Electrochemical Determination of 6-Benzylaminopurine (6-BAP) Using a Single-wall Carbon Nanotube-dicetyl Phosphate Film Coated Glassy Carbon Electrode

  • Herein, insoluble single-walled carbon nanotube (SWNT) was successfully dispersed into water in the presence of a special kind of surfactant-dicetyl phosphate (DCP), subsequently, a SWNT-DCP composite film coated glassy carbon electrode (GCE) was fabricated. The electrochemical behaviors of 6-benzylaminopurine (6-BAP) at the unmodified GCE and SWNT-DCP modified GCE were examined. It is found that the SWNT-DCP modified GCE remarkably enhances the oxidation peak current of 6-BAP, indicating great potential in the determination of trace level of 6-BAP. Finally, a sensitive and simple voltammetric method with a good linear relationship in the range of ${\times}5.0\;\;10^{-8}\sim 2.5\;{\times}\;10^{-6}$ mol/L, was developed for the determination of 6-BAP. The detection limit is as low as $2.0\;{\times}\;10^{-8}$ mol/L for 3-min accumulation. This newly-proposed method was successfully demonstrated with practical samples.

  • An efficient method of shoot regeneration of peanut is described. In vitro shoot organogenesis from the callus of cotyledon explants of Arachis hypogaea L. was stimulated by addition of Adenine sulphate (Ads) along with 6 - benzylaminopurine (BAP) and - napthalene acetic acid (NAA). Ads (13 ${\mu}{\textrm}{m}$) had a stimulatory effect on shoot bud differentiation when combined with BAP (13 ${\mu}{\textrm}{m}$) and NAA (2 ${\mu}{\textrm}{m}$). Shoot organogenesis was markedly higher (92%) from callus induced on Ads, BAP and NAA combined media than from those formed by the individual supplementation of Ads or BAP with NAA. The shoots elongated on the media with GA$_3$ (1 ${\mu}{\textrm}{m}$). Elongated plantlets rooted with MS media containing IBA (9 ${\mu}{\textrm}{m}$).

  • Kinetin(4.7$\times$10\ulcornerM) and 6-benzylaminopurine (2.2$\times$10\ulcornerM) were found to increase ca. 1.5-fold putrescice content in corn grown in medium containing kinetin and 6-benzylaminopurine(6-BAP) for 3days whereas kinetin was found to decrase ca. 30% spermidine and spermine, respectively. KCI (3$\times$10\ulcornerM) was found to decrease more than 50% putrescine content. After germination, ornithine decarboxylase activity was observed to increase constantly whereas arginine decarboxylase activity remained constant, suggesting involvement in putrescine biosynthesis. 6-benzylaminopurine was shown to increase more activities of arginine ornithine decarboxylase than kinetin when they were added to medium.

Effects of Mineral Media, Carbon Sources and Phytohormones on Micropropagation of Alnus hirsuta (물오리나무(Alnus hirsuta)의 기내증식에 미치는 기본배지, 탄소원 및 식물호르몬의 영향)

  • Shoot tip explants from germinated seeds of Alnus hirsuta were cultured on NT (Nagata and Takebe, 1971) mineral salts medium supplemented with 6% glucose, MS (Murashige and Skoog, 1962) vitamin mixture, polyvinylpyrrolidone (PVP) and $0-50\;\mu\textrm{M}$ 6-benzylaminopurine (BAP). Five $\mu\textrm{M}$ BAP was found to give the highest shoot multiplication rate. Accordingly about 200 shoots were obtained for further experiments by multiplying shoots on this medium for 4-5 months. Regardless of carbon sources, NT mineral medium produced 3-12 times of shoots than MS mineral medium did. On NT mineral medium, 3% sucrose, 3% glucose and 6% glucose yielded no significant differences. It was observed that media consisting of 1/4-1/2 strength NT mineral salts, 3% sucrose and $1-8\;\mu\textrm{M}$ IBA produced about 100% rooting rate. Almost 100% of the resulting plantlets survived after transfer to the soil by decreasing humidity stepwise.epwise.

