비차폐 환경에서의 고온초전도 SQUID 2차 미분기의 특성연구 (High-$T_c$ 2nd-order SQUID Gradiometer for Use in Unshielded Environments)

  • We have fabricated $∂^2$$B_{z}$ /$∂x^2$ type planar gradiometers and studied their properties in operation under various field conditions. $YBa_2$$Cu_3$$O_{7}$ film was deposited on $SrTiO_3$ (100) substrate by a pulsed laser deposition (PLD) system and patterned into a device by the photolithography with ion milling technique. The device consists of 3 pickup loops designed symmetrically Inner dimension and the width of the square side loops are 3.6 mm and 1.2 mm, respectively, and the corresponding dimensions of the center loop are 2.0 mm and 1.13 mm. The length of baseline gradiometer is 5.8 mm. Step-edge junction width is 3.0 $\mu\textrm{m}$ and the hole size of the SQUID loop is 3 $\mu\textrm{m}$ ${\times}$ 52 $\mu\textrm{m}$. The SQUID inductance is estimated to be 35 pH. The device was formed on a 20 mm ${\times}$ 10 mm substrate. We have tested the behavior of the device in various field conditions. The unshielded gradiometer was stable under extremely hostile conditions on a laboratory bench. Noise level 0.45 pT/$\textrm{cm}^2$/(equation omitted)Hz and 0.84 pT/$\textrm{cm}^2$/(equation omitted)Hz at 1 Hz for the shielded and the unshielded cases, which correspond to equivalent field noises of 150 fT/(equation omitted)Hz and 280 fT/(equation omitted)Hz, respectively. In spite of the short baseline of 5.8 mm, the high common-mode-rejection-ratio of the gradiometer, $10^3$, allowed us to successfully record magnetocardiogram of a human subject, which demonstrates the feasibility of the design in biomagnetic studies.

탐진강 및 도암만 지역 퇴적물의 퇴적지구화학적 특성과 환경영향 (Sedimentary Geochemical Characteristics and Environmental Impact of Sediments in Tamjin River and Doam Bay)

  • To examine the sedimentary geochemical characteristics of sediment in the Tamjin River and Doam bay, the analysis was conducted, using the sample obtained in February 2000, on the grain size and the contents of metallic elements and organic carbon. The factors that influence the geochemical behavior of metallic elements in the surface sediment are grain size, organism, surrounding soil and $CaCO_3$. To find out the pollution level of metallic elements, the enrichment factor (EF) and the index of geoaccumulation ($I_{geo}$) were researched. The majority of metallic elements sustain their values in natural state. The elements such as K, Ba, Zr, etc. appear to be rich in some places. The EF and $I_{geo}$ of P, Cu, Zn, and Pb, which belong to toxic heavy metals, are partly related with man-made pollution. P and Cu have a high EF, Pb has a high $I_{geo}$ and Zn is high in both EF and $I_{geo}$. The low contents of P and Cu are not likely to be related with the pollution of water environment. However, given the development of relative pollution, the research and the management regarding the pollutants are needed. Because Pb, naturally enriched by geological characteristics, has a large influence on water environment along with Zn, the adequate measures against man-made pollution should be worked out.

Characterizing LipR from Pseudomonas sp. R0-14 and Applying in Enrichment of Polyunsaturated Fatty Acids from Algal Oil

  • In this study, Pseudomonas R0-14, which was isolated from Arctic soil samples, showed a clear halo when grown on M9 medium agarose plates containing olive oil-rhodamine B as substrate, suggesting that it expressed putative lipase(s). A putative lipase gene, lipR, was cloned from R0-14 by genome walking and Touchdown PCR. lipR encodes a 562-amino-acid polypeptide showing a typical α/β hydrolase structure with a catalytic triad consisting of Ser153-Asp202-His260 and one α-helical lid (residues 103-113). A phylogenetic analysis revealed that LipR belongs to the lipase subfamily I.3. LipR was successfully expressed in Escherichia coli, purified, and biochemically characterized. Recombinant LipR exhibited its maximum activity towards p-nitrophenyl butyrate at pH 8.5 and 60℃ with a Km of 0.37 mM and a kcat of 6.42 s-1. It retained over 90% of its original activity after incubation at 50℃ for 12 h. In addition, LipR was activated by Ca2+, Mg2+, Ba2+, and Sr2+, while strongly inhibited by Cu2+, Zn2+, Mn2+, and ethylenediaminetetraacetic acid. Moreover, it showed a certain tolerance to organic solvents, including acetonitrile, isopropanol, acetone, methanol, and tert-butanol. When algal oil was hydrolyzed by LipR for 24 h, there was an enrichment of n-3 long-chain polyunsaturated fatty acids, including eicosapentaenoic acid (1.22%, 1.65-fold), docosapentaenoic acid (21.24%, 2.04-fold), and docosahexaenoic acid (36.98%, 1.33-fold), and even a certain amount of diacylglycerols was also produced. As a result, LipR has great prospect in industrial applications, especially in food and/or cosmetics applications.

