Inhibition of Starch Biosynthesis by Antisense Expression of cDNAs Encoding ADP-Glucose Pyrophosphorylase Small Subunit in Sweetpotato (고구마에서 ADP-Glucose Pyrophosphorylase Small Subunit cDNA의 Antisense 발현에 의한 전분생합성 저해)

  • Embryogenic calluses derived from shoot apical meristem explants of sweetpotato were subjected to particle bombardment to generate transgenic plants for antisense expression of cDNAs encoding two different AGPase small subunit (ibAGP1 and ibAGP2). Plants were generated via somatic embryogenesis. PCR and Southern analysis demonstrated that the incorporation of ibAGP1 and ibAGP2 into the genome in an antisense orientation. Immunoblot analysis confirmed reduced levels of AGPase small subunit in transgenic plant leaves. Plants with both ibAGP1 and ibAGP2 produced a lower level of the protein than plants with ibAGP1 alone. iodine test demonstrated that transgenic plant leaves and storage root accumulated reduced amounts of starch. Iodine staining of leaf tissues indicated that transgenic plants accumulated less amount of starch than control. In accordance with western blot analysis, plants with both ibAGP1 and ibAGP2 accumulated a lower amount of starch than plants with ibAGP1 alone. Both transgenic plants exhibited a severely retarded growth, resulting in bare survival. It is suggested that disrupted expression of the gene encoding AGPase small subunit is lethal to the growth of sweetpotato contrast to other species including potato.

The serological studies on infectious bursal disease (전염성 F낭병에 대한 혈청학적 연구)

  • This study was carried out to provide the fundamental information for development of proper vaccination program against infectious bursal disease(IBD) to the local chicken farms. The antigen detection was peformed from 8 samples of bursa of Fabricius with agar gel precipitation(AGP) and indirect immunofluorescent assay(IFA), And also, the antibodies in serum samples were detected by the various serological methods such as commercial ELISA assay, AGP and virus neutralization(VN) test. 1. The antigen detection rates were 25% for AGP which is 2 out of 8 farms and 10 out of 40 bursas, and 25% which Is 2 out of 8 farms and 20% 8 out of 40 bursas for IFA, respectively. 2. The mean titer of maternal antibody (>3,000) existed until 10 days of the age with ELISA-GMT. 3. The antibody positive rates which are over 80% showed until 5 days of the age with ELISA and at 10 days of the age with AGP except one, but none of them showed from 1 day of the age. This report came to conclusions that both the protective maternal antibody titers and the antigen positive rates were significant until at the 10 days of the age.

  • For the test organism of algal growth potential (AGP), the diatom in the genus Stephanodiscus which cause blooms in the Nakdong River was used instead of generally used strains of Selenastrum, Microcystis, or Anabaena. AGP results indicated that all the samples in the Nakdong River except for that from the Nakdan Bridge site were eutrophic state. Furthermore, the sample from Kumho River site was hypertrophic state. In the main stream Nakdong River, the value of AGP was lowest at the upstream Nakdan Bridge site and was highest at Koryoung Bridge site which is just downstream of Kumho River confluent point indicating the seriousness of pollution contributed by the Kumho River to the Nakdong River. Changes in the concentration of nutrients before and after the AGP tests and inter-relationship among the nutrients indicated that the growth of the Stephanodiscus in the AGP tests were mostly affected by the nitrate, silicate and phosphate. The limiting nutrient was identified by the nutrient addition experiments and the results showed that phosphate was the limiting nutrient for the growth of Stephanodiscus in the tested samples.

Soluble expression, purification and the role of C-terminal glycine residues in scorpion toxin BmK AGP-SYPU2

  • The existence of glycine residues in long-chain scorpion toxins has been well documented. However, their role as analgesics has not been evaluated. To address this issue, we investigated the functional role of glycines in the C-terminal end of Chinese-scorpion toxin from Buthus martensii Karsch (BmK AGP-SYPU2) using site-directed mutagenesis and analgesic activity assays. Recombinant BmK AGP-SYPU2 and its mutants were efficiently expressed in E. coli and purified to homogeneity using immobilized metal ion affinity chromatography (IMAC) and cation exchange chromatography. The mouse-twisting test was used to detect the analgesic activity of BmK AGP-SYPU2 and its mutants. As a result, we identified glycines at the C-terminal end that, when altered, significantly affected analgesic activity. Also, Mut6566 was significantly decreased compared to BmK AGP-SYPU2. These data indicate that the glycines at the C-terminal end are important for the analgesic activity of BmK AGP-SYPU2.

