This study was conducted in order to investigate the antioxidant and antimicrobial activity of Solanum nigrum L. fruit powder after undergoing different extraction solvent processes. The total phenolic content of Solanum nigrum L. fruit powder measured a 14.66 GAE mg/g after undergoing ethanol extraction, and the total flavonoid content measured at 201.23 mg CE/g when undergoing ethanol extraction. The ABTS radical scavenging activity was 160.38~209.53 TEAC umol/g, and the DPPH radical scavenging activity was 53.99~90.76 TEAC umol/g, which indicated a higher level of antioxidant power in the ethanol extract as opposed to in the water extract. The FRAP (ferric reducing antioxidant power) of Solanum nigrum L. fruit powder was 115.58~194.58 TEAC umol/g, and B. subtilis KCTC 2189 showed greater antimicrobial activity in the ethanol extract (concentration 200 ug/uL) as opposed to the water extract. Solanum nigrum L. fruit powder revealed differences in antioxidant and antimicrobial activity between the different extraction solvents. In particular, ethanol extract had higher antioxidant and antibacterial activity, meaning it is more favorable for usage as a functional food material.
This study was conducted to evaluate the antioxidant activities and the quality characteristics of 70% ethanol extracts from onion peels with different extraction methods (autoclave extraction, AE; low temperature high pressure extraction, LTPE; reflux extraction, RE; and stirrer extraction, SE). The yields of AE, LTPE, RE and SE were 9.00%, 5.39%, 13.21% and 12.41%, respectively. The total polyphenol and flavonoid contents in the AE were significantly higher than in the other extracts. The DPPH radical scavenging abilities with a concentration of 100 mg% (w/v) were : AE, 28.9%; RE, 26.07%; LTPE, 24.35%; and SE, 19.53%. The ABTS radical scavenging ability and the nitrite scavenging activity showed the same tendency as that of the DPPH radical scavenging ability. The angiotensin-converting enzyme (ACE) inhibitory activities of the LTPE and AE were higher than those of the RE and SE. The ${\alpha}$-Glucosidase inhibitory activity of the RE was higher than that of the extracts with other extraction methods. The nitrite scavenging activities with a concentration of 10 mg/mL were: AE. 33.97%; RE, 35.47%; LTPE, 21.86%; and SE, 21.71%. The ferrous ion chelating activity of the LTPE (54.73%) was significantly higher than that of the other extracts. These results suggest that AE is the superior method for the enhancement of anti-oxidant activity, and onion peel can be used as a natural antioxidant material for health foods and can be a good ingredient of functional foods.
The objective of this study was to investigate the antioxidative acitivities of various solvent extracts from haw (Crataegus pinnatifida Bunge) pulpy. The Freeze-dried pulpy of haw was extracted with five solvents (70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v/v), n-butanol, and ethyl acetate or EA), and the DPPH, reducing power, ferric-reducing antioxidant power (FRAP), ABTS, NO, hydroxyl radical scavenging activity were then measured. The DPPH radical scavenging activity of the 70% methanol and 70% ethanol extracts were 92.89% and 91.17% in the 0.8 mg/mL concentrations, respectively, which were higher than those of the other extracts. The reducing power and FRAP decreased in the following order: 70% methanol, 70% ethanol, n-butanol, EA, and CM extracts. The ABTS radical scavenging activity was high in the 70% methanol and 70% ethanol extracts and low in the n-butanol extract. The NO radical scavenging activity was high in the 70% methanol and 70% ethanol extracts but low in the CM extracts. On the other hand, the hydroxyl radical scavenging activity was high in the n-butanol and 70% methanol extracts but low in the EA extract. These results suggest that the 70% methanol and 70% ethanol extracts from haw will be useful as natural antioxidant and biohealth foods.