Direct Organogenesis in Geophila reniformis D. Don., an Important Medicinal Herb

  • Adventitious multiple shoots were developed from leaf, petiole and internode explants of Geophila reniformis D. Don. on MS medium supplemented with various concentrations of $N^6$-benzylaminopurine (BAP) or Kinetin (KIN) alone or in combination with indole-3-acetic acid (IAA). Leaf showed maximum organogenetic potential, followed by petiole and internode. Murashige and Skoog (MS) medium supplemented with 22.22 $\mu{M}$ BAP and 4.57 $\mu{M}$ IAA induced maximum shoot buds from leaf explants. Internodal segments showed low potential of direct organogenesis. The regenerated shoots rooted the best in presence of 10.75 - 13.44 $\mu{M}$ $\alpha$-naphthalene acetic acid (NAA) along with 2.22 $\mu{M}$ BAP, and were successfully established in the field with a survival rate of 89.11%.

Adventitious Shoot Formation and Plant Regeneration from Explants of Solanum nigrum L. (까마중(Solanum nigrum L.)의 유식물 절편체에서 부정아 형성 및 식물체 재분화)

  • In the present study, the effects of plant growth regulators on adventitious shoot and root formation of various explants of $in$ $vitro$ seedlings of $Solanum$ $nigrum$ L. were investigated to determine the optimum conditions for the high-efficiency plant regeneration of this species. The formation of adventitious shoots was higher in leaf explants than in cotyledon, hypocotyl, or epicotyl explants at low concentrations (0.5~2.0 mg $L^{-1}$ ) of 6-benzylaminopurine (BAP). The number of adventitious shoots and the shoot length were also higher in both leaf and cotyledon explants. In particular, 2.0 mg $L^{-1}$ BAP was most effective for stimulating the induction and multiplication of adventitious shoots. In terms of root formation and root development from shoots that were separated from multiple shoots, indole butyric acid (IBA) and indole acetic acid (IAA) were more effective than ${\alpha}$-naphthalene acetic acid (NAA). The percentage of rooting as well as the number of roots per shoot (4.0), root length (7.82 cm), and shoot length (8.76 cm) was highest on MS media supplemented with 0.05 mg $L^{-1}$ IAA. Furthermore, 100% of the regenerated plantlets survived when transplanted to compost soil. These results suggest that leaf explants are the best source for the high-efficiency regeneration of $S.$ $nigrum$ L., and that 2.0 mg $L^{-1}$ BAP and 0.05 mg $L^{-1}$ IAA are the best conditions for shoot and root induction, respectively.

Influence of Plant Growth Regulators on the Formation of Lateral Roots and Hypocotyl Enlargement in Soybean Sprouts (콩나물의 세근발생과 하배축 비대에 미치는 식물생장조절제의 처리 효과)

  • This study was carried out to identify the treatment methods for the inhibition of lateral roots and promotion of hypocotyl enlargement using plant growth regulators for the production of high quality soybean sprouts. Inhibition of lateral root formation was not significant from the $CA_3$ and NAA treatments. From the treatment of BAP, lateral roots were not occurred at all, and hypocotyl enlargement was promoted at the low concentration of $10\;mg{\cdot}L^{-1}\;and\;15\;mg{\cdot}L^{-1}$. Promotion of hypocotyl enlargement was higher as the BAP treatment time was shorter. The optimum time of BAP treatment was 6 hours. Occurrence ratio of lateral roots were low and hypocotyl enlargement was promoted from water supply every 24 hours after BAP treatment. In the indole B treatment, lateral roots occurrence was very low with the concentration of 1.1%, which is much lower than the optimum concentration of 4.2%.

