Fractionation of Anticarcinogenic Enzyme Inducer(s) from Roasted Perilla (볶은 들깨박으로부터 암예방효소계 활성성분의 분획)

  • Elevation of the activities of phase 2 enzymes such as quinone reductase(QR) provides protection against several types of neoplasia. In this study, we performed partial purification of QR inducer(s) from roasted and defatted perilla meal by solvent fractionation and thin layer chromatography. Cellular QR induction was most notable in chloroform fraction of roasted perilla extract, compared with other solvent fractions. QR inducer(s) was partially purified by TLC, with 0.8 of $R_f$ value in n-butanol : n-propanol : 2N-ammonium hydroxide(10 : 60 : 30). AHH-inducing activity in TLC fractions isolated from methanol extracts of roasted perilla comigrated with QR-inducing fraction, suggesting that QR and AHH are induced by the same compound. TLC fractions shown strong QR-inducing activity also had a potent antioxidative activity, suggesting that cellular QR enzyme is induced by antioxidant(s) present in roasted perilla.

Effect of Ginseng Extracts on the Binding to DNA of Benzo(a)pyrene Metabolites in uitro in Rats (DNA와 Benzo(a)pyrene 대사물질 결합형성에 미치는 인삼 추출물의 영향)

  • Reactive metabolites generated by benzo(a)pyrene(BP) monooxygenase(AHH) interact with nucleophiles in DNA and cause mutation and carcinogenesis. We studied the effect of Panax ginseng C.A. Meyer, which induce epoxide hydratase(EH) activity without concomitant induction of AHH activity, on the binding of BP metabolites to DNA in uitro in Sprague Dawley rats. DNA-BP metabolite adducts can be resolved into at least five distinct peaks by elution of a Sephadex LH-20 column with a water methanol gradieNt. These peaks are arbitrarily designated A(most polar) through I(least polar). Of the 5 peaks tentatively assigned to 7,8 biol-9,10-oxide(A),7,8·oxide(B),4,5-oxide(C), and further metabolites of 9-OH-BP(D & E), peaks A, C, D, and I were reduced to 70, 85, 80, and 30% of controls, respectively, and there was no significant change in peak B. In connection with this DNA binding study, BP metabolizing enzymes including AHH, EH, demethylase(DM) activity and cyt. P-450 contents were also investigated in order to compare the BP treated control with ginseng and BP treated test groups. The results showed that the EH activity was increased by 139% over the BP control, the Cyt. P-450 content was increased by 180% over the control value, and DM and AHH activities were also increased to some degree for the BP test group, but there was no significant effect of the ginseng treatment.

  • We studied the effects of a single, combined and mixed exposure of benzene, toluene and xylene on the activities of rat liver microsomal AHH, ADH and ALDH, and the excretion of their metabolites in urine. The AHH activities of the rats treated in combination and mixture were slightly higher and/or similar to those rats treated with single solvent, while the reverse effects were observed for ADH and ALDH. Similar effects were observed when the metabolites were examined in urine (p < 0.01). These results suggest that each solvent might interJkre the induction and action of ADH and ALDH, and decrease the excretion of their metabolites into urine.

In Vitro Enhancement of Microsomal Cytochrome P450-Dependent Monooxygenases by Organic Solvents in Rat Liver

  • In vitro effects of acetone, methanol, and dimethylsulfoxide (DMSO) on liver microsomal cytochrome P450 (P450) content, and P450-dependent arylhydrocarbon hydroxylase (AHH) and 7-ethoxycoumarin O-deethylase (ECOD) activities were studied in rats. Acetone at 1% (v/v) enhanced the content ofP450, assayed spectrally in 3-methylcholanethrene (MC)- and ${\beta}-naphthoflavone$ (BNF)-inducible microsomes by 18 and 7%, respectively. Methanol, up to 5% (v/v) applied, also showed enhancement effects on P450 content in liver microsomes from rats treated with phenobarbital (PB), MC, and BNF, as well as uninduced microsomes with similar but low strength. DMSO, however, did not show such enhancing effects at the ranges of the concentrations applied. AHH and ECOD activities in MC-inducible microsomes were also enhanced by acetone at 1%, which was in proportion to the increase in P450 content by the same concentration. However, the P450 content, and AHH and ECOD activities, were decreased by increasing the concentration of acetone. Methanol at the same concentration with acetone also enhanced ECOD activity but not AHH activity in MCinducible microsomes. The enhancing effect of acetone on the enzymes was negligible when the microsomes were pretreated with a specific monoclonal antibody of MC-inducible isozyme. The difference in the effects of these solvents on P450 system might be due to their different properties that cause the P450 active site to be exposed in milieu.