서울 북부지역에 거주하는 정상 아동의 모발 미네랄 함량 (Trace element concentrations profiles in the hair of normal children living in the northern area of Seoul)

  • 목 적 : 영양상태, 환경 오염원에 노출, 질병상태의 한 검진 방법으로서 모발 미네랄검사의 타당성에 대해 아직 많은 이견이 있으며 아직 국내 정상 소아의 참고치는 연구된 바 없다. 이에 저자들은 건강한 아동들의 모발 미네랄 함량의 정상 참고치를 산출하고 그 특성을 알아보고자 하였다. 방 법 : 서울시의 북부지역에 거주하는 3-12세 사이의 질병이 없는 건강한 정상 아동 223명(남아 110명, 여아 113명, 평균 연령 $8.8{\pm}2.2$세)을 대상으로 모발을 채취하여 유도결합플라즈마질량분석기(ICP-MS)를 통해 영양 미네랄 11가지(나트륨, 마그네슘, 인, 칼륨, 칼슘, 크롬, 망간, 철, 구리, 아연, 셀레늄)와 독성 미네랄 6가지(알루미늄, 비소, 카드뮴, 바륨, 수은, 납)의 함량을 측정하고 이를 분석하였다. 결 과 : 칼슘 및 마그네슘의 농도는 여아에서 남아보다 높았고 카드뮴, 납 및 크롬의 농도는 남아에서 여아보다 높았다. 아연, 칼슘, 마그네슘, 구리 및 수은은 연령이 증가할수록 모발 중 함량이 증가하는 상관관계를 보였다. 한국 아동의 모발 미네랄 함량의 참고치를 외국과 비교하였을 때 우리나라 아동은 아연치가 외국에 비해 낮은 경향을 보였다. 결 론 : 본 연구를 통해 국내 정상 아동의 모발 미네랄 함량의 기준치가 제시되었으며 본 연구에서 제시된 참고치가 한국 아동들의 여러 가지 질병과 모발 내 미네랄 함량과의 관계를 연구하는 데 유용한 기초 자료로 쓰일 것이라 사료된다.

Penicillium sp. CB-20이 생성하는 Polygalacturonase의 특성 및 작용양상 (Characteristics and Action Pattern of Polygalacturonase from Penicillium sp.CB-20)

  • Penicillium sp. CB-20 이 생성하는 polygalacturonase의 최대 효소활성을 위한 pH는 5.0, 최적온도는 4$0^{\circ}C$였으며, 이 효소는 약산성의 pH에서 안정성을 보였고, 온도에 의한 안정성은 3$0^{\circ}C$였으며 4$0^{\circ}C$이상에서는 급격한 효소단백질의 불활성화가 진행되었다. 금속이온 중 $Mg^{++}$, $Zn^{++}$, $Ba^{++}$ 등이 활성을 촉진시켰고, Ag$^{+}$, Cu$^{++}$, Pb$^{++}$, Fe$^{+++}$, $Ca^{++}$, $Na^{++}$, Mnc 등은 효소활성을 저해하였으며, 본 균주가 생성하는 호소의 $K_m$값과 V$_{max}$값, 활성화에너지는 2.13$\times$$10^{-2}$mol/$\ell$, 104.17 $\mu$mol/min, 2,499 Kcal/mol 이었으며, pectin보다 polygalacturonic acid를 특이 적으로 분해하였다. 효소저해제 중 EDTA, DNP, $H_2O$$_2$ 등에 의해 효소활성이 제해되어 효소분자 중의 금속이 활성에 관여하며, 말단 아미노기와 histidine의 imidazole기가 효소활성에 관여함이 입증되었던 반면에 효소분자 중 SH기는 활성에 관여하지 않음을 알 수 있었다. 이 효소의 기질분해 양상을 종이 크로마토그라피로 확인한 결과 반응초기에는 monomer, dimer oligomer 등이 생성되었고 시간이 경과할수록 oligomer는 사라지고 monomer dimer 만이 생성되는 것으로 보아 endo 형인 것으로 판단되었으며, 효소의 과즙청징도 측정에서는 약 8 unit 에서 거의 청징화되었다.

