Isolation of Serratia marcescens CK-3 against phytopathogenic fungi and its enzymatic properties (식물(植物) 병원류(病源惟) 사상균(絲狀菌)에 길항력(拮抗力)을 갖는 Serratia marcescens CK-3의 분리(分離) 및 효소적(酵素的) 성질(性質))

  • Serratia marcescens CK-3, decomposing chitin which is a mar component of cell wall in phyitopathogenic fungi, was isolated from the continuous cropping rhizosphere of pepper and cucumber and its enzymatic property was examined. S. marcescens CK-3 was found tn have an tagonistic effects against, Fusarium axysporum and Rhizoctonia solani and to have complex enzyme system such as chitinase, laminarinase, and proteinase. The preferable composition of the medium for production of chitinase was fond and was as follows : colloidal chitin 1.5%, tryptone 0.5%, glucose 1.0%, peptone 0.2%, $MgSO_4{\cdot}7H_2O\;0.1%,\;K_2HPO_4\;0.1%,\;and\;NaCl\;0.1%$(w/v), pH 6.8. The maximum enzyme production was observed after culture of 72 hours at $30^{\circ}C$ using a medium containing the above chemical composition. The optimal pH and temperature for in vitro activity of chitinase from S. marcescens CK-3 were pH 7.5 and $50^{\circ}C$, respectively. The enzyme activity in-creased by metal ions such as$Ag^+$ and $Mn^{++}$.

Result of Voice Analysis after Laryngeal Microsurgery for Vocal Polyp in Elderly (노인에서 성대 용종의 후두 미세수술 후 음성검사 결과)

  • Background and Objectives: Vocal polyps arc one of the most frequent benign laryngeal diseases. They arc usually found at the midpoint of the vocal fold. They are mainly caused by vocal overuse. Vocal polyps arc usually removed surgically. Generally, age-related changes to speech are attributed to change in anatomy and physiology of the speech mechanism. These changes result in increased variability in the acoustic properties of speech with age. Still, not 'all studies of age-related changes in speech have taken differences between the young group and adult group after laryngeal microsurgery into account. The aim of this investigation was to compare improvement of acoustic analysis in young patients and elderly patients with vocal polyps, before and after the laryngeal microsurgery. Materials and Method: One hundred and twenty-eight patients who underwent laryngeal microsurgery for vocal polyps from 2008 through 2011 were reviewed retrospectively. 105 of the 128 patients under age 60 were classified as adult group (AG), and remaining 23 patients as elderly group (EG). The speech of AG and EG were evaluated before and after surgery for identification of differences for age group across measures of fundamental frequency (F0), Jitter, Shimmer and Maximum phonation time (MPT). Results: There were not significant differences between two groups for improvement of F0, Jitter, Shimmer, NHR, and MPT before and after surgery. The findings suggest that elderly group compares quite well with adult group in effectiveness of surgery. However, comparison between elderly group and young group (Age under 40) there was significant difference of improvement in Jitter and Shimmer. Conclusion: In general, the results of the present research showed significant improvement in vocal quality after phonosurgery of vocal polyp in both elderly and adult group. However, comparison of improvement between elderly group and young group, there were significant differences of improvement in jitter and shimmer. Therefore, in treatment planning of elderly group, we should consider age related changes of vocal cord.

  • Screening and identification of genetic resources based on their phytoconstituents and morphological characters potentially provide baseline data for researchers, breeders, and nutraceutical companies who wish to formulate a nutrient-dense diet and health beneficial supplement. Thus, we evaluated the amount of total phenolic content and major phenolic compounds; examined if phenolic compounds could be used as distinguishing factors for perilla genetic resources; and investigated the relation between some quantitative and qualitative morphological characters with the contents of phenolic compounds in 360 accessions obtained from National Agrobiodiversity Center gene bank, Jeonju, Korea. Total phenolic content (TPC) was estimated using Folin-Ciocalteu colorimetric assay. Individual phenolic compounds were determined using an Ultra-High Performance Liquid Chromatography system equipped with Photodiode Array detector. Considerable variations were observed in TPC (7.99 to 117.47 mg GAE/g DE), rosmarinic acid (RA) (ND to 19.19 mg/g DE), caffeic acid (CA) (ND to 0.72 mg/g DE), apigenin-7-O-diglucuronide (ADG) (ND to 1.24 mg luteolin equivalent (LUE)/g DE), scutellarein-7-O-glucuronide (SG) (ND to 4.32 mg LUE/g DE), and apigenin-7-O-glucuronide (AG) (ND to 1.60 mg LUE/g DE). RA was the most dominant phenolic compound in most accessions (95.3%) followed by SG. The adaxial leaf color was light green, green and dark green in 13.8%, 65.0%, and 21.1 % of the accessions, respectively. 78.8% of the accessions had light green color at the abaxial side with the remaining being described as green. Most of the accessions (96.9%) were cordate shape, the remaining being eclipse. Intensities of green pigment at abaxial and adaxial leaf surfaces were correlated with contents of individual phenolic compounds and TPC whereas leaf length and width had no correlation with TPC, CA and RA, and negatively correlated with ADG, AG, and SG. Leaf shape was not related with content of phenolic compounds, color of leaves, or the length or width of leaves. Accessions IT57426, IT157434, IT267710, and IT267712 which contained relatively high contents of TPC and major phenolic compounds (RA and SG) could be used for further research in breeding and bioassay test. Our study result showed the contents of total phenolics and individual phenolic compounds along with the morphological characters could be useful distinguishing factors for perilla genetic resources.

