Dried Sugarcane Press Residue as a Potential Feed Ingredient Source of Nutrients for Poultry

  • Sugarcane press residue (SPR), a byproduct from the sugar industry was evaluated for it's nutrient and energetic quality in broilers and layers. The composition of SPR included (% DM): CP-11.76 (methionine-2.21, cystine-1.05, lysine-4.85, threonine-5.48% of CP), EE-7.87 (palmitic acid-30.3, stearic acid-4.1, oleic aicd-17.2, linoleic acid-38.0, linolenic acid-5.4% of EE), CF-10.08, TA-21.08 (Ca-3.87, P-1.10, Mg-0.95%, Fe-3500, Mn-284, Zn-113, Cu-61.5, Co-5.0 ppm and AIA-4.93%) and NFE-48.35% indicating that SPR is a valuable source of both organic and inorganic nutrients for poultry. The metabolic trials revealed the average ME of SPR as 749, 842 and 1,270 kcal/kg, respectively in broilers and 844, 936 and 1,031 kcal/kg in layers, at 10, 20 and 30% inclusion levels, respectively. Further, the fortification of SPR incorporated diets with biotechnological products viz., lipid utilizing agents (lipase and lecithin) or NSP degrading enzymes and their combination did not improve the ME content of such diets.

  • 본 연구에서는 BC 생산균주인 KJ-1을 UV와 NTG mutagenesis를 통하여 초기에 주입된 glucose를 효과적으로 이용하여 cellulose 수율이 향상된 균주로 계량하였다. Mutant들 중 수율이 2배정도 향상한 균주에서는 cellulose를 비롯한 gluconic acid, 2-keto gluconic aicd, 5-keto gluconic acid의 비율이 parent strain과 차이를 보였고, 시간에 따른 acid 농도 분포를 살펴보면 수율이 증가한 원인 중에 mutant가 parent strain보다 합성한 acid를 효율적으로 이용하고 있다고 추론 할 수 있었다.

Pantoea agglomeranso에 의한 Hydroxyapatite의 인산화

  • 불용성 인산염인 hydroxyapatite를 분해하여 인산을 생성하는 미생물인 P. agglomerans의 성장에 따른 인산의 생성량을 여러 산으로 처리하였을 때와 비교하여 보았다. P. agglomerans 의 성장에 있어서 필요한 인산의 양은 300mg/L에서 361mg/L 정도의 인산을 소모하였다. 또한 여러 유기 및 무기산으로 4% 의 hydroxyapatite를 처리하였을 때에 0.01N 의 시트르산과 옥살산으로 처리하였을 때 만이 인산의 생성량은 P. agglomerans 에서 보다 높은 각각 702와 537mg/L 이었고 배양후 48 시간이 지났을 때 P. agglomerans 의 유리인산의 최대가용화량은 465mg/L이었다.

  • In order to investigate the antioxidant activity of phenolic compounds contained in domestic Meju and Doenjang , the methanolic extract from defatted Meju and Doenjang was fractionated into the phenolic acid and the isoflavone fractions by alumina column and polyamide-6 column chromatography, respectively. Both phenolic acid and isoflavone fractions exhibited an identical antioxidative effect against the oxidation of linoleic acid. GC analysis revealed that the phenolic acid fractio contained vanillic , chlorogenic ,p-coumalic , ferulic, and caffeine acid. The content of caffic acid was greater than 70% in the phenolic acid fraction and the content of vanillic and chlorogenic acids were disappeared during fermentation, and thus, the facts are thought to be reasons for the low antioxidant activity of the phenolic aicd fraction from Doenjang. Similarly, the isoflavone fraction cotained daidzin, glycitin-65-0 glucosider and genistin and their aglycons such as daidzein, glyciten and genistein. The content of daidzin and genistin in Meju dramatically decreased at the early stage of fermentation ,whereas the content of daidzein an dgenistein rather increased ; however, these change in isoflavone contents did not affect the antioxidant activity of isoflavone fractions.

