Promotion of Aldehyde Oxidase Activities by Ethanol in Maize (Zea mays) Roots (옥수수 (Zea mays) 뿌리에서 에탄올에 의한 알데히드 산화효소의 활성 증가)

  • We observed that exogenously applied ethanol changed the activities of aldehyde oxidases (AO) in the primary roots of maize (Zea mays). The stimulatory effect of ethanol on the aldehyde oxidase activities was concentration dependent; the AO activities were slightly weaker with 0.2 - 0.4% ethanol and stronger with 0.8 - 1.0% ethanol than the level of control. The promotion of AO activities was not explained by the increased transcription of two AO genes in maize. In contrast, ethanol strongly increased the amount of AO proteins, indicating that ethanol enhanced AO activities by promoting the translation. Among three alcohols including ethanol, methanol and isopropanol, only ethanol promoted AO activities. These results suggested that enhancement of AO activities was specific to ethanol, whose level could be naturally increased when the plant roots drove fermentation to overcome low oxygen stresses.

Comparison of Chronic Disease Risk by Abdominal Obesity in Korean Adult Women - Using Data from the 2013~2014 Korea National Health and Nutrition Examination Survey - (한국 성인 여자에서 복부비만에 따른 만성질환 위험 비교 - 2013~2014년 국민건강영양조사 자료를 이용하여 -)

  • This study was conducted to investigate the risk of chronic disease as predicted by abdominal obesity in Korean adult females. Data on 2,738 adult females aged 40~64 yrs was obtained from the 2013~2014 Korean National Health Nutrition Examination Survey. The subjects were divided into normal (n=1,835), abdominal obesity (AO, n=73), and BMI-diagnosed abdominal obesity (BMI-AO, n=505) groups based on the NCEP-ATP III guidelines and by applying the KSSO definition regarding waist circumference. Triglyceride blood levels, fasting blood sugar levels, systolic blood pressure, and diastolic blood pressure were higher in the AO and BMI-AO groups than in the normal group. The HDL-cholesterol levels of subjects in the two abdominal groups were lower as compared to those of subjects in the normal group. The mean adequacy ratio was lower among subjects in the AO and BMI-AO groups than among those in the normal group. Moreover, the NAR and INQ scores of some micro-nutrients in both of the abdominal obesity groups were lower than those in the normal group. Both the AO and BMI-AO groups showed significantly higher risks of hypertriglycemia, hyperglycemia, hypertension, hypoHDL-cholesterolemia, and metabolic syndrome than were shown by the normal group. Notably, as compared to the normal group, the AO group showed higher risk of hypercholesterolemia, hyperLDL-cholesterolemia, hypertriglycemia, and metabolic syndrome than did the BMI-AO group. This finding suggests that it is essential to develop prevention programs including programs for those with abdominal obesity despite their having a BMI within the normal range.

Biological Characteristics of Recombinant Arthrobotrys oligospora Chitinase AO-801

  • Chitinase AO-801 is a hydrolase secreted by Arthrobotrys oligospora during nematode feeding, while its role remained elusive. This study analyzed the molecular characteristics of recombinant chitinase of Arthrobotrys oligospora (reAO-801). AO-801 belongs to the typical glycoside hydrolase 18 family with conserved chitinase sequence and tertiary structure of (α/β)8 triose-phosphate isomerase (TIM) barrel. The molecular weight of reAO-801 was 42 kDa. reAO-801 effectively degraded colloidal and powdered chitin, egg lysate, and stage I larval lysate of Caenorhabditis elegans. The activity of reAO-801 reached its peak at 40℃ and pH values between 4-7. Enzyme activity was inhibited by Zn2+, Ca2+, and Fe3+, whereas Mg2+ and K+ potentiated its activity. In addition, urea, sodium dodecyl sulfate, and 2-mercaptoethanol significantly inhibited enzyme activity. reAO-801 showed complete nematicidal activity against C. elegans stage I larvae. reAO-801 broke down the C. elegans egg shells, causing them to die or die prematurely by hatching the eggs. It also invoked degradation of Haemonchus contortus eggs, resulting in apparent changes in the morphological structure. This study demonstrated the cytotoxic effect of reAO-801, which laid the foundation for further dissecting the mechanism of nematode infestation by A. oligospora.

