Purification and Characterization of Chitinase from Paenibacillus illinoisensis KJA-424

  • A chitinase was purified from the culture supernatant of Paenibacillus illinoisensis KJA-424 by protein precipitation, DEAE-Sephadex anion-exchange chromatography, and Sephadex G-150 gel filtration. The molecular weight of the purified chitinase was 54 kDa on SDS-PAGE and activity staining. Optimal pH and temperature were pH 5.0 and 60$^{circ}$C, the presence of 10 ruM Ag$^{+}$ and Hg$^{2+}$ inhibited the activity by $92.1/%$ and $97.7/%$, and the K$_{m}$ and V$_{max}$ values were 1.12 mg chitin mrl and 1.48$\mu$mol GlcNAc min$^{-1}$, respectively. The enzyme hydrolyzed tetramer to dimer, pentamer to dimer and trimer, and hexamer to dimer, trimer and tetramer, indicating an endo-splitting mechanism. The chitinase had no hydrolytic activity toward dimer and trimer. The chitinase inhibited the mycelial growth of Rhizoctonia solani, suggesting an antifungal property.

A Study on the Recent Sediments of Han River -Grain Size, Heavy Minerals and Trace Elements- (한강유역(漢江流域)의 현생퇴적물(現生堆積物)에 관한 연구(硏究) -입도(粒度), 중광물(重鑛物) 및 유용원소(有用元素)를 중심(中心)으로-)

  • This paper deals with grain size analysis, heavy mineral analysis and trace element analysis of the recent sediments of Han River basin between Cheongpyong, Gyeonggi province and Seoul. For each location the samples are taken at river shoreline, river berm and river dune. The size analysis discloses that the mean values range from $-1.37{\phi}$ to $-1.60{\phi}$, sorting values range from 0.25 to 1.84, skewness values range from -0.44 to 0.51 and kurtosis values range from -0.1 to 1.75. Based on the textural parameters, the dune sand can be distinguished from the shoreline-berm sand. The content of heavy minerals of each sample ranges from 0.04 to 4.7%. The principal heavy minerals are ilmenite, magnetite, leucoxene, garnet, amphibole, pyroxene, kyanite, zircon, monazite, tourmaline, epidote, limonite, and minor minerals are apatite, sillimanite, andalusite and olivine. In general, dune sand samples contain more heavy minerals than the samples of shoreline or berm sand. This suggests that the heavy mineral concentration is affected by wind action more than by any other causes. The content of ilmenite and leucoxene decreases, whereas the content of zircon and epidote increases as it approaches the downstream region. The differences result from the variance of geological occurrences. The emission spectrochemical analysis and colorimetry analysis revealed that the content of Ni and V in the heavy minerals of the study area are higher than those of other stream sediments in Korea. On the other hand the content of Cu, Ph, Zn, Mo, W, P, Mn, Cr, Ag and Sn are lower in the study area. It has been observed also that the contents of all the elements except for Bi are higher in this area than the samples of marine sediments of Yellow Sea.

K-Ar Age of the Keumseongsan Volcanic Rocks and Mineralization in the Southeastern Part of Euiseong, Gyeongsangbuk-Do, Republic of Korea (경북·의성 동남부에 분포하는 금성산 화산암류의 K-Ar연대와 그주변의 광화시기)

  • The Keumseongsan caldera is composed of the Cretaceous sedimentary rocks of the Gyeonesang Supergroup, volcanic rocks of the Yucheon Group and basic dykes. The Keumseongsan caldera is formed by subsidence of volcanic rocks, and arc fault developed late. Also, synistral strike-slip fault ($N60^{\circ}W$) developed. Volcanic rocks belong to subalkaline rocks and calcalkaline magma series. First tuffaceous breccia erupted before 71.4 Ma and cavity of magma chamber caused subsidence, which formed arc fault. Basaltic lava erupted at 71.4 Ma and residual fluids containing Fe, As, Pb, Zn and Cu metal elements built the Ohto deposits, which are dated to be 70.5 Ma based on K-Ar age for sericite. Tuffaceous breccia and tuff erupted between 70.5 and 67 Ma. When volcanic eruption became weakened, cavity in site of magma chamber brought subsidence. Rhyolite intruded and erupted at 67 Ma, and intrusive rhyolite intruded according to arc faults, also. Hydrothermal fluids containing Fe, As, Pb, Zn, Cu, Sb, Bi, Au and Ag formed the Tohyeon deposits. K-Ar age for sericite from the Tohyeon mine gives 66.0 Ma. Results of field exploration, geochemical analyses of volcanic rocks support mineralization possibility by volcanism. Especially, age of volcanism and mineralization are well in coincidence with results of K-Ar age dating. By these results, Ohto Cu mineralization is regarded to be associcated with basaltic rocks, while Tohyeon Cu mineralization with rhyolitic rocks.

