Quality Characteristics of Commercial Fermented Skates (시판 발효 홍어의 품질특성)

  • This study was conducted to evaluate the quality characteristics of domestic as well as imported fermented skate. Three types of fermented skate products were analyzed for proximate composition, pH, VBN, ammonia-N, free amino acids, and fatty acids. The results indicated that the domestic fermented skate contained large amounts of TMAO. Also, the domestic and imported fermented skates each contained approximately 7.1 log CFU/g and $5.8{\sim}6.5$ log CFU/g of aerobic bacteria, respectively, and 585.9 mg and $384.1{\sim}398.5$ mg of total free amino acids, respectively; all samples contained high levels of taurine, anserine, lysine, alanine, glycine, proline, and ${\beta}-alanine$. For fatty acid composition, the domestic fermented skate contained 11 different types of saturated fatty acid and 16 types of unsaturated fatty acid, whereas the imported skate contained 8 types of saturated fatty acid and $10{\sim}15$ types of unsaturated fatty acid. Overall, the results suggest that domestic fermented skate is a better source of amino acids and essential fatty acids and contains more aerobic bacteria than imported fermented skate.

Application of the Microbial Process for Hydrogen Sulfide Removal and Bio-Sulfur Production from Landfill Gas (미생물 공법에 의한 매립가스 황화수소 제거 및 바이오황 생산)

  • Operational testing of the THIOPAQ® facility that removes H2S from landfill gas was performed for 746 days. The average H2S removal efficiency was 99.4%, and the input quantities of air, NaOH, and nutrients per sulfur load were 13.1 ㎥/ton, 1.5 ㎥/ton, and 28.7 L/ton, respectively. The purity of the bio-sulfur produced from the facility was 94.8%, with 3.3% impurities, except for moisture. X-ray photoelectron spectroscopy showed that the compositional contents of amino acids and free amino acids of the bio-sulfur surface were 5,308 and 728 mg/kg, respectively. The mean particle size was 3.41 ㎛, which was much smaller than that of chemical sulfur. Based on these results, a high H2S removal rate of more than 97% is feasible, and high value-added bio-sulfur, which is used as a fungicide because of its hydrophilic characteristics and small size, can be obtained at this facility.

  • Yeonsan Ogae has been known as supporting health and high efficacy of treatment. In recent days, as the efficacy of functional peptides has known, the optimization of oligo peptides production and its characteristics from Ogae legs has been performed. Response surface method was used to perform the optimizaion of enzyme hydrolysis. The range of processes was temperature ( 40, 50 and $60^{\circ}C$), pH( pH 6.0, 7.0 and 8.0 ), and enzyme( 1, 2 and 3% ). The degree of hydrolysis, amino acids, molecular weight of products were analyzed. The optimum process of enzyme hydrolysis were determined as temperature $58^{\circ}C$, pH 7.5, and enzyme concentration 3%. At optimum conditions, the degree of hydrolysis after 2 h reaction was 75-80%. The amino acid and were 168.131 mg/100 g, respectively. The molecular weight of products by using MALDI-TOF was ranged from 300 to 1,000 Da.

Studies on the Decomposition of Environmental Pollutants by Utilizing Microorganisms (미생물을 이용한 환경오염원의 분해에 관한 연구 II)

