Authentication and classification of strawberry varieties by analysis of their leaves using near infrared spectroscopy.

  • It is well known now that near infrared spectroscopy (NIRS) is a fast, no destructive, and inexpensive analytical technique that could be used to classify, identify, and authenticate a wide range of foods and food items. Therefore, the main aims of this study were to provide a new insight into the authentication of five strawberry (Fragaria x ananassa) varieties and to correlate them with geographical zones and the propagating methods used. Three weeks plants of five different strawberry varieties (F. x ananassa Duch. cv Camarosa, Seascape, Chandler, F. Chiloensis, and F. Virginiana) were cultivated in vitro first and then transferred to pots with special soil, and grown in a greenhouse at CINVESTAV, all varieties were acquired from California (USA). After 18 months, ten leaves from each variety were collected. Transmission spectra from each leave were recorded over a range of 10, 000-4, 000 cm$-^{1}$, 32 scans of each strawberry leave were collected using a resolution of 4 cm$-^{1}$ with a Paragon IdentiCheck FT-NIR System Spectrometer. Triplicates of each strawberry leave were used. All spectra were analyzed using principal component analysis (PCA) and soft independent modeling class analogy (SIMCA). The optimum number of components to be used in the regression was automatically determined by the software. Camarosa was the only variety grown from the same shoot but propagated by a different method (direct or in vitro). Five different classes (varieties) or clusters were observed among samples, however, larger inter class distances were presented by the two wildtype samples (F. Chiloensis and F. Virginiana). Camarosa direct and Camarosa in vitro displayed a small overlapping region between them. On the other hand, Seascape variety presented the smallest rejection percentage among all varieties (more similarities with the rest of the samples). Therefore, it can be concluded that the application of NIRS technique allowed the authentication of all strawberry varieties and geographical origin as well. It was also possible to form subclasses of the same materials. The results presented here demonstrate that NIRS is a very powerful and promising analytical tool since all materials were authenticated and classified based on their variety, origin, and treatment. This is of a tremendous relevance since the variety and origin of a plant material can be established even before it gives its typical fruit or flower.

Principal Discriminant Variate (PDV) Method for Classification of Multicollinear Data: Application to Diagnosis of Mastitic Cows Using Near-Infrared Spectra of Plasma Samples

  • In linear discriminant analysis there are two important properties concerning the effectiveness of discriminant function modeling. The first is the separability of the discriminant function for different classes. The separability reaches its optimum by maximizing the ratio of between-class to within-class variance. The second is the stability of the discriminant function against noises present in the measurement variables. One can optimize the stability by exploring the discriminant variates in a principal variation subspace, i. e., the directions that account for a majority of the total variation of the data. An unstable discriminant function will exhibit inflated variance in the prediction of future unclassified objects, exposed to a significantly increased risk of erroneous prediction. Therefore, an ideal discriminant function should not only separate different classes with a minimum misclassification rate for the training set, but also possess a good stability such that the prediction variance for unclassified objects can be as small as possible. In other words, an optimal classifier should find a balance between the separability and the stability. This is of special significance for multivariate spectroscopy-based classification where multicollinearity always leads to discriminant directions located in low-spread subspaces. A new regularized discriminant analysis technique, the principal discriminant variate (PDV) method, has been developed for handling effectively multicollinear data commonly encountered in multivariate spectroscopy-based classification. The motivation behind this method is to seek a sequence of discriminant directions that not only optimize the separability between different classes, but also account for a maximized variation present in the data. Three different formulations for the PDV methods are suggested, and an effective computing procedure is proposed for a PDV method. Near-infrared (NIR) spectra of blood plasma samples from mastitic and healthy cows have been used to evaluate the behavior of the PDV method in comparison with principal component analysis (PCA), discriminant partial least squares (DPLS), soft independent modeling of class analogies (SIMCA) and Fisher linear discriminant analysis (FLDA). Results obtained demonstrate that the PDV method exhibits improved stability in prediction without significant loss of separability. The NIR spectra of blood plasma samples from mastitic and healthy cows are clearly discriminated between by the PDV method. Moreover, the proposed method provides superior performance to PCA, DPLS, SIMCA and FLDA, indicating that PDV is a promising tool in discriminant analysis of spectra-characterized samples with only small compositional difference, thereby providing a useful means for spectroscopy-based clinic applications.

