Effects of Crude Protein Levels in Diets Containing MKP on Water Quality and the Growth of Japanese Eels Anguilla japonica and Leafy Vegetables in a Hybrid BFT-Aquaponic System (일인산칼륨(MKP)이 함유된 사료 내 단백질 수준이 Hybrid BFT 아쿠아포닉스(HBFT-AP)의 뱀장어(Anguilla japonica) 및 엽채류의 생산성과 수질변화에 미치는 영향)

  • This study investigated the effects of crude protein levels in diets containing monobasic potassium phosphate (MKP) on water quality and the growth of Japanese eels Anguilla japonica and leafy vegetables in a Hybrid BFT-Aquaponics (HBFT-AP) system. The first experiment (EXP1) was designed to verify the effects of the feed itself on leafy vegetable productivity using two diets (CP48 and CP30) with MKP and one commercial eel diet (COM58). The second experiment (EXP2) examined the effects of the three diets on productivity of the fish and leafy vegetables in the HBFT-AP for 6 weeks. After the 6 week feeding trial, the weight gain, feed efficiency, specific growth rate and protein efficiency ratio of fish fed COM58 and CP48 were higher than those of fish fed CP30 (P<0.05) and the growth of the four leafy vegetables was the highest with fish fed CP48. Water quality was measured six times per week using a portable water quality meter and reagent measurements and showed variance with time for TAN (0.01-0.09 mg/L), NO2-N (0.010-0.064 mg/L), NO3-N (5.52-27.15 mg/L), PO4-P (2.03-5.32 mg/L) and pH (7.86-6.15).

  • The effect of various packaging methods on Kimchi quality was investigated in order to develop the packaging techniques for preventing commercial Kimchi Products from ination and explosion due to fermentative gas evolved during storage and distribution. Kimchi was packaged in different methods; atmospheric (AP), check-valved (CV), double (DP) and vacuum (VP) packaging. The quality of Kimchi during storage at $10^{\circ}C$ was evaluated ill terms of gas composition, free volume, pH, titratable acidity, color index, lactic acid bacteria and sensory properties. $O_2$ concentration decreased to about 1% at $6{\sim}8$ days of optimal ripening time in all packages except DP, in which it remained around 20%. $CO_2$ concentration reached $70{\sim}90%$ after $6{\sim}8$ days in AP, CV and DP, while it stayed around 10% in DP. The free volume in AP had typical sigmoidal curve similar to $CO_2$ concentration changes. It remained almost constant in CV and DP, but started to increase at the late stage of storage In VP. There was no significant influence of packaging methods on the other quality attributes such as pH, titratable acidity, color index (L.b/a), lactic acid bacteria count and sensory evaluation score. As results, it could be proposed to employ CV and DP method as the effective packaging techniques for preventing commercial Kimchi products from inflation.

Identification and Functional Analysis of the putAP Genes Encoding Vibrio vulnificus Proline Dehydrogenase and Proline Permease

  • The pathogenic marine bacterium Vibrio vulnificus is the causative agent of food-borne diseases such as life-threatening septicemia. To better understand this organism's strategies to survive osmotic stress, a mutant that was more sensitive to high osmolarity was screened from a library of mutants constructed by a random transposon mutagenesis. By a transposon-tagging method, putAP genes encoding a proline dehydrogenase and a proline permease were identified and cloned from V. vulnificus. The amino acid sequences deduced from nucleotide sequences of putAP from V. vulnificus were 38 to $59\%$ similar to those of PutA and PutP reported from other Enterobacteriaceae. Functions of putAP genes were assessed by the construction of mutants, whose putAP genes were inactivated by allelic exchanges. When proline as the sole carbon or nitrogen source was used, the putA mutant was not able to grow to the substantial level, revealing the proline dehydrogenase is the only enzyme for metabolic conversion of proline into other amino acids. Although the growth rate of the putP mutant on proline as the sole carbon or nitrogen source was significantly reduced, the mutant still grew. This indicated that at least one more proline permease is produced by V. vulnificus. The putP mutant decreased approximately $2-log_10$ CFU/ml after a hyperosmotic challenge, while the parent strain decreased approximately $l-log_10$ CFU/ml. This result suggests that the gene product of putP contributes to the osmotic tolerance of V. vulnificus.