Objectives: We investigated the antioxidant and antibacterial activities of Pinus densiflora ethanol extracts (PDEE) treated with trypsine as a protease. Methods: Various antioxidant activities were evaluated by measuring total contents of polyphenol and flavonoid, DPPH electron-donating ability and $ABTS^+$ radical scavenging activity of test material. To compare the antibacterial activity, paper disc diffusion assay was performed against two resident bacteria in human skin (Staphylococcus aureus and Staphylococcus epidermidis). Results: As for the total contents of polyphenol and flavonoid, and the electron-donating ability and ABTS+ radical scavenging activity, both PDEE and trypsin-treated Pinus densiflora ethanol extract (T-PDEE) showed high antioxidant activity in dose-dependent manner. And the T-PDEE showed slightly higher activity than PDEE, which indicated protease treatment seemed to affect in antioxidant activity. In the result of paper disc diffusion assay, antibacterial activity was confirmed in all two types of skin resident bacteria. T-PDEE was more active than PDEE and it seems that treatment of protease may increase the antibacterial activity of PDEE. Conclusion: All of these results, we confirmed that treatment of protease to PDEE can increase the antioxidant and antibacterial activities, and it can be explained thought that this would be applicable as a cosmeceutical material in the future.
After preparation of acetone, ethanol, methanol, and water extracts (10 g/300 mL) of dried persimmon (Diospyros kaki cv. Fuyu) flower-buds, total phenolic contents (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (RSA), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) RSA, reducing power (RP), and tyrosinase inhibitory activity of the extracts were evaluated. The methanol extracts produced the highest TPC (113.39 mg gallic acid equivalents/g), DPPH RSA ($IC_{50}=40.25\;{\mu}g/mL$), ABTS RSA ($IC_{50}=58.17\;{\mu}g/mL$) and RP ($IC_{50}=69.43\;{\mu}g/mL$) activities while the water extracts generated the lowest values. The ethanol extract showed the highest tyrosinase inhibitor activity (88.90%) at a concentration of 1 mg/mL. These results indicated that persimmon flower-buds may be a useful source of natural antioxidants.
The potential use of 4 plant species, Astragalus membranaceus var. membranaceus, Senna occidentalis, Dianthus longicalyx and Plantago asiatica, as new sprout vegetables with high antioxidant function was examined in the present experiments. Seeds of above plants were allowed to germinate under light condition, and seedlings were maintained under dark condition for shoot growth in length for contain period of time. Then the seedlings were put under light for photosynthesis (greening treatment) for the period of 0${\sim}$3 days. Samples were collected to analyze the changes in antioxidant levels and activity, and it was observed that antioxidant substances were affected by greening treatments, depending on plant species. In A. membranaceus, the contents of total polyphenol was highest with no greening, total flavonoids with 3 days greening, DPPH radical scavenging effects with no greening, ABTS scavenging with 1 day greening, $Fe^{2+}$ chelating effects with no greening, and inhibitory activity against linoleic acid peroxidation with 3 day greening. In S. occidentalis, highest levels of antioxidant activity and radical scavenging effects were obtained by 2 day greening, $Fe^{2+}$ chelating effects by no greening and inhibitory activity against linoleic acid peroxidation by 1 day greening. In D. longicalyx, highest levels of antioxidant activity and $Fe^{2+}$ chelating effects were obtained by 2 day greening, $Fe^{2+}$ chelating effects by no greening and inhibitory activity against linoleic acid peroxidation by 1 day greening. In D. longicalyx, highest levels antioxidant activity and $Fe^{2+}$ chelating effects were observed with 3 day greening, and highest radical scavenging effects and inhibitory activity against linoleic acid peroxidation with no greening treatment. In P. asiatica, antioxidant activity and radicals scavenging effects were highest with 2 day greening, whereas highest chelating effects was obtained with no greening and highset inhibitory activity against linoleic acid peroxidation with 3 day greening. As the length of greening treatments influenced the antioxidant levels and function in plant species tested in this experiments, different culture methods are recommended for different plant species to get maximum health benefits out of sprout vegetables.

