Study of 8-Hydroxyquinoline Derivatives. The Acid Dissociation Constants of 5-Acetyl-8-Hydroxyquinoline (8-Hydroxyquinoline 誘導體에 關한 硏究 5-Acetyl-8-Hydroxyquinoline의 酸解離定數)

  • Acid dissociation constants of 5-acetyl-8-hydroxyquinoline have been determined by spectrophotometric and potentiometric methods at $25^{\circ}C$. $pK_1$, $pK_2$ values obtained by potentiometric method were 4.56, 8.20 respectively and $pK_1$ obtained by spectrophotometric method was 4.21. pK values were lower than those of 8-hydroxyquinoline. The lower basicity of 5-acetyl-8-hydroxyquinoline compared with 8-hydroxyquinoline was considered as the result of the electronic attraction of the acetyl group with aromatic ring.

  • 7-Nitro-8-hydoxyquinoline-5-sulfonate (NHQS) have been synthesized from 8-hydroxyquinoline-5-sulfonic acid by nitrosation in alkaline medium at temperature below $10^{\circ}C$ and the acid dissociation constants of NHQS have been determined by spectrophotometric and potentiometric methods. Almost same values are obtained by both methods but the values are lower than the corresponding 8-hydroxyquinoline. The lower basicity of NHQS is considered as the effect of electron withdrawing groups, especially adjacent nitroso group.

  • The properties of Fe(II)-7-nitrose-8-hydroxyquinoline-5-sulfonate have been studied spectrophotometrically at 700$m{\mu}$, $26^{\circ}C$. The absorbance of the complex shows a maximum at pH 6.0 and good thermal stability. Excess of the ligand and the reducing agent do not interfere the formation of the complex. The composition of the complex is found to be 3:1 ligand to metal species by molar ratio and continues variation methods. Calibration curve follow Beer's law over the range of concentratiion studied.

Genetic Toxicity Test of 8-Hydroxyquinoline by Ames, Micronucleus, Comet Assays and Microarray Analysis

  • 8-Hydroxyquinoline is used as antibacterial agent and antioxidant based on its function inducing the chelation of ferrous ion present in host resulting in production of chelated complex. This complex being transported to cell membrane of bacteria and fungi exerts antibacterial and antifungal action. In this study, we have carried out in vitro genetic toxicity tests and microarray analysis to understand the underlying mechanisms and the mode of action of toxicity of 8-hydroxyquinoline. TA1535 and TA98 cells were treated with 8-hydroxyquinoline to test its toxicity by basic genetic toxicity test, Ames and two new in vitro micronucleus and COMET assays were applied using CHO cells and L5178Y cells, respectively. In addition, microarray analysis of differentially expressed genes in L5178Y cells in response to 8-hydroxyquinoline were analyzed using Affymatrix genechip. The result of Ames test was that 8-hydroxyquinoline treatment increased the mutations in base substitution strain TA1535 and likewise, 8-hydroxyquinoline also increased mutations in frame shift TA98. 8-Hydroxyquinoline increased micronuclei in CHO cells and DNA damage in L5178Y. 8-Hdroxyquinoline resulted in positive response in all three tests showing its ability to induce not only mutation but also DNA damage. 783 Genes were initially selected as differentially expressed genes in response to 8-hydroxyquinoline by microarray analysis and 34 genes among them were over 4 times of log fold changed. These 34 genes could be candidate biomarkers of genetic toxic action of 8-hydroxyquinoline related to induction of mutation and/or induction of micronuclei and DNA damage. Further confirmation of these candidate markers related to their biological function will be useful to understand the detailed mode of action of 8-hydroxyquinoline.

Atomic Absorption Spectrophotometric Determination of Trace Cadimuim after Preconcentration by Extracting Its 8-Hydroxyquinoline Complex into Molten Benzophenone

  • A sensitive method for the determination of trace cadmium after the preconcentration by extracting its 8-hydroxyquinoline complex into a molten benzophenone was developed. Several experimental conditions such as the pH of solution, the amounts of 8-hydroxyquinoline and benzophenone, stirring time, and standing time were optimized. Trace cadmium in 100 mL water sample was chelated with 2.5 mL of 0.001 M 8-hydroxyquinoline at pH 8.0. After 0.07 g benzophenone was added, the solution was heated to about 70 ℃ and stirred vigorously for 1 minute to dissolve the complex quantitatively in a molten benzophenone, and stood for 30 minutes to reproduce the microcrystalline benzophenone. The benzophenone containing Cd-8-hydroxyquinoline complex was filtered and dissolved in acetone. Cadmium was determined by a flame atomic absorption spectrophotometry. The interfering effects of diverse concomitant ions were investigated and eliminated. This method could be applied to natural water samples and the recovery of more than 90% was obtained in the real samples.