  • Objective: The semen quality of stallions including sperm motility is an important target of selection as it has a high level of individual variability. However, effects of the molecular architecture of the genome on the mechanisms of sperm formation and their preservation after thawing have been poorly investigated. Here, we conducted a genome-wide association study (GWAS) for the sperm motility of cryopreserved semen in stallions of various breeds. Methods: Semen samples were collected from the stallions of 23 horse breeds. The following semen characteristics were examined: progressive motility (PM), progressive motility after freezing (FPM), and the difference between PM and FPM. The respective DNA samples from these stallions were genotyped using Axiom Equine Genotyping Array. Results: We performed a GWAS search for single nucleotide polymorphism (SNP) markers and potential genes related to motility properties of frozen-thawed semen in the stallions of various breeds. As a result of the GWAS analysis, two SNP markers, rs1141327473 and rs1149048772, were identified that were associated with preservation of the frozen-thawed stallion sperm motility, the relevant putative candidate genes being NME/NM23 family member 8 (NME8), olfactory receptor family 2 subfamily AP member 1 (OR2AP1), and olfactory receptor family 6 subfamily C member 4 (OR6C4). Potential implications of effects of these genes on sperm motility are herein discussed. Conclusion: The GWAS results enabled us to localize novel SNPs and candidate genes for sperm motility in stallions. Implications of the study for horse breeding and genetics are a better understanding of genomic regions and candidate genes underlying stallion sperm quality, and improvement in horse reproduction and breeding techniques. The identified markers and genes for sperm cryotolerance and the respective genomic regions are promising candidates for further studying the biological processes in the formation and function of the stallion reproductive system.

  • Let H be a separable complex Hilbert space and L(H) be the *-algebra of all bounded linear operators on H. For $T \in L(H)$, we construct a pair of semi-positive definite operators $$ $\mid$T$\mid$_r = (T^*T)^{\frac{1}{2}} and $\mid$T$\mid$_l = (TT^*)^{\frac{1}{2}}. $$ An operator T is called a semi-hyponormal operator if $$ Q_T = $\mid$T$\mid$_r - $\mid$T$\mid$_l \geq 0. $$ In this paper, by using a technique introduced by Berberian [1], we show that the approximate point spectrum $\sigma_{ap}(T)$ of a semi-hyponomal operator T is empty.

Changes in Chemical Properties and Effect on Germination of Radish Seed from Aeration of Co-digestate Fertilizers (통합 혐기소화액별 폭기처리에 따른 화학적 성분 변화와 무의 발아효과)

  • This study aimed to determine the minimum period of aeration treatment of co-digestate to develop it as liquid fertilizer and the chemical changes that occur in the aerobic liquefying process. The co-digestates were divided into three types depending on their additives: swine slurry anaerobic digestate (SS AD), swine slurry 70% + cow slurry 30% anaerobic digestate (SS + CS AD), and swine slurry 70% + apple pomace 30% anaerobic digestate (SS + AP AD). The pH of all co-digestates increased rapidly after 3 days of aerobic treatment, but had slightly decreased in SS AD after 9 days and in SS + CS AD and SS + AP AD after 15 days. All co-digestates showed a strongly reduced pH between 27 and 36 days of aeration treatment. SS AD had lower pH value, dissolved oxygen (DO), NH4-N, and NO3-N content under aerobic conditions than other co-digestates. To assess the fully decomposed liquid fertilizer, a germination test was performed on the undiluted and diluted co-digestate using the liquid fertilizer germination index (LFGI) method. The relative germination ratio, relative root elongation, and germination index of SS AD were higher than those of the others. When the LFGI method was used for the germination test, all co-digestates showed an appropriate germination index of 70 after 60 days of aeration treatment. Thus, we suggest that the minimum period of aeration treatment for co-digestates might be 60 days to develop the fully decomposed liquid fertilizer.

Therapeutic Effect of Injection-Acupuncture with Bee-Venom (Apitoxin) in Cases of Canine Otitis Externa

  • This study was carried out to determine the therapeutic effect of injection-acupuncture (AP) with bee-venom (apitoxin) in cases of canine otitis externa (COE). Fifteen dogs with naturally-acquired otitis externa were used in this study. The dogs were divided into the following antibiotics group (control group), apitoxin group (experimental group A) and apitoxin combined with antibiotics group (experimental group B). All groups were treated by ear cleaning with normal saline once on day 1. The control group was treated with susceptible antibiotics, and experimental group A was given injection-AP with apitoxin $(100{\mu}g/head)$ at TH17 (Yi Feng), SI19 (Ting Gong), GB03 (Shang Guan) and TH03 (Zhong Zhu) bilaterally. Experimental group B was treated with susceptible antibiotics and injection-AP with apitoxin at the same acupoints as experimental group A. All the groups were treated 3 times/week for 2 weeks. The identity of the causative agents, the changes in the clinical signs, otoscopic findings, bacterial count in the auricular discharges, and total WBC counts and neutrophil/lymphocyte (N/L) ratio in the peripheral blood were investigated in all groups. In bacterial isolation, Staphylococcus spp. combined with Streptococcus spp. was detected higher than other agents. The bacterial cell count in experimental group A was significantly decreased at 2 weeks (p<0.01), and those in experimental group B was significantly decreased at 1 week (p<0.01) and 2 weeks (p<0.01) compared by those of control group, respectively. The changes of clinical score in experimental group B were significantly decreased at 2 weeks (p<0.01) compared by those of control group, but, those of experimental group A was similar to those of control group. The changes of total WBC counts and neutrophil/lymphocyte (N/L) ratio were no significant difference found. In conclusion, injection-AP with apitoxin is an effective treatment for COE and might be an alternative method for treating COE.

