Reticulone, a Novel Free Radical Scavenger Produced by Aspergillus sp.

  • Bioassay-guided fractionation of the culture broth of Aspergillus sp. FN070449 (KCTC 26428) using a DPPH (2,2-diphenyl-1-picrylhydrazyl) assay led to the isolation of two compounds: reticulone (1) and reticulol (2). Their chemical structures were elucidated on the basis of UV, IR, NMR, and MS spectroscopic analyses. Compound 1 exhibited more potent free radical scavenging activity on $ABTS^{\cdot+}$ (2,2'-azino-bis [3-ethylbenzthiazoline-6-sulphonic acid]) and DPPH radicals than did butylated hydroxyanisole (BHA) and caffeic acid.

  • DPPH and ABTS radical scavenging, and tyrosinase inhibitory activities of bioactive compounds 1-4 isolated from Cedrela sinensis leaf were assessed. Structures of isolated compounds were established by UV, one- and two-dimensional NMRs, and mass spectroscopic methods.

Antioxidative Properties of Different Solvent Extracts from Persimmon (Diospyros kaki cv. Fuyu) Flower-Buds

  • After preparation of acetone, ethanol, methanol, and water extracts (10 g/300 mL) of dried persimmon (Diospyros kaki cv. Fuyu) flower-buds, total phenolic contents (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity (RSA), 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) RSA, reducing power (RP), and tyrosinase inhibitory activity of the extracts were evaluated. The methanol extracts produced the highest TPC (113.39 mg gallic acid equivalents/g), DPPH RSA ($IC_{50}=40.25\;{\mu}g/mL$), ABTS RSA ($IC_{50}=58.17\;{\mu}g/mL$) and RP ($IC_{50}=69.43\;{\mu}g/mL$) activities while the water extracts generated the lowest values. The ethanol extract showed the highest tyrosinase inhibitor activity (88.90%) at a concentration of 1 mg/mL. These results indicated that persimmon flower-buds may be a useful source of natural antioxidants.

비자 추출물의 식품부패균에 대한 항균효과 및 항산화활성 (Antimicrobial Effects on Food-Borne Pathogens and the Antioxidant Activity of Torreya Nucifera Extract)

  • This study investigates antimicrobial effects of food-borne pathogens and the antioxidant activity of Torreya nucifera extract. The growth of food-borne pathogens including Staphylococcus aureus, Pseudomonas aeruginosa, and Candida albicans was inhibited by the extract. The antimicrobial activity of the extract was highest for Staphylococcus aureus among seven gram-positive bacteria and for Pseudomonas aeruginosa among six gram-negative bacteria. The extract exhibited slightly lower DPPH radical-scavenging activity, but its ABTS radical-scavenging activity was higher than that of ${\alpha}$-tocopherol. The results demonstrate the extract's antimicrobial effects on food-borne pathogens as well as potent antioxidant capacity and suggest that Torreya nucifera may be used as a natural antibacterial agent and an effective antioxidant in food.

Antioxidant Activity-of Curcuma Longa L., Novel Foodstuff

  • The potential antioxidant activities of different fractions from methanolic extract of Curcuma longa L. were assayed in vitro. All of the fractions exception of n-hexane and $H_2O$ showed a strong antioxidant activity, especially the ethylacetate (EtOAc) fraction, which showed the highest 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging activity ($IC_{50}=9.86\;{\mu}g/mL$). The results of 2,2'-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity and ferric reducing antioxidant power (FRAP) assay showed concentration dependency, the EtOAc fraction demonstrating a better result than the other fractions at the same concentration in this studies. Additionally, when the total phenolic contents of fractions were measured, the EtOAc fraction contained the highest level. Meanwhile, correlation analysis indicated a high correlation between the antiradical activity and the total phenolic contents, suggesting that fractions obtained from the methanolic extract of Curcuma longa L. have wide potential for use as sources of antioxidant material.

