Requirement of Reactive Oxygen Species Generation in Apoptosis of MCF-7 Human Breast Carcinoma Cells Induced by Sanguinarine

  • Although sanguinarine, a benzophenanthridine alkaloid, possesses anti-cancer properties against several cancer cell lines, the molecular mechanisms by which it inhibits cell growth and induces apoptosis have not been clearly understood. In order to further explore the critical events leading to apoptosis in sanguinarine-treated MCF-7 human breast carcinoma cells, the following effects of sanguinarine on components of the mitochondrial apoptotic pathway were examined: generation of reactive oxygen species (ROS), alteration of the mitochondrial membrane potential (MMP), and the expression changes of Bcl-2 family proteins. We show that sanguinarine-induced apoptosis is accompanied by the generation of intracellular ROS and disruption of MMP as well as an increase in pro-apoptotic Bax expression and a decrease of anti-apoptotic Bcl-2 and Bcl-xL expression. The quenching of ROS generation with N-acetyl-L-cysteine, the ROS scavenger, protected the sanguinarine-elicited ROS generation, mitochondrial dysfunction, modulation of Bcl-2 family proteins, and apoptosis. Based on these results, we propose that the cellular ROS generation plays a pivotal role in the initiation of sanguinarine-triggered apoptotic death.

Paramyrothecium eichhorniae sp. nov., Causing Leaf Blight Disease of Water Hyacinth from Thailand

  • Paramyrothecium eichhorniae sp. nov. was observed and collected from Chiang Mai and Phetchaburi Provinces, Thailand. This new species is introduced based on morphological and molecular evidence. This fungus is characterized by its production of sporodochium conidiomata with a white setose fringe surrounding an olivaceous green to dark green slimy mass of conidia, penicillately branched conidiophores, and aseptate and cylindrical to ellipsoid conidia. Phylogenetic analyses of combined LSU rDNA, ITS rDNA, tef1, rpb2, tub2 and cmdA sequence data using maximum parsimony, maximum likelihood and Bayesian approaches placed the fungus in a strongly supported clade with other Paramyrothecium species in Stachybotryaceae (Hypocreales, Sordariomycetes). The descriptions of the species are accompanied by illustrations of morphological features, and a discussion of the related taxa is presented.

Upregulation of thioredoxin and its reductase attenuates arsenic trioxide-induced growth suppression in human pulmonary artery smooth muscle cells by reducing oxidative stress

  • The thioredoxin (Trx) system is an important enzymatic complex involved in cellular redox homeostasis. Arsenic trioxide (ATO; As2O3) is known to trigger cell death in vascular smooth muscle cells (VSMCs) via oxidative stress. In the present study, the effects of changes in thioredoxin 1 (Trx1) and Trx reductase1 (TrxR1) on cell growth, death, reactive oxygen species (ROS), and glutathione (GSH) levels were evaluated in ATO-treated human pulmonary artery smooth muscle cells (HPASMCs). ATO inhibited growth and induced cell death in the HPASMCs at 24 h. Overexpression of Trx1 and TrxR1 using adenoviruses attenuated cell growth inhibition caused by ATO and partially prevented cell death. ATO increased ROS levels including the mitochondrial superoxide anion (O2•-) at 5 min. Administration of adTrx1 or adTrxR1 reduced the increased mitochondrial O2•- level in these cells. HPASMCs treated with Trx1 or TrxR1 siRNA showed increases in ROS levels with or without treatment of ATO at 5 min. Although ATO transiently increased GSH levels at 5 min, Trx1 and TrxR1 siRNAs reduced the increased GSH levels in these cells. In addition, PX-12 (a Trx1 inhibitor) and auranofin (a TrxR1 inhibitor) diminished the cellular metabolism in HPASMCs at 4 h, accompanied by an increase in ROS level and a decrease in GSH level. In conclusion, upregulation of Trx1 and TrxR1 somewhat decreased cell growth inhibition and death in ATO-treated HPASMCs, which was accompanied by reduced oxidative stress.