Robust Design for Showerhead Thermal Deformation

  • Showerhead is used as a main part in the semiconductor equipment. The face plate flatness should remain constant and the cleaning performance must be gained to keep the uniformity level of etching or deposition in chemical vapor deposition process. High operating temperature or long period of thermal loading could lead the showerhead to be deformed thermally. In some case, the thermal deformation appears very sensitive to showerhead performance. This paper describes the methods for robust design using computational fluid dynamics. To reveal the influence of the post distribution on flow pattern in the showerhead cavity, numerical simulation was performed for several post distributions. The flow structure appears similar to an impinging flow near a centered baffle in showerhead cavity. We took the structure as an index to estimate diffusion path. A robust design to reduce the thermal deformation of showerhead can be achieved using post number increase without ill effect on flow. To prevent the showerhead deformation by heat loading, its face plate thickness was determined additionally using numerical simulation. The face plate has thousands of impinging holes. The design key is to keep pressure drop distribution on the showerhead face plate with the holes. This study reads the methodology to apply to a showerhead hole design. A Hagen-Poiseuille equation gives the pressure drop in a fluid flowing through such hole. The assumptions of the equation are the fluid is viscous-incompressible and the flow is laminar fully developed in a through hole. An equation can be expressed with radius R and length L related to the volume flow rate Q from the Hagen-Poiseuille equation, $Q={\pi}R4{\Delta}p/8{\mu}L$, where ${\mu}$ is the viscosity and ${\Delta}p$ is the pressure drop. In present case, each hole has steps at both the inlet and the outlet, and the fluid appears compressible. So we simplify the equation as $Q=C(R,L){\Delta}p$. A series of performance curves for a through hole with geometric parameters were obtained using two-dimensional numerical simulation. We obtained a relation between the hole diameter and hole length from the test cases to determine hole diameter at fixed hole length. A numerical simulation has been performed as a tool for enhancing showerhead robust design from flow structure. Geometric parameters for the design were post distribution and face plate thickness. The reinforced showerhead has been installed and its effective deposition profile is being shown in factory.

Results obtained demonstrate that the PDV method exhibits improved stability in prediction without significant loss of separability. The NIR spectra of blood plasma samples from two cows are clearly discriminated between by the PDV method. Moreover, the proposed method provides superior performance to PCA, DPLS, SIMCA md FLDA, indicating that PDV is a promising tool in discriminant analysis of spectra-characterized samples with only small compositional difference.