Hyperuricemic effects of Alpiniae Oxyphyllae Fructus extracts (고요산동물에서의 익지인의 요산저하 효과)

  • Objective : Hyperuricemia is a metabolic disease characterized by elevated blood uric acid levels, and its prevalence is rapidly increasing worldwide. Alpiniae Oxyphyllae Fructus (AO) belonging to Zingiberaceae is one of well-known traditional medicines in China and Korea, and has been used to treat intestinal disorders, urosis, diuresis, and chronic glomerulonephritis traditionally. However, the effect of AO has not been studied. In this study we investigated the anti-hyperuricemic effect of AO, and the mechanisms underlying the effect in potassium oxonate (PO)-induced hyperuricemic rats. Methods : To examine the anti-hyperuricemic effects of the AO extract, serum uric acid levels were analyzed in normal and PO-induced hyperuricemic rats. The mechanism underlying the effects of the AO extract on uric acid levels was studied through xanthine oxidase (XOD) activity test and uric acid uptake assay in vitro. The chemical finger printing of the AO extract was analyzed using HPLC-DAD. Results : The AO extract significantly reduced serum uric acid levels in normal as well as PO-induced hyperuricemic rats. It also significantly inhibited the uptake of uric acid in oocytyes and human embryonic kidney cells (HEK293) expressing urate transporter (URAT)1, but not XOD activity in vitro. The chemical finger printing analysis of the AO extract showed nootkatone as a main component. Conclusion : The AO extract exhibits anti-hyperuricemic effects, and these effect were accompanied by increasing excretion of uric acid in kidney. Therefore, the AO extract could be used for prevention or treatment of hyperuicemia and gout.

  • Age of Information (AoI) has been introduced in wireless networked control systems (WNCSs) to guarantee timely status updates. In addition, as the edge computing (EC) architecture has been deployed in NCS, EC close to sensors can be exploited to collect status updates from sensors and provide control decisions to actuators. However, when lots of sensors simultaneously deliver status updates, EC can be overloaded, which cannot satisfy the AoI requirement. To mitigate this problem, this paper uses actuators with computing capability that can directly receive the status updates from sensors and determine the control decision without the help of EC. To analyze the AoI of the actuation update via EC or directly using actuators, this paper developed an analytic model based on timing diagrams. Extensive simulation results are included to verify the analytic model and to show the AoI with various settings.

  • Fire and disaster broadcasting systems are divided into analog, digital, and network-based digital public address systems, and important specifications in network-based digital public address systems are low-latency audio, high sampling rate, and multi-channel input and output. In the past, it has been widely used to the AoE method for distinguishing based on the MAC address of the data link layer. However, this method has a problem of increasing complexity and cost. This proposal is an AoIP/UDP method, which allows communication to be easily distinguished by IP address without the need for a separate redundant network, so that the network can be freely used and configured, and cost can be reduced by reducing complexity. After implementing the AoIP/UDP method, the experimental results showed that the cost was improved with the equivalent performance with 2.66ms latency.

  • Adaptive Optics (AO) was first studied in the field of astronomy, and its applications have been extended to the field of laser, microscopy, bio, medical, and free space laser communication. AO modelling and simulation are required throughout the system development process. It is necessary not only for proper design but also for performance verification after the final system is built. In KASI, we are trying to develop the AO Python Package for AO modelling and simulation. It includes modelling classes of atmosphere, telescope, Shack-Hartmann wavefront sensor, deformable mirror, which are the components for an AO system. It also includes the ability to simulate the entire AO system over time. It is being developed in the Super Eye Bridge project to develop a segmented mirror, an adaptive optics, and an emersion grating spectrograph, which are future telescope technologies. And it is planned to be used as a performance analysis system for several telescope projects in Korea.

