Effect of Phenanthrene on Gill Biotransformation in Olive Flounder (Paralichthys olivaceus)

  • The potential of phenanthrene to induce xenobiotic responses was investigated. Olive flounder (Paralichthys olivaceus) was exposed to different levels of phenanthrene $(0.5,\;1\;and\;{\mu}M)$ for 4 weeks, Gill CYP450 content and EROD (ethoxyresorufin O-deethylation) activity were found to be significant in the flounders treated with the higher concentration of phenanthrene $(>1.0\;{\mu}M)$, however, there were no significant changes in gill PROD (penthoxyresorufin O-deethylation) activity in all treated group compared to the controls. This study demonstrated that phenanthrene has potential to induce gill cytochrome P450 and EROD enzyme in olive flounder.

  • CYP1 enzymes, CYP1A1, CYP1A2 and CYP1B1, are known to bioactivate procarcinogens particularly polyaromatic compounds. Flavonoids are a class of natural compounds that are present in edible plants. Structurally, these compounds are polyphenols with two aromatic rings (A, B) and a heterocycyclic ring (C). We observed the differential inhibition of 5,7-dihydroxyflavones which are different in numbers of B-ring-OH, to the activity of ethoxyresorufin O-deethylase (EROD) in human hepatic microsomes with the IC50 values, ie, 0.57 mM, 1.28 mM, and 3.62 mM, chrysin, apigenin, and Luteolin, respectively. Endosulfan이 흰쥐체내의 Cytochrome P-450 효소계에 미치는 영향 (Effects of Endosulfan on Cytochrome P-450 Enzymes in Mouse(Balb/c.))

  • Endosulfan이 흰쥐(Balb/c.) 체내의 cytochrome P-450 효소계에 미치는 영향을 조사하기 위해 endosulfan을 7.5 mg/kg 수준으로 복강주사하였다. Endosulfan을 복강주사 처리하여 48시간 후 적출한 흰쥐의 간에서는 cytochrome P-450 함량이 $3.3{\sim}4.2$배, cytochrome $b_5$ 함량이 $2.3{\sim}3.8$배, NADPH cytochrome P-450 reductase 활성이 $5.3{\sim}6.4$배 그리고 총 haem 함량이 $3.1{\sim}3.6$배씩 대조구의 그것들에 비해 증가하였다. Endosulfan은 대조구 흰쥐 간의 cytochrome P-450 효소계와 상호작용하여 파장 387와 389 nm에서 흡광도의 증가를 보였으며 파장 407 nm에서 넓은 흡수대를 형성하였다. 환원형 P-450-CO spectrum은 대조구의 경우 파장 451 nm에서 흡광도의 극대를 보인 반면 endosulfan으로 처리된 흰쥐 간의 그것은 파장 449와 450 nm에서 흡광도의 극대를 보였다. Endosulfan 처리로 흰쥐 간과 신장의 aldrin epoxidase 활성이 각각 2.8배와 2.1배씩 증가하였으며 7-ethoxyresorufin dealkylase 활성은 각각 1.7배와 1.8배씩 증가하였다.

PCBs노출에 의한 틸라피아, Oreochromis niloticus의 간 cytochrome P450 효소계의 변화 (Changes of Hepatic Microsomal Cytochrome P450 Monooxygenase System in Nile tilapia, Oreochromis niloticus Exposed to PCBs)

  • PCBs의 경구 투여에 의한 틸라피아, Oreochromis niloticus의 간 cytochrome P450 효소계의 변화를 30일간 관찰하였다. 30일 동안 PCBs 0.05, 0.25 및 0.50mg/kg을 투여했을 때, HSI는 각각 1.81, 1.72 및 $1.54\%$ 감소하였으나, 유의한 차이는 인정되지 않았다. PCBs 0.05mg/kg에 노출된 간 cytochrome P450 농도는 30일째 대조구에 비해 1.4배 유의한 증가를 나타냈고, 0.25mg/kg 이상에서는 실험 20일 이후부터 유의적으로 증가하였다. EROD 활성도는 PCBs 0.05 mg/kg에서 실험 30일째부터 유의하게 증가하여 약 8배 높게 나타났다. 또한, PCBs 0.25mg/kg이상에서는 실험 20일째부터 유의하게 증가하여 30일째부터는 각각 18 및 22배의 증가를 나타냈다. PROD는 PCBs $0.05\~0.50mg/kg$에 노출시킨 개체에서 $0.78\~1.93$pmole/mg protein이 측정되어 대조구에 비해 뚜렷한 변화는 관찰되지 않았다. 이상의 결과는 자연 수역에서 PCBs의 잔류에 따라 먹이를 통해 틸라피아의 체내에 적어도 0.05 mg/kg 이상이 30일간 이상 계속적으로 유입될 경우에 간 cytochrome P450농도 및 EROD 활성도는 증가될 것으로 추정된다.