과실 및 채소류의 저장에 있어서 Lactic Acid Bacteria의 이용 (Lactic held Bacteria for the Preservation of Fruit and Vegetables)

  • Lactic aicd bacteria(LAB)는 전통적으로 식품의 발효과정 중에 사용되는 미생물이다. Lactic acid bacteria를 이용한 bacteriocinogenic culture와 lactic acid bacteria에서 생산되는 산물은 식품의 부패에 원인이 되는 미생물과 병원성균에 대한 생육억제효과을 지니고 있다. 이러한 이유로 생선제품, 유제품. 육제품과 발효되지 않은 냉장식품 등의 식품저장에 lactic acid bacteria를 이용한 연구가 다양하게 수행되어져 오고 있었으나 과실 및 채소류에서의 lactic acid bacteria에 대한 효용가능성에 대해서는 아직까지 광범위하게 연구된 것이 없다. 따라서 lactic acid bacteria의 과일 및 채소류의 저장시 효과적이고 적용성이 넓은 보존제로서의 성공적인 이용을 위해서 식품저장과 lactic acid bacteria에 대한 특성, lactic acid bacteria의 대사산물, lactic acid bacteria의 안정성 등을 고려하여 과일 및 채소의 식품성분 및 구성요소와 미생물의 생육저해효과가 있는 대사산물과의 관계에 대한 많은 연구의 수행이 과일 및 채소제품에서 lactic acid bacteria의 식품보존제로서의 이용가능성을 평가하기 위해 필요한 점으로 사료된다.

Structural and Functional Importance of Two Glutamate Residues, Glu47 and Glu146, Conserved in N-Carbamyl D-Amino Acid Amodohydrolases

  • The mutant enzymes of N-carbamyl-D-amino aicd amidohydrolase (N-carbamylase) from Agrobacterium radiobacter NRRL B11291, showing a negligible activity, were selected from the library generated by random mutagenesis. From the sequence analysis, these mutants were found to contain the amino acids substitutions at Cys172, Glu47, and Glu146. Previously, Cys172 was reported to be necessary for the enzyme catalysis. The chemical modification of the N-carbamylase by carboxyl group specific chemical reagent, 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide(EDC), resulted in a loss of activity. The replacement of glutamic acids with glutamines by site-directed mutagenesis led to aggregation of the enzymes. Mutant enzymes fused with maltose binding protein (MBP) were expressed in soluble form, but were inactive. These results indicate that two glutamic acid residues play an important role in structure and function of the N-carbamylase. Multiple sequence alignment of the related enzymes revealed that Glu47 and Glu146 are rigidly conserved, which suggests that tese residues are crucial for the structure and function of the functionally related C-N hydrolases.

Lignin을 분해하는 Streptomyces strainsdop 의한 페놀화합물의 분해 (The Degradation of Phenolic Compounds by Lignolytic Streptomyces strains)

  • The purpose of this was to investigate the degradation efficiency of phenol compounds(catechol, ferulic acid, protocatechuic acid, syringic acid, vanillic acid) by Streptomyces halstedii scabies SAI-36, Streptomyces avendulas SA2-14, and Strptomyces badius(ATCC 39117, control group). The results were as follows: Catechol showed the degradation efficiency that is lower than 50% in three strains. Ferulic acid and vanillic acid showed high degradation efficiency of 98.8% and 94.5% respectively by Streptomyces lavendulas SA2-14. protocatechuic acid and syringicacid showed high degradation efficiency of 89.6% and 77.9%. The degradation efficiency of catechol by Streptomyces halstedii scabies SAI-36, Streptomyces lavendulas SA2-14 and Streptomyces badius(ATCC 39117) was low as 49.2%, 40.2% and 20.2% respectively. But the degradation of other phenolic compoumds except catechol by Streptomyces laven-dulas SA2-36 and Streptomyces badius(ATCC 39117). The results demonstrated that two experimental strains are superior ability to control group in degradation of phenol compounds and Streptomyces lavendulas SA2-14 was superior of two experimental strain. This results were consistent with previous research results that Streptomyces lavendulas SA2-14 was the best strain in degradation ability for lignin, decoloration abilities for variousdyes, and various enzyme production abilities. Therefore, it is suggested that lignin can be used as a indicator when selecting Actinomycetes for degradation of non-degradable materials such as phenol compounds.

