Antioxidant and pancreatic lipase inhibitory activities of Anemarrhena asphodeloides (지모 추출물의 항산화 및 pancreatic lipase 저해 활성 평가)

  • In this study, the antioxidant and pancreatic lipase inhibitory activities of aqueous methanolic (70% methanol) extract from the roots of Anemarrhena asphodeloides were investigated. The extracts of four solvent fractions (the n-hexane layer, EtOAc layer, n-BuOH layer, and $H_2O$ layer) of the 70% methanol extract were also investigated. Furthermore, the total phenolic content was quantified using a spectrophotometric method. All the tested samples showed dose-dependent radical scavenging and pancreatic lipase inhibitory activities. In particular, the pancreatic lipase inhibitory activity of the ethyl acetate soluble portion (the EtOAc layer) from the rhizomes of the A. asphodeloides was higher than that of the other solvent-soluble portions. The antioxidant property of the extracts was evaluated using radical scavenging assays with DPPH and $ABTS^+$ radicals. 1000 mg/ml of the n-BuOH layer extract showed 91.2% DPPH radical scavenging activity. The EtOAc layer extract and the n-BuOH layer extract showed $IC_{50}=20.5{\pm}1.7mg/ml$ and $IC_{50}=50.5{\pm}0.7mg/ml$ $ABTS^+$ radical scavenging activities, respectively. The anti-obesity efficacy of the A. asphodeloides extract was tested via porcine pancreatic lipase assay. A pancreatic lipase inhibitory activity ($IC_{50}$) of $31.3{\pm}0.1mg/ml$ was obtained from the EtOAc layer extract. These results suggest that A. asphodeloides can be considered a new potential source of natural antioxidant and anti-obesity agents.

The Evaluation on the Effectiveness of Zingiber mioga extract as a Cosmetic Material through Verification of Cosmeceutical Activations (양하(Zingiber mioga) 추출물의 화장품 약리활성 검증을 통한 화장품 소재로서의 유효성 평가)

  • The purpose of this study was to confirm the possiblility of Zingiber mioga as a cosmetic material. For this we carried out biologically activated material characteristic evaluation about anti-oxidant, anti-inflammatory, wrinkle reduction effects using Zingiber mioga extract. To carry out this experiment, we extracted Zingiber mioga extract from Zingiber mioga flowers exract (ZMF) and Zingiber mioga leaves exract (ZML) with 70% ethanol. In order to evaluate the anti-inflammatory effect, we tested the toxicity and the hindrance activity to nitric oxide of samples using macrophages (RAW 264.7 cells). After we measured DPPH radical scavenging activity, ABTS+ radical scavenging activity and superoxide dismutase (SOD)-like activity of the Zingiber mioga extracts, we knew that they increased depending on their concentration. ZMF showed higher antioxidant activity than ZML after the measurement of ABTS+ radial scavenging activity and superoxide desmutase (SOD)-like activity. According to the measurement result of Nitric oxide inhibition activity we knew that ZMF reduced NO productions in a concentration-dependent manner. After the measurement of the biosynthesis quantity of pro-collagen type-1, we knew that its excellent effect appeared 110% or more at the concentration of 25 ㎍/ml. And at the same concentration, the result of the measurement of metalloprotease (MMP)-1 inhibition effect showed the 20% activation. In conclusion, ZMF is expected to be applied as a cosmetic material for wrinkle reduction. Zingiber mioga is believed to be used as a natural cosmetic material because it has been proven to have antioxidant, anti-inflammatory, and wrinkle-improving effects.

Antioxidant Activities and Antimicrobial Effects of Extracts from Auricularia auricula-judae (목이버섯(Auricularia auricula-judae) 추출물의 항산화 활성 및 항균 효과)

  • This study investigated the antioxidant and antimicrobial activities of various solvents (acetone, ethyl acetate, and ethanol) for extraction of Auricularia auricula-judae. Antioxidant activity was evaluated by determining total polyphenol and flavonoid contents, 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical scavenging activity. Total polyphenol and flavonoid contents were not significantly different among the extracts, whereas DPPH radical scavenging activity and ABTS cation radical scavenging activity were significantly higher in ethanol and acetone extracts. DPPH radical scavenging activities of ethanol and acetone extracts showed high values (58.7% and 46.7%, respectively). The antimicrobial properties of these extracts were determined against six bacterial pathogens (Bacillus subtilis, Staphylococcus aureus, Micrococcus luteus, Escherichia coli, Pseudomonas aeruginosa, and Enterobacter cloacae) by the disc diffusion method. The acetone extracts showed antimicrobial activities against all tested bacteria, and all extracts showed the highest antimicrobial activity against B. subtilis.