Peroxidase Activity of Cytochrome c

  • The peroxidase activity of cytochrome c was studied by using a chromogen, 2,2'-azinobis-(2-ethylbenzthiazoline-6-sulfonate) (ABTS). Initial rate of ABTS oxidation formation was linear with respect to the concentration of cytochrome c between 2.5-10 ${\mu}$M and $H_2O_2$ between 0.1-0.5 mM. The optimal pH for the peroxidase activity of cytochrome c was 7.0-8.5. The peroxidase activity retained about 40% of the maximum activity when exposed at 60 $^{\circ}C$. for 10 min. The peroxidase activity showed a typical Michaelis-Menten kinetics for $H_2O_2$ which Km value was 29.6 mM. Radical scavengers inhibited the peroxidase activity of cytochrome c. The peroxidase activity was significantly inhibited by the low concentration of iron chelator, deferoxamine. The results suggested that the peroxidase activity was associated with iron in the heme of cytochrome c.

Laccase를 이용한 Chlorophene 산화전이에 관한 연구 (Laccase-Catalyzed Transformation of Chlorophene)

  • Laccase catalyzes the oxidation and polymerization of aromatic compounds in the presence of molecular oxygen. The oxidative transformation of chlorophene with laccase was conducted in a closed, temperature controlled system. The optimal pH for transformation of chlorophene was proven to be about 5-6. As the temperature rose up to $55^{\circ}C$, the transformation of chlorophene increased. The chlorophene transformation was not enhanced in the presence of soluble polymers. The toxicity of the reaction mixture was increased two times than that of initial reaction mixture after the enzymatic treatment. ABTS has enhanced chlorophene transformation at 0.1 mM and showed negative linear relationship with residual chlorophene by the reaction.

Influence of pH on the Antioxidant Activity of Melanoidins Formed from Different Model Systems of Sugar/Lysine Enantiomers

  • This study was to investigate the influence of pH on the antioxidant activity of melanoidins formed from glucose (Glc) and fructose (Fru) with lysine enantiomers in the Maillard reaction. Melanoidins formed from D-isomers were found to be effective antioxidants in different in vitro assays with regard to the ferrous ion chelating activity, 1, l-diphenyl-2-picryl-hydrazil (DPPH) radical scavenging activities, ferric reducing/antioxidant power (FRAP), and 2,2'-azinobis(3-ethylbenothiazoline-6-sulfonic acid) diammonium salt (ABTS) radical scavenging activity. In particular, the chelating activity of these melanoidins at a pH of 7.0 was greater than those with pH of 4.0 and 10.0. The chelating activity and DPPH radical scavenging activity of the melanoidins formed from the Glc systems were higher than those of the melanoidins formed from the Fru systems. However, the FRAP and ABTS radical scavenging activity of these melanoidins were not different according to pH level, with exceptions being the Fru systems.

In vitro free radical scavenging potential of acetone extract and sub-fractions of Albizia amara (Roxb.) Boiv. stem bark

  • In the present study, acetone extract and sub-fractions of Albizia amara stem bark were evaluated for their free radical potential. The results showed that the crude extract and all the fractions exhibited antioxidant and free radical scavenging activities under different in vitro assays. Among the different fractions, the ethyl acetate fraction exhibited higher DPPH and ABTS radical scavenging activities than the standard quercetin. A. amara stem bark might be valuable source of natural antioxidants that could be used for medicinal and food applications.

참당귀(Angelica gigas Nakai)잎 용매추출에 따른 생리활성 (Biological Activities of Extracts from Leaf of Angelica gigas Nakai)

  • Antioxidant properties and antioxidant activities were analyzed for water extracts and 50% and 70% ethanol extracts of the leaf of Angelica gigas Nakai. The polyphenol and flavonoid contents in water, 50%, and 70% ethanol extract of the leaf of Angelica gigas Nakai, it was found that the polyphenol contents were 18.75 mg GAE/g, 28.95 mg GAE/g, and 34.73 mg GAE/g, respectively, and flavonoid contents were respectively. The DPPH IC50 scavenging activity was 45.84 mg/mL, 36.44 mg/mL, 19.11 mg/mL, respectively, and theABTS+ radical scavenging ability (1 mg/mL) was 28.73%, 22.79%, and 12.70%, respectively. Tyrosinase inhibitory activity, 70% ethanol extract, 50% ethanol extract, and water extract 33.14%, and 4.53%, respectively. Nitrite scavenging activity, 70% ethanol extract, 50% ethanol extract, and water extract were in the order of 1 mg/mL scavenging activity, 36.43%, 34.80%, and 18.85%, respectively.
