당귀의 종별 생육특성 및 생산성 비교 (Growth Characteristics and Yield of the Three Species of Genus Angelica)

  • 당귀는 우리 나라는 참당귀 (Angelica gigas), 중국은 중국당귀 (A. sinensis) , 일본은 일당귀 (A.acutiloba)를 각각 같은 용도의 한약재로 사용하고 있다. 일당귀는 과거부터 우리나라에서도 일부 재배되고 있으며, 중국당귀는 ‘97년 이후 종자를 도입하여 재배하였으나 실패하여 농가의 피해가 발생한다. 이들 종의 생산성과 중국당귀의 국내 적응성을 수원 (해발 50 m), 진부 (해발 500 m), 태백 (해발 700 m) 3개지역에서 시험하였던 바 얻어진 결과를 요약하면 다음과 같다. 1. 참당귀, 일당귀는 3개 지역 모두 정상적으로 생육되고 수확이 되었다. 2. 중국당귀의 생존율은 테백 70.3%, 진부 45.2%, 수원 4%로 수원에서는 7월 이후 고온기에 고사되었다. 3. 중국당귀의 10a당 수량은 태백 160 kg, 진부 127 kg, 수원 8 kg으로 태백, 진부에서는 수확이 가능하였으나 수량이 극히 낮았으며, 수원에서는 생산이 불가능하였다. 4. 중국당귀는 해발 500 m 이하에서는 생존율이 극히 낮으며, 해발 700 m 이상의 높은 지역에서는 생존은 가능하나 생산성이 낮아 국내생산은 어려운 것으로 나타났다.

Identification of the Food Sources-Metabolism of the Pacific Oyster Crassostrea gigas using Carbon and Nitrogen Stable Isotopic Ratios

  • In order to understand food sources-metabolism for the pacific oyster (Crassostrea gigas), the stable isotope ratios of carbon (${\delta}^{13}C$) and nitrogen (${\delta}^{15}N$) of its gut, gill, and muscle as well as potential food sources (particulate organic matter, sedimentary organic matter, benthic microalgae, seagrass detritus) were determined in Dongdae Bay. Average ${\delta}^{13}C$ and ${\delta}^{15}N$ values reflect that oysters primarily fed on sedimentary organic matter as opposed to suspended organic matter during summer and winter seasons. However, the relatively enriched $^{15}N$ values of particulate organic matter (>$250{\mu}m$) and sedimentary organic matter in the summer may be due to the photosynthetic incorporation of $^{15}N$-enriched nitrogen (DIN) or the spawning events of bivalves. Specific oyster tissues (gut, gill, and muscle) revealed different metabolic pathways, which were determined through analysis of ${\delta}^{13}C$ and ${\delta}^{15}N$ in each organ. The present results suggest the determination of carbon and nitrogen stable isotopes to be a useful approach in ecological research related to the food sources- metabolism of Crassostrea gigas.

Identification of Angelica Species by Pyrosequencing

  • Objective : Angelica species are some of the most medicinally important materials in Oriental medicine. This study was performed to determine if Angelica species could be identified by genetic analysis and to verify Pyrosequencing analyses, which were used to assess genetic variation. Methods : The DNAs of Angelica acutiloba, Angelica gigas and Angelica sinensis were extracted. We have investigated the typing of single-base variations of Angelica species in DNA by using Pyrosequencing. Results : Angelica gigas showed a different pattern compared with Angelica acutiloba and Angelica sinensis. The peak of Angelica gigas was very weak in the second C nucleotide base compared with that of the others. The peak of Angelica acutiloba was present in the fourth C nucleotide base compared with that of the others. From these results we verified that our Angelica species-specific sequencing primer was well designed. Conclusion : Pyrosequencing analysis might be able to provide the identification of the Angelica species.

기후대별 산림환경에 따른 참당귀의 생육 및 지표성분 특성 (Characteristic of Growth and Active Ingredient in Angelica gigas Nakai according to Forest Environment by Climate Zone)

  • Background: Angelica gigas Nakai, that belong to the Umbelliferae family, is one of the traditional medicinal plants in Korea. Its roots have been used to treat gynecological diseases. In this study, growth characteristics and index components were compared with the forest microclimate at several forest sites. Methods and Results: A. gigas was planted in three climatic zones according to the temperature (southern temperature zone - Hamyang, central temperature zone - Bonghwa, and northern temperature zone - Jeongseon) and growth characteristics were investigated in comparison with the forest microclimate. Our results indicated that the root diameter and length, and fresh and dry weight were the highest in Jeongseon. The total content of decursin was the highest in Jeongseon (9.52%), followed by those in Hamyang (8.07%) and Bonghwa (7.48%), respectively. Additionally, the yield of decursin (1.39 g) was the highest in Jeongseon. Conclusions: The yield and index components were influenced by the microclimate in the forests, and it was assumed that high altitude and low temperature affected the increase in growth and index components. These results will be useful as basic data to study the correlation among environmental conditions, growth, and index components.

