BTG 1 (B-cell translocation gene 1) gene was first identified as a translocation gene in a case of B-cell chronic lympocytic leukemia. BTG1 is a member of the BTG/TOB family with sharing a conserved N-terminal region, which shows anti-proliferation properties and is able to stimulate cell differentiation. In this study, we identified and characterized the pacific oyster Crassostrea gigas BTG1 (cg-BTG1) gene from the gill cDNA library by an Expressed Sequence Tag (EST) analysis and its nucleotide sequence was determined. The cg-BTG1 gene encodes a predicted protein of 182 amino acids with 57% 56% identities to its zebrafish and human counterparts, and is an intron-less gene, which was confirmed by PCR analysis of genomic DNA. Maximal homologies were shown in conserved Box A and B. The deduced amino acid sequence shares high identity with other BTG1 genes of human, rat, mouse and zebrafish. The phylogenic analysis and sequence comparison of cg-BTG1 with other BTG1 were found to be closely related to the BTG1 gene structure. In addition, the predicted promoter region and the different transcription-factor binding site like an activator protein-1 (AP-1) response element involved in negative regulation and serum response element (SRE) were able to be identified by the genomic DNA walking experiment. The quantitative real-time PCR analysis showed that the mRNA of cg-BTG1 gene was expressed in gill, heart, digestive gland, intestine, stomach and mantle. The cg-BTG1 gene was expressed mainly in heart and mantle.
This study was conducted to investigate the effects of modification of a herbal recipe(Herb $Mix^{(R)}$) on the growth of pullet and laying performance of hens. The formula of Herb $Mix^{(R)}$, a mixture of Rehmannia glutinosa, Angelica gigas, Discorea japonica, Glycyrrhiza uralensis, Schisandra chinensis and Ligusticum jeholense, was modified in mixing ratio. A total of 1,120 pullets(Hy-Line Brown) of 14 wks old were assigned to seven treatments; control, Herb $Mix^{(R)}$(HM), R. glutinosa fortified HM, A. gigas fortified HM, D. japonica fortified HM, G. uralensis fortified HM, S. chinensis fortified HM, L. jeholense fortified HM and Flavomycin supplemented diet. Each treatment had 8 replicates of 20 birds each housed in 2 birds cages. Body weight at 10% egg production was significantly(P<0.05) influenced by treatments. Birds fed A. gigas fortified HM diet were heaviest followed by L. jeholense fortified HM, HM-original and D. japonica fortified HM, Flavomycin supplemented diet and R. glutinosa while those fed control diet were lightest. Also, age reaching 50% egg production and peak production was earliest in A. gigas fortified HM and latest in the control. Egg production, feed intake, feed conversion and egg weight were significantly influenced by treatments. Significant improvement in egg production and feed intake was shown in A. gigas fortified HM treatment. Feed conversion ratio was lowest in antibiotic(Flavomycin) treatment and egg weight was heaviest in L. jeholense fortified HM treatment. There were no significant differences among treatments in intestinal microflora but cfu of Cl. perfringnes and E. coli tended to be lower in HM treatments than the control. Among the leucocytes of blood, the HM treatments were lower than the control in counts of white blood cell and heterophils. It was concluded that modification of Herb $Mix^{(R)}$ fortifying with A. gigas, D. japonica and L. jeholense significantly influence growth and laying performance of birds.

Hormone replacement therapy (HRT) consists of highly effective prescription medications for treating menopausal symptoms; however, these agents have exhibited side effects including the risk of estrogen-induced carcinogenesis. Therefore, interest in phytotherapy-based materials as a natural source of alternatives to estrogen therapy has increased. However, some of these herbal medicines have been reported to increase the risk of estrogen-induced cancer. Herbal formulations composed of a combination of Cynanchum wilfordii Hemsley (CW), Phlomis umbrosa Turczaninow (PU), and Angelica gigas Nakai (AG) extracts (CPAE) have been used for treating menopausal symptoms. Therefore, in this study, we aimed to examine the safety of CPAE by determining its potential adverse estrogenic activity using the Organization for Economic Cooperation and Development (OECD) test guideline 455 (TG455) in a stably transfected transcriptionally activated human estrogen receptor ${\alpha}$ ($hER{\alpha}$)-HeLa9903 cell model. We found that CPAE did not how any estrogenic activity or stimulate promoters containing estrogen response elements in MCF-7 cells. In addition, CPAE showed no significant selective activity against $hER{\alpha}$ and $hER{\beta}$, non-selective activity against the ER, or effects on ER target gene expression. Furthermore, CPAE did not significantly induce MCF-7 cell proliferation and uterine weight increase in ovariectomized rats. These results demonstrate that CPAE can be used as beneficial herbal drug for prevention and therapeutic intervention of estrogen carcinogenesis in menopausal women.