Optimal conditions for adventitious root organogenesis from peony root explant callus cultures (작약(Paeonia lactiflora Pall.) 뿌리절편 유래 캘러스 배양으로부터 부정근발생을 위한 최적 배양조건)

  • The optimal culture conditions for root organogenesis from the callus of peonies (Paeonia lactiflora Pall.) were investigated. Root explants with vascular bundles were cultured in Murashige and Skoog (MS) medium combined with 0.5-4.0 mg/L auxins (indole acetic acid [IAA], naphthalene acetic acid [NAA], indolebutyric acid [IBA], and 2,4-dichlorophenoxyacetic acid [2,4-D]) and 0.0-2.0 mg/L cytokinins (kinetin, zeatin, and benzylaminopurine [BAP]) to induce callus formation. The callus was then cultured in MS medium combined with three concentrations (0.1, 0.5, and 1.0 mg/L) of IAA, NAA, IBA, kinetin, zeatin, and BAP in the dark for 6 weeks. Based on the results, the effects of dark and light conditions on the callus cultured in MS medium with combinations of 0.1-1.0 mg/L IBA and zeatin for 6 weeks were studied. Callus formation was most effective (>+++) in the medium with a combination of 1.0 mg/L NAA and 1.0 mg/L zeatin. A high number of long adventitious roots were observed in the mediums with 0.1 mg/L IBA (6.66 and 4.82 cm) and 0.5 mg/L zeatin (2.32 and 0.72 cm) among auxins and cytokinins, respectively. The highest number (14.06) of adventitious roots were formed from the callus cultured in light in the MS medium combined with 0.1 mg/L IBA and 0.5 mg/L zeatin. This same medium induced the formation of the longest adventitious root (5.45 cm) in the dark. Thus, optimization of in vitro culture conditions may be possible for the mass propagation of adventitious roots in peonies.

High Frequency Shoot Regeneration from leaf Explants of Cucumber

  • Leaf explants of the cucumber (Cucumis sativus L.) were cultured on Murashige and Skoog's (MS) medium supplemented with various concentrations of $\alpha$-naphthalene acetic acid (NAA) and 6-benzylaminopurine (BAP). Direct shoot orgnogenesis as well as callus formation with somatic embryos and multiple shoots was observed from leaf explants of cvs. Shinhukjinju and Chungjang. The highest frequency of shoot formation 80% was observed on MS medium supplemented with NAA/BAP (5.0 ${\mu}{\textrm}{m}$/2.5 ${\mu}{\textrm}{m}$), with explants forming 3-7 shoots. Shoots formation occured within 3 to 4 weeks. Only one subculture of calli was required for plant regeneration on normal growth regulator-free medium. Plantlets transferred to soil developed into plants of normal appearance, which flowered and set fruits.

High-frequency regeneration of plants in vitro from seedling-derived apical bud explants of Tilia mandshurica Rupr. & Maxim

  • This work describe an efficient method for the shoot induction and plant regeneration of seedling-derived apical bud explants of Tilia mandshurica Rupr. & Maxim. The highest rate of shoot induction (82.2%) was obtained when apical bud explants from juvenile seedlings (5 months old) were cultured on Murashige and Skoog (MS) medium containing 1.0 mg/L 6-benzylaminopurine (BAP). However, apical bud explants obtained from mature trees (12 years old) did not produce any shoots, even with BAP supplementation. Among the three cytokinins tested for shoot multiplication (BAP, zeatin, and kinetin), BAP was the most effective; the highest number of shoots per explant (2.1) was observed on MS medium supplemented with 1.0 mg/L BAP. In contrast, the longest average shoot length (3.0 cm) was observed after growth on MS medium with 2.0 mg/L zeatin. No multiplication occurred when apical bud explants were cultured with kinetin-supplemented media. During rooting of in vitro-elongated shoots, the highest rooting rate (100%) was observed in half-strength MS medium supplemented with 0.5 ~ 1.0 mg/L indole-3-butyric acid (IBA) or 3.0 mg/L 1-naphthaleneacetic acid (NAA). During the acclimatization process, plantlets that were rooted on the IBA (0.5 mg/L)-supplemented medium had the highest survival rate (100%) and maximum root length (18.5 cm). These findings suggest that a low concentration (0.5 mg/L) of IBA is appropriate for the rooting and acclimatization of T. mandshurica. Plants were successfully transferred to the greenhouse with a 100% survival rate. This protocol will be useful for the large-scale propagation of Tilia species.