Effects of Light on Activities of Antioxidative Enzymes in Hairy Root Cultures of phytolacca esculenta Houtte (자리공(Phytolacca esculenta van Houtte) 모상근배양에서 항산화효소의 활성에 미치는 광의 영향)

  • The effects of light on the activities of several antioxidative enzymes, catalase (CAT), superoxide dismutase(SOD), ascorbate oxidase(AO), and peroxidase(POD) were examined in the hairy root cultures of Phytolacca esculenta van Houtte induced by Agrobacterium tumefaciens $A_4$T. Activities of CAT, SOD, and AO were significantly decreased with incresing light intensity (500-2,000 lx). The activity of AO under high light condition (2,000 lx)was decreased by 92% compared to the dark condition. The activities of glutathoine peroxidase (GPO), ascorbate peroxidase (APO) and general POD were increased under lower light intensify below 500 lx. The activity of GPO under 2,000 lx was decreased by 85% compared to the dark condition. The activities of antioxidative enzymes were more decreased in blue light (400-500nm). The activities of antioxidative enzymes in blue light intensity were increased in lower light intensity below 30 lx, but decreased 21-70% under 200 lx. The activity of AO was decreased by 70% under 200 lx with increasing blue light intensity. Our results suggest that the activities of antioxidative enzymes in hairy roots might be inhibited by endogenous oxidants generated under the high blue light conditions.

A Report on a Crosstabulation Analysis of a Body Composition Test of Workers (건강검진을 받은 근로자들의 체성분검사 교차분석 보고)

  • Objectives: This study was designed to analyze the results of body composition test by Oriental medical examination. The subjects were workers in the automobile manufacturing industry. Methods: The subjects consisted of 24,077 industrial workers who had received Oriental medical examination and completed questionnaires about drinking history and smoking history, during 9 months from March to November. They were categorized by sex, age, job class, drinking history and smoking history. The percentage of body fat(%BF) and abdominal obesity(AO) were measured by using bioelectrical impedance analysis (Jawon Medical, Seoul, Korea). We analyzed the relationship between sex, age, job class, drinking history, smoking history and %BF and AO using x$^2$-test of the SPSS Ver. 14.0. Results: Regarding the relationship between sex and %BF, AO, there was statistically significant difference as men had a significant higher level of %BF and AO than women. There was a positive correlation between age and both %BF and AO. Workers in their 40s especially showed a high level of AO. In job class, office workers and sales personnel were apt to have a higher level of %BF and AO than factory workers. %BF and AO of the drinker group showed a higher level than the non-drinker group. On the other hand, %BF and AO of the non-smoker group were higher than those of the smoker group. Conclusions: Companies should manage obesity closely and educate the workers to quit smoking for their health promotion. Furthermore, continuous health checkups need to be done systematically.

A Comparison of Nutrient Removal Characteristics between (AO)2 SBBR and A2O SBBR ((AO)2 SBBR과 A2O SBBR에서 영양염류 제거 특성 비교)

  • This study was carried out to compare the performance of two types of sequencing batch biofilm reactors (SBBRs), anoxic-oxic-anoxic-oxic $(AO)_2$ SBBR and anoxic-oxic-anoxic $A_2O$ SBBR, on the biological nutrient removal. The TOC removal efficiency in $A_2O$ SBBR was higher than that in $(AO)_2$ SBBR. At the 1st non-aeration period, the release of ${PO_4}^{3-}-P$ in $A_2O$ SBBR was higher than that in $(AO)_2$ SBBR because of the high TOC removal. At the 1st aeration-period, the nitrification was not completed in $(AO)_2$ SBBR, however, it was completed in $A_2O$ SBBR and the nitrification rate in $A_2O$ SBBR was higher than that in $(AO)_2$ SBBR. The release and uptake of ${PO_4}^{3-}-P$ in $A_2O$ SBBR was much higher than in $(AO)_2$ SBBR. Also, the profiles of DO and pH in reactors were used to monitor the biological nutrient removal in two SBBRs. The break point in DO and pH curves at the aeration period coincided with the end of nitrification.