To characterize active cells during microspore culture of Angelica gigas, flow cytometric and epifluorescent techniques were applied. The knowledge obtained from these types of studies will give us insight into early stage in plant development and may lead to the application of microspore-derived from haploid plants for breeding in recalcitrant species. Viability of cultured microspore differed depending on the developmental stages. Frequencies of active cells from tetrad, uni-nucleate, bi-nucleate and matured pollen were 12.8, 49.3, 42.3 and 31.7%, respectively. Alive microspores have luminescent the green fluorescence stained with FDA and blue fluorescence stained with DAPI.
The hot water extraction(HWE) has many problems such as a low extract yield and a reduced flagrance by excessive heating during concentration process notwithstanding it has been the general method to get the extract from the traditional chinese medicines Astragalas manbranaceus Bunge and Angelica gigas Nakai. For that reason, adopted the alcohol extraction In this research and got the good results of the 65% and 75% extract yield of Astragalas manbranaceus Bunge and Angelica gigas Nakai respectively, 15% and 36% increased compare with 50% and 39% (w/w) of HWE. The differences of extraction process between the HWE and alcohol extraction is substituting alcohol for water of extraction were concentrated at the relatively low temperature 90$\^{C}$ compare with the thermal extraction temperature 104$\^{C}$ . This alcohol extract, has the outstanding effect collecting the original fragrance at the low temperature. Applying this extract to starch syrup and beverage, expected that those contain a sufficient flavor as well as fragrance without artificial spices.
Pacific oysters Crassostrea gigas (Thunberg) were collected on April, 1999 and MarchSeptember, 2000 from Goseung Bay along the southern coast of Korea. The oysters tested cp;;ected from a depth of 0.5-2 m in which they cultured by a long line hanging method. The oxygen consumption rates (OCR) of oysters held under constant temperature and darkness (CC), were determined using an automatic intermittent-flow-respirometer (AIFR). Depending on holding periods after oyster collection, the experiments were divided into two groups: Group 7-d (held to ambient temperature for ca. 7 days) and Group 2l-d (held to ambient temperature for ca. 21 days). The OCR for Group 7-d single oyster displayed two peaks every day under CC, while Group 2l-d single oyster showed one peak every day. It is likely that the rhythmic patterns 02.6-12.8 hours) of the OCR in the Group 7-d single oyster may have been influenced by tidal currents at the sampling site. The rhythmic patterns (24.3-24.7 hours) in the Group 2l-d single oyster may have been shifted from two peaks to one peak each day under CC. The present study concludes that the OCR rhythm of wild oysters in nature is governed by two lunar-day clocks (24.8 hours); one driving one peak and the other driving the second peak. When oysters are subjected to the long-term CC conditions, one of the two-clock systems is depressed or only intermittently becomes active. Jpwever. the OCR rhythms by two to three oysters occurred arrhythmic patterns during the experiments and exhibited some evidence of weak rhythmicity of compared to those of a single oyster. It could be partly due to differences group effects.

