사람의 Serine palmitoryl transferase II 및 ceramidase의 promoter에 대한 연구

  • Serine palmitoyl transferase(SPT) and ceramidase are the key enzymes in sphingolipid biosynthesis. To study sphingolipid metabolism, we have got the 5'-upstream regions of human serine palmitoyl transferase subunit II and acid ceramidase gene by using GenomeWalker kits(Clontech Co.). Human genomic DNA was purified from HT29, human colon canser cell line by using DNAzol. We got several bands after secondary PCR and subcloned them to T7bule vector. Human SPTII promoter which we got was 2690bp but we cut it with Bgl II and vector with Bgl II and BamH I, and subcloned 1782bp to pGL2-enhancer vector and pGL2-basic vector with luciferase reporter gene. Human acid ceramidase promoter which we got were 2028bp and 1034bp and subcloned to pGL2-enhancer vector and pGL2-basic vector. We transfected these promoters to HT29 cell and assayed luciferase activity. For measuring transfection efficiency, pRL-TK vector with seapancy luciferase reproter gene was cotransfected with these promoters.

Effects of Loess on the Mycelial Pellet Formation of Phosphate Dissolving Fungus, Penicillium sp. GL-101 in the Submerged Culture (유리인산 생성균 Penicillium sp. GL-101의 액침배양중 Pellet 형성에 미치는 황토의 영향)

  • In order to investigate effects of loess on the mycelial pellet formation a phosphate dissolving fungus, Penicillium sp. GL-101, was cultured in potato dextrose broth containing loess. The strain formed an amorphous pellet or loose aggregates agitated at a low speed(50rpm) while spherical and regular pellets at a high speed(150rpm). The higher concentration of loess, the smaller size of a pellet in the medium formed by the strain. Cultured in the medium supplemented with 1.5% loess the pellet size was reduced to a seventh compared to the control. In the case of addition of several insoluble salts, which are main components of loess, to the culture medium the higher concentrations of salts, the smaller sizes of pellet formed by the strain and the smallest pellet was formed by the addition of calcium sulfate.

  • Recently, the utilization of geospatial open platforms has been constantly increased and the interest in 3D geospatial data such as terrain, building and shopping mall has been increased significantly. In particular, rather than simplified 3D geospatial data, interest in high-precision 3D geospatial data which similarly represents the real world objects has increased significantly. In order to satisfy the demand for such the high-precision 3D geospatial data, various kinds of 3D geospatial open platforms has been developed and has provided services on the web. However, most of the 3D geospatial open platforms have been used plug-in module in order to ensure a fast 3D rendering performance on the web, despite the many problems such as difficulty of the installation, no supporting of cross browser/operating system and security issues. In addition, recently, the existing 3D geospatial open platforms based on plug-in module are facing a serious problem, by declaring the NPAPI service interruption in Chrome and Firefox browsers. In this study, we presents the design and implementation of a new 3D geospatial open platform based on HTML5/WebGL technology without the use of plug-ins. Such the new 3D geospatial open platform based on HTML5/WebGL may support cross browsers such as IE, Chrome, Firefox, Safari and cross OS platforms such as Windows, Linux, Mac and mobile OS platforms.

Atypical Fruiting Structure Formation of White Fruitbody-Forming Isolates in Ganoderma lucidum (백색자실체를 형성하는 영지 균주의 비정형(非定型) 자실체 구조의 형성)