Cloning of Farm Animals in Japan; The Present and the Future

  • 1. About fifty thousand of cattle embryos were transferred and 16000 ET-calves were born in 1999. Eighty percents of embryos were collected from Japanese Black beef donors and transferred to dairy Holstein heifers and cows. Since 1985, we have achieved in bovine in vitro fertilization using immature oocytes Collected from ovaries of slaughterhouse. Now over 8000 embryos fertilized by Japanese Black bull, as Kitaguni 7 -8 or Mitsufuku, famousbulls as high marbling score of progeny tests were sold to dairy farmers and transferred to their dairy cattle every year. 2. Embryo splitting for identical twins is demonstrated an useful tool to supply a bull for semen collection and a steer for beef performance test. According to the data of Dr.Hashiyada (2001), 296 pairs of split-half-embryos were transferred to recipients and 98 gave births of 112 calves (23 pairs of identical twins and 66 singletons). 3. A blastomere-nuclear-transferred cloned calf was born in 1990 by a joint research with Drs.Tsunoda, National Institute of Animal Industry (NIAI) and Ushijima, Chiba Prefectural Farm Animal Center. The fruits of this technology were applied to the production of a calf from a cell of long-term-cultured inner cell mass (1998, Itoh et al, ZEN-NOH Central Research Institute for Feed and Livestock) and a cloned calf from three-successive-cloning (1997, Tsunoda et al.). According to the survey of MAFF of Japan, over 500 calves were born until this year and a half of them were already brought to the market for beef. 4. After the report of "Dolly", in February 1997, the first somatic cell clone female calves were born in July 1998 as the fruits of the joint research organized by Dr. Tsunoda in Kinki University (Kato et al, 2000). The male calves were born in August and September 1998 by the collaboration with NIAI and Kagoshima Prefecture. Then 244 calves, four pigs and a kid of goat were now born in 36 institutes of Japan. 5. Somatic cell cloning in farm animal production will bring us an effective reproductive method of elite-dairy- cows, super-cows and excellent bulls. The effect of making copy farm animal is also related to the reservation of genetic resources and re-creation of a male bull from a castrated steer of excellent marbling beef. Cloning of genetically modified animals is most promising to making pig organs transplant to people and providing protein drugs in milk of pig, goat and cattle.

작물학 분야 프로테오믹스의 응용과 전망 (Application and perspectives of proteomics in crop science fields)

  • Thanks to spectacular advances in the techniques for identifying proteins separated by two-dimensional electrophoresis and in methods for large-scale analysis of proteome variations, proteomics is becoming an essential methodology in various fields of plant sciences. Plant proteomics would be most useful when combined with other functional genomics tools and approaches. A combination of microarray and proteomics analysis will indicate whether gene regulation is controlled at the level of transcription or translation and protein accumulation. In this review, we described the catalogues of the rice proteome which were constructed in our program, and functional characterization of some of these proteins was discussed. Mass-spectrometry is a most prevalent technique to identify rapidly a large of proteins in proteome analysis. However, the conventional Western blotting/sequencing technique us still used in many laboratories. As a first step to efficiently construct protein data-file in proteome analysis of major cereals, we have analyzed the N-terminal sequences of 100 rice embryo proteins and 70 wheat spike proteins separated by two-dimensional electrophoresis. Edman degradation revealed the N-terminal peptide sequences of only 31 rice proteins and 47 wheat proteins, suggesting that the rest of separated protein spots are N-terminally blocked. To efficiently determine the internal sequence of blocked proteins, we have developed a modified Cleveland peptide mapping method. Using this above method, the internal sequences of all blocked rice proteins (i. e., 69 proteins) were determined. Among these 100 rice proteins, thirty were proteins for which homologous sequence in the rice genome database could be identified. However, the rest of the proteins lacked homologous proteins. This appears to be consistent with the fact that about 30% of total rice cDNA have been deposited in the database. Also, the major proteins involved in the growth and development of rice can be identified using the proteome approach. Some of these proteins, including a calcium-binding protein that fumed out to be calreticulin, gibberellin-binding protein, which is ribulose-1,5-bisphosphate carboxylase/oxygenase activate in rice, and leginsulin-binding protein in soybean have functions in the signal transduction pathway. Proteomics is well suited not only to determine interaction between pairs of proteins, but also to identify multisubunit complexes. Currently, a protein-protein interaction database for plant proteins (http://genome .c .kanazawa-u.ac.jp/Y2H)could be a very useful tool for the plant research community. Recently, we are separated proteins from grain filling and seed maturation in rice to perform ESI-Q-TOF/MS and MALDI-TOF/MS. This experiment shows a possibility to easily and rapidly identify a number of 2-DE separated proteins of rice by ESI-Q-TOF/MS and MALDI-TOF/MS. Therefore, the Information thus obtained from the plant proteome would be helpful in predicting the function of the unknown proteins and would be useful in the plant molecular breeding. Also, information from our study could provide a venue to plant breeder and molecular biologist to design their research strategies precisely.