Muscle Length and Shortening Velocity Changes during the Different Types of Vertical Jumps (수직점프 동작시 근육길이와 수축속도 변화)

  • 본 연구의 목적은 각기 다른 수직 점프 동작 시 근육의 길이와 근육의 수축속도 변화를 비교 분석하는데 있다. 피험자의 운동학적 변인들을 분석하기 위해 2대의 고감도 카메라를 (60 Hz, Panasonic AG455) 사용하여 점프 동작을 촬영하였다. 대퇴직근, 내측광근, 외측광근, 중간광근, 대퇴이두근(단두), 내측과 외측 비복근의 길이와 근수축 속도는 Brand et al. (1982)에 의해 제시되어진 하지근 기시 정지점의 3차원 좌표값과 동작분석을 통한 하지 분절간의 회전 및 변환행렬을 사용하여 측정되어졌다. 일반적인 근육 길이와 수축속도의 변화 형태는 각기 다른 점프간에 매우 유사한 형태를 보였다. 상승기 초기에 대퇴사두근의 길이가 최대인것으로 나타났으며, 이에 반해 대퇴이두근과 내외측 비복근은 공중 동작이 발생하는 시점에 근의 길이가 최대인 것으로 나타났다. 근육의 길이 변화 범위는 대퇴직근이 35.9에서 47.5 cm, 외측광근이 29.4에서 38.8 cm, 중간광근이 31.5에서 38.0 cm, 내측광근이 30.9에서 38.6 cm, 대퇴이두근이 21.3에서 39.1 cm, 외측비복근이 31.4에서 33.5 cm, 내측비복근이 30.5에서 33.2 cm인 것으로 나타났다. SQ와 CMJ에서는 대퇴사두근의 최대 단축성 수축 속도와 대퇴이두근과 내외측 비복근의 최대 신장성 수축이 공중동작이 발생하기 바로 전에 이루어졌다. 대퇴사두근의 최대 신장성 수축과 대퇴이두근과 내외측 비복근의 최대 단축성 수축은 일반적으로 피험자가 착지하는 순간에 발생되어졌다. 그러나 HJ와 DJ에서는 대퇴사두근의 최대 신장성 추축과 대퇴이두근과 내외측 비복근의 최대 신장성 수축이 하강기 초반에 발생되어졌다.

  • 현재 압전재료로써 사용되고 있는 PZT계 세라믹스는 우수한 압전 및 유전특성으로 초음파 센서, 압전변압기, 액츄에이터, 필터, 레조네이터와 같은 여러 응용분야에 널리 사용되어지고 있다. 그러나 납을 60%이상 포함하므로 환경오염에 의하여 그 사용에 대한 규제가 강화되고 있어 최근에는 납을 함유하지 않은 무연 환경친화형 압전세라믹스가 주목받고 있다. 현재 무연 조성 세라믹스에는 텅스텐-브론즈 형, 비스머스 레이어형, 페로브스카이트 형 등이 이으며, 그 중 페로보스카이트 혈을 제외하고는 보통 소성법 조제시 낮은 압전특성을 갖는다. 그러므로 압전특성을 증가시키기 위하여 Hot pressing, Hot forging, RTGG(Reactive Template Grain Growth), SPS(Spark Plasma Sintering), 그러나 이는 무연 세라믹스의 대량생산 과정에서 어려운 문제를 가지고 있고, 저가격 관점에서 볼 때 보통소성법이 보다 바람직하다. 그래서 보통소성법으로 NKN세라믹스의 소결성을 향상 시키고 비교적 높은 압전특성을 얻기 위해서 $LiSbO_3$, $LiNbO_3$, $LiTaO_3$, alkaline-earth(Mg, Ca, Sr, Ba) 등을 첨가한 논문들이 보고되고 있으며, 이러한 재료들의 kp는 대략 0.3-0.4를 나타내고 있다. 따라서, 본 연구에서는 소결온도를 낮추고자 $Ag_2O$를 소결조제로 사용하였고 유전 및 압전특성을 높이고자 소결시간 변화시켜 시편을 제작하여 NKN 세라믹스의 유전 및 압전특성을 조사하였다.