LB법을 이용한 Arachidic acid LB막의 공기-물 계면현상과 전기적특성 연구 (Study on air-water interface phenomenon and electrical property of Arachidic acid LB films)

  • 본 연구에서는 Arachidic acid Langmuir-Blodgett (LB)막의 계면특성과 전기적 특성을 측정하였다. Arachidic aicd는 포화지방산으로 ($CH_3(CH_2)_{18}COOH$)의 구조를 가지며, 크기가 27.5[$\AA$]으로 $CH_3(CH_2)_{18}$의 소수기와 COOH의 친수기로 구성되어 있다. LB막은 박막제작시 배열과 배향의 제어가 용이하다. Chloroform을 용매로 하여 2[mmol/l]의 농도를 조성하여 ${\pi}-A$ 등온선을 통해 기체 상태, 액체 상태, 고체 상태를 관찰하였으며 편광각 현미경 (Brewster angle microscopy) 이미지를 통해 각 상태에서의 이미지를 관찰하였다. 또한 LB막의 제작시 사용되어진 ITO 기판은 친수처리 전 후의 접촉각의 측정을 통하여 막의 안정성을 확인하였다. 또한 LB막을 Metal/LB막/Metal 구조의 소자로 제작하여 전압-전류 특성을 측정하였다.

Searching for blue ocean of Alzheimer's disease drug discovery

  • Alzheimer's disease (AD) is an age-related neurodegenerative disorder. The pathological hallmarks of AD are senile plaques and neurofibrillary tangles in the brain. Major component of senile plaques is amyloid beta peptide(A$\beta$) which is derived from amyloid precursor protein (APP). A$\beta$ is generated through the sequential cleavage of App by $\beta$ - and $\gamma$-secretases. $\beta$-secretase excises the ectodomain of APP ($\beta$-APPs) to leave a 99-amino acid long C-terminal fragment (APP-C99-CTF) in the membrane. $\gamma$-secretase then cleaves this membrane-tethered APP-CTF within the transmembrane domain, so releasing A$\beta$ peptides and APP-intracellular domain (AICD). Thus, $\beta$- and $\gamma$-secretase are regarded to perform the key steps in the pathogenesis of AD and have become important therapeutic targets in the prevention and treatment of AD. Enormous efforts have been focused to develop the amyloid beta related drug for cure of AD becuase A$\beta$ is believed to be one of the major causes of AD. since major pharmaceutical companies in world wide base compete to develop new drug for AD, we have to be careful to choose the drug target to success the tough race. In the present talk, possible drug targets based on basic research results will be discussed. These molecules should be a good target for development of new drug for AD and be less competitive to have a good shape for world wide competition.

폐경 후 여성의 지방산 섭취실태와 혈청 지질농도와의 관련성 (Relationships between Fatty Acid Intake and Serum Lipids in Postmenopausal Women)

  • The intake of fatty acids, and the effects of fatty acid intake patterns on serum lipid concentrations was investigated in 112 postmenopausal women. Dietary intakes of fatty acids were assessed by means of a semi-quantitative food questionnaire with food models and other measuring tools. The intakes of saturated fatty acid(SFA), monounsaturated fatty acid(MFA) and polyunsaturated fatty acid(PUFA) were 10.85g, 11.89g and 12.09g, respectively. The ratios of P/M/S and $\omega$6/$\omega$3 fatty acid were 1.19 : 1.10 : 1 and 6.65 ; 1, respectively. Serum lipid concentrations were adjusted for age, menopausal period, BMI, stress scores, exercise habit and energy intake and the relationship between fatty acid intake and serum lipids was measured using Pearson's correlation coefficient. Serum cholesterol concentration was positively correlated with dietary intake of SFA (p<0.10), MUFA(p<0.05), palmitic acid(p<0.10), stearic acid(p<0.05), oleic aicd(p<0.05), and arachidonic acid(p<0.10). Serumk LDL-cholesterol concentration was positively correlated with MUFA(p<0.10), oleic acid(p<0.10), oleic acid(p<0.10), and arachidonic acid (p<0.10). Serum triglyceride concentration was positively correlated with arachidonic acid(p<0.10). However, serum HDL-cholesterol concentration was not found to be significantly correlated with any fatty acids.
