Effects of Saengjihwangeumja-gami on STZ-induced Diabetic Mice (생지황음자가미(生地黃飮子加味)가 Streptozotocin으로 유발된 당뇨생쥐에 미치는 영향(影響))

  • First, mice were dosed with 50mg/kg of streptozotocin(STZ) twice every 24 hours to cause high blood-sugar. Then, after three days, mice were injected with 100mg/kg of STZ again. Two different dosages of Saengjihwangeumja-gami were given to the experiment groups: SA group, 15mg/kg/day, and SB group, 90mg/kg/day, in order to determine the effects of Saengjihwangeumja-gami, which has been known to be good for DM(Diabetes Mellitus). By observing weight and blood-sugar level changes, blood tolerance, the numerical value of BUN(Blood Urea Nitrogen) and creatinine in blood, and through light-electronicmicroscopic and immunohistologic investigations of pancreas and kidneys, the following results were obtained: 1. The experiment groups showed a high suppressive effect of weight-loss. 2. The experiment groups' blood-sugar and blood tolerance showed an effective lowering of blood-sugar levels. 3. The experiment groups did not show any noticeable change in the numerical value of BUN and creatinine in blood compared with that of the control groups. 4. The experiment groups showed a higher Insulin positive reaction of pancreatic islets ${/beta}-cell$ than the control groups. 5. The experiment groups showed a higher immuno-reaction against IGF- II than the control groups. 6. Observation of apoptosis of the pancreatic islets showed that the cells of experiment groups were less injured compared with those of the control groups, and fewer apoptag-positive reaction cells were seen in experiment groups than in the control groups. 7. Uunder electron-microscopy, the insulin-containing granules in pancreatic islets ${/beta}-cells$ had increased more in the experiment groups than in the control groups. 8. Under light microscopy, the injury on the inner & outer membrane of the glomerulus and epithelial cells of capillaries and cells among vessels were fewer in the experiment groups than in the control groups. 9. More apoptag-positive reaction cells in the kidney were seen in the control groups than in the experiment groups. 10. PAS-positive reaction substances had increased more in the substrate among the vessels of a glomerulus belonging to the control group than those of the experiment group. 11. Uunder electron-microscopy, the nucleonic membrane, nucleoplasm and mitochondria of proximal and distal renal tubular were more injured in the control groups than in the experiment groups. In conclusion, strong evidence for the efficacy of Saengjihwangeumja-gami in lowering blood-sugar, and in recovery and generation of pancreatic tissues injured by DM was observed. Results suggest Saengjihwangeumja-gami is an effective treatment for DM. Further study of the principles of blood-sugar dropping effects of Saengjihwangeumja-gami are needed, as well as further study of recovery and regeneration of pancreatic tissues injured by DM.

Spatio-Temporal Patterns of Extreme Precipitation Events by Typhoons Across the Republic of Korea (태풍 내습 시 남한의 극한강수현상의 시.공간적 패턴)

  • In this study, spatio-temporal patterns of extreme precipitation events caused by typhoons are examined based on observational daily precipitation data at approximately 340 weather stations of Korea Meterological Administration's ASOS (Automated Synoptic Observation System) and AWS (Automatic Weather System) networks for the recent 10 year period (2002~2011). Generally, extreme precipitation events by typhoons exceeding 80mm of daily precipitation commonly appear in Jeju Island, Gyeongsangnam-do, and the eastern coastal regions of the Korean Peninsula. However, the frequency, intensity and spatial extent of typhoon-driven extreme precipitation events can be modified depending on the topography of major mountain ridges as well as the pathway of and proximity to typhoons accompanying the anti-clockwise circulation of low-level moisture with hundreds of kilometers of radius. Yellow Sea-passing type of typhoons in July cause more frequent extreme precipitation events in the northern region of Gyeonggi-do, while East Sea-passing type or southern-region-landfall type of typhoons in August-early September do in the interior regions of Gyeongsangnam-do. These results suggest that when local governments develop optimal mitigation strategies against potential damages by typhoons, the pathway of and proximity to typhoons are key factors.

