  • The structure of tuguaconitine, MP $196{\sim}198^{\circ}C$, a new diterpene alkaloid isolated from the roots of Aconitum sibiricum P., was studied. Its empirical formula is $C_{23}H_{35}NO_7$ and spectroscopic evidence shows that the base contains three methoxyl, three hydroxyl and one N-ethyl groups. The acute toxicities of most of Aconitum species were strong and the acute toxicity of the alkaloid fraction of A. sibiricum was decreased by Glycyrrhizae, black bean or Zingiberis extract.

A Study on the Acute Toxicity and Accumulation of Mercuric Chloride to Goldfish (Carassius auratus) (Mercuric Chloride가 붕어(Carassius auratus)에 미치는 급성독성 및 축적에 관한 연구)

  • This experiment was performed to study the acute toxic effects of mereuric chloride(HgCl$_{2}$) in goldfish (Carassius aurams). In the study, Lc$_{50}$ values were determined for each 24, 48, 72 ,and 96-hr and mercury bioassay during the tests were performed. in five parts gill, kidney, liver, muscle and egg. The results isummarized were as follows 1. In the range of acute lethal toxicity for goldfish was 0.15-1.50 mg/l and the Lc$_{50}$ values decreased with exposure time. 2. The Lc$_{50}$ values of HgCl$_{2}$ for goldfish at 24, 48, 72 and 96 hour were 0.64, 0.53, 0.46, and 0.39 mg/l, respectively. 3. In the 400 $\mu$g/l treatment group, the average levels of mercury residues in the gill, kidey, liver, muscle and egg during 96 hour were 107.02 $\mu$g/g, 8.65 $\mu$g/g, 6.87 $\mu$g/g, 0.61 $\mu$g/g, and 0.82 $\mu$g/g, respectively. 4. The mercuny residues of gill was up to 12-175 times greater than mercury contents found in the other tissues.

Pharmacological Studies on Ether Fraction of Corni Fructus (산수유 에텔분획물의 약리작용에 관한 연구)

  • The ether fraction obtained from dry fruit of Cornus officinalis was investigated for the anti-inflammatory action, acute toxicity and central nervous system activities. From several pharmacological examinations, it was found that the ether fraction suppressed considerably carrageenin edema at the dose of 600mg/kg p.o. in rats, inhibited the granuloma formation in rats as given 100mg/kg p.o. for 7 days and decreased the swelling of both of complete adjuvant injected and noninjected (contralateral) paws of the rats at the dose of 100mg/kg p.o. given for 14 days. The $LD_{50}$ of the fraction are estimated to be more than 2,000mg/kg p. o. and 642mg/kg i. p. in mice. And the fraction did not show any sedative, stimulative, analgesic and anticonvulsant action but exhibited hypothermic action. These results might be concluded that the ether fraction of Corni fructus showed anti-inflammatory action in both of acute and chronic type models without any considerable central nervous depressant activity and exhibited very weak acute toxicity in mice.

  • Single subcutaneous administration to S.D rats of both sexes was performed to investigate the acute toxicity of Syndella gel, a new topical drug containing deproteinised dialysate of calf's blood and micronomicin sulfate. $LD_{50}$ values for S. D rats were 23,047 mg/kg for male and 23,725 mg/kg for female. The death occurred within 24 hours after administration at doses over 19,200 mg/kg. The main cause of death seemed to be respiratory disturbance by acute shock. Major general symptoms induced by injection subcutaneously with Syndella gel were underactivity, decreased respiratory rate, salivation, tremor and loss of consciousness. No significant body weight changes and gross findings of internal organs in treatment groups in comparison with those of control groups was observed at any dose levels in Syndella gel.

  • A 47-year-old woman ingested about 12 mg of colchicine with suicidal intent. Colchicine, a highly poisonous alkaloid, is a commonly used treatment for gout, Bechet's disease, and familial Mediterranean fever. Despite the knowledge of its side effects, the risk of a significant overdose is under-appreciated. She suffered from acute multisystem toxicity, including gastrointestinal disorders, bone marrow suppression, alopecia, and probable pancreatitis, but she ultimately recovered with supportive therapy. We report a case of acute colchicine toxicity from a single overdose with a review of the literature.

Evaluation of Alternative Acute Toxicity Test Method with Daphnia magna (Toxkits에 의한 물벼룩 급성독성시험방법의 제도적용 적합성 평가)

  • This paper aimed to evaluate the results of alternative acute toxicity test on 83 wastewater samples. Each sample was tested by traditional method (using laboratory cultured D. magna as a test organism) and alternative method (using Toxikit which can be purchased from a specialized company). The relationship between Lab. culture toxicity and Toxkits toxicity indicated good relation ($r^2=0.84$, p<0.01, n = 83). Number of samples which showed a small difference of lower than 0.5 between two test methods were 52 and they account for 63 percent of collected samples. In addition, these 52 samples had a lower average toxicity of TU 0.5 (Lab. culture method) and TU 0.45 (Toxkits method). Whereas samples which indicated big difference of test results between two methods, had a tendency to show higher toxicity. From these results, alternative toxicity test method could be applied to the official test method, if samples would have a lower toxicity less than TU 2. Also, Toxikit standard toxicant test results indicated $EC_{50}$ values between 0.93 and 1.68 mg/L and these results were considered as valid for quality control standard.

