Two experiments were conducted to determine the standardized ileal digestible (SID) lysine (Lys) requirement and the ideal SID threonine (Thr) to Lys ratio for finishing barrows. In Exp. 1, 120 barrows with an average body weight of $72.8{\pm}3.6$ kg were allotted to one of six dietary treatments in a randomized complete block design conducted for 35 d. Each diet was fed to five pens of pigs containing four barrows. A normal crude protein (CP) diet providing 15.3% CP and 0.71% SID Lys and five low CP diets providing 12% CP with SID Lys concentrations of 0.51, 0.61, 0.71, 0.81 and 0.91% were formulated. Increasing the SID Lys content of the diet resulted in an increase in weight gain (linear effect p = 0.04 and quadratic effect p = 0.08) and an improvement in feed conversion ratio (FCR) (linear effect p = 0.02 and quadratic effect p = 0.02). For weight gain and FCR, the estimated SID Lys requirement of finishing barrows were 0.71 and 0.71% (linear broken-line analysis), 0.79 and 0.78% (quadratic analysis), respectively. Exp. 2 was a 26 d dose-response study using SID Thr to Lys ratios of 0.56, 0.61, 0.67, 0.72 and 0.77. A total of 138 barrows weighing $72.5{\pm}4.4$ kg were randomly allotted to receive one of the five diets. All diets were formulated to contain 0.61% SID Lys (10.5% CP), which is slightly lower than the pig's requirement. Weight gain was quadratically (p = 0.03) affected by SID Thr to Lys ratio while FCR was linearly improved (p = 0.02). The SID Thr to Lys ratios for maximal weight gain and minimal FCR and serum urea nitrogen (SUN) were 0.67, 0.71 and 0.64 using a linear broken-line model and 0.68, 0.78 and 0.70 using a quadratic model, respectively. Based on the estimates obtained from the broken-line and quadratic analysis, we concluded that the dietary SID Lys requirement for both maximum weight gain and minimum FCR was 0.75%, and an optimum SID Thr to Lys ratio was 0.68 to maximize weight gain, 0.75 to optimize FCR and 0.67 to minimize SUN for finishing barrows.
Two experiments were conducted to determine the effects of bird age on apparent ileal digestibility (AID) and standardized ileal digestibility (SID) of amino acids (AA) for 10-d-old Experiment (Exp. 1) and 22-d-old (Exp. 2) male broilers. This study investigated the effects of different broiler ages and feed ingredients on AID and SID of AA in corn and soybean meal (SBM). Four hundred and eighty (age = 7 d; initial body weight [BW] = 173.4 ± 12.65 g) and 192 (age = 18 d; initial BW = 772.2 ± 62.13 g) birds were allocated to three dietary treatments in a randomized complete block design with eight replicate cages per treatment. Two diets were formulated based on corn or SBM as the sole source of AA in the diet. A nitrogen-free diet was also formulated to measure basal endogenous losses of AA. Experimental diets were given for 3 and 4 days in Exps. 1 and 2, respectively. An interaction was observed (p < 0.05) between the age of birds and the type of ingredient for the AID of most AA, except for methionine, valine, cysteine (Cys), and tyrosine; however, the effects of age and type of ingredients were diminished in the SID of AA, except for histidine, isoleucine, leucine (Leu), phenylalanine, alanine (Ala), and glutamic acid (Glu). The AID of AA, except for Leu and Cys and the SID of AA, except for Leu, Ala, Glu, and Pro in SBM were greater (p < 0.05) than in corn. As the age of birds increased from 10 to 22 d, digestibility of all AA increased (p < 0.05), regardless of the expression of AA digestibility (i.e., AID and SID). In conclusion, the AID and SID of AA in both corn and SBM increased with increasing age, and the AID and SID of AA in SBM were greater than in corn.