  • Injection processing of denture base resin was introduced by Pryer in 1942, in an attempt to reduce processing shrinkage. More recently a continuous-pressure injection type technique has been developed (SR-Ivocap, Ivoclar AG, Schaan, Liechtenstein.), and it reduced processing error and increased resin density. The purpose of this study was to compare tensile bond strength of heat-cured, cold-cured, and light-cured denture base resin bonded to continuous-pressure injection type resin. To know it, 60 cylindrical resin specimens were fabricated, and tensile bond strength were measured. The results were as follows : 1. The mean tensile bond strength bonded to continuous-pressure injection type resin was lower than bonded to conventional heat cured resin. But tensile bond strength of conventional heat cured resin bonding with light cured resin was lower than continuous-pressure injection type resin. 2. Of the tensile bond strength bonded to continuous-pressure injection type resin, tensile bond strength bonding with continuous-pressure injection type resin was the greatest(but not significantly different from bonding with conventional heat cured resin), followed by cold-cured, light-cured resin. 3. Of the tensile bond strength bonded to conventional heat cured resin, tensile bond strength bonding with conventional heat cured resin was the greatest and followed by continuous-pressure injection type resin, cold-cured resin, light-cured resin. According to these results, bonding of continuous-pressure injection type resin with conventional heat cured resin or continuous-pressure injection type resin is acceptable, but bonding with light-cured resin is questionable.

Purification and Characterization of the External Invertase Constitutively Produced by Rhodotorula glutinis K-24 (Rhodotorula glutinis K-24에 의해 구성적으로 생산되는 세포외 Invertase의 정제 및 특성)

  • Rhodoto& ghtbth~ K-24 was found to produce external invertase in addition to internal and cell wall bound invertase. External invertase was purified to an electrophoretically homogeneous state and partitally characterized and was compared with internal and cell wall bound invertase of which procedures for purification and characterization were reported previously. The enzyme was purified by ethanol precipitation, column chromatographies on DEAE-Sephadex A-50 and SP-Sephadex C-50, and gel filtration on Sephadex G-100. The molecular weight and subunit molecular weight of external invertasGwere estimated to be 220,000 and 100,000, respectively. The isoelectric point of the enzyme was about pH 6.0. The optimum pH and temperature for enzyme activity were pH 4.0 and $60^{\circ}C$, respectively. The enzyme remained stable at the wide range, from pH 3.0 to 11.0 and stable up to $40^{\circ}C$, but was inactivated at temperatures above that. $HgC_12, AgN0_3, MnS0_4$, SDS and p-CMB inhibited the enzyme activity. The $K_m$ value of the enzyme for sucrose was $1.0\times 10^{-2}$M. From these results, the three isozymes from Rh. glutinis K-24 seem to have the similar enzymatic properties, but to differ in molecular and subunit weights.

Microstructure properties with variation of doped amount $Pr_{2}O_{3}$ of BSCT ceramics ($Pr_{2}O_{3}$ 첨가량에 따른 BSCT 세라믹의 미세구조 특성)

  • The barium strontium calcium titanate((Ba,Sr,Ca)$TiO_3$) powders prepared by the sol-gel method and $MnCO_3$ as acceptor were mixed oxide method. The microstructure was investigated with variation of $Pr_{2}O_{3}$ amount. The BSCT powder and $Pr_{2}O_{3}$ were mixed with organic vehicle(Ferro. B75001). BSCT thick films were fabricated by the screen-printing method on alumina substrates. The bottom electrode was Pt and upper electrode was Ag, respectively. All BSCT thick films were sintered at $1420^{\circ}C$, for 2h. The result of the differential thermal analysis(DTA), exothermic peak at around $654^{\circ}C$ due to the formation of the polycrystalline perovskite phase. In the X-ray diffraction(XRD) patterns, all BSCT thick films showed the typical perovskite polycrystalline structure and no pyrochlore phase was dbserved. The microstructure investigated by scanning electron microscope(SEM). Pore and grain size of BSCT thick films were decreased with increasing amount of $Pr_{2}O_{3}$ dopant. And the average grain size and thickness of BSCT thick films doped with 0.1 mol% $Pr_{2}O_{3}$ was $3.09{\mu}m$, $60{\mu}m$, respectively. The relative dielectric constant decreased and dielectric loss decreased with increasing amount of $Pr_{2}O_{3}$ dopant, the values of the BSCT thick films no doped with $Pr_{2}O_{3}$ were 7443 and 4 % at 1 kHz, respectively.

