Effect of the ethanolic extract of cactus pear (Opuntiaficus-indica) fruit on net handling stress in zebrafish (백년초 에탄올 추출물이 net handling stress를 유도한 제브라피쉬에 미치는 영향)

  • Objectives : In this study, the anti-stress effects of ethanolic extract of Opuntiaficus-indica (OF70E) were investigated. Methods : To determine the effects of OF70E on physical stress, changes in whole-body cortisol level or behavior were monitored in zebrafish. After treatment with 0.9% saline or OF70E for 6 min, all fish underwent net handling stress (NHS), which induced physical stress. And then, we conducted open field test (OFT) or sacrificed fish for collecting body fluid from whole-body. We used the cortisol enzyme-linked immunoassay kit to measure the amount of cortisol in each zebrafish sample. Results : In result, compared with normal group which were not treated by NHS, whole-body cortisol levels were significantly increased in stressed-control group. Compared with control group, pretreatment with OF70E at concentrations of 25, 50 and 100 mg/L for 6min significantly inhibited the increase of whole-body cortisol levels induced by NHS(p<0.05). To anti-stress effects of OF70E on behavior, we conducted OFT after the induction of NHS following pretreatment of OF70E. As results of OFT, compared with unstressed-normal group, distance moved was significantly decreased by induction of NHS in stressed-control group (p<0.05). OF70E-pretreatment blocked decreases of distance moved increased by NHS (p<0.05). And meandering movement, immobility and turn angle were significantly increased by NHS in stressed-control group compared with unstressed-normal group (p<0.05). OF70E-pretreatment prevented the increases of meandering movement immobility and turn angle by NHS (p<0.05). Conclusions : In conclusion these results suggest that OF70E-pretreatment may prevent stress responses.

Glycoprotein Isolated from Morus indica Linne Enhances Detoxicant Enzyme Activities and Lowers Plasma Cholesterol in ICR Mice (뽕잎 당단백질의 혈중지질 저하 효과 및 항산화 효과)

  • The objective of this study was to evaluate the effects of glycoprotein isolated from Morus indica L. (MIL) on plasma cholesterol levels and on the activities of hepatic detoxicant enzymes in ICR mice. MIL glycoprotein evidenced good scavenging activities against lipid peroxyl radicals. When the mice were treated with Triton WR-1339, the levels of total cholesterol (TC) and low-density lipoprotein (LDL)-cholesterol in plasma increased significantly by 53.9 and 47.5 mg/dL, respectively, as compared to the controls. However, when pretreated with MIL glycoprotein $(100{\mu}g/mL)$, ICR mice showed marked reductions to 55.4 and 47.0 mg/dL, as compared to Triton WR-1339 treatment alone. Interestingly, high density lipoprotein cholesterol levels were unchanged. These results indicate that the MIL glycoprotein is capable of scavenging lipidperoxyl radicals, lowering plasma lipid levels, and increasing the activities of detoxicant enzymes in the mouse liver.

Nutrient Composition in the Parts of Opuntia ficus-indica var. saboten and Opuntia humifusa (보검선인장과 저단선인장의 열매와 줄기 영양성분 조성)

  • This study was conducted to analyze the nutrient compositions of stem and fruit of Opuntia ficus-indica var. saboten (OF) and Opuntia humifusa (OH). The results of the two-way analysis of variance test indicated that the parts, varieties and parts*varieties had influence on nutrient content except crude protein, soluble dietary fiber, Fe, Se, I, vitamin E, niacin and vitamin C. Moisture and crude ash content was higher in stem than in fruit, while crude lipid and insoluble dietary content was higher in fruit than in stem. Mineral content revealed that K (1,313.67 mg% in OF, 1,351.38 mg% in OH) was the highest in fruit and Ca (5,146.29 mg% in OF, 1,388.19 mg% in OH) was the highest in stem. Vitamin C was the most abundant vitamin in the fruit of OF (199.98 mg%) and OH (187.12 mg%). Polyunsaturated fatty acid was the highest among fatty acids (66.9~70.1%), with higher content in the stem (753.89 mg%) than fruit (578.01 mg%) in OF, while higher in the fruit (1,093.63 mg%) than stem (475.07 mg%) in OH. Moisture, crude protein, Mg, Se and riboflavin was higher in OF than OH; whereas, crude lipid, insoluble dietary fiber, total dietary fiber and monounsaturated fatty acid were higher in OH than OF. These results indicated that OF and OH could be a good food source for Ca, K and Vitamin C. Also, different nutrient content by parts and varieties, can be helpful in choice of parts or varieties for consumer purposes.