Comparison of Algal Growth Potentials in the Large Reservoirs and River Mainstream of Naktong River Watershed (낙동강 수계 대형 인공호 및 하천본류의 조류성장 잠재력 비교)

  • Algal growth potential test (AGPT) has been used as a tool for assessing biological productivity potential in the aquatic ecosystems. This study was conducted to compare the productivity potentials of large reservoirs (Lakes Andong, Hapchon and Jinyang, and Naktong estuarine dam) and river sites (Sangju, Koryung, and Samlangjin) located in the Naktong River watershed. AGPT was conducted in both non-monsoon and mosoon season (February, April, July and September) of 2003, using Microcystis aeruginosa as a test alga. The AGPs in the reservoirs were relatively much lower than those of river sites. The river AGPs increased towards upstream close to the influent streams, while it generally decreased towards downstream. Immediately after the abrupt increase in influent discharge in summer, the AGP became similar between midstream and downstream sites. The water quality of river and reservoirs deteriorated during the drought period in accordance with AGP: it was the highest during this period. The AGPs showed the closest correlation with the P concentration, leading to the conclusion that bioavailable P is highly influential to the algal growth in both lentic and lotic ecosystems in the Naktong River watershed. Based on the AGPs, the water quality of tested sites was likely eutrophic. Our results suggest that AGPT be a useful tool in evaluating the productivity potential and trophic state of the water body as well as determining the nutrients that limit the growth of algae.

Evaluation of Water Quality after Rehabilitation of Cheonggye Stream using AGP Test (조류성장잠재력 조사를 이용한 청계천 복원 이후 수질 평가)

  • Algal growth potential (AGP) test was performed to evaluate the water quality and changes in phytoplankton communities before and after a heavy rain event at six sampling sites in Cheonggye Stream (St. 1 and 2), Jungnang Stream (St. 3 and 4), and Lower part of Han River System (St. 5 and 6) after rehabilitation of Cheonggye Stream, October 2005. To test AGP on each sampling site, cyanobacterium Microcystis aeruginosa was applied as a standard alga. Total nitrogen (TN) showed high values at Jungnang Stream, while St. 4 recorded highest values in this study. However, TN values of Cheonggye Stream and Lower Part of Han River showed similar levels. Total phosphate (TP) also showed high values at Jungnang Stream, while St. 4 recorded highest. However, TP in Cheonggye Stream were extremely low levels. Although chlorophyll-${\alpha}$ (chi-${\alpha}$) contents before the rain event were similar through the sampling sites, chl-${\alpha}$ after the rain increased dramatically at Jungnang Stream and Lower part of Ban River. In particular, after the rain, TP was the highest at St. 4, where Cyclotella sp. dominated the phytoplankton community. When compared with control, AGP values before the rain were comparatively low in all sites, while those after the rain highly increased with the dose-dependently of field water added, due perhaps to the increased nutrients by rainfall. Similar results were observed in Cheonggye Stream. Therefore, for the aesthetic fostering for the citizens, although Cheonggye Stream was Presently being sustained by treated water supply, they have a potential of outbreak of phytoplankton by the increased nutrients supply when a heavy rain comes.

Comaprison of Six Serological Methods for the Diagnesis of Bovine Brucellosis (브루셀라병(病) 검색(檢索)에 사용(使用)되는 여러가지 혈청진단법(血淸診斷法)의 비교연구(比較硏究))

  • Results obtained from six secological tests for diagnosing bovine brucellosis-standard plate agglutination test (SPT), standard tube agglutination test(STT), complement fixation test(CFT), Rivanol test (RT), agar gel precipitation test (AGP) and counterimmunoelectrophoresis(CIEP) were compared using 38 sera from brucella reactors and 222 sera from dairy and beef cattle in field. The SPT gave 1.6% apparent false negative reactions and 15.4% apparent false positive reactions when compared with STT which is an official test for bovine brucellosis in this country. The distribution of antibody titers determined by STT showed that 37.5% of 38 reactors had antibody titers ranging from 100 to 200, and the remaining 62.5% had antibody titers of 400 or higher. when 38 reactor judged by STT were tested by CFT and RT, 32 cattle(82.4%) were positive by CFT and 33 cattle (86.8%) were positive by RT, respectively. This results suggest that RT is comparable to CFT in the diagnosis of bovine brucellosis. The results also indicated that both AGP and CIEP were insensitive to detect brucella infection in cattle.