Studies on Efficacy of $Bayvarol^{(R)}$ against Mite(Varroa jacobsoni) and Safety of Bayvarol^{\circledR}$ in Honey Bees (바이바롤(${Bayvarol}^{(R)}$)의 꿀벌응애(Varroa jacobsoni)에 대한 효능 및 꿀벌에 대한 안전성에 관한 연구)

  • In order to determine the efficacy of $Bayvarol^{(R)}$ strips(Bayer AG) against the mite Varroa jacobsoni and the safety of $Bayvarol^{(R)}$ strips in honey bees, a series of trials was carried out under field condition in autumn and spring. The results obtained were as follows; 1. The average varroacidal rate of $Bayvarol^{(R)}$ strips was 92.7% in autumn and 93.7% in spring. However the average varroacidal rats of treated control group was 73.8 % in autumn and 75.1 % in spring, and that of untreated control group was 12.1 % in autumn and 16.1 % in spring. 3. The average number of dead bees in $Bayvarol^{(R)}$ group was 13.0 in recommended dosage(RD), 12.3 in 2 times of RD and 14.0 in 4 times of 70 in autumn, and 19.7 in RD,17.0 in 2 times of RD and 19.0 in 4 times of RD in spring. However the average number of dead bees in treated control groups was 38.3 in RD, 62.7 in 2 times of RD and 60.3 in 4 times of RD in autumn and 18.0 in RD, 43.3 in 2 times of RD and 102 in 4 times of RD in spring and that of untreated control of group was 12.7 in autumn and 19.3 in spring. 3. The varroacidal effects and the number of dead bees of all groups were higher in spring than in autumn. 4. It was revealed that $Bayvarol^{(R)}$ were highly saft and effective varroacides for honey bees.

Element Dispersion and Wall-rock Alteration from Daebong Gold-silver Deposit, Republic of Korea (대봉 금-은광상의 모암변질과 원소분산 특성 연구)

  • The Daebong deposit consists of gold-silver-bearing mesothermal massive quartz veins which fill fractures along fault zones($N10{\sim}20^{\circ}W,\;40{\sim}60^{\circ}SW$) within banded gneiss or granitic gneiss of Precambrian Gyeonggi massif. Ore mineralization of the deposit is composed of massive white quartz vein(stage I) which was formed in the same stage by multiple episodes of fracturing and healing and transparent quartz vein(stage II) which is separated by a major faulting event. The hydrothermal alteration of stage I is sericitization, chloritization, carbonitization, pyritization, silicification and argillization. Sericitic zone occurs near and at quartz vein and includes mainly sericite, quartz, and minor illite, carbonates and epidote. Chloritic zone occurs far from quartz vein and is composed of mainly chlorite, quartz and minor sericite, carbonates and epidote. Fe/(Fe+Mg) ratios of sericite and chlorite range 0.36 to 0.59($0.51{\pm}0.10$) and 0.66 to 0.73($0.70{\pm}0.02$), and belong to muscovite-petzite series and brunsvigite, respectively. Calculated $Al_{IV}-Fe/(Fe+Mg)$ diagrams of sericite and chlorite suggest that this can be a reliable indicator of alteration temperature in Au-Ag deposits. Calculated activities of chlorite end member are $a3(Fe_5Al_2Si_3O_{10}(OH){_6}=0.00964{\sim}0.0291,\;a2(Mg_5Al_2Si_3O_{10}(OH){_6}= 9.99E-07{\sim}1.87E-05,\;a1(Mg_6Si_4O_{10}(OH){_6}=5.61E-07{\sim}1.79E-05$. It suggest that chlorite from the Daebong deposit is iron-rich chlorite formed due to decreasing temperature from $T>450^{\circ}C$. Calculated $log\;{\alpha}K^+/{\alpha}H^+,\;log\;{\alpha}Na^+/{\alpha}H^+,\;log\;{\alpha}Ca^{2+}/{\alpha}^2H^+$ and pH values during wall-rock alteration are $4.6(400^{\circ}C),\;4.1(350^{\circ}C),\;4.0(400^{\circ}C),\;4.2(350^{\circ}C),\;1.8(400^{\circ}C),\;4.5(350^{\circ}C),\;5.4{\sim}6.5(400^{\circ}C)\;and\;5.1{\sim}5.5(350^{\circ}C)$, respectively. Gain elements (enrichment elements) during wallrock alteration are $K_2O,\;P_2O_5,\;Na2O$, Ba, Sr, Cr, Sc, V, Pb, Zn, Be, Ag, As, Ta and Sb. Elements(Sr, V, Pb, Zn, As, Sb) represent a potentially tools for exploration in mesothermal and epithermal gold-silver deposits.