  • 약초 당귀의 원산지를 판별하기 위하여 SAW 센서가 내장된 GC로 구축된 시스템을 이용하여 휘발성 성분을 분석하였다. 사용된 시료는 별도의 전처리 과정 없이 분석되어, 30초 이내에 얻어진 hertz 값(Frequency)과 이를 미분하여 표현한 크로마토그램을 $VaporPrint^{TM}$ 이미지 소프트웨어를 사용하여 패턴화하였으며 패턴인식을 통하여 비교한 결과, 수입산 당귀와 국내산 당귀의 패턴이 뚜렷하게 차이를 나타냄으로 원산지 판별을 할 수 있었다.

  • The proximate composition of 3 kinds of Umbelliferaeceae Wild Plants (Angelica gigas NAKAI., Ostericum sieboldii NAKAI. and Coriandrum sativum LINNAEUS) were analyzed and used for making pork sausages as a functional ingredient. In the proximate composition, crude protein content was the highest in Ostericum sieboldii, 37.87%, and crude lipid and crude ash contents were the highest in Coriandrum sativum. Vitamin C was contained in Angelica gigas at 0.30mg%. but was not detected in Ostericum sieboldii and Coriandrum sativum. $\beta$-carotene content in Angelica gigas was the highest at 19.96%, but selenium was not detected in any of them. Total phenol content in Coriandrum sativum was the highest as 1107.43mg%. TBA and VBN values of all sausages with wild plants increased gradually during four-week-storage and were lower than those of control. Coriandrum sativum-added sausage had the lowest TBA as well as VBN valuew. During the four-week-storage at 4$\^{C}$, TBA value and VBN value were in the very safe range at all sample. Total phenol contents in sausages appeared to have more antioxidative activity than vitamin C, $\beta$-carotene and selenium, and consequently decreased the TBA values of sausages.

감마선 조사 당귀(Angelica gigas Nakai)의 유효성분 안정성 및 유전독성학적 안전성 연구 (Evaluation on the Safety of ${\gamma}$-Irradiated Angelica gigas Nakai: Stability of Active Components and Safety in Genotoxicity Test)

  • In the present studies, we assessed the stability of active components and toxicological safety of irradiated Angelica gigas Nakai(Danggui). In order to confirm the stability of active components in the ${\gamma}$-irradiated roots of Danggui, the quantitative analysis of decursin and decursinol angelate of ${\gamma}$-irradiated sample was carried out by high performance liquid chromatographic (HPLC) methods using reverse phase columns and normal phase columns. From the root of Danggui, decursin and decursinol angelate were isolated by a silica gel column chromatography(toluene : ether (1 : 1), Hexane : EtOAc(15 : 1)). And then the structures were confirmed in the 1H and 13C-NMR analysis. The HPLC chromatograms of decursin and decursinol angelate in ${\gamma}$-irradiated Danggui were similar with those of non-irradiated sample. In the examination of in vitro genotoxicity of the water extract from ${\gamma}$-irradiated Danggui using Salmonella reversion assay(Ames test) and micronucleus test in Chinese hamster ovary (CHO) cells, mutagenicity was not exhibited in the two assays with or without metabolic activation. These resutls suggest that active components in the ${\gamma}$-irradiated Danggui should be stable and that the safety of ${\gamma}$-irradiated Danggui could be revealed in further test in vivo.

First Record of Mycoplasma-like Organism in Pacific Oyster (Crassostrea gigas) in Korea

  • During routine survey of Pacific oyster (Crassostrea gigas) collected from Tongyoung area in southern coast of Korea, histological examination revealed that a intracellular microorganisms infected the digestive gland of the oyster. They infected hepatopancreatic cells extensively. The size of intracellular microorganism was of 45 to 86nm in diameter and 200nm to more thar 500nm in length. They were pleomorphic. The morphological characteristic of intracellular microorganisms lacked cell wall and was bounded by the plasma membrane. They contained typical prokaryotic ribosomes and fibrillar DNA-like strands. No additional internal structure has been observed. Based on the lack of cell wall and the cellular localization, the intracellular microorganism is considered as a Mycoplasma-like organism.

한국산 당귀의 생약학적 연구 (Pharmacognostical Studies on the Dang Gui from Korea)

  • Chinese crude drug 'Dang Gui' has been used mainly as a tonifying the blood, promoting blood circulation, relieving pain and moistening the bowels. To clarify the botanical origin of 'Dang Gui' from Korea, the anatomical characteristics of Angelica gigas, Angelica sinensis and Angelica acutiloba were studied. As a result, it was clarified that 'Dang Gui' from Korea was the root of Angelica gigas.

The Effect of Angelica gigas Nakai on Immune Enhancement in Cyclophosphamide-induced Immune-suppressed Mice

  • Natural products are important sources for drug development because they have a wide variety of useful biological properties. Angelica gigas Nakai (AGN) has been used as an herbal medicine. The present study was designed to evaluate the immune-enhancing effect of AGN in the cyclophosphamide (CP) induced immune-suppressed mice. As the result, we found that CP induced the reductions of body ratio, spleen weights, hematopoietic parameter and cytokine productions in mice. However, AGN recovered immunosuppression-mediated decreased body ratio, spleen and thymus weights as well as regulation of hematopoietic parameter including white blood cell, lymphocyte, and neutrophil. According to histological study, AGN regenerated on CP-mediated injured spleen. Moreover, AGN increased the CP-induced reduction of cytokine expression in spleen tissue. Collectively, the findings provide experimental evidence that AGN may be a candidate for health-improving herbs.