Characteristics of Acetone Enhancement of Microsomal Cytochrome P45O-dependent B(a)P Hydroxylation in 3-Methylcholanthrene-inducible Rat Liver Microsomes

  • Previously, we showed that acetone enhanced aryl hydrocarbon hydroxylase (AHH) activity in only 3-methylcholanthrene (MC)- or $\beta$-naphtoflavone (BNF)-inducible microsomes of rat liver. In the present study, the possible mechanism underlying acetone action on AHH was investigated in the liver microsomes from MC-pretreated rats. Other n-alkylketones except acetone did not increase AHH activity, which rather decreased significantly with the length of alkyl side chain. Acetone had no effect on the activity of NADPH-cytochrome P450 reductase or inhibited the formation of 3-OH benzo(a)pyrene (B(a)P) in nonenzymatic model ascorbic acid system. However, in cumene hydroperoxide (CuOOH)-supported B(a)P hydroxylation, acetone enhanced its velocity remarkably by 30% at the optimal concentration (30 $\mu$M CuOOH and 1.0% acetone). From these results, we conclude that acetone may facilitate the formation of an activated oxygen species or the insertion of oxygen into B(a)P molecule in CYP1A rich microsomes.

  • Due to increasing marine pollution there is a great possibility that seaweed is contaminated with polycyclic aromatic hydrocarbons (PAHs). To investigate the effect of processing on PAM removal from Porphyra tenera (laver) contaminated PAH, laver was contaminated with fluoranthene known to have a strong photoinduced toxicity, followed by washings and drying, which are usual processes for dried laver preparation. Sample at each step was collected and its toxicity was evaluated using cultured animal cells as well as analyzing PAH contents. Fluoranthene level in laver was significantly lowered by sequential washings with sea water and distilled water, but not by drying. Fluoranthene content in raw laver right after contamination was 221 ppm and decreased to 130 ppm by washings with seawater plus distilled water while its level was not lurker lowered by drying process. Cytotoxicity and photoinduced cytotoxicity in mammalian cells were significantly elevated in laver extracts containing fluoranthene. Cellular arylhydrocarbon hydroxylase (AHH), one of the biomarkers for cellular accumulation of PAH, was greatly induced by laver extract contaminated with fluoranthene. These results suggest that photoinduced toxicity and AHH activity can be used to monitor contamination of seafood by PAHs. Fluoranthene accumulated in laver was efficiently removed by sequential washings with seawater and tap water for 24 hrs and 12 hrs, respectively.

  • 본 연구에서는 아이돌 음악 중 타 장르를 연상하게 하는 일부 기법이 혼용된 사례를 분석하였다. 해당 사례에 해당되는 곡 중 여러 차트의 10위권 이내에 기록된 트와이스의 'OOH-AHH하게'와 'Cheer up'으로 한정하였으며, 이 결과를 통해 아이돌 음악의 한 편곡의 특정 유형과 음악적 유행의 일정 부분을 파악할 수 있었다.

  • Unlike existent field flow fractionation, new method, osmotic sink field flow fractionation is introduced and used ultrafiltration hollow fiber membranes as separation channel. This hollow fiber osmotic sink field flow fractionation is called HF-OSFFF. A theory that describes the retention, relaxation, resolution, plate number for the system, has been developed and experimentally verified by separation model of po1ystyrene latex beads. At external field, it is measured that radial flow rates change according to various concentrations of PEG solutions. Concentration of PEG solution vs. radial flow rate is a linear relation. For diameter distribution of unknown polymer sample, HF-OSFFF compared with the commercial capillary hydrodynamic flow fractionation (CHDF).

  • Protective effects of dithiol malonate derivatives (DMDs), YH-100, YH-150 and YH-439 on carbon tetrachloride-induced hepatotoxicity were investigated in primary rat hepatocytes culture. Treatment of DMDs to hepatocytes culture did not affect total cytochrome P-450 content and ECOD and AHH activities. Protein and RNA synthesis was also similar to control. Meanwhile, DMDs significantly decreased LDH release and in vitro lipid peroxidation induced by $CCI_4$. Accumulation of cellular triglyceride and decreased secretion of VLDL from liver cells by $CCI_4$ treatment were also significantly protected.