A Study of Entomopathogenic Nematode at Heavy Metal Contents in the Polluted Soil of Kyungsangbuk-do Area (경상북도지역의 오염된 토양에서의 중금속 함량에 관한 곤충병원성 선충에 관한 연구)

  • The variety and density of entomopathogenic nematodes from the polluted soils of heavy metals were examined. In order to investigate the pollution of heavy metals in soil, 300 sites in kyungsangbuk-do were collected from March to October in 2001. We measured the contents of seven heavy metal elements (Cd, Cu, As, Hg, Pb, Cr^{6+}$, CN) and than cheesed soil of 25 sites with high concentration of heavy metals. The seven strains of nematodes were isolated from seven samples by silkworm host (Bombyx mori mori) and white trap. Isolated nematodes are composed of two families, one order. The members of Rhabditida were isolated in the soil with mean Cd content of 0.870 ppm. And they were isolated in the soil samples with As content less than 0.745 ppm. However they were isolated regardless of concentration of Cu and Pb. The members of Cylindrocorpidae were isolated in the soil samples with Cr^{6+}$ content less than 0.05 ppm. Any entomopathogenic nematode was not detected in the CN polluted soil. Isolated nematodes successfully cultured on the silkworm host and were confirmed the pathogenicity, multiplicity, and tolerance against various condition of preservation. Which proved its potential usefulness as biological agent.

EFFECTS OF AMYLASE ON THE DEMINERALIZATION IN HYDROXYAPATITE (Amylase가 Hydroxyapatite 탈회에 미치는 영향)

  • Salivary proteins which are produced in the saliary acinar cells have been known to be involved in the Calcium and phosphate metabolism. The acquired pellicle resulting from such metabolism is considered as a secondary defence membrane against tooth caries. In this respect, some proteins included in saliva probably play an important role in the prevention of demineralization in enamel. On the other hand, fluoride has long been known to prevent the demineralization of enamel by the inhibition of the growth of Streptococcus mutans(S. mutans) and by the chemical reaction with calcium and phosphate, Therefore, I have examined the roles of amylase and albumin in the demineralization of enamel and compared these preteins with fluoride in terms of anticariogenic effect. 1. The demineralization caused by S. mutans occurred slowly and progressively for the first 60 min, then the rate of demineralization was accelerated afterwards. 2. pH decreased continuously during the entire period of each experiment. 3. The demineralization was significantly inhibited by the preteatment of amylase and fluoride but albumin had little effect on it. 4. An addition of 0.1 mM lactic acid (final concentration 0.1 ${\mu}M$) caused a rapid increase in calcium concentration reaching a maximum within 10 min. 5. pH decreased rapidly by the addition of 0.1 mM lactic acid and reached a minimum within a few seconds followed by an increase in pH. pH reaced a plateu with 10 min. 6. Fluoride, amylase and albumin played little role in the 0.1 mM lactic acid-induced demineralization. 7. A slow infusion of 0.1 M lactic acid at a rate of 5 ${\mu}l/min$ caused a slower increase in calcium concentration compared with the bolus addition of lactic acid. 8. Fluoride had an inhibitory effect on the calcium release caused by slow infusion of lactic acid while amylase and albumin had no effect on it. These results suggest that fluoride inhibits demineralization by protecting the HA from the acid attack whereas amylase has a direct effect on S. mutans to prevent demineralization.

Experimental Studies on the Anti-tumor and the Immunomodulatory Effects of Jiaweicitaowan(加未慈桃丸) (가미자도환(加味慈桃丸)의 항암(抗癌) 및 면역증강효과(免疫增强效果)에 관한 부험적(實驗的) 연구(硏究))