The antioxidant activity of 70% methanol, 70% ethanol and chloroform-methanol (CM, 2:1, v/v) extracts from turmeric (Curcuma longa L.) was investigated in this study. The antioxidant potential of various extracts of turmeric was evaluated by using different antioxidant tests, namely DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity, ABTS [2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, ferric reducing antioxidant power and ${\beta}$-carotene bleaching assays as well as total phenol content and total flavonoid content. All extracts exhibited abundant total phenol content, total flavonoid content and strong antioxidant activity. Particularly, higher activity was exhibited by CM extract with $46.32{\pm}0.35mgCAE/g$ and $15.58{\pm}0.39 mgQE/g$ of total phenol content and total flavonoid content. Generally, antioxidant activity of various extracts from turmeric increased in the following order: 70% methanol extract, 70% ethanol extract and CM extract, respectively. Results suggested that turmeric could be widely used as a source of antioxidant and a kind of functional material in food.
This study evaluated the effects of heat treatment on antioxidant activity of Rehmannia radix Libosch (RRL). RRL was heated at various temperatures ($110-150^{\circ}C$) for various times (1-5 hr), and the total polyphenol, flavonoid content, and antioxidant activity were investigated. With increased heating temperature and exposure time, total content of polyphenol, flavonoid, as well as antioxidant activity increased. The highest total polyphenol and flavonoid contents were 21.65 and 3.56 mg/g, respectively, these values were occurred after heating for 3 hr at $150^{\circ}C$ (RRL was 5.09 and 0.83 mg/g, respectively). The 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity was highest value of 83.46% after heating for 3hr at $150^{\circ}C$. The 2,2-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) cation radical scavenging activity was highest value of 20.43mg ascorbic acid (AA) eq/g after heating for 2 hr at $150^{\circ}C$. There were highly significant differences in the total polyphenol, flavonoid content, and antioxidant activity among heating temperatures and times (p<0.001), with heating temperature having the greater effect.
This study was conducted to develop HTHP ginseng (high temperature and high pressure ginseng) with improved antioxidative activity and phenolic acid composition by high temperature and high pressure process. The HTHP ginseng extract was analyzed for the total phenol content, DPPH radical scavenging activity and phenolic acid composition. The total phenol content was increased in HTHP ginseng (14.76 mg/g) compared to raw ginseng (3.59 mg/g) and red ginseng (3.93 mg/g). DPPH radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 4.8~78.4%, 1~47.4% and 1.8~56.5% at $1{\sim}100mg/m{\ell}$ concentration. Also ABTS radical scavenging activities of HTHP ginseng, raw ginseng and red ginseng extracts were 8.9~99.8%, 3.4~96% and 1.2~96.5% at $1{\sim}100mg/m{\ell}$ concentration. In HPLC analysis, amounts of measured phenolic acid of HTHP ginseng greatly increased than raw ginseng and red ginseng, but salicylic acid was not detected in HTHP ginseng. In addition, DPPH radical scavenging activity of phenolic acid from HTHP ginseng was increased. Consequently, we believe high temperature and high pressure process is better method than existing method to increase the bioactivity of ginseng.
Present studies were conducted to investigate the effects of autoclaving on antioxidant activities of Achillea alpina and Solodago virgurea. At early April, young leaves of 2 species were collected, subjected to autoclaving ($121^{\circ}C$, 1.2 atmospheric pressure, 15 minutes), freezed-dried, grinded, and extracted with 80% ethanol. The same process was repeated with unautoclaved control. Total polyphenol and flavonoid contents, scavenging activities on DPPH and ABTS radicals and ferrous ion chelating effects were analyzed. Extraction yield of autoclaved S. virgurea was 39.55% and A. alpina 28.15%. In both species, autoclaving significantly reduced scavenging activities on DPPH and ABTS radicals. On the contrary, ferrous ion chelating effects increased after autoclaving, especially in young leaves of A. alpina. Autoclaving resulted in decrease of polyphenol and flavonoid contents, especially in the A. alpina. The present experiments demonstrated that autoclaving had negative effects on antioxidant activities of A. alpina and S. virgurea, except in ferrous ion chelating effects. Young leaves of former species were unstable to heat treatment, resulting in big lose of antioxidant activity.