The Development of Korean Traditional Wine Using the Fruits of Opuntia ficus-indica var. saboten - II. Characteristics of Liquors - (손바닥 선인장 열매를 이용한 전통주 개발 - II. 침출주의 특성 -)

  • Different amounts of the Opuntia ficus-indica var. saboten fruit were soaked in different alcohol concentrations of a Korean rice wine distillate for 4 months. Changes in pH, brix degree, contents of reducing sugar and polyphenol compound, alcohol concentration, turbidity, and color were analyzed during the soaking period. Quality of the final product was determined through the analysis of the volatile flavor compounds and sensory evaluation. During the soaking periods, pH, alcohol concentration, and contents of polyphenol compound decreased, whereas the content of reducing sugar increased. These changes were affected more by the content of the fruit than the alcohol concentration of the soaking media. Turbidity of the wine increased with higher fruit contents, while the increase of alcohol concentration resulted in the turbidity decrease. As the soaking period increased, lightness and yellowness increased but redness decreased. Acetaldehyde, acetyl acetone, ethyl alcohol, guaiacol, thymol, and acetic acid phenyl ester were detected in all liquors. Significant differences in all sensory attributes test were shown by six experimental groups (p<0.05), and the best overall acceptability was obtained from the liquor made of 67% fruits and 30% alcohol concentration.

Identification of Major Blast Resistance Genes in Korean Rice Varieties(Oryza sativa L.) Using Molecular Markers

  • The 13 major blast resistance(R) genes against Magnaporthe grisea were screened in a number of Korean rice varieties using molecular markers. Of the 98 rice varieties tested, 28 were found to contain the Pia gene originating from Japanese japonica rice genotypes. The Pib gene from BL1 and BL7 was incorporated into 39 Korean japonica varieties, whereas this same gene from the IRRI-bred indica varieties was detected in all Tongil-type varieties. We also found that 17 of the japonica varieties contained the Pii gene. The Pii gene in Korean rice varieties originates from the Korean japonica variety Nongbaeg, and Japanese japonica varieties Hitomebore, Inabawase, and Todorokiwase. The Pi5 gene, which clusters with Pii on chromosome 9, was identified only in Taebaeg. Thirty-four varieties were found to contain alleles of the resistance gene Pita or Pita-2. The Pita gene in japonica varieties was found to be inherited from the Japanese japonica genotype Shimokita, and the Pita-2 gene was from Fuji280 and Sadominori. Seventeen japonica and one Tongil-type varieties contained the Piz gene, which in the japonica varieties originates from Fukuhikari and 54BC-68. The Piz-t gene contained in three Tongil-type varieties was derived from IRRI-bred indica rice varieties. The Pi9(t) gene locus that is present in Korean japonica and Tongil-type varieties was not inherited from the original Pi9 gene from wild rice Oryza minuta. The Pik-multiple allele genes Pik, Pik-m, and Pik-p were identified in 24 of the varieties tested. In addition, the Pit gene inherited from the indica rice K59 strain was not found in any of the Korean japonica or Tongil-type varieties tested.

Hibernation and Seasonal Occurrence of the Cotton caterpillar, Palpita indica(Lepidoptera: Pyralidae), in Watermelon (수박에서 목화바둑명나방(Palpita indica)의 월동과 발생소장)

  • The population density of the Cotton caterpillar, Palpita indica (Saunder) larvae was gradually increased until 10th October and then decreased rapidly after that time at the watermelon culture of rain sheltered house. After 15th October, many larvae went to soil and molted to pupae for the hibernation. Most of pupae were found in pF 2.4 soil moisture condition, but none in pF 1.5 hydro-morphic soil. Most of the pupae were found below 5cm and there was no pupa below 10cm from the soil surface. The sex ratio of the overwintering pupae was 1.09 : 1.00. For the monitoring of the cotton caterpillar adults using sex pheromone compounds, different mixture ratios of each pheromone compound were investigated with (E)-11-hexadecenal and (E, E)-10,12-hexadecadienal. Seven versus three ratio of (I)-11-hexadecenal and (E, E) -10,12-hexadecadienal was more attractive than any other ratio, and then followed by 6 : 4, 8 : 2, 9 : 1, and 5 : 5 mixtures. As a result of monitoring with 7 : 3 mixture of (E)-11-hexadecenal and (E, E)-10,12-hexadecadienal, occurrence peaks of cotton caterpillar adults showed 4 times per year. The first time of adult appearance was late July in 2000 and middle July in 2001 and the highest number was collected on late September.