  • 최근 개발되고 있는 의료기기들은 사용자의 상호 복합적인 요구사항을 반영하기 위하여 모듈(Module)화되는 추세이다. 이는 환자의 상태를 관찰, 진단, 처리를 위해 여러 기기들로부터 데이터를 수집하고, 이에 따르는 동작을 제어하기에 가장 효율적인 방법이기 때문이다. 이와 같은 모듈화 경향은 여러 개의 단일 시스템들을 쉽게 통합 관리할 수 있기 때문에 더욱 일반화되고 있다. 본 연구에서는 모듈화된 의료장비들을 하나의 통신 시스템으로 묶어 중앙 집중형 제어를 할 수 있는 통신 프로토콜을 구현하였다. 본 연구에서 구현된 시스템은 모든 기기들을 제어하고 통신을 관장하는 하나의 마스터(Master) 모듈과 각각의 의료기기들에 해당되는 여러 개의 슬레이브(Slave)모듈로 구성된다. 각 모듈간 통신은 데이터의 정확한 송, 수신을 위해 여러 동기 직렬 통신방식 중 SPI(Serial Peripheral Interface)를 사용하였다. 모든 통신은 패킷(Packet)형태로 이루어지고, 통신중 에러를 검출할 수도 있다. 또한, 마스터 모듈은 자동으로 특정 슬레이브 모듈외 연결 및 동작 유무를 알 수 있는 PNP(Plug And Play)기능이 있다. 본 연구에서 구현된 프로토콜온 컴퓨터와 연결하여 데이터 전송 확인을 통해, 1Mbps이상의 빠른 속도에서 정화한 송, 수신이 이루어지며, 인공호흡기와 같은 실시간 데이터의 송, 수신이 이루어져야하는 의료장비에서도 모듈이 수행하는 본래의 일에 방해하지 않고 실시간 통신이 이루어지는 것을 실제 호흡기 시스템에 적용하여 확인하였다. 또한, 중앙 집중형 제어에 의한 다양한 기능을 편리하게 구현할 수 있었다.이다.ure scheme based on KCDSA which is a domestic digital signature scheme and it apply a electronic cash system.   • 1994년 처음으로 국내 지반공학자들에게 소개된 수압파쇄 시험법은, 측정장비 및 해석방법에 있어서 많은 발전을 이루어 왔다. 파이프라인 타입이었던 제1세대 수압파쇄 시험장비는 다루기가 쉽지 않았고 파이프 연결부위에서의 압력 누설 문제가 대두되면서, 제2세대라 불리는 와이어라인 타입으로 보완된 바 있으나, 기존의 시스템에 비해 많은 장점을 가짐에도 불구하고 공기압축기를 별도로 설치해야 하는 문제점을 여전히 안고 있어, 2004년 이후 이를 모두 포함하는 현재의 제3세대 all-in-one 시스템으로 보완된 바 있다. 장비의 발전과 함께, 응력해석을 위한 소프트웨어의 발전도 꾸준히 진행되어온 바, 불확실한 결정값으로 인해 가장 다루기 어려운 균열폐쇄압력에 대하여 여러 가지 기법을 적용함으로써 가장 합리적인 값을 도출해내는 방법의 개발이 그 한 예가 될 것이다. 이러한 지난 10여년 간의 기술발전과 함께 국내 대부분의 지역에서 수행된 수압파쇄시험을 통하여 대한민국의 거의 전 지역에 대한 측정결과들을 축적할 수 있었으며, 현재 이 결과값은 터널굴착을 위한 기초설계단계에서 널리 활용되고 있다. 향후 해저터널의 건설과 관련하여 해상에서의 수압파쇄 초기응력측정법 또한 꾸준히 개발되고 있으나, 지금까지 축적된 내륙에서의 초기응력 측정값이 해저터널의 안전한 설계를 위해 직간접적으로 사용되어지길 기대하며, 현재까지 수압파쇄시험을 통해 얻어진 한반도의 초기응력의 평균 크기와 방향을 지질학적 특성결과와 관련 지어 제시하고자 한다.