  • This experimental study was carried out to evaluate the anti-tumor and the immunomodulatory effects of Jiaweicitaowan(加未慈桃丸) against cancer. The in vitro anti-tumor effects were evaluated by MTT assay. The cytotoxicity, extension of survival days, the effect of inhibition solid tumor which was induced sarcoma 180, and the changes of body weight were evaluated for in vivo effects of anti-tumor. To evaluate the immunomodulatory effects of Jiawei- citaowan(加未慈桃丸), delayed type hypersensitivity, hemagglutinin, hemolysin titers for humoral immune response, rosette forming cells for cell-mediated immune response, natural killer cell activity, proliferation of lymphocyte, productivty of Interleukin-2, and carbon clearance were measured with methotrexate treated mice. The results were as follows; 1. In the case of existence ability of tumor cell, IC50 had an anti-tumor ativity resulted 2.52mg/ml to SNU-C4. 0.41mg/ml to SNU-396, resulted to 0.09mg/mlSNU-1. 2. The groups of Jiaweicitaowan(加未慈桃丸) 10mg/ml, 20mg/kg had no body weight loss. reduction in intake of water and feed, so these had no toxicity. 3. In the case of the effect of extention of existence. the group of 20mg/kg Jiaweicitaowan(加未慈桃丸) extract treated group was showed 250% in ILS. 4. The effect of inhibition solid tumor was significantly decreased in both 10mg/kg, 20mg/kg of Jiaweicitaowan(加未慈桃丸) extract treated groups as compared with control group S. The groups of 10mg/kg, 20mg/kg Jiaweicitaowan(加未慈桃丸) had significant effect of body weight change compared to control group. 6. Delayed type hypersensitivity was not significant in both Jiaweicitaowan(加未慈桃丸) extract treated groups as compared with control group. 7. Hemagglutinin and Hemolysin titers were significantly increased by dose-dependent. so these results showed that the humoral immume respose was activated. 8. For the effect of rosette formimg cells was not significant in hoth Jiaweicitaowan(加未慈桃丸) extract treated groups as compared with control group. 9. Natural killer cell activity was significantly increased in both Jiaweicitaowan(加未慈桃丸) extract treated groups as compared with control group in the ratio of 100: 1, 50: 1 of effector and target cells, but in the ratio of 10:1, the Jiaweicitaowan(加未慈桃丸) extract treated groups were not significant. 10. The proliferation of lymphocyte and productivty of Interleukin-2 were significantly increased by dose-dependent in both 10mg/ kg, 20mg/ kg of Jiaweicitaowan(加未慈桃丸) extract treated groups as compared with control group. 11. In the phagocytic effect, the 20mg/kg of Jiaweicitaowan(加未慈桃丸) extract treated group showed the increasing effect with significance as compared with control group. According to the results, we can suggest that Jiaweicitaowan(加未慈桃丸) has the antitumor and the immunomodulatory effects.

Development of Adsorbents for Edible Oil Refining using Agricultural Byproducts (농산부산물을 이용한 식용유지 정제용 흡착제 개발)

  • The purpose of this study was the development of adsorbents for the refining of edible oil using agricultural byproducts such as rice hull, barley hull, and soybean hull as well as evaluation of their adsorptive effects against free fatty acids and lutein, the major impurities of soybean oil. Ash-type and carbon-type adsorbents were produced from the hulls. Ash-type adsorbents such as rice hull ash (RHA), barley hull ash (BHA) and soybean hull ash (SHA) were effective for the removal of free fatty acids; the acid value of degummed soybean oil was decreased by 86% upon treatment with 5% SHA. However, carbon-type adsorbents such as rice hull carbon (RHC), barley hull carbon (BHC), and soybean hull carbon (SHC) were effective for removing lutein, resulting in a 52% decrease in the lutein content of degummed soybean oil upon treatment with 5% SHC. Whereas ash-type adsorbents were composed of mesopores or macropores with small surface areas and total pore volume, carbon-type adsorbents were mainly composed of micropores with large surface areas and total pore volume.

  • Male rats of the Albino strain received methylene blue in the dose of 40mg/kg by subcutaneous injection and were subjected to total body X-irradiation, 300 roentgen, at 30 minutes after the injection. The protective effect of methylene blue against the single total body X-irradiation was studied for 24 days after X-irradiation with regard to the levels of liver glycogen, blood glucose, and electrolytes in serum. 1. Total body X-irradiation generally casued an increase in the levels of the liver glycogen and blood glucose in both methylene blue treated and control group during the entire experiment. 2. Methylene blue has been shown to delay slightly the increase of the levels of the liver glycogen and blood glucose when comparing with both groups which were given methylene blue and control saline injection before irradiation in the rats. 3. The delay in the increase in the levels of liver glycogen, in experimental group injected with methylene blue, significantly came in two phases. The first phase appeared at there days after the exposure, the second followed at eighth day. It appeared that the recovery phase was at nineteenth day. 4. During the experimental days the levels of the blood glucose increased generally, methylene blue, however, caused delay in two phases; the first at fifth day, the second at eighteenth day after the exposure to X-rays. 5. In electrolytes, there was not a significant difference. The levels of chloride were, however, slightly decreased in both groups, levels of potassium appeared different in two phases at first day and twelfth day, and the levels of sodium appeared to show irregular changes at the same levels, but there was no significant difference. 6. It may be considered that methylene blue greatly reduces the sensitivity of rats to X-rays, provided that methylene blue is given before the exposure.