The Influence of Temperature and Water Stress on the Varietal Reactions of Rice to the Inoculum of the Blast Infected Leaves (BIL) (도열병 이병엽접종원에 대한 수도 품종의 반응에 미치는 온도 및 Water Stress의 영향)

  • Using the milled, blast infected leaves (BIL) as an inoculum source on the screening for the resistance to blast of rice plant was a simple and useful technique. The temperature with high (25^\circ C\sim 35^\circ C) and low (15^\circ C\sim 28^\circ C) and the water stressed or not, was conditioned of to the inoculation with the BIL to the test varieties in seedling stage. In low temperature, most of the varieties were more infected with blast, however the Indica-Japonica hybrids were more infected in high temperature conditions. The water stressed was more infected with blast than the not stressed. The interaction of variety with water stress was not so much as that of variety with temperature. Resistant reaction to blast (BIL) was not affected by the temperature and water stress, but the moderately resistant or susceptible one was much affected by them. Inoculum of BIL was virulent to the newly bred Indica-Japonica hybrid cultivars such as Tongil, Nopung, etc, but not virulent to the Japonica cultivars such as Nongbaek, Jinheung, etc. The discrete, mixed or variable lesions were observed mainly in the moderately resistant or susceptible cultivars such as Kanto 51, Yashiromochi, Ishikari-shiroke, etc.

Studies on Screening Rice Cultivars for Drought Resistance I. Screening at Seedling Stage (수도 한발저항성 품종 선발에 관한 연구)

  • Seven hundred and forty eight rice cultivars were screened for drought tolerance at seedling stage in a special green house. A number of cultivars such as Wonkiljo, Hanyangdo, YR 52-$_{x}\textrm{BF}_7-67, Satominori, Mamoriaka, Akanomochi 114, Thkebenimochi, IR 937-55-3, IR2735-F_3B-6-2, IR 2871-53-2, $_{x}\textrm{BFI76}^9/Dwan, Khoo lliok Mali-4-2-105, Os6, Palawan, IRAT10, TOS 2583 and H-l05 seemed to be tolerant to drought at seedling stage. There was negative correlation between drought resistance score and growth of plant height and water content of culm and leaf after drought treatment, and highly positive correlation between resistance to blast and resistance to drought at seedling stage. There was no relationship between drought resistance and germination speed, plant height before drought treatment.

  • Physicochemical properties of rice starches from Paldal(japonica type) and Tongil(indica type) were investigated. There were no significant differences in water-binding capacity, blue value and amylose content between the two starches. Paldal starch showed a higher value for-swelling power than Tongil starch. Amylograph data showed that both Paldal and Tongil starches had similar paste viscosities except setback in which Tongil starch showed a higher value. No significant differences were observed for intrinsic viscosity and glucose units per segment between Paldal and Tongil amylopectin fractions. However, the intrinsic viscosity for Tongil amylose was considerably higher than Paldal amylose. The rate of retrogradation of Tongil starch gels at $21^{\circ}C$ was faster than Paldal starch gels.

Optimization of Demucilage Process of Opuntia ficus-indica var. saboten Fruit using High Hydrostatic Pressure Enzyme Dissolution (고압 효소 액화 장치를 이용한 백년초 점질물 분해 공정의 최적화)

  • This study aimed to develop and optimize a demucilaging process of Opuntia ficus-indica var. saboten (OFI) fruit to increase its usability as functional food ingredient and food additive. Viscozyme and Novozym 33095 as multienzyme complex having a broad spectrum of carbohydrases and pectolytic enzymes, respectively, were used in enzymatic dissolution along with high hydrostatic pressure liquefaction. To optimize the liquefaction process using high hydrostatic pressure liquefying extractor, response surface methodology with 3-factor central composite design was employed with reaction factors such as temperatures (25, 32, 40, 48, and $55^{\circ}C$), pressures (20, 40, 60, 80, and 100 MPa), and times (15, 30, 45, 60, and 75 min). At optimum conditions ($25^{\circ}C$, 100 MPa, and 58.275 min) for high hydrostatic pressure liquefaction process, the processed OFI fruit juice was predicted to have viscosity at 2.917 poise, partly due to the release of free sugars such as fructose and glucose detected using HPLC-ELSA system. The results above suggests that the OFI fruit juice with decreased viscosity may be used for various manufacturing processes of food, beverage, ice cream, and cosmetics.