Mitochondrial COI sequence-based population genetic analysis of the grasshopper, Patanga japonica Bolívar, 1898 (Acrididae: Orthoptera), which is a climate-sensitive indicator species in South Korea

  • Patanga japonica Bolívar, 1898 (Orthoptera: Acrididae) is listed as a climate-sensitive indicator species in South Korea and is called southern group of insects in that the main distributional range is southern region of South Korea and Asian continent. In South Korea, thus, the species was distributed mainly in southern region of South Korea including southward a remote Jeju Island, but recently the species has often been detected in mid to northern region of South Korea, implying northward range expansion in response to climate change. Understanding the characteristics of the changes in genetic diversity during range expansion in response to climate change could be a foundation for the understanding of future biodiversity. Thus, in this study, we attempted to understand the changing pattern of the genetic diversity of the P. japonica in newly expanded regions. For the purpose of study, we collected 125 individuals from seven localities throughout South Korea including two newly distributed regions (Pyeongtaek and Yeongwol at ~37° N). These were sequenced for a segment of mitochondrial cytochrome oxidase subunit I (COI) and analyzed for genetic diversity, haplotype frequency, and population genetic structure among populations. Interestingly, northward range expansion accompanied only haplotypes, which are most abundant in the core populations, providing a significant reduction in haplotype diversity, compared to other populations. Moreover, genetic diversity was still lower in the expanded regions, but no genetic isolation was detected. These results suggest that further longer time would take to reach to the comparable genetic diversity of preexisting populations in the expanded regions. Probably, availability of qualified habitats at the newly expanded region could be pivotal for successful northward range expansion in response to climate change.

Effects of Soil Environment on the Growth of Pinus Thunbergii and Zelkova Serrata at the Reclaimed Seaside (임해매립지의 토양환경이 곰솔과 느티나무의 생육에 미치는 영향)

  • The purpose of thus paper is to provide the knowledge on preparing for the planting soil and planting method, and maintenance at the reclaimed seaside. Based on the collected data from the field work, the soil environment, the growth of height, inter-node, tree ring and roots of the two species had been analyzed. The determinant of soil factors, affecting the growth of trees, turned out to be six elements such as soil hardness, soil acidity, potassium, calcium, magnesium and total nitrogen. Because the variances of both growth of tree height and tree ring are greater than that of root, the growth characteristics of ground parts of the species by the individual tree species is more dynamical than those of underground parts. From the mean difference test the growth of height, root between Pinus thunbergii and Zelkova serrata, have been turned out to be statistically significant at 5 percent level. Pinus thunbergii is a sapling, so it grows faster than Zelkova serrata while Pinus thunbergii has better roots system than Zelkova serrata. From the correlation analysis, it showed the very strong correlation between tree height growth and potassim, while the lowest correlation coefficient was between soil hardness and potassim as 0.744. From the multiple regression analysis, both soil hardness and magnesium affect to the tree growth, soil hardness and potassium to the tree growth, potassium and calcium to the rot growth, respectively. Using this research results, we can be use the planting plan including revegetation, construction and maintenance of the reclaimed seaside. In the future, the planting method including the ground preparation and tree species selection for the reclaimed seaside should be accompanied in advanced through the soil survey and relevant analysis.

Ginsenoside Rg1 ameliorates chronic intermittent hypoxia-induced vascular endothelial dysfunction by suppressing the formation of mitochondrial reactive oxygen species through the calpain-1 pathway

  • Background: As the major pathophysiological feature of obstructive sleep apnea (OSA), chronic intermittent hypoxia (CIH) is vital for the occurrence of cardiovascular complications. The activation of calpain-1 mediates the production of endothelial reactive oxygen species (ROS) and impairs nitric oxide (NO) bioavailability, resulting in vascular endothelial dysfunction (VED). Ginsenoside Rg1 is thought to against endothelial cell dysfunction, but the potential mechanism of CIH-induced VED remains unclear. Methods: C57BL/6 mice and human coronary artery endothelial cells (HCAECs) were exposed to CIH following knockout or overexpression of calpain-1. The effect of ginsenoside Rg1 on VED, oxidative stress, mitochondrial dysfunction, and the expression levels of calpain-1, PP2A and p-eNOS were detected both in vivo and in vitro. Results: CIH promoted VED, oxidative stress and mitochondrial dysfunction accompanied by enhanced levels of calpain-1 and PP2A and reduced levels of p-eNOS in mice and cellular levels. Ginsenoside Rg1, calpain-1 knockout, OKA, NAC and TEMPOL treatment protected against CIH-induced VED, oxidative stress and mitochondrial dysfunction, which is likely concomitant with the downregulated protein expression of calpain-1 and PP2A and the upregulation of p-eNOS in mice and cellular levels. Calpain-1 overexpression increased the expression of PP2A, reduced the level of p-eNOS, and accelerated the occurrence and development of VED, oxidative stress and mitochondrial dysfunction in HCAECs exposed to CIH. Moreover, scavengers of O2·-, H2O2, complex I or mitoKATP abolished CIH-induced impairment in endothelial-dependent relaxation. Conclusion: Ginsenoside Rg1 may alleviate CIH-induced vascular endothelial dysfunction by suppressing the formation of mitochondrial reactive oxygen species through the calpain-1 pathway.