Antioxidant Contents and Antioxidant Activities of White and Colored Potatoes (Solanum tuberosum L.)

  • This study was performed to evaluate and compare the antioxidant substance content and antioxidant activities of white (Superior) and colored (Hongyoung, Jayoung, Jasim, Seohong, and Jaseo) potatoes. The potatoes were extracted with 80% ethanol and were evaluated for the total polyphenol, flavonoid, and anthocyanin contents and for 1,1-diphenyl-2-picrylhydrazyl (DPPH)/2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity, reducing power, and ferrous metal ion chelating effect. The total polyphenol, flavonoid, and anthocyanin contents of Hongyoung and Jayoung were higher than white and other colored potatoes. All colored potato extracts, except for Jaseo and Seohong, showed higher ABTS radical scavenging activities than the general white potato extract. Hongyoung and Jayoung had the highest ABTS and DPPH radical scavenging activities. Optical density values for the reducing power of Jayoung and Jaseo at concentration of 2 mg/mL were 0.148 and 0.090, respectively. All colored potato extracts had lower ferrous metal ion chelating effect than the white potato. A significant (P<0.05) positive correlation was observed between total polyphenol content and total flavonoid content (r=0.919), anthocyanin content (r=0.992), and ABTS radical scavenging activity (r=0.897). Based on these results, this research may be useful in developing the Hongyoung and Jayoung cultivars with high antioxidant activities.

A Comparison of Antioxidant Activity from Schisandra chinensis Water Extracts depending on Stir-frying and Stir-frying with Liquids Process (청초와 주정 전처리에 따른 오미자 물 추출물의 항산화 활성 비교)

  • Schisandra chinensis (Sc), a species of the family Schisandraceae used fruits as medicinal herbs. In the study, we performed to determine a comparison of antioxidant activity from Sc water extracts depending on stir-frying and stir-frying with liquids by 30% ethanol pretreatments. The contents of polyphenol and flavonoid, DPPH radical scavenging, ABTS radical scavenging and reducing power activity were measured to evaluate the antioxidant activity. Total polyphenol and flavonoid contents of Sc extracts were increased in stir-frying and 30% ethanol stir-frying pretreatments compared to a control. In particular, contents of stir-frying pretreatments Sc were highest increased at $220^{\circ}C$ treatments. The DPPH radical scavenging activity were highest increased in stir-frying pretreatments at $220^{\circ}C$. And result that measurement of the ABTS radical scavenging, it showed higher activity in 30% ethanol stir-frying pretreatments at $180^{\circ}C$ and $220^{\circ}C$. But Sc in the $140^{\circ}C$ showed higher activity in stir-frying pretreatments. In the reducing power activity, stir-frying pretreatments increased higher than 30% ethanol stir-frying pretreatments at $220^{\circ}C$. As in results, antioxidant activity of Sc water extracts in stir-frying and stir-frying with liquids process increased higher than a control, and it was most effectively in stir-frying pretreatments at $220^{\circ}C$.

Effects of medium supplemented with persimmon peels on antioxidant activity of Pleurotus eryngii (감과피 첨가배지가 큰느타리 버섯의 항산화활성에 미치는 영향)

  • In this study, we investigate the potential use of persimmon peels (PP) in mushroom culture medium for the production of functional mushrooms. Pleurotus eryngii was cultivated in medium supplemented with PP (SMPP) at the following concentrations: 0% SMPP (control), 5% SMPP, 10% SMPP, 15% SMPP, 20% SMPP, or 30% SMPP. The total polyphenol content, DPPH radical scavenging ability, ABTS cation scavenging ability, and reducing power of P. eryngii cultivated in SMPP were investigated. P. eryngii cultured in 20% SMPP produced the highest values for all four measurements. Total polyphenol content, DPPH radical scavenging activity, ABTS cation scavenging ability, and reducing power all increased upon the addition of PP. Based on our results, we can conclude that persimmon peels are a highly valuable supplement for functional mushroom culture medium.

