A fluorogenic method for measuring enteropeptidase activity: spectral shift in the emission of GD4K-conjugated 7-amino-4-methylcoumarin

  • Enteropeptidase is a serine protease secreted by the pancreas and converts inactive trypsinogen to active trypsin. Enteropeptidase cleaves the C-terminal end of the substrate recognition sequence Asp-Asp-Asp-Asp-Lys ($D_4K$). The assay for enteropeptidase has utilized $GD_4K$-conjugated 2-naphthylamine ($GD_4K$-NA) as a fluorogenic probe over the last 30 years. However, no other $D_4K$-conjugated fluorogenic substrates of enteropeptidase have been reported. Furthermore, naphthalene is known as carcinogenic to humans. In this study, we used shift in the emission spectrum of $GD_4K$-conjugated 7-amino-4-methylcoumarin ($GD_4K$-AMC) as a fluorogenic method to measure enteropeptidase activity. The kinetic analysis revealed that enteropeptidase has a $K_M$ of 0.025 mM and a $k_{cat}$ of 65 $sec^{-1}$ for $GD_4K$-AMC, whereas it has a $K_M$ of 0.5 to 0.6 mM and a $k_{cat}$ of 25 $sec^{-1}$ for $GD_4K$-NA. The optimum pH of $GD_4K$-AMC hydrolysis was pH 8.0. Our data indicate that $GD_4K$-AMC is more suitable as a substrate for enteropeptidase than $GD_4K$-NA.

Synthesis of the Low-Hygroscopic Polyimide for 2-Layer Flexible Copper Clad Laminate (2층 연성동박적층판용 저흡습 폴리이미드의 합성)

  • In this study, nine kinds of polyimides were synthesized from 1,2,4,5-benzenetetracarboxylic dianhydride (PMDA), 4,4'-(4,4'-isopropylidenediphenoxy)bis(phthalic anhydride) (BPADA), m-pheny lenediamine (m-PDA) and 4,4'-oxydianiline (ODA) by controlling molar ratio of monomers. Synthesized polyimides were used as insulator films for 2-layer Flexible Copper Clad Laminate(FCCL) which were manufactured by the casting method. Glass transition temperature and thermal degradation temperature for 5% weight loss of the polyimide film were improved by increasing contents of m-PDA and PMDA, respectively. Water absorption of polyimide film was reduced by increasing contents of ODA and BPADA which have relatively long structure, respectively. Peel strength of 2-layer FCCL was improved by increasing contents of ODA and BPADA.

Comparison of Detectable Levels for Screening Residual Antibacterial Agents by Bioassay (잔류 항균물질에 대한 미생물학적 간이검사법의 검출감도 비교)

  • Minimun-detectable levels to 28 antibacterial agents used for the prevention and the treatment of fish diseases were determined to establish optimal detective method of bioassay in fish by the EEC 4-plate method, the modified method of EEC 4-plate and the standard method of analysis in food safety regulation. The test organisms used in the methods of bioassay were as follows: Bacillus subtilis BGA (B. subtilis) and Micrococcus luteus ATCC 9341 (M. luteus) in the EEC 4-plate method, B. subtilis, M. luteus and Bacillus cereus var. mycoides ATCC 11778 (B. cereus) in the modified of EEC 4-plate, and B. subtilis, M. luteus, B. cereus and Bacillus stearothermophilis var. calidolactis C-953 (B. stearothermophilis) in the standard method. The standard method showed predominant sensitivity in the detection of penicillins (PCs), and was also highly sensitive to aminoglycosides (AGs). The sensitivity of standard method in the detection of tetracyclines (TCs), marrolides (MLs), nitrofuran derivatives(NFs) and quinolones (QNs) was very low, and against sulfonamides (SAs), however, was extremely low. The modified method of EEC 4-plate showed very high sensitivity to TCs. Both the EEC 4-plate and the modified method of EEC 4-plate showed competitively high sensitivity in the detection of PCs, MLs, NFs, QNs and SAs. All the methods studied in the experiment showed very low sensitivity against chloramphenicol (CMs). Consequently, the modified method of EEC 4-Plate was the best bioassay method with a wide range of sensitivity for the optimal detection of the residual antibacterial agents in fish.

Development of pressurizing support tunneling method and case study of its field application (가압지보 터널공법 개발 및 현장적용 사례 분석)

  • The pressurizing support tunneling method has been developed that overcomes shortcomings of conventional trenchless methods and applied to the field. The main concept of the new method is the pressurization system which, by means of pressurization bag between outer flange of steel ribs and excavated perimeter, applies higher pressure than the pressure relaxed by excavation to the ground to prevent ground displacement. The stability of the support members and effect of displacement control of the new method were verified through 3D numerical analyses. The new method was applied to the construction of a 10.7 m wide, 7.9 m high and 85 m long ramp tunnel that passes under ${\bigcirc}{\bigcirc}$ Expressway. By applying the new method, the tunnel construction was successfully completed in 13.5 months which decreases construction time to 35% compared to conventional methods, and ground displacement was almost negligible.