The distribution of phytoplankton in Yeong-il Bay, Korea (迎日 의 植物플랑크톤 分布)

  • The interrelationships between distribution of phytoplankton and hydrographical conditions were studied from December 1983 to July 1984 in Yeongil Bay. The temporal variation of phytoplankton abundance seems to be mainly influenced by the vertical stability of water column and the freshwater run-off accompanied with possible nutrient input. The temporal progression of dominant species shows general succession pattern, especially skeletonema costatum was found a dominant species in each month. Although the differences between concentration of chlorophyll a and abundance of phytoplankton may be caused by cell size variation, during the study period they show felatively good linear manner. The phytohydrographic region based on multivariate analysis reflects the relationships between hydrographical condition and distribution pattern of phytoplankton.

  • Cobitis sinensis-longicorpus complex considered as hybrid origin between C. sinensis and C. longicorpus coccurred ommonly in the upper streams of the Nakdong River, Korea. Histological examinations of their gonad were accompanied with 272 individuals of C. sinensis-longicorpus complex collected. Most of fishes collected were females, however, only 6 individuals were found males. The ovarian tissues of females are completely fertile undergoing normal oogenensis. In the male gonads, testicular lobule structure with abnormal vacuolar tissues were observed. Spermatogonia and spermatocytes were also observed of their testis however spermatids or sperms were not shown in their developmental stages. From these facts, we infer that female population of C. sinensis-longicorpus complex may be unique reproductive hierarchy accomplishing their reproduction with participation of males of their closely related bisexual species.

  • The present investigation was undertaken to find out the anticancer activity of monoterpene compounds. Monoterpenes showed generally the inhibitory effect on the proliferation o f L1210 cancer cells (cytotoxicity). Geraniol was found to exibit the most potent cytotoxic effect on L1210 cells with an IC50 values of $0.67{\mu}g/ml$. On the other hand, geraniol proved to be capable of stimulating the macrophage proliferation (135% of control). When the life prolonging activity of geraniol by daily oral administration of 0.1~10${\mu}g/10{\mu}l/20$ g body weight to Sarcoma 180 bearing ICR mouse was examined, there was also a significant elevation of survival (best result of 134% of control). The contradictory effects of geraniol on the proliferation of L1210 cells and macrophages proved to be accompanied by the coincident alterations of RNS (reactive nitrogen species) related enzymes activities such as inducible nitric oxide synthase (Inos) in macrophages and ROS (reactive oxygen species) related enzymes activities such as superoxide dismutase (SOD) in L1210 cells, respectively.

Reactive Oxygen Species are Involved in Y-27632-induced Neurite Outgrowth in PC12 Cells

  • Inhibition of Rho-associated coiled coil-containing kinase (ROCK) has been reported to promote differentiation of neuronal cells. Here, we examined the effect of Y-27632, a ROCK inhibitor, on the outgrowth of neurites in PC12 cells. Y-27632 caused a rapid induction of neurite outgrowth in PC12 cells in a time-dependent manner. The neurite outgrowth, triggered by Y-27632, was accompanied by Rac1 activation, and was attenuated by Rac1 inhibitor NSC23766, in a concentration-dependent manner. Y-27632 also induced an increase in the production of reactive oxygen species (ROS). Pretreatment with N-acetylcysteine, an ROS scavenger, inhibited the ROS generation and neurite outgrowth in response to Y-27632. These results indicate that the activation of Rac1 and the generation of ROS contribute to the neurite outgrowth triggered by Y-27632 in PC12 cells.