Effect of Nitrogen Fertilization on the Forage Growth, Yield and Quality of Native Reed (Phragmites communis) (갈대 생육지에서 질소 시비가 사초의 생육과 수량 및 사료가치에 미치는 영향)

  • This study was carried out to investigate the effect of nitrogen (N) fertilization on the forage growth, production and quality of native reed (Phragmites communis) grasses. Field experiments were conducted in Cheonan and in Ansan, 2012. Treatments were control (no N fertilization), 50 kg/ha and 100 kg/ha in Cheonan plots (fertilization on April $30^{th}$, and harvest on June $21^{st}$). Treatments in Ansan plots were control (no N fertilization) and 60 kg/ha (fertilization on May first, and harvest on August first). Forage growth and leaf colors were improved in N fertilized plots. However, the drymatter (DM) percentage was slightly decreased with N fertilization. Forage yields, in terms of DM, crude protein (CP) and digestible DM (DDM), were significantly increased with N fertilization in both sites. In Cheonan, DM, CP and DDM yields per ha were 4,026 kg, 235 kg and 1,850 kg, respectively, in the control plot, and were 4,658 kg, 306 kg and 2,388 kg, respectively, in the N 50 kg plot, and 5,622 kg, 446 kg and 3,143 kg, respectively, in the N 100 kg plot. In Ansan, DM, CP and DDM yields per ha were 2,802 kg, 177 kg and 1,288 kg, respectively, in the control plot, and were 3,876 kg, 294 kg and 1,853 kg, respectively, in the N 60 kg plot. Forage quality in terms of CP content, in vitro DM digestibility (IVDMD) and relative feed value (RFV) were also increased with N fertilization in both sites. In Cheonan, the CP content, IVDMD and RFV were 5.85%, 45.96% and 64.5 (grade 5), respectively, in the control plot, 6.58%, 51.27% and 72.3 (grade 5), respectively, in the N 50 kg plot, and 7.94%, 55.91% and 72.7 (grade 5), respectively, in the N 100 kg plot. In Ansan, the CP content, IVDMD and RFV were 6.30%, 45.98% and 70.2 (grade 5), respectively, in the control plot, and 7.59%, 47.80% and 78.3 (grade 4), respectively, in the N 60 kg plot. In conclusion, N fertilization of 60~100 kg/ha was desirable for greater forage production, with a higher quality of native Phragmites communis achievable. This should only be applied if the fertilization area is not located at a riverside/streamside or in riparian land where there is a high risk of water pollution by fertilization.

Antimutagenic and Cytotoxic Effects of Fagopyrum esculentum Moenech Noodles Extracts (메밀 국수 추출물의 항돌연변이원성 및 세포독성 효과)

  • This study was performed to determine the antimutagenic and anticancer effects of Fagopyrum esculentum Moenech noodles (FEMN) extracts using Ames test and cytotoxicity, respectively. FEMN made buckwheat wet noodles (BWN), buckwheat extruded noodles (BEN) and buckwheat dehydrated noodles (BDN) by 60% buckwheat flour and 70% buckwheat flour. The inhibitory effects of FEMN extracts on cell proliferation in A549, Hep3B, MCF-7, AGS and HeLa were investigated by SRB assay. The cytotoxic effects of FEMN against the cell lines with human lung carcinoma (A549), human gastric carcinoma (AGS), human hepatocellular carcinoma (Hep3B), human cervical adenocarcinoma (HeLa) and human breast adenocarcinoma (MCF-7) were inhibited with the increase of the extract concentration. The treatment of 1.0 mg/mL FEMN of 60% BEN extracts showed strong cytotoxicities of 74.7%, 75.3% and 70.5% against AGS, A549 and HeLa, respectively. The inhibition rate of 70% BWN of FEMN extracts in the S. Typhimurium TA100 strain showed 41% against the mutagenesis induced by MNNG. The inhibition rate of 70% BEN of FEMN extracts in the S. Typhimurium TA98 strain showed 45% against the mutagenesis induced by 4NQO.