Antioxidant and Neuronal Cell Protective Effects of Eugenia caryophyllata Thunb. by Extraction Solvent (용매별 정향 추출물의 항산화 및 신경세포 보호 효과)

  • This study examined the antioxidant and neuronal cell protective effects of the water and methanol extracts of Eugenia caryophyllata Thunb. The total polyphenol content was significantly higher in the methanol extract than in the water extract. The DPPH radical scavenging activity in the water extract was similar to Vit. C at a concentration of $100{\sim}200{\mu}g/mL$. The ABTS radical scavenging activity in the water and methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The superoxide dismutase (SOD)-like activity in the methanol extract was similar to Vit. C at a concentration of $800{\sim}1,000{\mu}g/mL$. The DPPH, ABTS radical scavenging and (SOD)-like activity increased with increasing extract concentration. In a cell viability using MTT, the water extract (50 and 100 ppm) and methanol extract (100 ppm) had a protective effect against $H_2O_2$-induced neurotoxicity.The result ssuggest that the extract of E. caryophyllata Thunb. has antioxidant activities and may be useful for treating neurodegenerative disorders.

Antioxidative Activities of Methanol Extracts from Different Parts of Chrysanthemum zawadskii (구절초의 부위별 메탄올 추출물의 항산화활성)

  • The major objective of this study was to investigate the antioxidant activities of methanolic extracts from different parts (flower, leaf stem, and root) of Chrysanthemum zawadskii by employing various in-vitro assay systems. The extraction yields from the flower, leaf stem, and root were 18.347, 12.93, and 11.33-----, respectively. The total polyphenol content was highest in the flower (17.16 mg/100 g) and lowest in the root (11.33 mg/100 g). The antioxidant activities were raised within creasing amounts of extracts, and the extracts from the flower showed the highest effect on the superoxide anion radical scavenging, metal chelating on ferrous ions and reducing power. In addition, the leaf stem also showed good antioxidant activity in various systems. These results suggest that the methanolic extracts from the flower and leaf stem possess excellent antioxidant activities and may thus serve as potential sources of natural antioxidants.

Antioxidant Effects of Isoflavones from the Stem Bark of Cudrania tricuspidata

  • Isoflavones 4'-O-methylalpinumisoflavone (1) and 5, 7, 4'-trihydroxy-6,8-diprenylisoflavone (2) were isolated from stem bark of Cudrania tricuspidata (Carr.) Bureau. These compounds were the first reported from this plant. Antioxidant activities 1 and 2 were evaluated by measuring their ability to scavenge DPPH and ABTS radicals. Diprenylisoflavone 2 showed strong scavenging activity against ABTS ($IC_{50}\;=\;16.3\;{\mu}M$), three-fold higher compared to genistein, which do not possess prenylated group which indicates strong scavenging activity against ABTS radical of isoflavone 2 was due to prenyl group.

Antioxidant Effects of Ethanol Extracts from Flower Species of Compositae Plant (국화과 식물 중 꽃 에탄올추출물의 항산화효과)

  • The present experiments were carried out to develop new antioxidants from EtOH extract of flower resources of Compositae plant concerning total polyphenol and flavonoid content, scavenging activities on DPPH and ABTS radicals, ferrous ion chelating effects, and inhibition activity on lipid peroxidation of linoleic acid. EtOH extracts from Matricaria recutica, Cosmos bipinnatus, Synurus deltoides, and Aster pilosus showed higher level of total polyphenol content, and the flavonoid content was the highest in C. bipinnatus. However, scavenging activity on DPPH radicals was the highest in EtOH extract of Hieracium pilosella-1.1 times higher than BHT control. The activity on ABTS radical scavenging was the highest in EtOH extracts of M. recutica and S. deltoides-2.0 and 1.2 times higher than ascorbic acid, and 2.2 and 1.3 times higher than BHT, respectively. Ferrous ion chelating effects was also the highest in M. recutica, but the level was much lower than EDTA. Inhibition activity on lipid peroxidation of linoleic acid, measured at every 4 days for 32 days, was superior and longer lasting with M. recutica and Achillea alpina, compared to that of BHT. In conclusion, antioxidant activity was different depending on species, so selection of proper plant species for the development of potential antioxidant is very important.