Three experiments were conducted to evaluate the metabolizable energy (ME) value, standardized ileal digestibility (SID) of amino acids (AA) of soybean meal (SBM), soy protein concentrate (SPC) and fermented soybean meal (FSBM), and the application of these products in early-weaned piglets. In Exp. 1, four barrows with initial body weight (BW) of $14.2{\pm}1.4$ kg were used in a $4{\times}4$ Latin square design. The diet 1 contained corn as the only energy source. The other three diets replaced 25% of corn in diet 1 with one of the three soybean products, and the digestable energy (DE) and ME contents were determined by difference. In Exp. 2, four barrows (initial BW of $18.2{\pm}1.5$ kg) were fitted with ileal T-cannulas and allotted to a $4{\times}4$ Latin square design. Three cornstarch-based diets were formulated using each of the soybean products as the sole source of AA. A nitrogen-free diet was also formulated to measure endogenous losses of AA. In Exp. 3, ninety six piglets (initial BW of $5.6{\pm}0.9$ kg) weaned at $21{\pm}2$ d were blocked by weight and assigned to one of three treatments for a 21-d growth performance study. The control diet was based on corn and SBM, the two treatments' diets contained either 10% SPC or FSBM and were formulated to same SID lysine to ME ratio of 3.6 g/Mcal. The results showed that the ME content of SPC was greater than SBM (p<0.05). The SID of most AA in SPC was greater than the SID of AA in SBM (p<0.05). For the essential AA, the SID of histidine, isoleucine, leucine, lysine and threonine in FSBM were greater than in SBM (p<0.05). Even though they were fed same SID lysine to ME ratio of 3.6 g/Mcal diets, pigs fed SPC and FSBM diets had greater weight gain, G:F (p<0.05) and better fecal score (p<0.05) than pigs fed SBM diet. In conclusion, SPC showed a higher ME content and SID of AA than the SBM. SID of some essential AA in FSBM was higher than SBM and was similar with SPC. But the lower antigenic proteins and anti-nutritional factors content in SPC and FSBM may be the main factors affecting the performance of early-weaned piglets rather than the increased ME content and SID of AA.
This study is to figure out the amount of primary X-ray generated in SID 50cm, 1m, and 2m penetrating protective aprons in X-ray radiography for hands, skull, and lumbar spine. Results are as follows: Firstly, the exposure dose of primary X-ray which is low such as that of hand X-ray may be reduced by 270 times if protective aprons are worn, but it still slightly penetrates 0.3mm thick Pb protective aprons at SID 50cm, 1m, and 2m. Secondly, the exposure dose of primary X-ray which is moderate such as that of skull X-ray may be reduced by 22 times if protective aprons are worn, but it still fairly penetrates 0.3mm thick Pb protective aprons at SID 50cm, 1m, and 2m. Thirdly, the exposure dose of primary X-ray which is very high such as that of lumbar spine X-ray may be reduced by 13 times if protective aprons are worn, but it still penetrates a lot 0.3mm thick Pb protective aprons at SID 50cm, 1m, and 2m. Therefore, people in X-ray room should not only wear protective aprons at any spaces that the primary X-ray can reach, but also need to stand behind the thick Pb shield to protect the body if it is inevitable to stay in the room.
To develop vector plasmid for the bacteriophage P2-P4 system which is a useful experimental tool for the study of viral capsid assembly, we constructed a new P4-derived vector plasmid starting from P4 ash8 sid71 With recombinant DNA technology, a portion of P4 genome was deleted and tetracyclin resistance gene (terR) was introduced into P4 genome to give P4 selectivity. Resulting P4 ash8(sid71) terR was 12.09 kb long and could be converted to a viable bacteriophage with P2 infection. The burst size of induced bacteriophage form of P4 ash8(sid71) terR was determined. The CsCl buoyant equilibrium density gradient experiment of new P4 derivative suggested the upper limit of packaging capacity in P2-size head.
