeJA, cold stress, drought stress and salt stress treatments. To study the role of CPFI in plant, transgenic Arabidopsis and tobacco plants which express higher level of pepper CaPFl were generated. Global gene expression analysis of transgenic Arabidopsis by cDNA microarray indicated that expression of CaPFl in transgenic plants affect the expression of quite a few GCC box and DRE/CRT box-containing genes. Furthermore, the transgenic Arabidopsis and tobacco plant, expressing CaPFl showed tolerance against freezing temperature and enhanced resistance to Pseudomonas syrnigae pv. tabaci. Taken together, these results indicated that CaPFl is a novel EREBP/AP2 transcription factor in hot pepper plant and it may has a significant role(s) in regulation of biotic and abiotic stresses in plant.

Ethanol Extract of Oenanthe javanica Modulates Inflammatory Response by Inhibiting NF-${\kappa}B$ Mediated Cyclooxygenase-2 Expression in RAW 264.7 Macrophage

  • Effect of Oenanthe javanica ethanol extract (OJE) on nuclear factor-${\kappa}B$ (NF-${\kappa}B$)-mediated inflammatory reaction in RAW 264.7 macrophage cells was investigated. The OJE dose-dependently inhibited secretions of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and prostaglandins $E_2\;(PGE_2)$ from lipopolysaccharide (LPS)-stimulated RAW 264.7 cells and blocked LPS-induced expression of cyclooxygenase-2. To clarify mechanistic basis for its inhibitions of NF-${\kappa}B$ and activator protein-1 (AP-1) activations, effects of OJE on activations of NF-${\kappa}B$ and AP-1 genes by luciferase reporter activity were examined. The LPS-stimulated activations of NF-${\kappa}B$ and AP-1 were significantly blocked by 400 and $600\;{\mu$}g/mL of OJE, implicating that OJE might regulate gene expression through more than one signaling pathway. Cytosolic degradation of I-${\kappa}B{\alpha}$ was inhibited by OJE dose-dependently, indicating that the nuclear translocation of p65 was inhibited by OJE. These findings suggest that the inhibition of LPS-stimulated COX-2 expression by OJE is due to its inhibition of NF-${\kappa}B$ activation by blocking I-${\kappa}B{\alpha}$ degradation, which may be mechanistic basis of anti-inflammatory effects of OJE.