Effects of Caffeic Acid Methyl Ester from Mentha arvensis Linne var. piperascens on Function of E6 Oncoprotein of HPV 16 (박하에서 분리한 Caffeic Acid Methyl Ester가 자궁경부암 바이러스 발암단백질 E6의 기능에 미치는 영향)

  • Cervical cancer is one of the leading causes of female death. Viral oncoproteins E6 and E7 are selectively retained and expressed in carcinoma cells infected with HPV (Human pa pilloma virus) type 16 and cooperated in immotalization and transformation of primary keratinocyte. E6 and E7 oncoproteins interfere the functions of tumor suppressor proteins p53 and retinoblasoma protein (pRb), respectively. Among a lots of natural products, Mentha arvensis Linne var.piperascens have inhibitory effects on bindings between E6 oncoprotein and tumor suppressor p53, E3 ubiqutin- protein ligase (E6AP). HPV oncoprotein inhibitors from Mentha piperita L. were isolated by solvent partition and column chromatography (Silica gel, RP-18) and inhibitory compounds were finally purified by HPLC using an ELISA screening system based on binding between E6 and E6AP. The aim of this study is to identify the structure of inhibitory compounds and to investigate whether these compounds have inhibitory effects on functions of E6 oncoprotein. We investigated whether caffeic acid methyl ester (CAM) extracted from Mentha piperita L. could inhibit the function of E6 oncoprotein. CAM inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53 and also inhibited the proliferation of human cervical cancer cell lines (SiHa and CaSKi) in a dose response manner. These results suggest that CAM inhibited the function of E6 oncoprotein, suggesting that it can be used as a potential drug for the treatment of cervical cancers infected with HPV.

Evaluation of Scattered Rays of Jelly Type Shielding Body by L-spine AP using X-ray (L-Spine X-선 촬영에서의 Jelly type 차폐체의 산란선 차폐평가)

  • There have been continuous controversies on medical X-ray protection and numerous researchers have been trying to prevent unnecessary exposure to radiation. As X-ray passes through the patient and obtains an image, it creates scattered ray due to interactions such as photoelectric effect and Compton scattering with the subject. As a result, both medical radiation staff and patient are exposed to unnecessary radiation on areas other than the target area. In response, this study will be assuming a body of a female, radiating X-ray on the phantom under the conditions of lumbar spine AP test, and measuring scattered ray around breasts and thyroid glands. Then, The experiment results were as follows. After application of non-shielding material, the average of scattered ray was 0.88 mR in thyroid measurement, 3.34 mR, Lt Axillary 3.54 mR, and Rt Axillary 3.03 mR in mamonary measurement but, After application of shielding material, the average of scattered ray was 0.16 mR in thyroid measurement, 0.60 mR, Lt Axillary 0.64 mR, and Rt Axillary 0.54 mR in mamonary measurement showing average scattered ray protection effect of about 82%. This study suggested the manufacturing method of a Jelly-type shielding material, identified the possibilities of researches on mixing various substances with radiology field, and verified the usability of the Jelly-type shielding material as a substitute for existing protection tools.

Treatment of Canine Cervical and Lumbar Disc Disease by Injection-Acupuncture (개 경부 및 요부 디스크의 수침치료)

  • Two cases of canine intervertebral disc disease (IVDD) were treated twice a week with injection-acupuncture (injection-AP) using 0.1 ml/acupoint. In case 1, a paralysed dog with cervical intervertebral disc disease (C3-C4), was submitted to injection-AP with dexamethasone (1 mg/ml) at session 1-2 and thiamine (25 mg/ml) at session 3-5. Injected acupoints included GV-16, GB-20, BL-10, LU-7, LI-4 and SI-6. The acupoints GV-6, GV-20 and SP-6 were added at session 2-5. Trigger point (TP) therapy with 0.2 ml of 2% lidocaine was used in session 3-5 at TP in the infraspinatus and triceps muscles. Treatment was stopped when paralysis disappeared after 5 injection-AP treatments; there were no recurrent symptoms in the follow-up period of 5 months. In case 2, an ataxic dog with lumbar IVDD (L1-L2), was submitted to injection-AP with dexamethasone at session 1 and 2 and thiamine at session 3-4. Injected acupoints included GV-6 as the main point, ST-36, GB-30, ST-40, GB-34, ST-41 and BL-40. TP therapy with 0.2 ml of 2% lidocaine was used at TP in the ileocostorum lumborum and quadriceps muscles. Treatment was stopped when ataxia disappeared after 4 treatments; there were no recurrent symptoms In the fallow-up period of 5 months. Injection-AP using dexamethasone and thiamine, combined with TP therapy using lidocaine, effectively alleviate the symptoms of canine cervical and lumbar IVDD.