  • Using M-ary QAM and PSK, various modulation methods are researched so far. In this paper, a new constellation method of $4L^2$-PSK that normalized circular multi-layer method is proposed with the advantage of those modulation methods. These replacements of signal points complement weak points of an nonlinear radius increment as a layer increases in M-ary QAM and a low efficiency in PSK compared with QAM. But, $4L^2$-PSK have some problems that in the power efficiency and difference of BER in each layer, So in this paper suggest that advanced $4L^2$-PSK also.

  • A gas chromatography-mass spectrometric method was developed for the determination of formaldehyde in urine and saliva. In a 20 mL glass tube, 0.2 mL of urine or saliva was taken. Further, 1.8 mL of 0.1 M HCl, 0.1 mL of 2,000 mg/L 2,4-dinitrophenyl hydrazine and $20{\mu}l$ of 500 mg/L acetone-$d_6$ as internal standard were added in the tube and sealed tightly with cap. The solution was shaken for 20 min at room temperature and extracted using 4 mL of toluene. The extract was concentrated and redissolved with $100{\mu}l$ of acetonitrile, and then measured by gas chromatography-mass spectrometer (selected ion monitoring). The detection limit was 2.0 ng/mL and 0.5 ng/mL in saliva and urine, respectively. The calibration curves showed good linearity with r = 0.997 and 0.998 for saliva and urine, respectively. The method was used to analyze formaldehyde in rat urine after oral exposure. The developed method may be use ful to the monitoring for formaldehyde exposure in human.

Conservative Visibility Preprocessing by Expressing 4-D visibility Information on 2-D Spaces (2차원 평면상에 4차원 가시성 정보의 표현을 통한 포괄적 가시성 전처리)

  • Visibility preprocessing is a useful method to reduce the complexity of scenes to be processed in real-time, and so enhances the overall rendering performance for interactive visualization of virtual environments. In this paper, we propose an efficient visibility preprocessing method. In the proposed method, we assume that navigatable areas in virtual environments are partitioned into rectangular parallelpiped cells or sub-worlds. To preprocess the visibility of each polygon for a given partitioned cell, we should determine at least the area-to-area visibility. This is inherently a four-dimensional problem. We efficiently express four-dimensional visibility information on two-dimensional spaces and keep it within a ternary tree, which is conceptually similar to a BSP(Binary Space Partitioning) tree, by exploiting the characteristics of conservative visibility. The proposed method is able to efficiently handle more general environments like urban scenes, and remove invisible polygons jointly blocked by multiple occluders. The proposed method requires O(nm) time and O(n+m) space. By selecting a suitable value for m, users can select a suitable level of trade-off between the preprocessing time and the quality of the computational result.

  • The characteristic of calcined BaTiO3 powder and sintered PTC ceramics was investigated varing with calcination temperature and time of BaTiO(C2O4)2.4H2O synthesized from BaCl2.2H2O, TiCl4, oxalic acid and ethanol by oxalic acid method. When the particle size was less than 0.1 ${\mu}{\textrm}{m}$ by controlling calcination temperature and time, the resistance at room temperature was measured very high (above M{{{{ OMEGA }}). However, when the calcined particle sizes ranged from 0.2 to 0.3 ${\mu}{\textrm}{m}$, the resistance was 100 {{{{ OMEGA }} (After sintering, the grain size was 10~30 ${\mu}{\textrm}{m}$ homogeneously with the addition of dopant in sintering, the resistivity, resistance, $\alpha$ value and jumped to were 110{{{{ OMEGA }}.cm, 24$^{\circ}C$/% and 106{{{{ OMEGA }}, respectively.

Efficient Implementation of Finite Field Operations in NIST PQC Rainbow (NIST PQC Rainbow의 효율적 유한체 연산 구현)

  • In this paper, we propose an efficient finite field computation method for Rainbow algorithm, which is the only multivariate quadratic-equation based digital signature among the current US NIST PQC standardization Final List algorithms. Recently, Chou et al. proposed a new efficient implementation method for Rainbow on the Cortex-M4 environment. This paper proposes a new multiplication method over the finite field that can reduce the number of XOR operations by more than 13.7% compared to the Chou et al. method. In addition, a multiplicative inversion over that can be performed by a 4x4 matrix inverse instead of the table lookup method is presented. In addition, the performance is measured by porting the software to which the new method was applied onto RaspberryPI 3B+.

A DIFFERENCE SET METHOD FOR CIRCULANT DECOMPOSITIONS OF COMPLETE PARTITE GRAPHS INTO GREGARIOUS 4-CYCLES

  • The complete multipartite graph $K_{n(m)}$ with n $ {\geq}$ 4 partite sets of size m is shown to have a decomposition into 4-cycles in such a way that vertices of each cycle belong to distinct partite sets of $K_{n(m)}$, if 4 divides the number of edges. Such cycles are called gregarious, and were introduced by Billington and Hoffman ([2]) and redefined in [3]. We independently came up with the result of [3] by using a difference set method, and improved the result so that the composition is circulant, in the sense that it is invariant under the cyclic permutation of partite sets. The composition is then used to construct gregarious 4-cycle decompositions when one partite set of the graph has different cardinality than that of others. Some results on joins of decomposable complete multipartite graphs are also presented.
