참당귀(Angelica gigas Nakai)의 체력증진 및 면역조절효과 (The Regulatory Effect of Angelica gigas Nakai on Immune Enhancement and Cytokine Production in vivo and in vitro)

  • 본 연구에서는 다양한 실험모델을 활용하여 참당귀 추출물의 면역조절 효능을 평가하였다. 특히 강제수영부하실험에서 참당귀 추출물의 경구투여는 부동시간의 감소 및 신장기능 지표인 BUN 및 근육의 피로관련 생화학적 지표인 LDH 활성 조절로 체력증진 효과를 나타내는 것으로 확인하였다. T세포를 활용한 참당귀 추출물의 면역조절 효능 평가를 위하여 IL-2, IL-4 및 IFN-𝛾 mRNA 발현 변화에 대한 영향을 측정하였다. 실험결과, 참당귀 추출물은 IL-2, IL-4 및 IFN-𝛾 세포 활성물질의 생성 조절을 통하여 면역조절 효능이 있음을 확인하였다. 본 연구결과를 통해 규명된 결과는 참당귀 추출물의 체력증진 및 면역조절을 위한 건강 기능성 제품개발에 활용 가능성을 제시하였다.

  • 식물세포의 장기보존방법 연구를 위하여 참당귀(Angelica gigas Nakai) 현탁세포를 이용하여 동결보존을 수행하였다. 최적의 예비성장배지로는 0.7 M sucrose를 첨가하여 6일간 배양한 것이 가장 좋았으며, 동결보호제로는 1 M sucrose, 1 M glycerol, 1 M DMSO를 혼합하여 사용하였을 때 가장 좋은 결과를 나타내었다. 동결 시에는 처음 8시간 동안 deep freezer에서 천천히 냉동을 한 후 액체질소에 넣는 것이 가장 좋은 것으로 나타났다. 또한 여러 가지 삼투압 증진제를 사용한 실험을 수행한 결과 sucrose가 가장 좋은 결과를 나타내었는데 이것은 sucrose가 삼투압 증진제 역할뿐만 아니라 cell membrane을 보호하기 때문으로 사료된다.

Gametogenic Cycle and Fine Structure of Ripe Germ Cells in the Pacific Oyster, Crassostrea gigas on the South Coast of Korea

  • The gonadal development and the gametogenic cycle and the fine structure of ripe germ cells of the cultured Pacific oyster, Crassostrea gigas were investigated using oysters monthly collected from the southern coast of Korea from October 2000 to September 2001. Monthly changes in the condition index were similar to that of meat weight rate and the highest value was observed in between April and May, and the lowest value in August. The external colors of the testis and the ovary were milky white and yellowish, respectively. The spawning period of the Pacific oyster was continued from May to September, with a peak in July. The gametogenic cycle could be classified into five successive stages: multiplicative stage (December to March), growing stage (March and April), mature stage (April to June), spawning stage (June to August) and resting stage (August to January). Variety of egg yolk granules, lipid granules, mitochondria, and endoplasmic reticula were observed in cytoplasm of ripe oocyte. The spermatozoon consisted of the head, middle piece and tail; including cap-shaped acrosome with domed structure, elliptical shaped nucleus, four mitochondria, two centrioles and flagellum.

Serotonin 처리 농도에 따른 굴, Crassostrea gigas의 산란유발 효과 (Effects of serotonin concentration on spawning induction in Pacific oyster Crassostrea gigas)

  • 굴의 종패생산 시 수정란을 대량으로 얻기 위하여 성숙한 굴 어미를 대상으로 신경전달 물질인 serotonin의 최적 처리 농도를 파악하기 위한 연구가 수행되었다. Serotonin 주사 후 방란까지 소요되는 시간은 7.6분부터 27분까지 였으며, 농도가 낮아짐에 따라 소요시간은 유의하게 길어졌다 (P < 0.05). 방정시간 역시 방란시간과 비슷한 경향을 보였다. Serotonin 농도에 따른 반응률 역시 처리 농도가 높을수록 반응률이 높아졌다. Serotonin 처리 후 산란량을 조사한 결과, $10^{-2}M$ 에서 2,664천개로 $10^{-5}M$ 농도에 비해 유의하게 많았으나 (P < 0.05), $10^{-4}$$10^{-3}M$ 처리구는 유의한 차이를 나타내지 않았다 (P > 0.05). Serotonin 처리 후 산란된 알의 GVBD율, 수정률 및 부화율은 농도에 영향을 받지 않았다.

Comparison of Extractive Nitrogenous Constituents between the Diploid and the Triploid of Oyster Crassostrea gigas Whole Body

  • In order to investigate the composition of extractive nitrogenous components in the diploid and the triploid oysters, Crassostrea gigas, cultured at the south coast of Korea, the whole edible part (whole body) was analyzed into extractive nitrogen, free amino acids, oligopeptides, ATP and its related compounds, quaternary ammonium bases, and guanidino compounds using specimens collected from April to May of 1992. The major free amino acids in the diploid and the triploid were taurine, proline, alanine, glycine, glutamic acid hypotaurine, glutamine, arginine, aspartic acid, and $\beta-alanine$. There was no conspicuous difference in the constituents of free amino acids between the diploid and the triploid. A lot of hypotaurine was detected in the diploid and the triploid of oyster and the contents of them were 107 mg and 123 mg/100g, respectively. The compounds, glycinebetaine, homarine and trigonelline were found in both the diploid and the triploid. Among them, glycinebetaine was the most prominent in all the samples. The amount of protein, glycogen, extractive nitrogen, oligopeptides, ATP and its related compounds, and free amino acids in the triploid was higher than that of the diploid (p<0.10)