  • 콩나물의 형태는 상품성에 커다란 영향을 미친다. 본 연구는 상면살수 방식과 하면담수 방식에서 압착이 콩나물의 생장과 형태에 미치는 영향을 조사하고자 상면살수 방식에서는 상하로 재배통을 포개어 이를 매일 교환하거나, 계속 압착하는 방법과 하면담수 방식에서는 4일부터 압착을 가하거나 계속 압착하는 방법으로 처리를 가하여 시험을 수행하였던 바 그 결과를 요약하면 다음과 같다. 1. 개체당 형성된 세근수는 상면살수 및 하면담수 방식 모두 압착 강도가 강할수록 적었다. 2. 상면살수 방식에서 하배축은 계속 압착시 가장 짧았던 반면, 뿌리는 압착하지 않거나 계속 압착시 가장 길었다. 하면담수 방식에서는 하배축 길이에서만 무압착에서 가장 길고 계속 압착시 가장 짧았다. 3. 관수방식에 관계없이 자엽 바로 아래의 hook 직경은 압착 처리간 차이가 없었던 반면, 하배축 중간부분의 직경은 압착강도가 강할수록 굵어지는 경향을 보였다. 4. 생산수율과 관련된 개체당 전체생체중은 상면살수 방식에서는 무압착시 가장 적었으며, 압착 강도가 강할수록 증가하여 계속 압착시 가장 많았다. 그러나 하면담수 방식에서 전체생체 중은 압착처리간 차이가 없었다.

LPS로 활성화된 복강 대식세포에서 세신 추출물의 항염증 효과 (Anti-inflammatory effect of extract of Asarum sieboldii in LPS-stimulated Murine peritoneal macrophage)

  • Objectives : The purpose of this study was to investigate the anti-inflammatory effects of extract Asarum sieboldii(AS) on the peritoneal macrophage. Methods : To evaluate of anti-inflammatory of AS, we examined cytokines production in lipopolysacchride (LPS)-induced macrophages. Furthermore, we checked molecular mechanism using western blot. Results : 1. Extract from AS reduced LPS-induced Nitric oxide (NO), tumor necrosis factor-a (TNF-a), interleukin (IL)-6 and IL-12 production in peritoneal macrophages 2. Extract from AS itself does not have any cytotoxic effect. AS inhibited the activation of mitogen-activated protein kinases (MAPKs) such as p38, extracelluar signal-regulated kinase B a (IkBa) in the LPS-stimulated peritoneal macrophages Conclusion : AS down-regulated LPS-induced NO and cytokines production, which could provide a clinical basis for anti-inflammatory properties of AS

남성 입원환자들의 맥파속도에 따른 요통 호전도의 비교 연구 (Comparison of improvement on Low back pain depending on male inpatient's Pulse wave velocity)

  • Objectives : The aim of this study is to compare the improvement of Low back pain (LBP) depending on male Inpatient's Brachlalankle Pulse Wave Velocity (baPWV), Method : We evaluated 35 LBP inpatients who took pulse wave velocity test during admission at Jaseng hospital from November 2008 to september 2009. We used applanation tonometry method to measure pulse wave velocity and numerical rating scale to measure patient's improvement. Result : At admission, standard deviation of normal group's NRS was $7.44{\pm}1.67$ and high risk group's was $7.57{\pm}2.09$(P=0.678). After 5 days of admission, standard deviation of normal group's NRS was $5.67{\pm}1.94$ and high risk group's was $6.00{\pm}2.17$(P=0.680). After 10 days of admission, standard deviation of normal group's NRS was $4.00{\pm}1.80$ and high risk group's was $4.95{\pm}1.96$(P=0.281). After 15 days of admission, standard deviation of normal group's NRS was $2.89{\pm}1.62$ and high risk group's was $4.10{\pm}1.92$(P=0.124). At discharge, standard deviation of normal group's NRS was $5.11{\pm}1.69$ and high risk group's was $4.86{\pm}2.08$(P=0.504). Comparison between admission and discharge, standard deviation of normal group's NRS was $5.11{\pm}1.69$ and high risk group's was $4.86{\pm}2.08$(P=0.504) Conclusion : Low back patients with high Brachialankle Pulse Wave Velocity, showed slower improvement rate compare to patients within normal rate. But statically, had no significance.

