Microbiological Assay of Vitamin B group in Panax Ginseng Roots I.Assay of Nicotinic acid and Folic acid roots (인삼중 Vitamin B 군의 미생물학적검정 I Vitamin $B_{12}$, Nicotinic acid 및 Folic acid 의 검정)

  • The purpose of this investigation is to observe tha variation of vitamin $B_{12}$, nicotinic acid and folic acid contents in relation to the growing years of Panax Ginseng roots. The contents of the vitamins were estiamted microbiologically with Lactobacillus leichmannii, Lactobacillus arabinosus and Streptococcus faecalis, respectively. It is found that the content of vitamin $B_{12}$ in Panax Ginseng roots somewhat increases according to their growing years except 3-year-old roots. It is probable that the lower content in these roots should be due to the cultivating soil. The content of nicotinic acid in all the roots is not significantly different. The result of paper chromatography using the concentrated extract of the roots suggests that there exists some other substance besides nicotinic acid and nicotinamide, of which Rf value is 0.5 in comparison with the Rf 0.75 of nicotinic acid and nicotinamide. It is thought that this substance stimulate the growth of L. arabinosus. The content of folic acid is significantly different. The content level is the highest in the 4-year-old roots and the lowest in the 6-year-old roots.

Surveys on Ginseng Damage by Insect and Other Animal Pests (인삼 포장에서 발생하는 해충의 종류와 피해 양상)

  • Surveys were conducted in 66 ginseng fields damaged by insect and other an~mal pests from 1984 to 1993. Holohichio rnoroso, Holotrichio dromphalio, Holotrichia titonis Malodera orientaliq Ectinus sericeus, Gyllotalpa ofricana, Teleog~iluse mmo, Ostrinio furnacoii* Agrotis tokionis, Marnestro brassicae, Hydrellia griseolo, unidentified stem maggo$, Pseudococcus comstocki (13 species of insects). Deroceras uarions (slug), Acusta despecta sieboldiona (snail), probably two species of rats. and pheasant species were ascertained to damage ginseng plants M them, Holotrich~a morosa, Holotrichia diomphalia, Gryliotaipo africanq Deroceras uarions, Acusto despech siebaldiano showed higher frequencies. Underground (root) damage occul~ed mainly in spring penod (MayJune) and fall period (September-Odober) in 2-year-old glnseng fields at slopes, and aboveground (leaf and stem) damage occurred mainly in spring period in 3 or more-year-old ginseng iields mulched with rice straws at plains. Three ginseng fields were abolished due to heavy underground damage.

Induction of Ginseng Hairy Roots And Their Possible Application To Large Scale Culture

  • Ginseng(Panax ginseng C. A. Meyer) is important medicinal plant but requires 4-year cultivation for root harvest because of slow growth. In contrast, ginseng hairy roots induced by introducing Ri-plasmid of Agrobacterium rhizogenes into genomic DNA of plant cells show vigorous growth, and the hairy roots produce the same or more saponins than natural ginseng roots. Therefore, hairy roots can be used for commercial purposes. The present study was carried out to induce hairy roots with both active growth and high saponin contents. Numerous hairy roots of Panax ginseng were obtained after root disks of three-year old roots were infected with Agrobacterium rhizogenes R1000 A4T in dark condition after one month of culture. About 3 hundred lines of hairy roots were selected according as morphological characters on medium with carbenicillin. After pre-selection of fifteen lines of hairy roots with active growth, KGHR-l and KGHR-8 lines were finally selected which had characters of high content of ginsenoside-Rd and ginsenoside-Re, respectively. The optimum growth of hairy roots was achieved in the culture of 1/2 MS liquid medium in dark (22 $^{\circ}C$) under 60 rpm gyratory shaking. Hairy roots grew well in 5L Erlenmeyer flasks, lL roller drums, 10L jar-fermenters, and especially in 20L air-lift culture vessels.

  • Lime-bordeaux mixture has been used to prevent diseases in the field of ginseng. The purpose of this study is to investigate the efficacy of lime-bordeaux depending on the concentration and treatment time on major diseases of ginseng such as Alternaria blight and anthracnose, and to evaluate the root growth of ginseng. Lime-bordeaux caused damage on leaf when it was sprayed on ginseng between April and early May. No difference was found in root growth by spraying lime-bordeaux mixture between ratio 4-4 and 8-8 ratio in concentration. Plot of 6-6 raito and 8-8 ratio appeared to be similar efficacy compared to that of practical chemical control. However, the plot of 4-4 ratio showed lower than that of chemical control.