  • 1. When Chong Ju and Chung Ju soils possessing different physicochemical properties were treated with 500 ppm of TOK and incubated in flooded anaerobic condition for 2, 4, and 6 months, respectively, they produced 4-Chloro-4'-amino diphenyl ether, 2,4-Dichloro-4'-amino diphenyl ether(amin-TOK), N-[4'-(4-Chlorophenoxy)] phenyl acetamide, and N-[4'-(4-Chlorophenoxy)] phenyl formamide as the metabolities. This result indicates that TOK undergose the reduction of its $NO_2\;to\;NH_2$ group, dechlorination, acetylation, and formylation under this condition. The cleavage of ether linkage does not occur. In addition, TOK degrades more readily in Chung Ju soil which is characterized by pH 6.43 and higher contents of $Ca^{++}$ and C.E.C. than in Chong Ju soil which is lower in pH, $Ca^{++}$, and C.E.C. 2. In the aerobic incubation of TOK of 25ppm in Chung Ju soil suspension for 21 days, the ratio of the resulting metabolites, TOK : amino-TOK : 4-Chloro-4'-amino diphenyl ether was 100 : 130 : 76. Meanwhile, in the 42 day incubation, the ratio was 100 : 19 : 5, which indicates that TOK in aerobic condition dose not necessrily degrade as a function of the incubation period. 3. The citrate buffer extract of Chung Ju soil has the capability of degrading TOK, which was verified to be due to the action of the microorganisms involved. 4. Twelye strains of soil bacteria were isolated from the TOK-treated soils. In the incubation of TOK in pure cultures of the respective isolates, the strain T-1-1 isolated from Chong Ju soil had almost no degradability whereas the strain T-2-3 was the most potent. The degradation of TOK by the isolates constituted mostly the reduction of the nitro group to amino group. 5. In a test for the degradability of TOK by some selected microorganisms, Pseudomonas species were more potent than fungi. Yet, Isolate B which had been isolated from Chung Ju soil suspension was the most potent.

Effect of Amino Acids on Oxygen Consumption of the Mang-Kong Frog (Gallula tornieri VOGT) Liver (맹꽁이 간의 산소소비량에 미치는 수종 아미노산의 영향)

  • The present experiment was conducted to investigate the rate of oxygen consumption in relation to the several amino acids in the Mang-Kong frog ,Gallula tornieri VOGT. 1. When the liver tissue was suspended in the Ringer's solution (0.85% NaCl), the amount of oxygen consumption per hour was 0.043$\mu$l/mg , whereas when it was suspended in M/15, pH 5.8 phosphate buffer the oxygen consumption reached 0.93$\mu$l/mg. 2. When 0.5ml of 1% alanine and 0.5ml of 0.5% arginine were used as respiratory material the amount of oxygen consumption increased remarkably, while on the other hand glycine showed little influence on the amount of oxygen consumption. 3. When 0.5ml of 0.1% , 0.5% alanine-arginine mixture, 0.5ml of 0.1% , 0.5% alanine-glycine mixture and 0.5ml of 1 % arginine-glycine were added the oxygen consumption in the liver tissue increased somewhat in comparison to the case in which single amino acid was used. 4. Glycine appeared to have influence on the respiration of the liver tissue but the amount of oxygen consumption increased as the concentration of arginine-glycine mixture increased, whereas the rate of oxygen consumption increased remarkably as alaninie-glycine mixture decreased. It was evidenced that in the combination of alanine-arginine the rate of oxygen consumption increased as the concentration of the mixture decreased and vice versa.

Isolation and Characterization of D-$\alpha$-Amino-$\varepsilon$-Caprolactam Utilizing Bacteria (D-$\alpha$-Amino-$\varepsilon$-Caprolactam 자화균의 분리 및 특성)

  • A bacterium which grows on D-$\alpha$-Amino-$\varepsilon$-Caprolactam as sole carbon, energy and nitrogen source was isolated from the sludge of industrial areas in Taegu, and identified as Alcaligenes eutrophus. The optimum pH, temperature and concentration of D-$\alpha$-Amino-$\varepsilon$-Caprolactam for the growth were 6.0, 3$0^{\circ}C$ and 0.2% respectively. The bacteria could utilize glucose and fructose as a carbon source, and utilize ammonium chloride, ammonium nitrate, ammonium sulfate and sodium nitrate as a nitrogen source, and utilize L-Iysine and L-glutamate as a carbon and nitrogen source. It was found with thin layer chromatography and polarimeter that D-$\alpha$-Amino-$\varepsilon$-Caprolactam was converted to L-Iysine by the cell-free extracts of Alcaligenes eutrophus A52.

Degradation of Amaranth by Microorganisms (미생물(微生物)에 의한 Amaranth의 분해(分解))

  • Fundamental study was carried out to elucidate the mechanisms of biological degradation of dyestuff in environments. A few bacterial strains which were capable of degrading amarnath were obtained from soil through an extensive screening program and identified by microbiolological properties. Conditions for bacterial growth and amaranth degradation were characterized and optimized, and the degradation products were identified. The results were as follows. 1. The most active strain A12-1 to be capable of degradation of amaranth was identified as Pseudomonas sp. 2. Optimal conditions for growth of the strain A12-1 were:$35^{\circ}C$ and pH 7.5, and growth was markedly increaesd by aeration. 3. Degradation of amaranth by the strain was accessed under similiar conditions for growth, however significantly inhibited when the culture was aerated. 4. Both bacterial growth and amaranth degradation were gradually decreased with increased concentration of amaranth in the culture. 5. Reaction of the crude enzyme from the strain A12-1 was optimal at $35^{\circ}C$ and pH 7.5 for degrading amaranth. 6. Sodium naphthionate and R-amino salt were found to be the products of amaranth degradation by the strain A12-1.