  • Five white fruitbodies of Ganoderma lucidum found from two different mushroom farms, and the characteristics of atypical fruiting structure formation of these strains were described. The white fruitbodies were spontaneously generated on Quercus-log during the cultivation. They did not differentiate to the normal fruitbodies with pileus, hymenium, stipe and coloration, and fruitbodies remained non-laccateed even after 3 months. Dikaryotic mycelia isolated from the five white fruitbodies differed from wild-type strains in the mycelial growth rate, colony color, and the capacity of atypical fruiting structure (AFS) formation on agar media. These white mutants readily induced brown colored AFSs on the colonies under ventilation and illumination conditions. Both isolates Gl-010 and Gl-011 that were obtained from a normal and white fruitbody, respectively, did not form AFSs in the dark and/or under black light blue (BLB) light illumination, but induced under the visible light. They required dim light for the AFS formation, and the AFS formation was inhibited up to $0.5{\mu}mol\;m^{-2}\;S^{-1}$ in light intensity. However, the other four isolates induced AFSs even in the dark and BLB illumination, although their parent strain, isolate Gl-030, did not form AFSs under any light conditions. The monokaryotic mycelia derived from basidiospores of the AFSs of the white mutants were compatible with the original culture (dikaryon) on a dual culture.

Storage Stability of Baikseolgi (백설기의 저장성 연구)

  • The effects of storage temperature, moisture content and the concentration of additives, such as sucrose fatty acid ester(SE), isomaltooligosaccharide(IO) and glycerin(GL), on texture properties, hardness(HA), cohesiveness(CO) and chewiness(CH) of Baikseolgi after 7 days storage were analyzed by response surface methodology(RSM). The contour values of HA of SE added Baikseolgi at 20, 50 and $80^{\circ}C$ of storage temperate were $1500{\sim}3200,\;500{\sim}1300$ and $100{\sim}400\;g_f$, respectively. The HA of IO or GL added Baikseolgi decreased with increased storage temperature, moisture content and additive concentration. The storage temperature was the most significant factor affecting the HA of Baikseolgi. However, the second and third significant factors were moisture content and additive concentration, respectively. These results imply that the control of storage temperature is the most effective method to increase the storage stability of Baikseolgi. The CO of IO or GL added Baikseolgi was increased by the change of strage temperature from $20{\circ}\;to\;50{\circ}$. While, there was no significant difference between $50{\circ}\;and\;80{\circ}$ of storage temperature. The CO of IO or GL added Baikseolgi was maximized around 40% of moisture content and that of GL added Baikseolgi was minimized around 0.5% of GL concentration. The storage temperature, additive concentration and moisture content were the first, second and third affacing factors on the CO of Baikseolgi, respectively. The CH of Baikeolgi was decreased by increasing storage temperature, moisture content and additive concentration.   • Glycyrrhizin (18$\beta$-glycyrrhetic acid $\beta$-D-glucuronyl a-D-glucuronic acid, GL), a main component of liquore extract (Glycyrrhiza glabra), is ingested orally as a component in the oriental medicine. By human intestinal bacteria, glycyrrhizin (18$\beta$-glycyrrhetinic acid $\beta$-D-glucuronyl a-D-glucuronic acid, GL) was metabolized to glycyrrhetinic acid (GA): main pathway metabolizing GL to GA by glucuronidases of Bacteroides J-37 (Kim et al., 1997) and Eubacterium sp strain GLH (Akao et al., 1987) and minor pathway metabolizing GL to GA via 18$\beta$-glycyrrhetic acid D-glucuronic acid (GAMG) by $\beta$-glucuronidase of Streptococcus LJ-22 and glucuronidases of Bacteroides J-37 / E. coli. $\beta$-Glucuronidase from Streptococcus LJ-22 hydrolyzed GL to GAMG, not GA. $\beta$-Glucuronidase of Streptococcus LJ-22 hydrolyzed $\beta$-glucuronic acid conjugates of polysaccharides rather than aglycone-$\beta$-glucuronides Optimal pH of Streptococcus LJ-22 $\beta$-glucuronidase was 5-6 and its molecular weight was 250 kDaltons. Km for GL was 0.37mM.

  • Microorganism producing glutaryl 7-aminodeacetoxycephalosporanic acid (GL-7-ADCA) acylase was screened from soil. The microorganism was identified as Alcaligenes sp. J-421 by its morphology and biochemical properties. Cultural conditions of Alcaligenes sp. J-421 were investigated for the production of GL-7-ADCA acylase. Optimum medium composition was 1% glucose, 1% beef extract, 0.5% yeast extract, 0.2% monosodium L-glutamate, 0.1% glutaric acid, 0.2% NaCl, 0.5% $K_2$ $HPO_4$, and 0.05% $CuSO _4{\cdot}5H_2O$. Optimum cultivation conditions for the production of the enzyme in 5 l jar fermentor were $37^{\circ}C$, tip speed 300 rpm, aeration 1 vvm. Optimum reaction pH of the enzyme was 8.0 and the enzyme was stable at pH7.0-11.0.

