The objective of this study was to evaluate the contents of reducing sugar and total polyphenol and the antioxidant activity of freeze-dried ginseng (Panax ginseng C.A. meyer cv Yun-poong). Ginseng root consists of the main root (MR), lateral root (LR) and fine root (FR). These roots were sorted into 3~4 groups (MR 1~4 groups, LR 1~4 groups, FR 1~3 groups) depending on the diameter of center region. The reducing sugar content has no influence on the diameter, but the reducing sugar content was decreased by subdivision. Total polyphenol contents of ginseng were the highest in FR-3 among all others. The antioxidant activity was measured using DPPH radical scavenging activity and ABTS radical scavenging activity. The $IC_{50}$ (50% inhibitory concentration) value of the hydrogen donating activity was the highest in FR-3 as 7.03 mg/mL and the lowest in MR-2 as 37.02 mg/mL. Overall, hydrogen donating activity of FR was higher than the main root and lateral root significantly (p<0.05). The ABTS radical scavenging activity in FR-3 showed the highest radical scavenging activity as 25.49%. This study's findings suggest that the total polyphenol contents and antioxidant activity of ginseng were the highest in FR-3, and that polyphenol contents and antioxidant activity of ginseng were related to root area and diameter.

In order to improved use of Persicaria tinctoria seeds and to get basic information, general composition, fatty acid, mineral, amind acid analysis and physiological activity of methanol extract of Persicaria tinctoria seeds were investigated. Total calories of Persicaria tinctoria seeds were 348.00 kcal/100 g, general composition, carbohydrate, crude protein, crude fat and crude ash consisted of 7.85%, 67.90%, 10.10%, 4.00% and 10.15%, respectively. The amount of saturated and unsaturated fatty acids was showed 0.9048 g/100 g and 2,714 g/100 g, respectively. Minerals contained 100g of Persicaria tinctoria seeds were followed by K (549.5 ㎎), Mg (264.4 ㎎), Ca (216.2 ㎎), Fe (12.1 ㎎), Zn (3.0 ㎎). Total 15 kinds of amino acids were detected, these amino acids displayed higher value in the alanine (1,432.6 ㎎/100 g) and glutamic acid (1,088.8 ㎎/100 g). Total polyphenol and flavonoid contents were 11.08 ㎎/L and 3.56 ㎎/L, respectively. DPPH radical scavenging and ABTS radical scavenging activity in the methanol extract of 1,000 ㎎/L was showed 86.74% and 61.74%, respectively.

In the present study, antioxidant activity and protective effect of extracts from Sambucus williamsii var. coreana stems (SWC) were evaluated on tert-butyl hydroperoxide (t-BHP) induced oxidative stress in human liver (Chang) cells. Antioxidant activities of the SWC extracts were determined by various radical scavenging activities, such as DPPH, ferric reducing antioxidant power (FRAP), 2,2'-azinobis-(3-ethybenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity and oxygen radical absorbance capacity (ORAC) assay. SWC extracts showed strong antioxidant effect on various assay. To determine the hepatoprotective effects of SWC on t-BHP induced oxidative damage, cell viability was measured using MTT assay. Pretreatment of SWC extracts showed increasing cell viability, decreasing ROS and restoring mitochondria membrane potential on t-BHP induced oxidative stress in Chang cells. Our findings suggest that SWC extracts may be considered a potential agent for therapeutic protective effect from oxidative stress through its antioxidant activity.
The polysaccharides from fruit body of Auricularia auricula and Tremella fuciformis were extracted using hot water, and partially purified through ethanol precipitation and dialysis. Free radical scavenging activities of the crude and purified polysaccharides were examined and compared each other. Free radical scavenging activities of the partially purified polysaccharides were higher than those of crude polysaccharides. DPPH free radical, ABTS radical and SOD-like activities of partially purified polysaccharide at 1 mg/mL of concentration from A. auricula were 61.7, 9.6 and 38.9%, respectively, while those of T. fuciformis were 9.6, 5.7 and 15.3%, respectively. Results of site and non-site specific hydroxyl radical scavenging activities indicated that the partially purified polysaccharide fractions from A. auricula and T. fuciformis exhibited the hydroxyl radical scavenging effect by hydrogen donating ability and iron ion chelating ability. Also, reducing powers of A. auricula and T. fuciformis were 77.1 and 14.7% of BHT (0.1%) as standard, respectively. It was suggested that antioxidant activities of A. auricula were about 1.4~6.4 times higher than those of T. fuciformis due to different levels of polyphenol content.