유전자총과 아그로박테리움을 이용한 여러 가지 한국 잔디류의 형질전환체계 확립 (Establishment of a transformation protocol combination particle bombardment with Agrobacterium tumefaciens in different zoysiagrass cultivars)

  • 다양한 Zoysiagrass 4가지 품종들을 식물재료로 사용하여 Agrobacterium만 이용한 방법 그리고 particle bombardment로 배발생캘러스에 상처를 낸 후, Agrobacterium으로 공동배양 시키는 2가지 다른 형질전환 방법을 비교하였다. 예비실험에서 일반적으로 형질전환에 널리 사용되는 kanamycin과 PPT(phospinitricin)의 적적선발농도에 대해서 실험하였는데, kanamycin의 경우 300mg/l 그리고 PPT의 경위 50mg/l의 농도에서 가장 효과적인 선발 효율을 나타내었다. Agrobacterium을 이용한 형질전환은 Agrobacterium을 2일간 배양시킨 다음, 박테리아 농도를 O.D 600nm=1.0-1.2로 맞추고, 배발생캘러스를 30분간 간염 시키는 방법이 효과적이었는데, particle bombardment를 이용하여 캘러스에 상처를 유발시킨 후, Agrobacterium으로 감염시키면 3배 이상 높은 형질전환 수율을 획득할 수 있었다. 이상의 결과는 한국잔디 형질전환에 있어서 particle bombardment과 Agrobacterium을 병행하여 실시한 최초의 보고이고, 이러한 시스템을 기반으로 하여 향후 한국잔디를 포함하여 다른 난지형 및 한지형 잔디의 품종개량에 널리 이용되리라 생각된다.

자도르노프 작품 속에 나라난 러시아 유머의 의미군조 (The semantic structure of the Russian humor in the works of Michael Zadornov)

  • In this article the structure of modern Russian humor is analyzed on the basis of some theories: bi-sociation theory (Koestler 1964), semantic script theory of verbal humor, using the concept of semantic presupposition, pragmatic felicity condition (Searle 1969; Levinson 1983) and grammatical rules (Chomsky 1965). Up to now the listed former theories were not examined and less analyzed by the semantic structure in the study of the structure of Russian humor(HcaeBa 1969; 3 $a_{OPHOB}$ 1991; 1992). Kreps (1981), who analyzed the works of Zoschenko, presented 21 types of humor, using the term 'humoreme'(Kpenc 1981, 36-37). These types are the list of the available means of humor that work not in the base of semantic criteria, but in the base of means of literary rhetoric. Kreps presented types of humor means, such as contradiction, antonymic substitution, macaronic speech and correlation of humoremes in the various types of humor. Apart from Kreps, Manakov (MaHaKOB 1986, 61-79) also studied these problems. He also set the system of the basic types of humor. Manakov introduced the linguistic means of humor of some Russian writers: Gogol, Tchechov. The means that Manakov showed with detailed examples, are trope, epithet, comic comparison, comic metaphor, comic periphrasis, euphemism, pun, zeugma, comic toponym, comic onomatopoeia, mania of foreign vocabulary, folk etymology, dialect etc. But these studies don't explain why these means make the works humorous. An, B.p tried to answer this question (안병팔 1997 a; b). An B.p. explains contexts of humor through the Release theory, the Superiority theory and the Incongruity theory. An, B.p. explained the process of deviation from the grammatical norms through morpho-syntactic and lexical means. But in these studies the humor was not analyzed by the semantic criteria. In order to linguistically evaluate various means of humor formation, it is necessary to elicit its deep structure, which makes it possible to research the formation and interpretation of humor. For this purpose this article, being based on the Incongruity theory, defined the structure of humor as negation of presupposition. Of course the former traditional studies also well shared the concept of 'contradiction' and 'contrast' of humor structure, but they didn't explain the structure by semantic differential features. This study, analyzing the works of' Zadornov, M., tried to note that through the negation of semantic presupposition the structure of contradiction is formed with semantic differential features on the semantic, syntactic or lexical dimensions.