  • Inordinate stress causes disorders of various systems in humans and activates defense mechanisms to maintain homeostasis in the body. Sleep is a vital, highly organized process regulated by complex systems of neuronal networks and neurotransmitters. Sleep is an essential biological process whose underlying regulating involves numerous anatomical structures and biochemical substances that can be compromised by stress and by the immune system. Gamma-amino butyric acid (GABA) is the main inhibitory neurotransmitter of the central nervous system, and activation of GABAA receptors is known to favor sleep. This study was conducted to evaluate the possible application of Lactobacillus brevis BJ20 fermentation to improve the functional qualities of sea tangle Saccharina japonica and oyster Crassostrea gigas. Antioxidant activity was determined by assaying levels of radical scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH) and superoxide. L. brevis BJ20 fermentation of sea tangle and oyster enhanced both antioxidant and antiinflammatory activities. These results suggested that L. brevis BJ20 fermented sea tangle and oyster could be used for alleviation of stress and to promote sleep.

당귀가 유산균의 생육에 미치는 영향 (The Effect of Angelica gigas Water Extract on the Growth of Lactic Acid Bacteria)

  • This study was carried out to survey the effect of medicinal herbs on the growth of 3 strains of lactic starter cultures in MRS broth by the addition of 0, 1, 3, 5, and 10% water extract, after 30 hrs incubation. The pH, titratable acidity and O.D. of lactic acid bacteria were investigated to obtain fundamental knowledge for the development a new product. The effects of medicinal herbs extracts on the growth of lactic acid bacteria were variable depending upon the species of lactic acid bacteria and medicinal herbs extract. Growth of Str. thermohilus was activated by addition of 1 and 3% Angelica gigas water extract, but the basal medium plus 5 and 10% extract and growth of L. acidophilus and Bif. longum were depressed by the addition of extract. The pH and acid production ability dropped slightly in the basal medium plus 1 and 3% of extract. The other samples following the same trend as the control.

삼배체와 이배체 굴, Crassostrea gigas의 형태 및 생리학적 특성 비교 (Morphological and physiological comparison between triploid and diploid Pacific Oyster, Crassostrea gigas)

  • The morphological relations and physiological characteristics of the triploid and diploid oysters (Crassostrea gigas) in Taean area, west coast of Korea, were investigated from May 2012 to April 2013. Mophometric analysis indicated that the triploid oysters have the same shell length to shell height ratio but higher shell depth to shell height ratio than diploids. Consistent with morphological characteristics, triploid oysters showed greater values of fatness, condition index and RNA/DNA ratio during the period of experiment. The DNA concentration in adductor muscle and mantle of triploid were either lower or equal to the nucleic acids of diploid. However, RNA/DNA ratio were significantly higher than diploid. It appears that RNA/DNA ratio could be a useful indicator of health condition of triploid and diploid oysters when taken in correlation with the morphological indices.

당귀의 수경재배에서 LED 광원에 의한 생장 증가와 형광등에 의한 기능성물질 축적 (Increased Growth by LED and Accumulation of Functional Materials by Florescence Lamps in a Hydroponics Culture System for Angelica gigas)

  • 당귀는 미나리과에 속하는 약용식물로서 한국, 일본, 중국 등에서 재배된다. 본 실험의 목적은 형광등과 LED 광조사가 당귀 생육의 발달과 기능성 물질로 알려진 Decursinol angelate와 Decursin의 생성에 미치는 영향을 분석하고자 수행하였다. 당귀 '만추' 종자를 우레탄 스폰지에 파종 후 4주간 유리온실에서 육묘하였다. 본엽이 3매 전개된 120주를 저면관수 시스템에 이식하였다. LED [적색: peak wavelength 660nm, 청색: peak wavelength 455nm, white = 3:2:4 비율]와 형광등 조건에서 최상부 군락에서 광합성유효광량자속은 $180{\pm}7{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$로 동일하였으며 16:8의 일장조건에서 5주간 재배하였다. LED 처리에서 당귀의 엽수는 유의적 차이는 없었지만, 형광등 처리와 비교하여 13.5% 증가하였으며, 형광등 처리에서 당귀의 엽장과 엽폭의 비율과 초장은 각각 24, 13% 증가하였으며 최대 근장은 두 그룹이 유사하였다. LED 처리한 당귀의 지상부 생체중과 건물중은 50, 42%씩 증가하였으며, 지하부는 125, 45%씩 증가하였다. 형광등에서 재배된 당귀의 Decursin과 Decursinol angelate는 LED처리한 당귀보다 지상부에서 단위 건물중당 188, 27% 증가하였으며, 지하부에서 78, 8% 각각 증가하였다. LED와 형광등 처리에서 재배된 당귀의 한 개체당 Decursin과 Decursinol angelate의 함량은 132과 122mg 이었다. 결론적으로 기능성 물질은 형광등에서 증가되고 생장은 LED 조건에서 증가되는 것으로 나타났다.

  • The effects of salt solution and chlorella on the quality of canned oyster, Crassostrea gigas, were evaluated to obtain basic data regarding the processing of two canned oyster products. In canned oyster processing, the shucked oyster meat was steamed for 20 min and then drained. Then, each can (301-3) was filled with 90 g boiled oyster in 60 mL 1.5% salt solution for the control samples or 30 mL 1.5% salt solution and 30 mL chlorella culture medium for the experimental samples. All canned products were sealed using a vacuum seamer and then sterilized to Fo values of 6-12 min in a steam retort system at 118℃. The viable bacteria count, proximate composition, pH, salinity, yield, volatile basic nitrogen (VBN), amino-nitrogen, thiobarbituric acid (TBA), mineral, color value, free amino acid levels, hardness, and sensory evaluation of the two canned products were measured under various sterilization conditions. There were no significant differences in the physical or chemical factors and little difference in the overall acceptance of the control and experimental samples.