Growth and Ginsenoside Content of One Year Old Ginseng Seedlings in Hydroponic Culture over a Range of Days after Transplanting (수경재배 시 1년생 묘삼 이식 후 경과일수에 따른 인삼의 생육 및 Ginsenoside 함량)

  • Background: Ginseng produced by hydroponics can be cultivated without using agricultural chemicals; thus, it can be used as a raw materials for functional foods, medicines, and cosmetics. This study aimed to determine the optimal harvesting time to obtain the highest levels of ginsenoside and ginseng, as this was not previously unknown. Methods and Results: One-year-old organic ginseng seedlings were transplanted and cultivated using hydroponics for 150 days in a venlo-type greenhouse, using ginseng nursery bed soil and a nutrient solution ($NO_3{^-}-N$; 6.165, P; 3.525, K; 5.625, Ca; 4.365, Mg; 5.085, S; $5.31mEq/{\ell}$). Ginsenoside content and fresh and dry weights were higher at 120 days after transplanting than at 30, 60, 90, and 150 days. Total ginsenoside content was 11.86 times higher in the leaf and stem than in the root at 120 days after transplanting. Ginsenosides F1, F2, F3, and F5 were detected in ginseng leaves and stems. These chemical compounds are known to be effective in altering skin properties, including whitening, anti-inflammation, and anti-aging. Conclusions: Optimal harvesting time for ginseng cultivated using hydroponics was 120 days after transplanting when the biomass and ginsenoside content were highest.

Effect of Green Manure Incorporation and Solarization on Root Rot Disease of 3-year-old Ginseng in Soil of Continuous Cropping Ginseng (녹비작물 토양환원과 태양열 소독에 의한 3년생 인삼의 뿌리썩음병 억제효과)

  • Background: Ginseng root rot disease, caused by Cylindrocarpon destructans and Fusarium solani is a major cause of replant failure in continuous cropping ginseng. Methods and Results: To control replant injury in soil infected with C. destructans and F. solani, biosolarization was performed by covering the plot with transparent polyethylene film after adding green manure of maize and sunflower for the summer season. Per 10 a, fresh and dry weight of maize was 10.1 and 2.5 tons, respectively, and that of sunflower was 8.1 tons and 1.2 tons, respectively. Mean maximum temperature at 20 cm depth was $33.2^{\circ}C$, $41.5^{\circ}C$ and $41.8^{\circ}C$ in the control, maize-incorporated and sunflower-incorporated plots, respectively. The elapsed time over $40^{\circ}C$ was 36.4 h in the maize-incorporated plot and 77.3 h in the sunflower-incorporated plot. Biosolarization increased $NO_3$ content in soil, while content of organic matter, Ca, and Mg was decreased. Electrical conductivity, $NO_3$ and $P_2O_5$ in soil significantly increased after two years of biosolarization. The number of spores of C. destructans in soil was significantly decreased by biosolarization, and sunflower treatment was more effective than maize treatment in decreasing the number of spores. Root yield of 3-year-old ginseng was significantly increased by biosolarization, however, there was no significant difference between maize and sunflower treatments. Rate of root rot in 3-year-old ginseng decreased to 16.5% with the incorporation maize and 5.0% with the incorporation of sunflower, while that in control 25.6%. Conclusions: Biosolarization was effective in inhibiting ginseng root rot by decreasing the density of root rot disease and improving soil chemical properties.

Ginsenosides analysis of New Zealand-grown forest Panax ginseng by LC-QTOF-MS/MS

  • Background: Ginsenosides are the unique and bioactive components in ginseng. Ginsenosides are affected by the growing environment and conditions. In New Zealand (NZ), Panax ginseng Meyer (P. ginseng) is grown as a secondary crop under a pine tree canopy with an open-field forest environment. There is no thorough analysis reported about NZ-grown ginseng. Methods: Ginsenosides from NZ-grown P. ginseng in different parts (main root, fine root, rhizome, stem, and leaf) with different ages (6, 12, 13, and 14 years) were extracted by ultrasonic extraction and characterized by Liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry. Twenty-one ginsenosides in these samples were accurately quantified and relatively quantified with 13 ginsenoside standards. Results: All compounds were separated in 40 min, and a total of 102 ginsenosides were identified by matching MS spectra data with 23 standard references or published known ginsenosides from P. ginseng. The quantitative results showed that the total content of ginsenosides in various parts of P. ginseng varied, which was not obviously dependent on age. In the underground parts, the 13-year-old ginseng root contained more abundant ginsenosides among tested ginseng samples, whereas in the aboveground parts, the greatest amount of ginsenosides was from the 14-year-old sample. In addition, the amount of ginsenosides is higher in the leaf and fine root and much lower in the stem than in the other parts of P. ginseng. Conclusion: This study provides the first-ever comprehensive report on NZ-grown wild simulated P. ginseng.