  • In order to develop a disease-resistant root stock for the growth of watermelon, an efficient regeneration system of the gourd(Lagenaria leucantha Duch.) inbred line GO701-2 via organogenesis was established in this experiment. Using proximal parts of cotyledon explant excised from germinated seedling in vitro, maximum adventitious shoot formation (39%) was achieved on MS medium where cytokinin (BA) and auxin (IAA) were added at a concentration of 3mg/L and 0.1mg/L, respectively. Roots of the elongated shoots were successfully formed on MS medium without adding any plant growth regulators. The cucumber CsGolS1 gene known as a resistance gene against biotic and abiotic stresses, was constructed into the binary vector pBI121 under the control of CaMV 35S promoter. When the gene was introduced into the genome of gourd by Agrobacterium-mediated transformation, putative transgenic plants were obtained with the transformation efficiency of approximately 20 percent.

Bioequivalence of Tigrin Capsule to Disgren Capsule (Triflusal 300 mg) (디스그렌 캅셀(트리플루살 300 mg)에 대한 티그린 캅셀의 생물학적 동등성)

  • Triflusal is a new antithrombotic agent which inhibits both platelet cyclooxygenase and c-AMP phosphodiesterase activity. The purpose of the present study was to evaluate the bioequivalence of two triflusal capsules, $Disgren^{TM}$ (Myung-In Pharmaceutical Co., Ltd.) and $Tigriri^{TM}$ (Hana Pharmaceutical Co., Ltd.) according to the guidelines of Korea Food and Drug Administration (KFDA). Eighteen normal male volunteers, $22.94{\pm}1.83$ in age and $63.7l{\pm}10.43$ kg in body weight, were divided into two groups and a randomized $2{\times}2$ cross-over study was employed. After one capsule containing 300 mg of triflusal was orally administered, blood was taken at predetermined time intervals and the concentrations of triflusal in serum were determined using HPLC method with UV detector. Pharmacokinetic parameters such as $AUC_t$ $C_{max}$ and $T_{max}$ were calculated and ANOVA test was utilized for the statistical analysis of the parameters. The results showed that the differences in $AUC_t$ $C_{max}$ and $T_{max}$ between two capsules were -0.30%, 0.81 % and -3.03%, respectively when calculated against the $Disgren_{TM}$ capsule. The powers $(1-{\beta})$ for $AUC_t$ $C_{max}$ and $T_{max}$ were 98.29%,84.73% and 81.02%, respectively. Minimum detectable differences $({\Delta})$ at ${\alpha}=0.1$ and $1-{\beta}=0.8$ were all less than 20% (e.g., 12.91%, 18.46% and 19.65% for $AUC_t$ $C_{max}$ and $T_{max}$ respectively). The 90% confid,ence intervals were all within ${\pm}20%$(e.g., $-8.97{\sim}8.37$, $-11.58{\sim}13.22$ and $-16.23{\sim}10.17$ for $AUC_t$ $C_{max}$ and $T_{max}$, respectively). All of the above parameters ($1-{\beta}, {\Delta}$ and 90% confidence intervals) met the criteria of KFDA for bioequivalence, indicating that $Tigriri^{TM}$ capsule is bioequivalent to $Disgren^{TM}$ capsule.

Characterization of Antibacterial Substance - Producing Bacillus subtilis Isolated from Traditional Doenjang (전통 된장으로부터 분리한 향균물질 생산 Bacillus subtilis의 특성)

  • A bacterium which has high enzymatic activities such as amylase, cellulase and protease was isolated from Korean traditional soybean food, doenjang. The isolated bacterium was identified to Bacillus subtilis HS25 by the test of morphological and biochemical properties according to Bergey's Manual of Systematic Bacteriology and API 50 CHL kit, and by the 16S rDNA sequence. The isolated B. subtilis HS25 had a potent antibacterial activity against food born causative or pathogenic bacteria. B. subtilis HS25 is endospore forming cell and contained flagella and abundant viscous material at the out layer of cell wall. It was rod type bacterium $(0.5{\sim}0.8{\times}3{\sim}5{\mu}m)$ having biochemical characteristics such as gram staining(+), catalase(+), oxidase(-) and hydrolysis of esculin(+). The optimal medium compositions for production of antibacterial substance in the B. subtilis HS25 were 1% of soluble starch, 0.5% of yeast extract, 0.5% of peptone and 0.05% of MgCl$_2{\cdot}6H_{2}O$. The optimum temperature and pH of the growth of the B. subtilis HS25 was 35$^{\circ}C$ and pH 7.5, respectively. The antibacterial activity was more high in neutral to a little alkaline pH (6.5-10.5) than in acidic pH. The optimal shaking speed to grow and to produce antibacterial substance of the B. subtilis HS25 was 160${\sim}$200 rpm. The optimal culture time for antibacterial activities of the bacterium were shown to be in the range of 12-36 hr.