In this study, we investigate the potential use of persimmon peels (PP) in mushroom culture medium for the production of functional mushrooms. Pleurotus eryngii was cultivated in medium supplemented with PP (SMPP) at the following concentrations: 0% SMPP (control), 5% SMPP, 10% SMPP, 15% SMPP, 20% SMPP, or 30% SMPP. The total polyphenol content, DPPH radical scavenging ability, ABTS cation scavenging ability, and reducing power of P. eryngii cultivated in SMPP were investigated. P. eryngii cultured in 20% SMPP produced the highest values for all four measurements. Total polyphenol content, DPPH radical scavenging activity, ABTS cation scavenging ability, and reducing power all increased upon the addition of PP. Based on our results, we can conclude that persimmon peels are a highly valuable supplement for functional mushroom culture medium.
This study was carried out to evaluate the possibility of unripe apple peel water extracts as cosmetic materials and to evaluate the biological activities of the antioxidant and whitening effects of the samples. The antioxidative properties of the samples were confirmed by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) cation radical scavenging ability. To evaluate the whitening effect of the samples, several analytical techniques were used, including toxicity evaluations of the samples by MTT assays. Measurements of the inhibition rates of cellular tyrosinase, melanin synthesis rates, and expression rates of whitening-related proteins and genes were confirmed using melanoma (B16F10 cell). At equivalent unripe apple peel water concentrations ($1,000{\mu}g/ml$), the DPPH radical scavenging and the ABTS cation radical scavenging activities were 77.3% and 93.1%, respectively. The whitening activity evaluation showed that tyrosinase activity and melanin synthesis were inhibited by 19.8% and 17.3%, respectively, at unripe apple peel water extract concentrations of $50{\mu}g/ml$. In B16F10 cells induced by ${\alpha}$-MSH, the expression of tyrosinase, TRP-1, and TRP-2 decreased. Also, the activity of the transcription factor MITF was inhibited. In real-time PCR experiments, the expression of related genes at the upstream signal level was also found to be progressively lowered as the concentration of unripe apple peel water extracts increased. From these results, it was confirmed that the unripe apple peel water extracts showed excellent whitening efficacy and could be used as safe, natural, raw cosmetic material in the future.
Objectives : To evaluate the radical scavenging and acetylcholinesterase (AChE) inhibitory activities of the ethylacetate (EtOAc)-soluble portion of a methanolic extract of Aster yomena, three different assay systems were performed. Methods : The antioxidant activity of A. yomena extract was tested as its capacity to scavenging free radicals of DPPH and $ABTS^+$, which has been widely used to evaluate the antioxidant activity of natural products from plant sources. AChE inhibitory activity was tested against mouse brain AChE by spectrophotometric method of Ellman using ELISA microplate reader. Results : The methanolic extract of A. yomena was fractionated and the EtOAc-soluble portion showed significant AChE inhibitory and free radical scavenging effects. Also the EtOAc-soluble portion revealed the highest phenolic contents as compared to the other extracts. Conclusions : These results indicate that phenolic compounds may be important constituents that give rise to the anti-AChE and antioxidative activities of A. yomena extract. Further phytochemical studies on this plant, for nutraceutical or pharmaceutical application, are warranted.