Due to the oil spill incident in December 2007, every facility of oyster culture was removed in western coast especially in Taean and Seosan. To restore oyster resources in Taean and Seosan, we brought oyster seeds from southern area and monitored their growth. In addition we monitored the culture environment throughout the year, and observed the number of oyster larvae and attached spats on collectors during summer. The factors of water environment were appropriate for oyster culture in both study area. The growth of shell height was larger in Uihang-ri, Taean than Jungwang-ri, Seosan. Spawning was more intensive in a short time in Jungwang-ri, Seosan than Uihang-ri, Taean. The number of oyster larvae and spats of collectors were much more in Jungwang-ri, Seosan than Uihang-ri, Taean. This study showed that transplantation of healthy oyster seeds from southern area can be a way of restoration of oyster resources in western coast. In addition, systematic approaches are necessary by building a better understanding of regional characteristics to restore and enlarge the oyster culture farms in western coast. In summary Uihang-ri, Taean will be appropriate for cultivation farms and Jungwang-ri, Seosan for seedling grounds to increase oyster culture productivity.

This study assessed the safety of raw oysters Crassostrea gigas for consumption during processing in a processing plant. Bacterial contamination (e.g., viable cell counts, coliform groups, Escherichia. coli and pathogenic bacteria) and chemical contamination (e.g., heavy metals and shellfish toxins) were measured on raw oysters, a processing equipment, employees and work areas. No total mercury, lead, paralytic shellfish poison, diarrheic shellfish poison or norovirus was detected in any post-harvested oyster samples. However, the cadmium level ranged from 0.1-0.2 mg/kg. The viable cell count, E. coli and coliform group levels in post-harvested oysters ranged from 4.00-4.54 log CFU/g, ND-210 MPN/100 g and 110-410 MPN/100 g, respectively. The viable contaminating cell counts on employees, equipment and work areas were in the range of $0.90-3.46log\;CFU/100cm^2$. Airborne bacteria in the work areas ranged from 0.60 to 1.81 log CFU/plate/15 min. Thus, no significant health risks were detected in the processing plant.
This study examined microbial contamination during seasoned and dried squid Dosidicus gigas processing, including the apparatus, machines, and employee's gloves at each step in processing at two companies. The numbers of bacteria floating in air in each processing area were also examined. The numbers of Staphylococcus aureus (3.6-6.0 log CFU/g) and Escherichia coli (1.3-1.4 log MPN/100 g) in domestic and imported daruma (a semi-processed product of seasoned and dried squid) at companies A and B exceeded the regulatory limits of the Food Sanitary Law of Korea (S. aureus, ${\leq}2.0$ log CFU/g; E. coli, negative). S. aureus in both daruma was reduced to below the detection limit or 3.6 log CFU/g after the roasting step, but increased again to 3.3 and 5.5 log CFU/g after the mechanical tearing step at companies A and B, respectively. E. coli showed similar tendencies at both companies. The surfaces of the apparatus, machines, and employee's gloves that contacted daruma were also contaminated with S. aureus (1.0-5.5 log CFU/$m^2$) and E. coli (negative-to 3.5 log MPN/$m^2$). The numbers of bacteria floating in air were high (1.7-5.1 log CFU/$m^3$) at both companies. These results suggest that sanitation standard operating procedures (SSOP) must be developed to control of microbial cintamination in seasoned and dried squid.
Objectives: The purpose of this study was to investigate the effects of Angelica gigas Nakai extract(AGNE) and Bacillus polyfermenticus KJS-2 (BP2) on atopic dermatitis (AD) induced by 2, 4-dinitrochlorobenzene (DNCB) in mice. Methods: In the experiment, we divided mice into four groups: a control group, a DNCB group, an AGNE group, and an AGNE+BP2 group. Then we examined the changes in scratching frequency, clinical aspects on dorsum skin, immunoglobulin (IgE), cytokines ($TNF-{\alpha}$, IL-6) and expression of COX-2. Resutls: From the experiment, the scratching frequency was significantly dropped in AGNE group and AGNE+BP2 group. Clinical observations of dorsum skin, there were a severe keratotic lesion and drop of dead skin cell in DNCB group, but symptoms of AD were decreased 39.6% in AGNE group and 49.6% in AGNE+BP2 group during 3 weeks. IgE, $TNF-{\alpha}$ and IL-6 were decreased significantly in both AGNE and AGNE+BP2 group. Expression of COX-2 was also decreased significantly in both groups. Conclusions: In conclusion, these data suggest that AGNE can decrease symptoms of AD and BP2 makes AGNE more effective. So AGNE can be useful herbal therapy for AD.