Effect of Dietary Supplementation of the Combination of Gallic and Linoleic Acid in Thigh Meat of Broilers

  • This study was performed to investigate the combined effect of dietary supplementation of gallic and linoleic acid (GL) on the antioxidative effect and quality of thigh meat from broilers. Broilers received 3 dietary treatments: i) commercial finisher diet (control), ii) 0.5% GL (gallic:linoleic acid = 1 M:1 M), and iii) 1.0% GL during the 22 to 36 d. The pH value of broiler thigh meat was increased by GL supplementation. Water holding capacity of the thigh meat was enhanced by the 1.0% dietary GL supplementation. Antioxidative effect (total phenolic content, DPPH radical scavenging activity, $ABTS^+$ reducing activity, reducing power, and TBARS value) in the thigh from the broilers improved significantly with 1.0% GL. Linoleic acid and docosahexaenoic acids were higher in the broilers fed both levels of dietary GL. However, volatile basic nitrogen content and microbiological quality was not shown to be different between control and treated group. Results indicate that 1.0% dietary supplementation of GL can improve the antioxidant activity of broiler thigh meat and may enhance the meat quality.

The Effects of gl, gh and wx Gene on the Grain Yield and Yield Components of Rice Plant (수도의 gl, gh 및 wx유전자가 수도수량 및 수량구성요소에 미치는 영향)

  • Isogenic lines for glabrous leaf (gl), hull color(gh) and glutinous endosperm (wx) were used to clarify the effects of the gl, gh and wx gene on the grain yield and yield components in rice plant. Eight genotypes: glgl ghgh wxwx, glgl ghgh WxWx, glgl GH-wxwx, glgl GH-WxWx, GL-ghgh wxwx, GL-ghgh WxWx, GL-GH-wxwx and GL-GH-WxWx were evalutated in the backcrossed $F_2$ generation. Significant effects were measured for gl genotypes on heading date, gh genotypes on grain fertility, and wxwx genotypes on number of panicles per hill and 1, 000 grain weight. The grain yield of straw hull and non glutinous lines were higher than gold hull and glutinous lines. The interaction of three genes, gl, gh and wx was significant for heading date, but not significant in other characters. The average value of isogenic lines which were combined with two recessive genes and one dominant gene were lower and the C.V. value was higher than the isogenic lines which were combined with two dominant genes and one recessive gene in all characters except plant height and number of grains per plant. The average value of isogenic line combined with three recessive genes was lower and the C.V. was higher than the isogenic line combined three dominant genes in all characters measured.

  • Lipids extracted and purified from Herbs Capsellae leaf ana root were studied, To investigate those, silicic acid column chromatography, TLC, GLC was used for analysis of lipids and Fatty acids composition. Following results were obtained. 1. From result of total lipid classes in Herba Capsellae leaf and root, NL were obtained 62.6%, 58.5% respectly. GL 20.8%, 17,8%. PL 16.6%, 23.7%, respectively. 2. Among the NL in Herba capsellae leaf and root, TG were predominant 77.5%, 80.3%, respectly. SE 9.2%, 8.4%. FFA 4.8%, 3.4%. 1.2-DG 4.1%, 3.6%. FS 2.2%, 2.2%. and 1, 3-DG 1.3%, 1.0%, in turn. 3. The major fatty acids in the lipid classes were linoic acid, palmitic acid, oleid acid, in all. And total unsaturated fatty acid was NL 78.0%, GL 66.6%, and PL 75.2% in Herbs capsellae leaf, and NL 66.1% GL 63.6% and PL 61.2% in Herbs capsellae root.