  • Smoke toxicity is the test for the toxicity evaluation of smoke and hazardous gas, caused by combustion of building materials and finishing materials. Smoke toxicity can be evaluated by the mean incapacitation time of mice. This test result can be influenced by the health status of mice and test condition. In acute inhalation toxicity test of hazardous gas, no typical clinical findings and histopathologic abnormalities were observed. Tracheitis and bronchitis as well as acute lung inflammation around terminal bronchiole in some mouse of the highest dose group. Through this study, we established the method for inhalation toxicity test of hazardous gas as well as the SOP of inhalation toxicity test. However, in the future studies, the concentration control methods for inhalation technologies on hazardous gas will be needed to improve continuously and also further studies on other gas inhalation toxicity will be needed to conduct.

Toxicity Assessment and Decomposition Characteristics of Triclosan in an E-beam Irradiation Process (전자빔 공정을 적용한 Triclosan의 제거특성 및 독성평가)

  • The abjective of this study was to study the degradation behavior and acute toxicity assessment of Triclosan and acute toxicity under E-beam irradiation. The experiments were conducted to investigate the efficts of the degradation efficiency in the initial concentration of Triclosan and the irradiation capacity of E-beam and the degree of mineralization based on a change of scavenger gas. The biological toxicity test by using on green algae, Pseudokirchneriella Subcapitata was conducted to lead the reducing toxicity. Degradation efficiency of Triclosan was improved when E-beam irradiation intensity was higher. Additionally, the % of TOC removal in each Radical scavenger gas was increased as the follows orders: $N_2O$ > $O_2$ > $N_2$, The toxicity test showed that the toxicity effect after 4 days(96hrs) was decreased by increase of E-beam irradiation intensity.

Single oral toxicity test and safety classification for Kaempferia parviflora (흑생강의 단회투여독성시험과 안전성등급화)

  • Objectives : Kaempferia parviflora Rhizome is black ginger indigenous to Laos and Thailand. It has been used as a folk medicine to improve blood flow and promote vitality and longevity with good health and well being. For these reasons, Kaempferia parviflora Rhizome has been focused on developing it as a food or food supplement. In addition, Kaempferia parviflora Rhizome could be under consideration of new prescription based on its characteristic compounds, polymethoxyflavonoids. However, it needs to be certified as safe before it can be used. Here, a single-oral toxicity test and safety classification was carried out to identity acute information of the toxicity of Kaempferia parviflora Rhizome powder and to make sure of its safety in clinical applications. Methods : Test substance was orally administered to male and female SD-rat at dose levels of 5000 mg/kg to estimate approximate lethal dose(ALD). Based on the acute information of the toxicity, the safety classification was estimated using the HED(human equivalent dose)-based MOS(margin of safety). Results : At 14 days after treatment with test substance. there were no of test substance related with mortalities and clinical signs. In addition, no changes in the body or organ weights and no gross or histopathological findings were observed. Thus, the ALD of Kaempferia parviflora Rhizome powder was considered over 5,000 mg/kg in both female and male mice. Conclusions : Based on the single oral toxicity test using the highest and limit dose, 5,000 mg/kg and the decision guideline for safety classification based on HED-based MOS, it was estimated that Kaempferia parviflora Rhizome powder is classified as "Specified class B" indicating that clinical dose is not limited to patients as safe as food.

Comparing Acute and Swimming Endpoints to Evaluate the Response of Two Freshwater Midge Species, Chironomus yoshimatsui and Chironomus riparius to Heavy Metals (요시마쯔깔따구와 리파리깔따구(파리목: 깔따구과)의 중금속에 대한 급성독성 및 유영능력 비교)

  • The relative sensitivity of two freshwater non- biting midges, Chironomus yoshimatsui Martin and Sublette and C. riparius Meigan, was examined for lead, cadmium, and mercury in water- only exposures. Two endpoints were compared to assess toxicity 48 h and 96 h after exposure: Acute toxicity ($50\%$ lethal concentration: $LC_50$) and behavioral toxicity ($50\%$ effective concentration: $EC_{50}$). for the behavioral toxicity, reduction of swimming performance of two midge species in the treated conditions was compared to that in the untreated control. The sensitivities differed depending on the species and heavy metals, although some trends emerged. $LC_50$ values in C. yoshimatsui to cadmium and lead were always higher than those in C. riparius with increasing toxicity, regardless of the exposure times. The opposit was true for the mercury treatment. Similar trends were observed in the $EC_{50}$ values. The $EC_{50}$ values were always lower than the $LC_50$ values in all the treatment cases (midge species, heavy metals, and exposure times). These results indicate that the two midge species respond to the heavy metals differently: C. riparius is sensive to cadmium and lead and C. yoshimatsui to mercury. Behavioral toxicity such as swimming performance can be an effective endpoint for assessing heavy metal toxicity in water.