  • 이 연구의 목적은 글로벌 해운항만물류에 있어 창조형 서비스산업으로서 금융특화와 금융지원을 통한 해양금융의 허브화와 해양물류 클러스터를 구축하여 국제경쟁력강화를 통한 국가경제발전에 이바지함에 있다. 효과적인 목적달성을 위해 이 연구는 통합적 접근법을 채택하여 효율적인 정책이행에 적용하였으며, 정책수행의 도구로서 창조형 해양금융발전 단계를 수립하여 정책이행시기의 가이드라인을 다음과 같이 제시하였다. 제1단계(조성과 성장정책): 2013~2016(창조형 해양금융산업집적지구축), 제2단계(육성/활성화정책): 2017~2019(창조형 해양금융산업클러스터구축), 제3단계(지속적 발전정책): 2020년 이후 장기과제(창조형 해양금융혁신클러스터구축). 세계해양금융의 경쟁력 열위에 있는 우리나라는 해양금융의 유동성확보를 위한 전략적 접근이 절실한 바, 근래 글로벌 금융위기 이후 실물거래를 수반하는 금융 거래의 새로운 대안으로 이슬람금융이 급부상하고 있다. 아울러 본 연구에서는 해양금융의 실무상 우리나라의 잠재형 슬랙을 창출하기 위해 이슬람금융권과의 컨소시엄의 구성을 초기단계에서의 보다 용이한 정책이행의 단초로 제안하고 있다.

지능망서비스의 발전방향에 관한 연구 (A study on the new developmental direction for Intelligent Network Service)

  • 미래 정보사회에는 이용자들의 다양한 서비스가 요구되며, 이용자들의 요구를 충족시키기 위해서는 기존의 공중통신망으로는 어렵다는 것이다. 따라서 통신사업자는 소비자의 다양한 서비스에 부흥하기 위해 지능망서비스의 수준을 향상시켜야 할 것이다. 본 연구는 지능망서비스의 향상을 위해 현행 문제점을 두 가지 측면에서 분석하였다. 첫째, 기술적 측면의 문제점은 새로운 기능이나 신규서비스의 추가 및 서비스 제어기능 모두의 해당 소프트웨어를 변경해야 하는 문제점, 각 서비스 별로 다른 서비스 제어기능을 사용하거나 동일 서비스 제어기능을 사용하더라도 데이터베이스 구조를 각각 달리하고 있어 서비스의 운용 및 관리의 복잡성 문제, 두 가지 이상의 서비스를 조합해서 제공하는 복합 서비스의 경우 호 접속 지연의 문제점, 대규모의 데이터베이스 및 신속한 호처리를 필요로 하는 서비스에 대한 효과성의 문제, 둘째, 서비스 측면의 문제점은 지능망서비스의 시장지향적 서비스 개발 및 고객욕구 서비스 개발의 미비한 점, 지능망서비스의 통신요금은 통신시설의 확장을 위한 투자 재원 마련과 정부의 공공요금 인상 억제정책 및 독점적 경향에 의해 결정되는 문제점, 지능망서비스의 번호체계를 보면 너무 복잡성 문제점이 나타났다. 이러한 문제점을 해결하기 위해서는 첫째, 지능망서비스의 체계적인 개발 둘째, 지능망서비스에 적합한 시설 확충 셋째, 유선망 무선망·인터넷망을 통합 및 멀티미디어 시대의 서비스 지원 넷째, 고객중심의 서비스 제공 다섯째, 원가개념 도입에 따른 요금 결정 여섯째, 지능망서비스 번호체계의 합리화를 추구해야 지능망서비스의 미래 정보사회에 맞게 발전시킬 수 있을 것이다.ure scheme based on KCDSA which is a domestic digital signature scheme and it apply a electronic cash system.