In this study, we studied the anti-inflammatory and physiological activities Abelmoschus esculentus extracts by hot water (AEW) and 70% ethanol (AEE) and confirm the possibility to use it as a natural ingredient. For this study measure the antioxidative activity, total polyphenol content was measured, and DPPH and ABTS scavenging activity assays were conducted. Total polyphenol content of AEW and AEE was measured, and the results showed that they were 126.76 mg TAE/100 g and 144.21 mg TAE/100 g, respectively. DPPH and ABTS radical scavenging activities were measured to determine the antioxidative activity, and the results indicated that DPPH and ABTS radical scavenging activities increased in both extracts concentration-dependently. The moisturizing effect was measured by measuring the amount of hyaluronic acid (HA) produced within HaCaT cell using the ELISA kit. AEW and AEE increased the amount of HA production in a concentration-dependent manner. In order to determine the anti-inflammatory activity of AEW and AEE, the NO assay was conducted, and the inhibitory effects were found to be 11.46% and 25.28%, respectively in 100 ㎍/ml. In order to measure the anti-inflammatory activity, nitric oxide (NO) inhibitory activity was measured, and the inhibition of expression of iNOS, COX-2 proteins was measured and shown. Furthermore, inhibition of expression was found in inflammatory inducing factors iNOS and COX-2 proteins showing concentration-dependent inhibition. This study found the excellent effects of antioxidative, moisturizing effect, anti-inflammatory activity in Abelmoschus esculentus extracts, which indicates that can be used as functional materials for aesthetics, food and functional cosmetics.
Objectives: Reactive Oxygen Species(ROS) are continuously produced at a high rate as a by-product of aerobic metabolism. Since tissue damage by free radical, ROS such as hydrogen peroxide($H_2O_2$), nitric oxide(NO) increases with age. Several lines of evidence provided that ROS appears to cause to develop aging-related various diseases such as cancer, arthritis, cardiovascular disease. In this study, we have conducted to investigate the pharmacological effects of red ginseng for the development possibility to pharmacopuncture drug sources or healthy aid foods. Methods: For our aims, it was investigated the biological activities of Red Ginseng ethanol extracts (RGEE) by measuring total polyphenol contents, total flavonoid contents, DPPH radical scavenging activity, ABTS radical scavenging activity and cell viability of MCF 10A and SK-MEL-2 in vitro with MTT assay method. Results: The total polyphenol contents of RGEE was 3.06${\pm}$0.11mg/g in 10mg/ml, the total flavonoid contents of RGEE was 1.35${\pm}$0.01mg/g in same concentration. The ABTS radical scavenging activity was about 80% and that of DPPH activity was 65% in 50mg/ml of RGEE. The cell viability of SKMEL-2, skin cancer cell line was decreased and that of MCF 10A, skin normal cell line was increased. Conclusions: We conclude that RGEE may be useful as potential functional foods or pharmacopuncture drug sources on the diseases induced by oxidant stress.
In this studies, contents and activities of antioxidative substances in the fruits of a pear cultivar and five Korean wild pear lines were investigated. The content of total phenolic compounds was highest with Pyrus pyrifolia var. culta "Shingo" (7.68 $mg{\cdot}g^{-1}$, D.W.) while total flavonoids was highest with P. ussuriensis var. acidula (5.66 $mg{\cdot}g^{-1}$, D.W.). The contents of total polyphenols and flavonoids varied according to plant species, but in general pyrifolia lines had higher total polyphenols than total flavonoids whereas in ussuriensis lines total flavonoid contents were higher than total polyphenols. The scavenging effects of DDPH radicals was highest with highest with P. ussuriensis var. seoulensis ($RC_{50}$=1.45 $mg{\cdot}mL^{-1}$) and that of ABTS radicals was highest with P. ussuriensis var. acidula ($RC_{50}$=0.69 $mg{\cdot}mL^{-1}$). A pear cultivar "Shingo" showed highest total polyphenol content, but scavenging activity of DDPH and ABTS radicals was lowest. The fruits of P. ussuriensis var. seoulensis contained low level of phenolic compounds, but their scavenging effects on two radicals was superior. Ferrous ion chelating effect was high in P. pyrifolia var. culta (98.93%), and in P. ussuriensis var. sinensis (68.71%), but two species had low scavenging activity. The content of phenolic compounds in wild pears was not always directly proportional to the radical scavenging activity. Neither radical scavenging activity was to ferrous ion chelating activity. In conclusion the fruits of P. ussuriensis var. acidula, P. ussuriensis var. seoulensis, and P. ussuriensis var. sinensis had great potential for the development of new antioxidative functional food because their antioxidative activity is higher or at least similar to P. pyrifolia var. culta. However, due to their different levels of antioxidative effects, the development of methods of using them jointly, not singly, to obtain maximum level of antioxidants is recommended.