Effects of 2,4-D and Kinetin on the Production of Saponin in Ginseng Tissue Culture (인삼근 유래 칼루스조직의 사포닌 함량에 미치는 2,4-D와 키네틴의 영향)

  • In the present study effects of 2,4-D and kinetin on the callus tissue growth of Korean ginseng(Panax ginseng C. A. Meyer), in relation to the synthesis of saponin were investigated. The saponin synthesis in the callus culture of ginseng root was enhanced by 2,4-D and kinetin. The total saponin content of callus grown on the optima growth conditions, that is, 5mg/l of 2,4-D and 2mg/l of kinetin, was about three times as high as that of the 6 year-old ginseng roots commercially used as herbs. The kinetin specifically increased the synthesis of protopanaxadiol group ginsenoside and decreased the syntehsis of protopanaxatriol gropu in callus cultures, while 2,4-D caused to an increase in the synthesis of protopanaxatriol group ginsenoside and decrease the synthesis of protopanaxadiol group.

Phenolic Compounds Contents of Rusty-Root Tolerance Ginseng Lines in 6-Year Old Root (6년생 적변내성 인삼계통의 Phenolic Compounds 함량 특성)

  • Experiments were carried out to select marker for rusty tolerance ginseng root using high rusty degree lines(HRL) and low rusty degree lines (LRL) in ginseng plant. A strong positive correlation was detected between degree of rusty-root in 4-year-root and that in 6-year-root. The contents of phenolic compounds among samples were not different in stele and branch & fine roots. The contents of phenolic compounds of rusty-roots was higher than that of healthy-roots in cortex, but those of high 겨sty degree lines (HRL) were not different compared with low rusty degree lines (LRL) in cortex using same rusty-degree samples. These suggest that phenolic compounds in cortex tissue were not adequate as a marker to select rusty tolerance ginseng roots. The contents of phenolic compounds of rusty-roots were higher than that of healthy-roots in epidermis, and those of HRL were higher than LRL in epidermis using same rusty-degree samples. These suggested that the contents of phenolic compounds in epidermis tissue might be a potent marker to select rusty tolerance ginseng roots.

Effect of Fludioxonil, Flutolanil, and Thifluzamide on Suppression of Damping-off Caused by Rhizoctonia solani on Panax ginseng (인삼 잘록병(Rhizoctonia solani) 발생억제에 미치는 Fludioxonil, Flutolanil 및 Thifluzamide의 효과)

  • In vitro and in vivo effectiveness of fungicides were evaluated for the control of damping off caused by Rhizoctonia solani on Panax ginseng. Fludioxonil(67 mg a. i./L), flutolanil(75 mg a. i./L), thifluzamide(35 mg a. i./L), and mepronil (750 mg a. i./L) were selected from 9 fungicides, which were based on inhibition of mycelial growth of R. solani (isolate Rh 9801) and duration of fungicidal effectiveness against the pathogen in vitro. Field trials were made twice in the year of 2003 and 2004. Experimental plots $(54m{\times}0.9m)$ of 4-year-old ginseng fields were artificially infested with 5kg and 14 kg in fresh weight of inoculum in 2003 and 2004, respectively. The fungicides were drenched at a volume of 8l in $3.6m{\times}0.9m$ with 3 replications. Fludioxonil, flutolanil, thifluzamide and mepronil reduced the incidence of damping off by $73\%,\;69\%,\;69\%\;and\;43\%$, respectively. In the 2004 trial, fludioxonil, flutolanil, and thifluzamide showed similar result as reducing the incidence by $85\%,\;84\%,\;and\;82\%$, respectively, in the plot where the inoculum was applied 2.8 times more than the 2003. The disease incidences in untreated control were $12\%$ in 2003 and $47\%$ in 2004.