  • The carbon tetrachloride($CCl_4$) has been demonstrated to have a hepatotoxic effect in human or many other species. To investigate the enzyme induction of mixed function oxygenases in liver of male Sprague-Dawley rats a single 0.1, 0.5 mι/kg dose of carbon tetrachloride were given. At 24 hr after a single dose of 0.1 mι CC1$_4$/kg weight, methanol extract of Hedera rhombea leaves was administered with 100, 500 mg/kg weight. Assays of 7-ethoxyresorufin-Ο-deethylation(EROD),7-benzyloxyresorufin-Ο-deathylation(BROD),4-nitro-phenol-UDP-glucuronosyltransferase(UDPGT), Western blot and RNA slot blot were used as representatives of the activities of cytochrome P-450 enzymes. The change of the activity of CYP1A1 form measured by EROD assay and Western analysis using 1-7-1 monoclonal antibody was not observed. The activity CYP2B1 form by BROD assay and using 2-66-3 monoclonal antibody was remarkably increased. Elevated level of CYP2B1 mRNA was shown by slot hybridization with 2B1-specific probe. Administration of methanol extract of Hedera rhombea leaves reversed the enzyme activity and the level of mRNA, which suggest the chemoprotective effect of methanol extracts of Hedera rhombea leaves to carbon tetrachloride hepatotoxlcity.

  • It is it well known fact that the most relevant toxic emissions such as polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), dioxin-1ike polychlorinated biphenyls (PCBs) are produced from incinerators. Induction of cytochrome P450IA1 catalyzed 7-ethoxyresorufin 0-deethylase (EROD) activity in mammalian cell culture (EROD-microbioassay) is a selective and sensitive parameter used for the quantification of dioxin-like compounds.   • Previous results from several laboratories have demonstrated that tumor necrosis factor-alpha (TNFalpha) depressed cytochrome P-450 (P-450)-dependent drug metabolism in vivo. However, there is some debate whether the action of TNFalpha is mediated by its direct effects on hepatocytes, or is indirectly mediated through the release of other mediators like IL-1 from macrophages. In the present studies, we investigated the effects of TNFalpha on P-450-dependent drug metabolizing enzyme as measured by 7-ethoxyresorufin O-deethylase (EROD) activity.

  • In mammalian, cytochrome P4501A1 (CYP1A1) is very important for metabolism of xenobiotics such as PAHs(Polycyclic aromatic hydrocarbon) and heterocyclic amine, and it is induced by environmental contaminants such as PAHs, TCDD(2,3,7,8-tetrchlorodibenzo-p-dioxin) and 3-MC (3-methylcholanthrene). In fish, like mammalian, when it is exposed to environmental contaminants, they cause specific and sensitive induction of CYP1A. Therefore, induction of CYP1A in fish and mammalian is widely used as a biomarker for exposure of environmental contaminants. In this study, to compare the function of Cyp1a1 in fish with it in mammalian, we have used rainbow trout(Oncorhynchys mykiss) hepatoma cells (RTH-149) and mouse hepatocyte (Hepa-I). in order to examine induction of Cyp1a1 by TCDD, we have used the bioassay system. We examined effects of TCDD on the Cyp1a1-luciferase reporter gene activity, 7-ethoxyresorufin O-deethylase(EROD) activity and Cypa mRNA level.

세포배양 생화학적 기법에 의한 수환경오염 평가 (Cell Culture Microbioassay for the Water Pollution Monitoring)

  • So far, investigation of environmental pollution has been achieved in field study. This remains the most exhaustive approach, current dimensions of environmental researches and their inherent complexity require that relatively inexpensive and simple laboratory procedures are developed to make possible the screening of large numbers of sites and samples. At this point. microbioassay has been high-lighted. The purpose of this study is to evaluate the water pollution using microbioassay. Two microbioassay methods were optimized and validated for the sensitive and quantitative determination of total toxic effects in the water. EROD(Ethoxyresorufin-O-deethylase) microbioassay was focused to detect PARs, PCBs and dioxinlike components in the water and E-screen assay to xenoestrogens. The EROD microbioassay was executed in rat hepatoma cell line, H4IIE and E-screen assay in MCF7-BUS cell line. Kumho river was selected for this study. 5ι of river water was extracted using combined solid-phase extraction in static adsorption mode with soxhlet extraction. Pollutants adsorbed to the XAD-4 resin were recovered by elution with ethyl acetate and methylene chloride (1 : 9). Toxic effects of extracts were determined by EROD-microbioassay and E-screen assay. EROD activities of water samples were 7.24-72.24 ng/ι MEQ. The estrogenic effect of various water samples is quantitatively evaluated by EEQ. The EEQ of samples range from 0.05 to 6.07 ng-EEQ/ι. These results suggested that Kumho river was highly polluted with organic toxic chemicals.