  • Rice Sikhye was fermented by Saccharomyces cerevisiae for 10 day at 29$^{\circ}C$. Fermentable sugars such as glucose, maltose and maltotriose in rice Sikhye were converted into ethanol by the yeast, but limit dextrin was remained after the fermentation. Rice Sikhye wine was found to contain 5.3% of limit dextrin, 6.5% of ethanol, 2.9 $\mu$mol/ml of amino acid, 457$\mu\textrm{g}$/ml of protein, and the acidity of the Sikhye showed 3.1, respectively, and its pH was 3.67. Limit dextrin in rice Sikhye wine showed both signal of $\alpha$-1,4- and $\alpha$-1,6-glucosidic linkage wit its estimation ratio of 5.6:1 by 1H-NMR analysis. The taste of rice Sikhye wine was similar that of wine.

  • This study was carried out to investigate the antimutagenic affect of crude and heated comfrey extract on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG), benzo${(\alpha)}$pyrene (B${(\alpha)}$P) and 3-amino-1, 4-dimethyl-5H-pyri-do [4,3-b] indole (Trp-P-1). In spore rec-assay using Bacillus subtilis $H17(rec^+)$ and $M45(rec^-),$ crude comfrey extract showed strong antimutagenic effects on MNNG in the concentration of $40{\mu}l/disc$(p<0.01). In the Ames test using Salmonella typhimurium TA98 and TA100, the crude comfrey extract suppressed about 43% and 52% in the mutagenesis induced by $B{(\alpha)}P.$ However, the heated comfrey extract strongly suppressed about 75% and 76% in the mutagenesis in Salmonella typhimurium TA98 and TA100 induced by Trp-P-1(p<0.01).

Arabinoxylo- and Arabino-Oligosaccharides-Specific α-ʟ-Arabinofuranosidase GH51 Isozymes from the Amylolytic Yeast Saccharomycopsis fibuligera

  • Two genes encoding probable α-ʟ-arabinofuranosidase (E.C. 3.2.1.55) isozymes (ABFs) with 92.3% amino acid sequence identity, ABF51A and ABF51B, were found from chromosomes 3 and 5 of Saccharomycopsis fibuligera KJJ81, an amylolytic yeast isolated from Korean wheat-based nuruk, respectively. Each open reading frame consists of 1,551 nucleotides and encodes a protein of 517 amino acids with the molecular mass of approximately 59 kDa. These isozymes share approximately 49% amino acid sequence identity with eukaryotic ABFs from filamentous fungi. The corresponding genes were cloned, functionally expressed, and purified from Escherichia coli. SfABF51A and SfABF51B showed the highest activities on p-nitrophenyl arabinofuranoside at 40~45℃ and pH 7.0 in sodium phosphate buffer and at 50℃ and pH 6.0 in sodium acetate buffer, respectively. These exoacting enzymes belonging to the glycoside hydrolase (GH) family 51 could hydrolyze arabinoxylo-oligosaccharides (AXOS) and arabino-oligosaccharides (AOS) to produce only ʟ-arabinose, whereas they could hardly degrade any polymeric substrates including arabinans and arabinoxylans. The detailed product analyses revealed that both SfABF51 isozymes can catalyze the versatile hydrolysis of α-(1,2)- and α-(1,3)-ʟ-arabinofuranosidic linkages of AXOS, and α-(1,2)-, α-(1,3)-, and α-(1,5)-linkages of linear and branched AOS. On the contrary, they have much lower activity against the α-(1,2)- and α-(1,3)-double-substituted substrates than the single-substituted ones. These hydrolases could potentially play important roles in the degradation and utilization of hemicellulosic biomass by S. fibuligera.