Cyanobacterial Bioassay (AGP test) on the Water Fertility of Treated Wastewater Effluents Discharged into Euiam and Paldang Reservoirs, Korea (의암호 및 팔당호에 유입되는 주요 하수처리 방류수의 수질 비옥도 생물검정: Algal Growth Potential(AGP) Test)

  • Euiam and Paldang Reservoirs have often been facing water quality problems, such as eutrophication, algal blooms and off-flavors by treated wastewater effluent (TWE) in the North-Han and the Han River basins, but little is examined on the direct biological effect of TWE. This study tested algal growth potential (AGP) of four TWEs discharged into Euiam and Paldang Reservoirs to evaluate water fertility in September 2014 and March and September 2015. Test alga was used Anabaena circinalis isolated from Paldang Reservoir. Mean concentration of T-N and T-P in TWEs was $3,956.7{\mu}g\;N\;L^{-1}$ and $50.8{\mu}g\;P\;L^{-1}$, and the proportion of $NO_3-N$ and $PO_4-P$ to the total fraction was 72.1% and 40.8%, respectively. Both N and P were high in TWEs, but much higher N than P concentration indicates strong P-limitation. As a consequence, the maximum AGP was determined by $PO_4-P$ concentration (r=0.998, p<0.01). Mean AGP value was $15.4mg\;dw\;L^{-1}$ among four effluents indicating its eutrophic condition. Due to the establishment of tertiary (advanced T-P) treatment method in the studied plants recently, P concentration was significantly decreased in TWEs compared to the years prior to 2012. However, P concentration seems to be still high enough to cause eutrophication and algal blooms. Therefore, wastewater treatment to P-free level needs to be considered if effluents are directly discharged into the drinking water resources.

Serological Diagnosis for Swine Cysticercus Cellulosae Infection in Korea (돼지의 유구낭충(有鉤囊蟲)에 관(關)한 혈청학적시험(血淸學的試驗))

  • The agar gel precipitation(AGP), indirect hemagglutination(IHA) and indirect enzyme immunoassay(IEIA) tests were used to detect antibodies in pigs naturally infected with Cysticercus cellulosae in Jeju. The results obtained were summarized as follows: 1. Sera collected from pigs naturally infected with Cysticcrcus cellulosae did not react in AGP test. 2. In the IHA test for swine cysticercosis, the peak titers observed were between 1:20 and 1:160 and non-specific reaction was recognized with a few samples among control sera. 3. In the IEIA test, optical density(OD) values were obtained the best results under the condition of OPD-substrats are reacted at room temperature for 5minutes. The OD values of greater than 0.2 were determined as positive and the high titers in positive sera ranged from 1:40 to 1:1,080. 4. Antibodies to swine cysticercosis were detected by IHA and IEIA tests but the latter was more sensitive and specific than the former. 5. In the preparation of Cystisercus antigens, saline extract which was prepared the precipitate of internal membrane treated ultrasonicator were better results than other antigens for serological tests. 6. Amounts of protein in antigens was not related in direct proportion to results of serological reaction.

Comparison of serological methods for detection of avian influenza virus antibodies (가금인플루엔자 바이러스 항체검출을 위한 혈청학적 진단법 비교)

  • An enzyme-linked immunosorbent assay (ELISA) using purified hemagglutinin of swine influenza virus (H1N1) as antigen was developed for detection of antibody to avian influenza virus (AIV). The sensitivity and specificity of a developed and commercial available ELISA kits were compared with those of agar gel precipitation (AGP) test and hemagglutination inhibition (HI) test using sera collected from chickens under condition of field exposure. The concentration of antigen, serum dilution and concentration of enzyme-conjugated secondary antibody in developed ELISA (S-ELISA) were 0.5ug/100ul, 1:200 and 0.03ug/100ul, respectively. The correlation coefficients between S-ELISA and commercial ELISA and HI titers were 0.419 and 0.533, respectively. A significant correlation (p < 0.01) was not found between HI and ELISA titers. The S-ELISA was found to be as more sensitive and specific than the AGP test, showing 86.8% sensitivity and 85.3% specificity. It is suggested that the ELISA using the SIV as antigen may be useful method as an investigating tool for AIV serological surveillance.