A Study on the Knowledge and Awareness of Dental Hygienists and Dental Hygiene Students about Hepatitis B (치과위생사와 치위생과 학생의 B형 간염에 대한 지식과 인식에 관한 조사연구)

  • The purpose of this study was to survey the knowledge and awareness of dental hygienists and dental hygiene students about hepatitis B in an attempt to lay the solid foundation for the prevention of hepatitis B and infection control, as dental hygienists were highly likely to be exposed to HBV during job performance. The findings of the study were as follows: 1. 32.5% of the dental hygienists and 15.6% of the dental hygiene students were aware of their own HBsAg and HBsAb states. The dental hygienists who were cognizant of the states statistically significantly outnumbered the dental hygiene students who were (p=.000). 2. As for vaccination against hepatitis B, 71.8% of the dental hygienists and 47.3 % of the dental hygiene students were inoculated against it. The dental hygienists who were vaccinated against it outnumbered the dental hygiene students who were, and the disparity between the two was statistically significant (p=.000). 3. In the event of those who were vaccinated against hepatitis B, the dental hygienists were better cognizant than the dental hygiene students of the right time for the vaccination, required vaccination frequency (three times) and confirmation of the formation of hepatitis B antibody (p=.000). 4. Regarding awareness of hepatitis B infection route, the dental hygienists knew significantly better than the dental hygiene students that hepatitis B might be infected via blood (p=.030), sexual relations (p=.000), contaminated needle sticks (p=.000), mothers with hepatitis B positive during delivery (p=.000), toothbrushes/razors (p=.000) and exchange of drinking cups (p=.000). 5. As to the relationship between health status and knowledge on hepatitis B infection route, the respondents who were in bad shape had the best knowledge about that, followed by those in an average state of health and healthy respondents. And the dental hygienists had a significantly better knowledge than the dental hygiene students (p=.001). 6. Just a small number of the dental hygienists and students knew about the hepatitis B-related past experiences of their families, and the gap between the two was insignificant.

Conventional and Inverted Photovoltaic Cells Fabricated Using New Conjugated Polymer Comprising Fluorinated Benzotriazole and Benzodithiophene Derivative

  • A new conjugated copolymer, poly{4,8-bis(triisopropylsilylethynyl)benzo[1,2-b:4,5-b']dithiophene-alt-4,7- bis(5-thiophen-2-yl)-5,6-difluoro-2-(heptadecan-9-yl)-2H-benzo[d][1,2,3]triazole} (PTIPSBDT-DFDTBTz), is synthesized by Stille coupling polycondensation. The synthesized polymer has a band gap energy of 1.9 eV, and it absorbs light in the range 300-610 nm. The hole mobility of a solution-processed organic thin-film transistor fabricated using PTIPSBDT-DFDTBTz is $3.8{\times}10^{-3}cm^2V^{-1}s^{-1}$. Bulk heterojunction photovoltaic cells are fabricated, with a conventional device structure of ITO/PEDOT:PSS/polymer:$PC_{71}BM$/Ca/Al ($PC_{71}BM$ = [6,6]-phenyl-$C_{71}$-butyric acid methyl ester); the device shows a power conversion efficiency (PCE) of 2.86% with an open-circuit voltage ($V_{oc}$) of 0.85 V, a short-circuit current density ($J_{sc}$) of 7.60 mA $cm^{-2}$, and a fill factor (FF) of 0.44. Inverted photovoltaic cells with the structure ITO/ethoxylated polyethlyenimine/ polymer:$PC_{71}BM/MoO_3$/Ag are also fabricated; the device exhibits a maximum PCE of 2.92%, with a $V_{oc}$ of 0.89 V, a $J_{sc}$ of 6.81 mA $cm^{-2}$, and an FF of 0.48.