  • The quantitative determination of chromium(VI) by separation from chromium(III) complex of 8-hydroxyquinoline using solvent extraction has been studied. The reaction conditions for chromium(III) complex of 8-hydroxyquinoline and the solvent extraction of complex were investigated in detail. The chromium(III) complex was extracted with organic solvent (n-hexane) and residual chromium(VI) was determined by ICP-AES in aqueous layer. This technique is quantitative in the pH range of 8-9 and the limitations such as interfering ions were discussed.

  • Stabilities of chelates of 7-nitroso-8-hydroxyquinoline-5-sulfonate have been determined for divalent transition metal ions, Mn(Ⅱ), Fe(Ⅱ), Co(Ⅱ), Ni(Ⅱ), Cu(Ⅱ), and Zn(Ⅱ) by means of the Calvin-Bjerrum technique. Comparison of these stability constants with those obtained for 8-hydroxyquinoline, and 8-hydroxyquinoline-5-sulfonate shows that the observed differences are essentially the results of the lower basicity of the sulfonated group and different metal-ligand bond. The divalent metal chelate stability sequence is not in agreement with the reported metal orders for other chelating agents. The stabilities were found to follow the order Mn(Ⅱ) < Fe(Ⅱ) ${\approx}$ Co(Ⅱ) > Ni(Ⅱ) < Cu(Ⅱ) > Zn(Ⅱ).

Study on Optical Characteristics of 8-Hydroxyquinoline Synthesized Derivative as Sensing Material of the Fiber-Optic Copper Ion Sensor in Aqueous Environment (수질환경에서 광섬유 센서의 구리 이온 감지 물질로서 8-Hydroxyquinoline 합성유도체의 광학적 반응 특성 연구)

  • An 8-hydroxyquinoline compound that was synthesized with 8-hydroxyquinoline-2-carboxaldehyde and 4-aminoantipyrine was investigated for use as the sensing material of a fiber-optic copper ion sensor in an aqueous environment. The experiment was conducted with a fiber-optic measurement system, in order to evaluate the relationship between the absorbance peak and copper ion concentration. The synthesized derivative exhibited a (highly selective) chromogenic phenomenon for copper ions among various metal ions in an aqueous environment and showed a specific absorbance peak at a wavelength of 530 nm for copper ions. The effect of mercury ions was investigated to evaluate the selectivity of the prepared synthesized derivatives toward Cu ions. The absorbance was measured at various concentration ratios of Cu and Hg ions (Cu:Hg ratios from 0.05 to 20), and it was found that the absorbance at 530 nm tended to increase with increasing Cu ion concentration. The experimental results also showed the linear relationship between the logarithmic concentration of copper ions and the specific absorbance peak at a wavelength of 530 nm. These results indicate that the synthesized 8-hydroxyquinoline compound has selectivity for copper ions and can be used as a sensing material for fiber-optic copper ion sensors.

2-Hydroxyquinoline and Its Structural Analogs Show Antidiabetic Effects against α-Amylase and α-Glucosidase

  • This study investigated the inhibitory activities of 2-hydroxyquinoline and its analogs against ${\alpha}$-glucosidase and ${\alpha}$-amylase. Based on the $IC_{50}$ values of 2-hydroxyquinoline analogs tested against ${\alpha}$-glucosidase and ${\alpha}$-amylase, 2-hydroxyquinoline had potent inhibitory activity (64.4 and $130.5{\mu}g/mL$, respectively), while 2-methyl-8-hydroxyquinoline showed weakly inhibitory activity (90.7 and $215.4{\mu}g/mL$, respectively). 2-Methylquinoline demonstrated no activity against ${\alpha}$-glucosidase and ${\alpha}$-amylase. In conclusion, 2-hydroxyquinoline analogs, with the existence of a methyl group and hydroxyl on quinoline, can be useful as a new diabetes treatment.

  • Organic electroluminescent devices have attracted a great deal of attention due to thier potential application to full-color flat-panel displays. The 8-hydroxyquinollne Zinc(Znq$_2$) were synthesized successfully from zinc chloride(ZnCl$_2$) and zinc acetate(Zn(C$_2$H$_3$O$_3$)$_2$) as green omitting material. A double-layer ELD consist of an emitting layer of B-hydroxyquinoline Zinc(Znq$_2$) and a hole-transport layer of tai-phenylene diamine(TPD) derivatives sandwiched between an Aluminium(Al) and Indium-Tin-Oxide(ITO) electrodes omitted green light resulting from Znq$_2$. The electroluminescent devices (ELD) exhibited a maximum luminance of 1000cd/$\textrm{cm}^2$ at a driving voltage of 8V and a driving current density of 0.4mA/$\textrm{cm}^2$.