Objective: The present experiment aimed to determine standardized ileal digestible (SID) lysine (Lys) requirements for pregnant sows individually housed under commercial farm conditions. Methods: Two hundred multiparous sows (parity = 5.1±2.0) on day 42 of gestation were randomly allocated to five dietary treatments with a balanced parity. Experimental diets were formulated to contain 0.22%, 0.32%, 0.42%, 0.52%, and 0.62% of SID Lys for the mid-gestation period (days 42 to 76) and 0.36%, 0.46%, 0.56%, 0.66%, and 0.76% of SID Lys for the late gestation period (days 77 to 103). All indispensable amino acids except Lys were provided at 110% of their requirement estimates. Daily feed allowance per sow was determined based on the back-fat thickness and body condition score at the second pregnancy check and on day 90 of gestation. Three different statistical models were used to estimate the SID Lys requirement. Results: Total born piglets alive per litter increased linearly and quadratically (p<0.001) as dietary SID Lys increased. For total born piglets alive per litter, the SID Lys requirement estimates ranged from 9.69 to 12.4 g/d for the mid-gestation period (1.19 to 1.52 g/Mcal metabolizable energy; 0.39% to 0.49%) and 14.6 to 17.4 g/d for the late gestation period (1.62 to 1.93 g/Mcal metabolizable energy; 0.52% to 0.62%). Conclusion: The mean values of the SID Lys requirement for the mid-gestation period and the late gestation period are 11.1 and 16.1 g/d (1.36 and 1.79 g/Mcal metabolizable energy; 0.44% and 0.58%), respectively, for maximal total born piglets alive per litter.
An experiment was conducted to determine the standardized ileal digestibility (SID) of amino acids (AA) in four sources of full-fat soybeans (FFSB) and in one source of soybean meal (SBM). The FFSB had different concentrations of trypsin inhibitor units (TIU) and included two sources of conventional FFSB, and two sources of a soybean variety that was selected for a reduced concentration of the Kunitz trypsin inhibitor. The conventional FFSB was either low temperature-processed (LT-FFSB-CV; 37.7% CP, 35.4 TIU/mg) or high temperature-processed (HT-FFSB-CV; 40.5% CP, 4.4 TIU/mg). The low-Kunitz FFSB was also either low temperature-processed (LT-FFSB-LK; 36.2% CP, 23.5 TIU/mg) or high temperature-processed HT-FFSB-LK; (38.2% CP, 4.0 TIU/mg). The SBM contained 47.5% CP and 3.20 TIU/mg. Twelve weanling barrows (initial BW: $11.1{\pm}1.3\;kg$) were fitted with a T-cannula in the distal ileum. Pigs were allotted to a replicated $6{\times}6$ Latin square design with six diets and six periods per square. Five diets were prepared using each of the soybean sources as the only source of AA in the diet. An N-free diet was also included in the experiment to measure basal endogenous losses of AA. The two low temperature-processed FFSB had lower (p<0.05) AID and SID values for all indispensable AA than the two high temperature-processed FFSB and SBM. The SID values for all indispensible AA except Trp were greater (p<0.05) in LT-FFSB-LK than in LT-FFSB-CV, but the SID of AA in HT-FFSB-CV and HT-FFSB-LK were not different. The SID of AA in SBM were not different from the SID in HT-FFSB-CV and in HT-FFSB-LK. Results of this experiment show that a reduction of the TIU from 35.4 to 23.5 TIU/mg will improve the SID of AA, but this reduction is not sufficient to completely ameliorate the negative impact of trypsin inhibitors. Results also show that the SID of AA in high temperature-processed FFSB is similar to that in de-hulled SBM.