  • CdTe crystals were grown by the vertical Bridgman method. P-type DcTe crystals were grown with Cd:Te= 1:1.001 wt. % ratio, while n-type CdTe crystals were 1:1 Also, CdTe:In crystals were investigated, Lattic constants were 6.489${\AA}$ for p-type 6.480${\AA}$for n=type and 6.483${\AA}$ for CdTe:In EPD was 10$\^$-3/-10$\^$4/cm$\^$-2/ for n-, p-type CdTd, 10$\^$4/-10$\^$5/cm$\^$-2 for Cd:Te:In using by E-Ag solution for (111) plane The carrier concentration, the resistivity and the Hall carrier mobility measured by the van der Pauw method were p=5.78${\times}$10$\^$15/cm$\^$-3/, $\rho$=20.2$\Omega$cm, ${\mu}$$\sub$p/=75.6cm$\^$-2/ V$\^$-1/ sec$\^$-1/ for p-typem n=2.98${\times}$10$\^$16/cm$\^$-3/, $\rho$=0.214$\Omega$cm, ${\mu}$$\sub$p/=978.9cm$\^$-2/ V$\^$-1/ sec$\^$-1/ for n-type and n=7.45${\times}$10$\^$16/cm$\^$-3/, $\rho$=1.54 ${\times}$10$\^$3/$\Omega$cm, ${\mu}$$\sub$p/=658.4 cm$\^$-2/ V$\^$-1/ sec$\^$-1/ for CdTe:In crystals, Transmittance of p-type CdTe was 61% that of n-type was 65%, Cd:Te:In showed 60% IR transmittance.

Geology and Ore Deposits of Kubong Gold Mine (구봉광산(九峯鑛山)의 지질(地質)과 광상(鑛床))

  • Kubong Gold Mine is located in Kuryongri, Sayang-myun, Chungyang-gun, Choongchung-Namdo.(latitude $36^{\circ}24^{\prime}N$. longitude $126^{\circ}45^{\prime}30^{{\prime}{\prime}}E$) The mine was begun to work soon after the inhabitants of this village had accidently discovered the outcrops in April 1908. It is one of the largest gold mines in Korea which produces 4,500 tons of crude ore a month. The geology in the area consists of granitic gneiss, banded gneiss, augen-gneiss, mica schist, limesilicate of Pre-Cambrian series and sedimentary rocks(sandstones & conglomerates) of Daedong series. Basic dikes intrude the former formations. The country rock of the ore deposit is a group of the metamorphic rocks mentioned above. Gold-silver bearing quartz vein contains small amounts of pyrite, chalcopyrite, arsenopyrite, galena and sphalerite in which gold and silver occur as native state. The vein strikes $N30^{\circ}{\sim}60^{\circ}E$ and dips $20^{\circ}{\sim}50^{\circ}S$ and the average width of the vein is estimated 1 to 1.5m. Average grade of ore is Au:6~8gr/t and Ag:5~6gr/t. The ore shoot continues from the outcrop to the depth of -1760ML with dip of $20{\sim}25^{\circ}$ and strike extension reaches to 400m at the depth of -1440 ML and to more or less 200m at below. Highgrade of ore vein was found at the lowest level of the ore shoot at the time of recent field survey at the end of August 1970. Its average grade was estimated as Au:20gr/t and its width 1~2.5M in average. A series of futher prospecting for other new ore shoot or parallel veins are urgent and crosscut prospecting along the horizontal level is strongly recommended.

Purification and some Properties of Keratinolytic Protease Produced by Pseudomonas sp. KP-364. (Pseudomonas sp. KP-364가 생산하는 Keratinolytic Pretense의 정제 및 성질)

  • A keratinolytic protease was purified from the culture medium of Pseudomonas sp. KP-364 by use of an assay of the hydrolysis of feather keratin. Membrane ultrafiltration and DEAE-cellulose ion-exchange resin and Sephadex G-150 gel chromatographies were used to purify the enzyme. The specific activity of the purified keratinolytic protease relative to that in the original medium was approximately 72-fold high. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Sephadex G-150 chromatography indicated that the purified keratinolytic protease is monomeric and has a molecular weight of 36 kDa. The optimal pH and temperature of the keratinolytic protease activity were 6.6 and 37 C, respectively, and the keratinolytic protease was relatively stable at pH value from 3.0 to 10.0 at 37 C for 1hour. The keratinolytic protease was inhibited by EDTA and EGTA, indicating that the keratinolytic protease was a kind of metalloprotease that require Li+ for cofactor.