Wrist PA-grip view is used to diagnose triangular fibrocartilage complex (TFCC) tear because it can easily diagnose damage to the surrounding wrist ligaments. However, despite advances in radiology equipment, distortion of images due to geometric elements still has many limitations. In this paper, we propose a method that can minimize the distortions of images by analyzing the distortions occurring in the wrist PA-grip view. A source of image distance (SID) were set at 130 cm and 150 cm for comparison with 110 cm. Depending on the SID, the phantom of wrist was moved at 0, 2, 4, 6, 8, and 10 cm in the X-axis and Y-axis directions, respectively. For quantitative evaluation, the difference of distance between the radius and ulna was measured in picture archiving and communication system (PACS) system. As a qualitative evaluation, survey was conducted among 20 radiologic technologists who examined the Wrist PA-grip view. The Kruskal Wallis test was performed to compare the distortion according to the phantom movement in the X-axis and Y-axis directions based on the SID, and the Tukey test was performed as a post-test. In the quantitative evaluation results, the measured values obtained in the X-axis was not significantly different in all groups (p>0.05). The measured values obtained in the Y-axis was significantly different in the most groups (p<0.05). Therefore, to reduce distortion while maintaining image quality, we recommend what examine the SID at 150 cm than 110 cm.
Objective: The experiment aimed to determine the standardized ileal digestibility (SID) of crude protein (CP) and amino acids (AA) in 10 brown rice samples fed to pigs, and to construct predictive models for SID of CP and AA based on the physical characteristics and chemical composition of brown rice. Methods: Twenty-two cannulated pigs (initial body weight: 42.0±1.2 kg) were assigned to a replicated 11×3 incomplete Latin square design, including an N-free diet and 10 brown rice diets. Each period included 5 d adaptation and 2 d ileal digesta collection. Chromic oxide was added at 0.3% to all the diets as an indigestible marker for calculating the ileal CP and AA digestibility. Results: The coefficients of variation of all detected indices for physical characteristics and chemical composition, except for bulk weight, dry matter (DM) and gross energy, in 10 brown rice samples were greater than 10%. The SID of CP, lysine (Lys), methionine, threonine (Thr), and tryptophan (Trp) in brown rice was 77.2% (62.6% to 85.5%), 87.5% (80.3% to 94.3%), 89.2% (78.9% to 98.9%), 55.4% (46.1% to 67.6%) and 92.5% (86.3% to 96.3%), respectively. The best prediction equations for the SID of CP, Lys, Thr, and Trp were as following, SIDCP = -664.181+8.484×DM (R2 = 0.40), SIDLys = 53.126+6.031×ether extract (EE)+0.893×thousand-kernel volume (R2 = 0.66), SIDThr = 39.916+7.843×EE (R2 = 0.41), and SIDTrp = -361.588+4.891×DM+0.387×total starch (R2 = 0.85). Conclusion: Overall, a great variation exists among 10 sources of brown rice, and the thousand-grain volume, DM, EE, and total starch can be used as the key predictors for SID of CP and AA.
The term "P2 sir-associated helper inefficiency" has been used to define the inefficient helper capability of P2 sir mutants for their satellite bacteriophage P4. The aim of this study was to investigate the factors overcoming P2 sir-associated helper inefficiency. At first, we verified whether the P2 cos region containing P4 sid71 cosP2 could overcome P2 sir-associated helper inefficiency with P2 sir3. The result was that P4 sid71 cosP2 could not overcome P2 sir-associated helper inefficiency with P2 sir3. Instead of cos region of P2, the size of the DNA packaged into a $P2_{sir}$-sized head seems to be important for overcoming P2 sir-associated helper inefficiency. In the present work, three kinds of P4 derivatives with packaged DNA sizes between those of P4 ost1 and P4 ost2, were constructed through DNA manipulation. In one P4 derivative, P4 sid71 delRI::apr, the size of the packaged DNA was identified with a CsCl buoyant equilibrium density gradient experiment. According to the burst sizes of the P4 derivatives, they could overcome P2 sir3-associated helper inefficiency. The size of the P4 derivative DNA suitable for packaging into a $P2_{sir3}$-sized head was 28-29 kb.
